Titel: ïiieuwe dithiënylpiperidinen; werkwijze voor de bereiding daarvan; geneesmiddel; werkwijze voor de bereiding van een geneesmiddel; toepassing van de nieuwe dithiënylpiperidinen.
Uit het Amerikaanse octrooischrift 2.739*968 zijn verbindingen met spasmolytische, analgetische en antihistaminewerking bekend, welke voldoen aan formule 5 van het formuleblad, waarin b.v. Eg een methyl-of ook ethylgroep is, R en twee thiënyl-(2/-groepen voorstellen, 5 waterstof is en Y waterstof of een hydrcccy lgroep voorstelt, of ook samen met de piperidinering een dubbele binding kan vormen.
Uit het Duitse Offenlegungsschrift 2.128.808 zijn verbindingen met formule 6 bekend, waarin R een lage alkylgroep en X een anionische groep van een farmaceutisch aanvaardbaar zuur is. Voor deze verbindingen 10 wordt eveneens een spasmolytische werking, die vergelijkbaar is met die van atropinesulfaat, aangegeven.
De uitvinding wordt nu gekenmerkt door verbindingen met formule 1, waarin R^ en Rg waterstof voorstellen of samen een tweede binding tussen de koolstofatcmen die de substituenten R^ en Rg dragen, 15 vormen, R^ bovendien ook een hydroxy lgroep kan zijn, R^ waterstof, een cycloalkylgroep met 3-8 koolstofatcmen of een alky lgroep met 1-20 koolstofatcmen, die eventueel ook een of twee hydroxylgroepen kan bevatten, voorstelt, en de groepen R^, R^, Rg en R^ gelijk of verschillend zijn en waterstof, alkylgroepen met 1-6 koolstofatcmen of 20 halogeenatcmen voorstellen, hun N-oxiden, hurquartaire zouten,
hun zuuradditiezouten, waarbij tenminste een thiënyl-(3)-groep aanwezig is, wanneer R^ een niet gesubstitueerde alkylgroep met 1-6 kool-stofatamen voorstelt. Voorts heeft de uitvinding betrekking op een werkwijze voor het bereiden van deze verbindingen, welke gekenmerkt 25 wordt doordat men een verbinding met formule 2, die ook trigeconjugeer-de dubbele bindingen in de ring kan bevatten, waarin R^ de aangegeven betekenissen heeft (R^ ontbreekt wanneer een pyridinering aanwezig is), en X
A) de groep -C0Z voorstelt, waarin Z een halogeenatocm of een alkoxy-30 groep met 1-6 koolstofatcmen voorstelt of een eventueel een-tot tweevoudig door halogeenatcmen en/of alkylgroepen met 1-6 koolstofatcmen gesubstitueerde thiënylgroep voorstelt, of (B) lithium of de groep -MgHal voorstelt, waarin Hal chloor, broom of jood is, 8000235

-2- ·* hetzij in geval A) met een verbinding met formule 3, waarin X’ lithium of de groep -MgHal voorstelt en de thiënylgroep ook een- tot -tweevoudig door halogeenatomen en/of alkylgroepen met 1-6 koolstofatomen kan zijn gesubstitueerd, laat reageren, 5 hetzij in geval B) met een verbinding met formule ^ laat reageren, waarbij in een van de bovenstaand genoemde uitgangsmaterialen tenminste thiënyl(3)-groep aanwezig moet zijn, wanneer de substituent R^ en van formule 2. een niet gesubstitueerde alkylgroep met 1-6 koolstof at omen voorstelt, en de verkregen verbindingen eventueel met reductiemiddelen 10 en/of dehydrateringsmiddelen behandelt en/of door de groep R^ alky-leert en/of in het N-oxide omzet of b) uitgaat van een verbinding met formule 1 en in het geval R1 de hydroxylgroep en Rg waterstof is, deze met dehydrateringsmiddelen en/of reductiemiddelen behandelt en/of de groep R^ tegen andere 15 substituenten binnen de voor R^ aangegeven betekenissen uitwisselt en eventueel de verkregen verbindingen in het N-oxide cmzet.
Bij voorkeur zet men de verkregen verbindingen in de zuuradditie-zouten resp. de quartaire zouten cm.
Voorts betreft de uitvinding geneesmiddelen die als werkzame 20 stof een of meer van deze nieuwe verbindingen bevatten tezamen met een gebruikelijke farmacologische drager en/of een verdunningsmiddel.
Ook betreft de uitvinding een werkwijze voor het bereiden van een geneesmiddel, welke gekenmerkt wordt doordat men een of meer van de nieuwe verbindingen met gebruikelijke farmaceutische drager-25 materialen of verdunningsmiddelen resp. andere fysiologisch aanvaardbare hulpstoffen tot farmaceutische preparaten verwerkt resp. in een voor farmceutische toepassing geschikte vorm brengt.
Ook betreft de uitvinding de toepassing van de nieuwe verbindingen voor de bereiding van geneesmiddelen.
30 De in de verbindingen met formule 1 optredende alkylgrcepen kunnen recht of vertakt zijn. In het geval de groepen R^, R^, Rg en/of R^ alkylgroepen zijn, bestaan deze bij voorkeur uit 1-1 koolstofatcmen. Liefst gaat het cm de methylgroep. Wanneer de groepen
Ri,, R._, R^ en/of R_ halogeenatomen zijn, gaat het b.v. cm fluor, *+ 5 o ! 35 chloor of brocm. Wanneer de groep R^ een alkylgroep is, die een 8000215 ψ -3- * hydroxylgroep bevat, béuindt deze bydroxylgroep zich bij voorkeur op de 2-plaats van de alkylgroep (de 1-plaats van de alkylgroep is de plaats die verbonden is met het stikstofatocm van de piperidinering).
De verbindingen volgens de uitvinding zijn farmacodynamisch 5 werkzaam en bezitten b.v. een antiischemische en bloeddrukverhogende werking; voorts bewerken zij een verhoging van de périfere doorbloeding en verlagen vaak de hartfrequentie (bradycardie). Enkele verbindingen, in het bijzonder die waarin een hydroxy alkylgroep, b.v. de 2-hyclroxy-ethylgroep voorstelt, bezitten ook een analgetische werking. Doel van 10 de uitvinding is derhalve verbindingen met gunstige fanaacodynamische eigenschappen, die als geneesmiddel toepasbaar zijn, ter beschikking te stellen.

Werkwijze a) wordt bij voorkeur uitgevoerd in een temperatunr- gebied tussen -100 en +150°C, liefst -75 tot+100°C of tot +50°C. Wan-15 neer men een thiënyl-(3)-metaalverbinding (in het bijzonder thiënyllithium) of een thiënyl-(3)-grignardverbinding toepast, wordt bij voorkeur bij lage temperaturen, in het bijzonder’beneden -50°C in een inert middel gewerkt. Bij voorkeur wordt in dergelijke gevallen de reactie uitgevoerd bij temperaturen tussen -70 en -8o°C. In het bijzonder geldt 20 het temperatuurgebied van -70 tot -80°C voor thiënyl-(3 )-metaalver-bindingen. Als oplosmiddelen zijn b.v. verzadigde alifatische symmetrische of niet symmetrische diëthylethers met aikylgroepen van b.v.
