Claims (6)
1. Een werkwijze voor het induceren van rijst ter verbetering van de tolerantie voor zout, welke werkwijze de volgende stappen omvat: Stap 1: het desinfecteren van de oppervlakken van rijstzaden, en vervolgens het oplossen van 5 - hydroxy - L - norvaline met een 3 - 10wt.%o0 NaCl oplossing om een 5 - hydroxy - L - norvaline oplossing te bereiden met een concentratie van 20 - 400 mg IL; Stap 2: het inweken van de rijstzaden met de in stap 1 bereide 5 - hydroxy - L - norvaline oplossing gedurende 16 - 24 uur bij 20 - 28°C om de behandelde rijstzaden te verkrijgen, zodat de rijstzaden normaal kunnen ontkiemen en groeien in een omgeving waar zoutstress heerst.
2. De werkwijze voor het induceren van rijst ter verbetering van de tolerantie voor zout volgens conclusie 1, waarbij het desinfecteren van de oppervlakken van de rijstzaden in stap 1 specifiek de volgende stappen omvat: — het selecteren van rijpe rijstzaden met volle zaden en zonder meeldauwvlekken, — het2 tot 3 keer spoelen van de rijstzaden met een 70 - 75 wt.% ethanoloplossing, waarbij het spoelen elke keer 