1-6 koolstofatanen, alkylethers (alkyl met 1-6 koolstofatcmen) van onverzadigde cycloalkanolen en alkyl gesubstitueerde cycloalkanolen 25 geschikt, waarbij de cycloalkanolringen telkens uit 3, ^» 5 of 6 koolstof atanen bestaan; verzadigde alifatische met 5-9 koolstofatanen of cycloalifatische met 3-7 koolwaterstoffen, waarbij de laatste bij voorkeur door aikylgroepen met 1-¾ koolstofatcmen enkel- tot drievoudig gesubstitueerd kunnen zijn; tetrahydrofuran; benzeen; benzeen dat door 30 alkylgroepaimet 1-3 koolstofatanen is gesubstitueerd. In het bijzonder gaat het bij de ethers en alifatische resp. cycloalifatische koolwaterstoffen cm die, welke in het gebied tussen -80 en +20°C vloeibaar zijn.
Voorbeelden van de in aanmerking komende oplosmiddelen zijn di-35 ethylether, diïsopropylether, methylcyclopentylether, hexaan, cyclohexaan, 8000285 -1μ- tolueen, xyleen, methylcyclohexaan, methylcyclopentaan, ethylcyclo-hexaan, dimethylcyclohexaan. De aangegeven oplosmiddelen kunnen ook gemengd worden toegepast. Men kan b.v. een oplosmiddelmengsel toepassen, dat uit een. verzadigde ether en een door alkylgroepen met 5 1-3 koolstofatcmen enkelvoudig gesubstitueerd benzeen bestaat.
Dergelijke oplosmiddelmengsels zijn b.v. beschreven in het Duitse Offenlegungsschrift 2.800.536.
Een passende overmaat aan metaalorganische verbinding is steede nodig wanneer de andere reactie component actieve waterstof (amino-, 10 hydroxylgroep, zout) bevat. Vaak: is het echter in het algemeen aan te bevelènu- een overmaat aan metaalorganische verbinding toe te passen, cmdat hiervoor betere opbrengsten worden verkregen.
Wanneer Z in de groep -C0Z (formule 2) een halogeenatoom is, gaat het in het bijzonder om chloor, broom of jood.
15 Uitgangsmaterialen met formule 7, waarin de thiënylgroep ook door halogeen of lage alkylgroepen kan zijn gesubstitueerd, kunnen b.v. door omzetting van de overeenkomstige thiënyllithium- resp. thiënylgrignardverbinding met de verbinding met formule 8 of door omzetting van het overeenkomstige thiënylcyanide resp. thiënyl-20 carbonylchloride met de verbinding met formule 9 (waarin Hal chloor of broom voorstelt; en R^ ontbreekt wanneer een pyridinering aanwezig is) in een oplos- of suspendeermiddel zoals gebruikelijk is voor Grignardreacties (b.v. lage verzadigde alifatische ethers, benzeen, methyl gesubstitueerde benzeen) bij temperaturen tussen -80 en +100°C 25 worden verkregen.
Voorts kunnen dergelijke uitgangsverbindingen ook uit een verbinding met formule 8 en het overeenkomstige thiofeen bij aanwezigheid van AlCl^ volgens Friedel-Crafts worden verkregen.
Uitgangsmaterialen met formule U kunnen b.v. door Friedel-30 Crafts acylering van een verbinding met formule 10 (waarin Hal chloor of broom voorstelt) met een verbinding met formule 11 bij aanwezigheid van AlCl^ in een oplosmiddel zoals dichloorethaan, nitro-methaan, bij temperaturen tussen 0 en +100°C worden verkregen.
Verder kunnen ze door een Grignard-reactie van een verbinding 35 met formule 12 of een verbinding met formule 13, waarin R een lage 3000285 * * -5- alkylgroep voorstelt, met een verbinding met formule ik, waarin M lithium of MgCl of MgBr voorstelt, in een oplos- resp. suspendeeimiddel zoals voor Grignard-reacties gebruikelijk is, bij temperaturen tussen -80 en +100°c vorden verkregen.
5 Uitgangsmaterialen met formule 2, waarin X de groep -MgHal voor stelt, kunnen b.v. uit verbindingen met formule 2 waarin X chloor, broom of jood is, door gebruikelijke Grignard- of lithium-Grignard-reactie onder toepassing van magnesium resp. metallisch lithium in de hiervoor gebruikelijke oplosmiddelen (b.v. tetr ahydrofuran, lage 10 alifatische ethers, lage alkylbenzeenverbindingen) bij temperaturen tussen 20 en 120°C worden verkregen. Eventueel moet de Grignard-reactie met jood + dibrocmethaan worden opgestart.
De waterafsplitsing uit verbindingen met formule 1, waarin S1 de hydroxylgroep en Rg waterstof is (de overige symbolen kunnen de 15 aangegeven betekenissen hebben), wordt bij voorkeur bij hogere temperaturen uitgevoerd, b.v. in een temperatuurgebied van 20-150°C.
Bij voorkeur worden oplosmiddelen zoals ijsazijn, benzeen, dioxan, lage alifatische alcoholen, toegepast.
Als wateraf splitsende middelen komen b.v. in aanmerking: anorga-20 nische zuren, zoals zwavelzuur of halogeenwaterstofzuren; organische zuren zoals oxaalzuur, mierezuur; thionylchloride; zinkchloride; tinchloride; boortrifluoride; kaliumwaterstofsulfaat; aluminium-chloride; fosforpentoxide; aluminiumoxide; zuurchloride; rode fosfor + jood bij aanwezigheid van water.
25 Vaak vindt een partiële waterstofsplitsing reeds bij de op- werking van de door omzetting van verbindingen met formule 2 met verbindingen met formule 3 resp. k verkregen reactieprodukten plaats.
De reductie van verbindingen met formule 1, waarin R^ en Rg de dubbele binding vormen of R1 de hydroxylgroep en Rg waterstof is, 30 alsmede van produkten, die in de piperidinering drie dubbele bindingen bevatten (de overige symbolen hebben de aangegeven betekenis) kan b.v. met waterstof bij aanwezigheid van hydrogeneringskatalysatoren , bij voorkeur in een oplosmiddel zoals alcoholen, dioxan, tetrahydro-furan, benzeen, azijnzuur, azijnzuuresters enz. worden uitgevoerd.
35 Als hydrogeneringskatalysatoren komen in het bijzonder edel metaal- 8000285 -6- katalysatoren zoals palladium, platina, enz. resp. sulfidische katalysatoren zoals palladiumsulfide, platinasulfide, rheniumheptasulfide e.d. in aanmerking. De katalysatoren kunnen met of zonder drager worden toegepast. Als dragermiddelen zijn b.v. "bariumsulfaat, aluminiumoxide 5 e.d. geschikt. De hydrogenering wordt "bij voorkeur "bij temperaturen tussen 20. en 100°C "bij normale druk of verhoogde druk tot h.v. 100 "bar uitgevoerd. Een druk van 2-20 bar wordt geprefereerd.
Voorts kamen als reductiemiddelen nascerende waterstof, b.v. metallisch natrium in een lage alcohol (b.v. ethanol) met of zonder 10 toevoeging van water, natrium in vloeibaar ammoniak, natriumamalgaam bij aanwezigheid van een zuur zoals verdund zoutzuur, verdund zwavelzuur of azijnzuur, in aanmerking. Hierbij wordt in het algemeen bij kamertemperatuur of verhoogde temperatuur tot ongeveer 150°C gewerkt.
Bovendien is b.v.· ook een electrolytische reductie of reductie 15 met andere waterstofleverende middelen, zoals complexe metaalhydriden, b.v. alkaliboorhydriden, lithiumalanaat, natrium-bis-(2-methcxyethoxy)-aluminiumhydride bij aanwezigheid van hydrogeneringskatalysatoren mogelijk.
Bij de reductie, in het bijzonder bij katalytische hydro-20 genering (Pd-CaCO^) of ook electrolytische reductie kunnen eventueel halogeenatamen op de 2,5-plaats aan de thiofeenring worden verwijderd. Een selectieve afsplitsing van halogeenatamen in de thiofeenring is b.v. met zink/ijsazijn mogelijk.
Wanneer als uitgangsmaterialen verbindingen met formule 1 aan-25 wezig zijn, waarin de hydroxylgroep en Rg waterstof is, is het vaak aanbevelenswaardig tegelijkertijd dehydrateringsmiddelen toe te voegen. Als dehydrateringsmiddelen komen b.v. in aanmerking: anorganische zuren zoals zwavelzuur of halogeenwaterstofzuren; organische zuren zoals oxaalzuur, mierezuur; thioylchloride; aluminiumchloride; 30 zinkchloride; tinchloride; boortrifluoride; kaliumwaterstofsulfaat; aluminiumoxide; fosforpentoxide; zuurchloride. In het bijzonder zal men als reductiemiddel naseerende waterstof in zuur medium toepassen.
De reductie kan in oplossing of suspensie worden uitgevoerd. Als oplosmiddelen kernen b.v. de reeds genoemde oplosmiddelen in aanmerking.
35 Wanneerd de produkten in de pyridinering drie geconjugeerde 8000285 -7- dubbele bindingen bevatten (pyridine ring) komen voor de kernhydroge-nering van deze pyridinering vooral platina-, rhodium- en ruthenium-katalysatoren (PtOg); rhodium/kool; rutheniumdioxide) bij temperaturen tussen 20 en 50°C en normale druk tot 10 bar in aanmerking.
5 Als oplosmiddelen zijn hiervoor in het bijzonder lage alcoholen, dioxan, tetrahydrofuran, ijsazijn, alcoholisch zoutzuur geschikt. Vaak is het doelmatig de overeenkomstige hydrochloriden te hydrogeneren. Een hy-drogenering van de pyridinering is echter b.v. ook .mogelijk met alkali-metalen (natrium) in lage alcoholen (ethanol) bij 20-150°C.
10 De omzetting van verbindingen met formule 1 in de overeenkom stige N-cociden kan b.v. in inerte oplosmiddelen zoals chloroform of andere chloorkoolwaterstbffen, benzeen, tolueen, aceton of azijnzuur-ethylester met waterstofperoxide, een gebruikelijk alifatisch of aromatisch perzuur (perazijnzuur, benzoperzuur, m-chloorbenzoperzuur) 15 of andere enkelvoudige substitutieprodukten van het waterstofperoxide, zoals alkylperoxiden (b.v. tert.butylperoxide) bij temperaturen tussen 0 en 150°C, bij voorkeur 0-100°C worden uitgevoerd.
Volgens werkwijze b) gaat men uit van verbindingen met formule 1. Verbindingen met formule 1, waarin de hydraxygroep en Rg water-20 stof is, en de overige symbolen de aangegeven betekenissen hebben, kunnen door behandeling met dehydrateringsmiddelen in-verbindingen met formule 1 worden omgezet, waarin R1 en Rg tezamen een tweede binding tussen de substituenten R^ en Rg dragende koolstofatcmen vormen.
Deze dehydratering geschiedt op de bovenstaand aangegeven wijze.· 25 Verbindingen met formule 1 waarin Rf en Rg de tweede binding tussen de de substituenten R1 en Rg dragende koolstof atomen vormen of waarin R.j de hydraxylgroep en Rg waterstof is, en de overige symbolen de aangegeven betekenissen hebben, kunnen door behandeling met reductie-middelen in verbindingen worden cmgezet, waarin R1 en Rg waterstof 30 zijn. Omstandigheden voor deze reductie zijn bovenstaand reeds aangegeven. De omzetting in de N-axiden geschiedt eveneens op de wijze zoals bovenstaand is aangegeven.
Verbindingen met formule 1, waarin R^ waterstof is, en de overige symbolen de aangegeven betekenissen hebben, kunnen door cmzet-35 ting met een verbinding met de formule R^-W waarin R^ de aangegeven 3000235 -8- * ·. * a betekenissen "behalve waterstof heeft en ¥ een chloor, broom of jood-atocm is of tezamen met de op een na dichtstbij liggende Cïï2-groep van ook een ethyleenoxidering kan vormen, of waarin ¥ de groep RSOgO voorstelt, waarin R een alkylgroep met 1-6 koolstofatcmen 5- of een eventueel door een of meerdere lageuaikylgroepen gesubstitueerde fenyl- of naftylgroep (b.v. een tosylgroep) is, worden verkregen.
Deze reactie kan met of zonder oplosmiddel bij temperaturen tussen 20 en 200°C, bij voorkeur 50-150°C worden uitgevoerd. Als oplosmiddelen resp. dispergeemiddelen kernen b.v. in aanmerking: 10 aranatische koolwaterstoffen, b.v. benzeen, tolueen, xyleen; ali-fatische ketonen, b.v. aceton, methylethylketon; gehalogeneerde koolwaterstoffen, b.v. chloroform, tetrachloorkoolstof, chloorbenzeen, methyleenchloride; alifatische ethers, b.v. butylether; cyclische ethers, b.v. tetrahydrofuran, dioxan; sulfoxiden, b.v. dimethylsul-15 foxide; tertiaire zuuramiden, b.v. dimethylformamide, N-methylpyrro-lidon; alifatische alcoholen, b.v. methanol, ethanol, isopropanol, amylalcohol, tertiair butanol; cycloalifatische koolwaterstoffen, b.v. cyclohexaan e.d. Ook kunnen waterige mengsels van de genoemde oplosmiddelen worden toegepast. Vaak werkt men bij terugvloeitem-20 peratuur van het toegepaste oplosmiddel resp. dispergeeimiddel.
In het algemeen worden de reactieccmponenten in molaire hoeveelheden cmgezet. Eventueel kan door de omzetting ook bij aanwezigheid van zuurhindende middelen, b.v. alkalicarbonaten (potas, soda), alkali -hydroxiden of tertiaire aminen (b.v. triëthylamine) worden uitge-25 voerd. Het laatste geldt in het bijzonder wanneer de overeenkomstige halogeniden worden toegepast.
De verbinding met formule 1 kan b.v. in de vorm van een me-taalzout, in het bijzonder alkalizout (natrium- of kaliumzout b.v.) worden toegepast. In het bijzonder geldt dit wanneer de andere reac-30 tieccmponent een halogenide is.
Bij de uitvoering van de reactie kan als ethyleenchlorideux tgangsverbinding in plaats van de ethyleencocideverbinding ook het overeenkomstige halogeenhydrine of een mengsel van deze beide verbindingen (ruw syntheseprodükt) worden toegepast'.

voorstelt (in het hij zonder de methyl- of ethylgroep) kunnen op een hiervoor bekende wijze door dealkylering in verbindingen worden augezet, waarin waterstof is (zie Houben-Weyl, Methoden der Organischen Chemie, Band XI/I, blz. 976-990 (1957)» Tetradedron Letters Nr. 18, blz. 1567-5 1570 (1977); J. Org. Chem. 39, 1507 (197*0; Helvetica Chimica Acta 59, 866 (1976)).
Deze dealkylering kan b.v. met chloormierezuuresters, b.v. chloor-mierezuurethylester, chloozmierezuurvinylester, chloormierezuurfenylester, in een inert oplosmiddel, b.v. aromatische koolwaterstoffen (tolueen, 10 xyleen, benzeen), lage alifatische halogeenkoolwaterstoffen, b.v. 1,2-dichloorethaan tusse -50 en +100°C en een daaropvolgende zure of basische verzeping met b.v. alcoholisch alkali (KOH/butanol) of alcoholische anorganische zuren (HC1, HBr, HgSO^) bij temperaturen tussen 50 en 150°C worden uitgevoerd.
15 De verbindingen volgens de uitvinding worden in het algemeen als racematen verkregen. De optisch actieve antipoden verkrijgt men door toepassing van optisch actieve uitgangsstoffen of door racemaatsplitsing via de zouten van optisch actieve zuren, b.v. L-(+)-wijnsteenzuur, D-(-)-wijnsteenzuur, (+)-0,0'-dibenzoyl-D-wijnsteenzuur, (-)-0,0’-di-20 benzoyl-L-wijnsteenzuur, (-)-0,0’-di-p-toluoyl-D-wijnsteenzuur, (+)-0,Of-di-p-toluoyl-D-wijnsteenzuur, (+)-kamfer-10-sulfonzuur en andere.
De verbindingen met formule 1 kunnen volgens bekende methoden in de zouten worden cmgezet. Als anionen voor deze zouten komen de bekende en therapeutisch toepasbare zuurresten in aanmerking. Voorbeel-25 den voor dergelijke zuren zijn, HgSO^., fosforzuur, halogeenwaterstofzuren, ethyleendiaminetetraazijnzuur, sulfaminezuur, benzeensulfonzuur, p-tolueensulfonzuur, kamfersulfonzuur, methaansulfonzuur, guajazuleensul-fonzuur, malelnezuur, fumaarzuur, bamsteenzuur, wijnsteenzuur, melkzuur, ascorbinezuur, glycolzuur, salicylzuur, azijnzuur, propionzuur, glucon-30 zuur, benzoëzuur, citroenzuur., acetaminoazijnzuur, axyethaansulfonzuur.
Uit de zouten van de verbindingen kunnen op gebruikelijke wijze weer de vrije basen worden bereid, b.v. door behandeling van een oplossing in een organisch middel, b.v. alcoholen (methanol) met soda of natronloog.
35 De omzetting in de quartaire zouten geschiedt door omzetting van de overeenkomstige secundaire resp. tertiaire aminoverbindingen met 3000285 * -10- lage alkylhalogeniden (l-6 koolstofat omen; chloriden, hraniden, jodiden) in een oplosmiddel resp. suspendeermiddel zoals aromatische koolwaterstoffen (benzeen, tolueen, xyleen), gehalogeneerde alifatische koolwaterstoffen (CHCl^» CHgClg), verzadigde alifatische ethers, cyclische 5 ethers (dioxan), dimethylsulfaat bij temperaturen tussen 0 en 100°C.
De verbindingen vólgens de uitvinding zijn geschikt voor de bereiding van farmaceutische, preparaten. De farmaceutische preparaten resp. medicijnen kunnen een of meerdere van de verbindingen volgens de uitvinding of ook mengsels daarvan met andere faunaeeutisch werkzame 10 stoffen bevatten. Voor de bereiding van farmaceutische preparaten kunnen de gebruikelijke farmaceutische dragers en hulpstoffen worden toegepast. De geneesmiddelen kunnen enteraal, parenteraal, oraal of perlinguaal worden toegediend. De toediening kan b.v. plaatsvinden in de vorm van tabletten, capsules, pillen, dragees, zetpillen, zalven, geleien» 15 cremes, poeders, liquida, stuifpoeders of aerosolen plaatsvinden.
Als liquida komen b.v. in aanmerking: olieachtige of waterige oplossingen of suspensies, emulsies, injecteerbare waterige en olieachtige oplossingen of suspensies.
Voorbeeld I
20 (Di-3-thiënyl)-(N-methyl-l+-piperidyl)-carbinol met de formule 15.
a) Men bedekte H,86 g (0,2 g-atocm) magnesium met absolute tetrahy- . . 3 .
drofuran, mengde met een korreltje jood en 0,5 cm dibrocmethaan.
Zodra de reactie een aanvang had genemen, werd binnen 15 minuten 26,5 g (0,198 mol) k-chloor-N-methylpiperidine, opgelost in 30 cm absolute 25 tetrahydrofuran toegedruppeld. Vervolgens werd gedurende 2 uur onder 3 terugvloeikoeling gekookt en daarna met 120 cm absolute tetrahydrofuran verdund. Hieraan werd bij kamertemperatuur 19,1+ g (0,1 mol) f-dithiënyIketon in kleine porties toegevoegd.
Daarna liet men gedurende 2 uur bij het kookpunt nareageren.
30 Ter hydrolyse werd het reactiemengsel uitgegoten over ijs/ammonium- chloride (250 g/25 g). De organische fase werd afgescheiden, de waterige fase meerdere malen met chloroform uitgeschud. Na drogen en afdestilleren van het oplosmiddel onder verminderde druk werd het gewenste pre-dukt door droge kolcmchrcmatografie over kiezelgel (elueemniddel:ether/ 35 methanol 90$/10$) geïsoleerd. Cm een zout te vormen werd de verkregen 8 0 0 0 2 S 5
It -11- vrije base in ethanol opgelost en met een equimolaire hoeveelheid maleine-zuur, eventueel opgelost in ethanol gemengd. Het maleaat werd uit ethanol herkri st alii seer d. Het vriespunt van het maleaat was l89-191°C. De opbrengst bedroeg 25$ (betrokken op £-dithiênylketon).
q q 5 b) Aan een mengsel, bestaande uit 350 cnr dilsopropylether en. 200 cm butyllithium (20^-ige oplossing in hexaan) werd 63,5 g (0,38 mol) 3-broan.thiofeen, opgelost in 100 cm^ dilsopropylether, bij -75°C toege-druppeld. ïïa de toevoeging werd gedurende nog 2 uur nageroerd, voordat een oplossing van 33 g (0,19 mol) N-methyl-U-piperidinecarbonzuurethyΙ-ΙΟ ester in 50 cm^ dilsopropylether druppelsgewijze bij -75°C werd toegevoegd. Bij alle bewerkingen werd ervoor gewaakt dat de reactietemperatuur van -70°C niet werd overschreden. De hydrolyse vend plaats door toevoe-ging van 250 cm water. Men extraheerde meerdere malen met chlorofoim.
Ia gebruikelijke droging werd de oplossing ingedampt en gekoeld. Het 15 uitgekristalliseerde carbinol werd afgezogen en gedroogd en zonder verdere zuivering aan dehydratering onderworpen (zie voorbeeld II).
De opbrengst bedroeg 73$.

U—(di —3-thieny1-methyleen)-N-methylpiperidine met de formule 16.
20 Men loste lU'g (0,0½ mol) (di-3-thiëny1)-(I-methy1-U-piperidy1)- 3 3 carbinol (ruw produkt) op in 200 cm methanol, mengde met 20 cm 8 I isopropanolisch zoutzuur en verhitte gedurende 1 uur. Het oplosmiddel werd onder verminderde druk verwijderd en het verkregen hydrochloride uit isopropanol herkristalliseerd. Het vriespunt van het 25 hydrochloride bedroeg 230°C. De opbrengst was 79$·
Het uitgangscarbinol werd analoog aan voorbeeld Ib verkregen.
Voorbeeld III
U-/3-thiënyl~(2,5-dimethyl-3-thiënyl)-methyleen7-N-methylpiperidine met de formule 17.
30 Men dehydrateerde b g 2,5-dimethyl-3-thiënyl)-(l-methyl-4- 3 3 piperidyl)-3-thiënyl)-carbinol in 50 cm methanol met 10 cm 8 I isopropanolisch HC1 analoog aan voorbeeld II. Ter zuivering werd uit het verkregen olieachtige hydrochloride eerst de base met ammoniak in vrijheid gebracht. Deze werd door droge kolcmchraaatografie over 35 kiezelgel (elueeimiddel : ether/methanol 95$/5$) geïsoleerd, in aceton 3000285 τ -12- opgelost en met een equimolaire hoeveelheid oxaalzuur gemengd. Het verkregen oxalaat werd uit azijnester/ethanol herkristalliseerd. Het vriespunt van het oxalaat was 155-157°C. De opbrengst bedroeg 56$.
Het uitgangscarbinol werd analoog aan voorbeeld Ia uit 12,2 g 5 (0,055 mol) (2,5-dimethyl-3-thiënyl)-3-thiënylketon, 1^,7 g (0,11 mol) U-chloor-ET-methylpiperidine en 2,67 g (0,11 g atoom) magnesium verkregen.
Voorbeeld IV
U-(di-3-thienyl-methyleen )-N-cyclohexylpiperidine met de formule 18.
10 Men dehydrateerde 5 g (di-3-thiënyl )-(H-cyclohexyl-U-piperidyJ;)- 3 3 carbinol in 50 cmJ methanol met 10 cm 8 IT isopropanolisch HC1 analoog aan voorbeeld II en werkte op. Het hydrochloride werd uit isopropanol herkristalliseerd. Het vriespunt van het hydrochloride bedroeg 232°C.
De opbrengst was bl%. Het uitgangscarbinol werd analoog aan voorbeeld Ib 15 bereid.
Voorbeeld V
U-(di-3-thiëny3methyleen)-piperidine met de formule 19·
De oplossing van 122 g (0,^3 mol) !+-(di-3-thiënylmethyleen)-H-methylpiperidine in UOO cm^ tolueen werd bij 80°C bij een mengsel van

20 100 g (0,886 mol) chloormierezuurethylester en 200 cm tolueen gedruppeld. Ha de toevoeging werd het reactiemengsel gedurende nog 3 uur bij 80°C geroerd en door afdestilleren van het oplosmiddel onder verminderde druk ingedampt. Hierbij kristalliseerde het ^-(di-3-thiëryl-metbyleen)-N-carbethoxy-piperidine uit en werd uit isopropanol her-25 kristalliseerd. De opbrengst bedroeg 98$. Het vriespunt was 109-111°C.
Een mengsel van l63 g (0,U8U mol) van deze carbethaxyverbinding, 120 g (2,2 mol) kaliumhydroxide en 1200 cm n-butanol werd onder terug-vloeikoeling gekookt, totdat volgens een dunnelaagchramatografische controle de uitgangsverbinding volledig was angezet. Het butanol werd 30 onder verminderde druk verwijderd, het residu met water gemengd en meerdere malen met metbyleenchloride uitgeschud. De zoutvorming geschiedde met maleïnezuur in aceton als oplosmiddel. De opbrengst was 68$. Het vriespunt van het maleaat was 173-17^°C.

Men loste 6,8 g (0,026 mol) 4-(di-2-thiênyl-methyleen)-piperidine

en 12,6 g (0,156 mol) 2-chloor-ethanol op m 100 cm xyleen en kookte hij aanwezigheid van 21,6 g (0,156 mol) kaliumcarbonaat gedurende 2 uur onder terugvloeikoeling. Na toevoegen van water, afscheiden van de or-5 ganisehe fase, meerdere malen extraheren van de waterige fase met chloroform, verenigen en drogen van. de organische fasen werd het oplosmiddel ander verminderde druk afgedestilleerd en het hydrochloride met iscpranolisch zoutzuur en aceton als oplosmiddel verkregen.
De herkristallisatie vond plaats uit isopropanol. Het vriespunt van 10 het hydrochloride was 196°C. De opbrengst bedroeg 63%.
De bereiding van de thiënyluitgangsstof vond plaats analoog aan voorbeeld V.
Voorbeeld VII
4- (di-3-tbiënyl-methyleen) -N-( 2-hydr oxy-ethyl) -piperidine.
15· De bereiding geschiedde analoog aan voorbeeld VI uit 6,8 g 4-(di-3-thiënyl-methyleen)-piperidine en 12,6 g 2-chloor-ethanol. Het vriespunt van het hydrochloride was 170-lTl°C(uit isopropanol); de opbrengst 48#.
Voorbeeld VIII
20 4-(di-3-thiënyl-methyleen)-N-/2-hydroxy-propyl-(1 )7-piperidine.
De bereiding geschiedde analoog aan voorbeeld VI uit 6,8 g 4-(di-3-thiënyl-methyleen)-piperidine en 4,9 g 1-chloor-2-hydroxy-propaan. Het vriespunt van het hydrochloride (uit isopropanol) was 209°C; de opbrengst 22$.
25 Voorbeeld IX
4- /J.i -2-thi ëny1-met hyleen/-N-/2., 3-dihydroxy-propyl- (1 )ƒ-piperidine met de formule 21.
Men lost 10 g (0,038 mol) 4-(di-2-thiënyl-methyleen)-piperidine en 5,6 g (0,076 mol) glycide op in 50 cm ethanol en kookte gedurende 30 8 uur onder terugvloeikoeling. Daarna werd het oplosmiddel verwijderd en de gewenste glycidylverbidding door droge kolcmchrcmatografie over kiezelgel (elueermiddel:chloroform) geïsoleerd. Ter verdere zuivering werd het hydrochloride bereid en twee keer uit isopropanol herkristalliseerd. Het vriespunt van het hydrochloride bedroeg l4l-l43°C; 35 de opbrengst 33%· 00 o 0 2 δ 5 f -]Α- ' Analoog aan voorbeeld IX werden de in tabel A vermelde verbin dingen met de fonmile 22 bereid. Telkens werd 0,038 mol U-(di-3-thiënyl-methyleen)-piperidine met 0,076 mol van het met de alcohol R^-OH overeenkomende glycide omgezet.

(di-3-thiënyl-methyleen) -N-dimethyl-piperidinium j odide met formule 23.
Men mengde 30 g (0,11 mol) ^-(di-3-thiënyl-methyleen)-N-methyl-piperidine in 200 cm ether met 28,1 g (0,20 mol) methyljodide. Na 25 meerdere uren staaain een koelkast werden de neergeslagen kristallen afgezogen en uit methanol/water herkristalliseerd. Het vriespunt was 272:°C, de opbrengst 56% ·
Voorbeeld XV
^-(di-3-thiënyl-methyleen)-N-methyl-piperidine-oxide met de formule 2b.
30 Men loste 5·>5 g (0,02 mol) b-(di-3-thiënyl-methyleen)-N-methyl- . ... 3 piperidine m 50 cm methanol op en mengde druppelsgewijze met 7,2 g 30#-ig waterstofperoxide onder koelen met ijs en liet gedurende 8 dagen op kamertemperatuur staan. Overmaat waterstofperoxide werd door toevoegen van Pt-kool vernietigd. Na filtreren en verwijderen van het op-35 losmiddel bij kamertemperatuur werd het amineoxide in azijneater tot 8000235 -15- kristallisatie gebracht. Het vriespunt bedroeg 92-9é>°C; de opbrengst 775.
Voorbeeld XVI
k-(di-3-thiënyl-methyl)-N-methyl-piperidine met de formule '25.
5 Men loste 9,5 g (0,03 mol) ^-(di-3-thiënyl-methyleen)-N-methyl- piperidine-hydrochloride in 270°C op en hydrogeheerde bij aanwezigheid van 9,5 g Pd-BaSO^ (9,23! Pd) bij Ê5°C en 5 bar. De zuivering vond plaats door droge kolamchramatografie over kiezelgel (elueermiddel: chloroform/methanol 95%f3% ) en herkristallisatie uit chloroform/ 10 petroleumether. Het vriespunt van het hydrochloride was 251-253°C, de opbrengst 11%.
Farmacologische en farmaceutische gegevens.
De verbindingen volgens de uitvinding zijn geschikt voor de bereiding van farmaceutische preparaten en samenstelling. De farma-15 ceutische samenstellingen resp. geneesmiddelen bevatten als werkzame stoffen een of meerdere van de verbindingen volgens de uitvinding, eventueel gemengd met andere farmacologisch resp. farmaceutisch werkzame stoffen. De bereiding van de geneesmiddelen geschiedt op bekende wijze, waarbij de bekende en gebruikelijke farmaceutische 20 hulpstoffen en andere gebruikelijke drager- en verdunningsmiddelen kunnen worden toegepast. rietsuiker, of melksuiker, lecithine, pectine, zetmeel (b.v. maïszetmeel), alginezuur, tylose, talk, lycopodium, kiezelzuur (b.v. colloïdaal kiezelzuur), cellulose, cellulosederivaten (b.v. cellulose-ethers, waarbij de cellulose-hydroxylgroepen gedeeltelijk met lage 35 verzadigde alifatische alcoholen en/of lage verzadigde alifatische 8000235 -ι6- oxyalcoholen zijn veretherd, b.v. methyloxypropylcellulose), stearaten, magnesium- en calciumzouten van vetzuren met 12-22 koolstof at amen, in het "bijzonder de verzadigde (b.v. stearaten), emulgatoren, oliën en vetten, in het bijzonder plantaardige (b.v. aardnotenolie, ricinusolie, 5 olijfolie, sesamolie, katoenzaadolie, maïsolie, tarwekiemolie, zonne-bloemzaadolie, kabeljouw-leverolie, mono-, di- en triglyceriden van verzadigde vetzuren en mengsels daarvan), farma ceutisch. aanvaardbare een- of meerwaardige alcoholen en' polyglycolen, b.v. polyethyleenglycolen en derivaten daarvan, esters van alifatische 10 verzadigde of onverzadigde vetzuren (2-22 koolstofatcmen, in het bijzonder 10-18 koolstofatcmen) met eenwaardige alifatische alcoholen (1-20 kool-stofatamen) of meerwaardige alcoholen zoals glycolen, glycerine, di-ethyleenglycol, pentaerythrièt, sorbiet, manniet e.d., die eventueel ook veredeld kunnen zijn, benzylbenzoaat, dioxalanen, glycerinefor-15 malen, tetrahycbrofurfurylalcohol, polyglycolethers met alcoholen met 1-12 koolstofatcmen, dimethylaceetamide, lactamiden, lactaten, ethyl-carbonaten, siliconen (in het bijzonder middelviskeuze dimethylpoly-silóxanen) magnesiumcarbonaat en dergelijke.
Voor de bereiding van oplossingen kernen b.v. water of fysio-20 logisch aanvaardbare organische oplosmiddelen in aanmerking, b.v. ethanol, 1,2-propyleenglycol, polyglycolen en derivaten daarvan, dimethylsulfoxide, vetalcoholen, triglyceriden, partieelesters van glycerine, paraffine e.d.
Bij de bereiding van de preparaten kunnen bekende en gebrui-25 kelijke oploshulpmiddelen, resp. emulgatoren worden toegepast.
Als oploshulpmiddelen en emulgatoren komen b.v. in aanmerking: poly-vinylpyrrolidon, sorbitanvetzuuresters zoals sorbitantrioleaat, lecithine, acacia, tragacanth, polyoxyethyleerd sorbitanmonoöleaat, polyoxyethyleerde veten, polyoxyethyleerde oleotriglyceriden, gelino-30 liseerde olèotriglyceriden, polyethyleenoxidecondensatieprcdukten van vetalcoholen, alkylfenolen of vetzuren. Polyoxyethyleerd betekent hierbij dat de betreffende stoffen polyoxyethyleenketens bevatten, waarvan de polymerisatiegraad in het algemeen tussen 2 en ^0 en in het bijzonder tussen 10 en 20 ligt. D.ergelijke polyoxyethyleerde 35 stoffen kunnen b.v. door omzetting van hydroxylgroepenheudende ver- 8000235 -17- bindingen (b.v. mono- of diglyceriden of onverzadigde verbindingen, zoals b.v. verbindingen die oliezuurresten bevatten) met ethyleenaxide worden verkregen (b.v. HO mol ethyleenaxide per mol glyceride). Voorbeelden voor oleotriglyceriden zijn olijfolie, aar dnotenolie, rieinus-5 olie, sesamolie, katoenzaadolie, maïsolie (zie ook Dr. H.P. Fiedler, "Lexikon der Hilfsstoffe fur Phaxmazie, Kosmetik und angrenzende Gebiete 1971, blz. 191-195).
Bovendien is de toevoeging van conserveringsmiddelen, stabilisatoren, buffermaterialen, b.v. calciumwaterstoffosfaat, colloïdaal 10 aluminiumhydroxide, smaakverbeteraars, antioxidantia en complexvormers (b.v. ethyleendiaminotetraazïjnzuur) en dergelijke mogelijk. Eventueel wordt ter stabilisering van het werkzame stofmolecuul met fysiologisch aanvaardbare zuren of buffers ingesteld op een pH-gebied van ongeveer 3-7- In het algemeen wordt een zo neutraal mogelijke tot 15 zwakzure (tot pH 5) pH-waarde geprefereerd.
Als antioxidantia komen b.v. natriummetabisulfiet, ascorbine-zuur, galluszuur, galluszuur-alkylesters, butylhydraxyanisool, nordi-hydroguajareetzuur, tocoferolen en tocoferolen + synergïsten (stoffen die zware metalen door ccmplexvorming binden, b.v. lecithine, ascorbine-20 zuur, fosforzuur) in aanmerking. De toevoeging van synergïsten verhoogt de antioxygene werking van de tocoferolen aanzienlijk.
Als conserveringsmiddelen kernen b.v. sorbinezuur, p-hydraxy-benzoëzuuresters (b.v. lage alkylesters), benzoëzuur, natriumbenzoaat, trichloorisobutylalcohol, fenol, cresol, benzethoniumchloride en 25 fozmalinederivaten in aanmerking.
De farmacologische en gallenische handhaving van de verbindingen volgens de uitvinding geschiedt volgens gebruikelijke standaardmethoden. Werkzame stoffen en hulp- resp. dragerstoffen worden b.v. door roeren of hemogeniseren (b.v. met behulp van colloidmolens, 30 kogeknolens) goed gemengd, waarbij in het algemeen bij temperaturen tussen 20 en 80°C, bij voorkeur 20-50°C wordt gewerkt.
De toediening van de werkzame stoffen resp. de geneesmiddelen kan op de huid of slijmhuid of in het inwendige van het lichaam plaatsvinden, b.v. oraal, enteraal, pulmonaal, rectaal, 35 nasaal, vaginaal, linguaal, intraveneus, intraarterieel, intracardiaal, 5000285 . ·» -18- s intramusculair, intraperitoneaal, intracutaan, subcutaan.
In het bijzonder is ook de toevoeging van andere -werkzame ge-neesmiddelstoffen mogelijk resp. gunstig.
De verbindingen volgens de uitvinding tonen bij onder narcose 5 gebrachte en aan thoracotcmie onderworpen honden, waarbij door vernauwing van een coronaire arterietak een locaal begrensde storing van de bloed-verzorging werd veroorzaakt, welke met behulp van epicardiale electro-cardiogramsignalen (ECG-signalen) wat plaats en tijd betreft nauwkeurig kon worden bepaald, een goede antiangineuze werking. De bepaling van 10 deze werking geschiedt b.v. volgens de methode van Skekeres et al, J. Pharmacol, exper. Ther. 19$, 15 (197$) met de volgende verandering:
De stenose wordt aan het coronairvat zelf ingesteld. De basiswerking komt tot uiting in een vermindering van de door de stenose bewerkte veranderingen in het karakteristieke gedeelte van de ECG-signalen, 15 die van het oppervlak van het hart worden afgeleid. B.v. wordt bij bovengenoemde proefkethode na een dosis van 3 mg/kg hond intraveneus de verandering in het electrocardiogram in het ischemiegebied met 62% verminderd. Deze therapeutische werking is verbonden met een daling van de frequentie en matige stijging van de bloeddruk.
20 Deze antiangineuze werking is vergelijkbaar met die van het bekende geneesmiddel nitroglycerine.
De laagste reeds antiangineus werkzame dosis bij bovengenoemde dierproef is b.v. 1 mg/kg oraal, 1 mg/kg sublinguaal, 0,1 mg/kg intraveneus.
25 Als algemeen dosisgebied voor de antiangineuze werking (dier proef zoals boven) komt b.v. in aanmerking: 1-50, in het bijzonder 3-20 mg/kg oraal, 1-50, in het bijzonder 3-20 mg/kg sublinguaal, 0,1-20, in het bijzonder 1-10 mg/kg intraveneus.
Indicaties waarvoor de onderhavige verbindingen in aanmerking 30 kunnen komen zijn angina pectoris, hartinfarct, tachycarde ritme-storingen.
De farmaceutische preparaten kunnen b.v. tussen 10 en 500 mg van de actieve componenten volgens de uivinding bevatten. De toedieningsvormen kunnen b.v. zijn tabletten, capsules, pillen, dragees, 35 zetpillen, zalven, geleien, cremes, poeders, stuifpoeders, aerosolen of η η 6 0 ? pi 5

-19- vloeistoffen. Als vloeibare toedieningsvormen kamen b.v. in aanmerking olieachtige of alcoholische resp. waterige oplossingen alsmede suspensies en emulsies. De voorkeur hebben tabletten, die b.v. tussen 50 en 500 mg of oplossingen die b.v. tussen 1 en 5$ aan actieve stof bevatten.
5 De enkelvoudige dosis van de actieve componenten volgens de uitvinding kan b.v. liggen a) bij orale geneesmiddelvormen tussen 200 en 500 mg, b) bij parenterale geneesmiddelvormen (b.v. intraveneus, intramusculair) tussen 100 en 300 mg, 10 c) bij geneesmiddelvormen voor rectale of vaginale toediening tussen 200 en 500 mg, d) bij geneesmiddelvormen voor locale applicatie op de huid en slijm-huid, (b.v. in de vorm van oplossingen, lotions, emulsies, zalven enz.) tussen 1 en 5%.
15 (De doses zijn telkens betrokken op de vrije base).
Bij wijze van voorbeeld kunnen driemaal daags 1 tot 3 tabletten met een gehalte van 100-200 mg werkzame stof of b.v. bij intraveneuze . . . . . 3 injectie een- tot driemaal daags een ampul met een inhoud van 2-10 cm met 50-100 mg stof worden aanbevolen. Bij orale toediening is de mini-20 male dagelijkse dosis b.v. 200 mg; de maximale dagelijkse dosis bij orale toediening dient niet hoger dan 1,5 g te zijn.
De acute toxiciteit van de verbindingen volgens de uitvinding bij de muis (uitgedrukt door het LD 50 mg/kg; methode volgens Miller en fainter: Proc. Soc. Exper. Biol. a. Med. (19^Μ 2βΐ ligt b.v.
25 bij orale toediening tussen 300 en 1000 mg/kg.
De geneesmiddelen kunnen in de mensgeneeskunde, de diergeneeskunde en de landbouw alleen of gemengd met andere farmaceutisch actieve stoffen worden toegepast.
Voorbeelden voor farmaceutische preparaten 3 0 Voorbeeld voor tabletten
Men mengde 8 kg werkzame stof (verbinding volgens voorbeeld VI) met 5 kg melksuiker en 3 kg microkristallijne cellulose en granuleerde met een oplossing van 0,3 kg polyvinylpyrrolidon in 1,2 kg water op gebruikelijke wijze. Na bijmengen van 3,^5 kg microkristallijne cellulose, 35 2 kg maïszetmeel, 0,05 kg hoogdispers kiezelzuur en 0,2 kg magnesium- 8000285 « -20- stearaat werden tabletten met een gewicht van 220 mg, een diameter van 9 mm en een bollingsstraal van 13,5 mm geperst. De hardheid van de tabletten bedroeg 5-7 kg (Heberlein-hardheidstester).
Elk tablet bevatte 80 mg werkzame stof.
j. Voorbeeld voor ampullen
Men loste U00 g werkzame stof (verbinding volgens voorbeeld VII) op in een mengsel van h kg 1,2-propyleenglycol en 15 kg water voor injectiedoeleinden, bracht de totale oplossing met water voor injectiedoeleinden op 20 1 en filtreerde. Na inbrengen van de oplossing 10 in ampullen tot 2 cm vond sterilisatie op gebruikelijke wijze bij 120°C, 1 bar gedurende 20 minuten plaats. Een ampul bevatte bO mg 3 werkzame stof m 2 cm .

- 4 9 * -21- CONCIUSIES:
- 1. Verbindingen met foimule 1 van het formuleblad, waarin en R^ waterstof voorstellen of samen een tweede binding tussen de koolstof-atomen die de substituenten R^ en Rg dragen vormen, R^ bovendien ook een hydroxylgroep kan zijn, R^ waterstof, een cycloaliylgraep met 3-8 5 koolstofatcmen of een alkylgroep met 1-20 koolstofatcmen, die eventueel ook een of twee hydroxyIgroepen kan bevatten, voorstelt en de groepen R^, R,-, Rg en Rj gelijk of verschillend zijn en waterstof, alkyl-groepen met 1-6 koolstof atcmen of halogeenatcmen voorstellen, hun ïï-oxiden, hun quartaire zouten en hun zuuradditiezouten, waarbij 10 tenminste een thiënyl-(3)-groep aanwezig is, warneer R^ een niet gesubstitueerde alky lgroep met 1-6 koolstof atcmen is.
- 2. Werkwijze voor het bereiden van verbindingen met foimule 1, waarin R^ en R^ waterstof voorstellen of samen een tweede binding vormen tussen de koolstofatcmen die de substituenten R^ en dragen,
- 15 R^ bovendien ook een hydroxylgroep kan zijn, R^ waterstof, een cyclo-alky lgroep met 3-8 koolstof at amen of een alkylgroep met 1-20 koolstof atomen, die eventueel ook een of twee hydroxyIgroepen kan bevatten, voorstelt, en de groepen R^, R^, Rg en Rj gelijk of verschillend zijn en waterstof, alkyIgroepen met 1-6 koolstofatcmen of halogeenatcmen 20 voorstellen, hun lï-oxiden, hun quaternaire verbindingen en hun zuuradditiezouten, waarbij tenminste een thiënyl-(3)-groep aanwezig is, wanneer een niet gesubstitueerde alkylgroep is, met het kenmerk, dat men a) een verbinding met foimule 2, die ook drie geconjugeerde dubbele 25 bindingen in de ring kan bevatten, waarin R^ de aangegeven betekenissen heeft (R^ ontbreekt wanneer een pyridinering aanwezig is) en X A) de groep -C0Z voorstelt, waarbij Z een halogeenatoaa of een alkoxygroep met 1-6 koolstofatomen of een eventueel een- tot 30 tweevoudig door halogeenatomen en/of alkyIgroepen met 1-6 kool- stofatcmen gesubstitueerde thiênylgroep is, of B) lithium of de groep -MgHal voorstelt, waarin Hal chloor, brocm of jood is, hetzij in geval A) met een verbinding met foimule 3 waarin X' 35 lithium of de groep -MgHal is en de thiënylgroep ook een-tot twee- 6000285 « -22- voudig door halogeenat omen en/of alkylgroepen met 1-6 koolstof-atcrnen kan zijn gesubstitueerd, cmzet, hetzij in geval B) met een verbinding met formule U cmzet waarbij in een van de bovenstaand genoemde uitgangsverbindingen tenminste 5 l-thiënyl-(3)-groep aanwezig moet zijn, wanneer de substituent R^ in formule 2 een niet gesubstitueerde alkylgroep met 1-6 koolstof-atamen is, en de verkregen verbindingen eventueel met reductiemid-delen en/of dehydrateringsmiddelen behandelt en/of door de groep R^ alkyleert en/of in het N-cocide cmzet of 10 b) uitgaat van een verbinding met formule 1 en wanneer R^ de hydroxy 1-groep en R^ waterstof zijn, deze met dehydrateringsmiddelen en/of rednctiemiddelen behandelt en/of de groep R^ tegen andere sub-stituenten binnen de voor R^ gegeven betekenissen uitwisselt en eventueel de verkregen verbindingen cmzet in het N-oxide.
- 3. Werkwijze volgens conclusie 2, met het kenmerk, dat men de verkregen verbindingen in de zuuradditiezouten resp. de quartaire zouten cmzet. h. Geneesmiddel, met het kenmerk, dat het als werkzame stof een of meer verbindingen uit de conclusies 1-3 tezamen met een gebruikelijke farmacologische drager en/of een verdunningsmiddel bevat.
- 5. Werkwijze voor het bereiden van een geneesmiddel, met het kenmerk, dat men een of meer van de verbindingen uit de conclusies 1-3 met gebruikelijke farmaceutische dragerstoffen of verdunningsmiddelen resp. andere fysiologisch aanvaardbare hulpstoffen verwerkt tot farmaceutische preparaten resp. in een farmaceutisch geschikte toedieningsvorm brengt.
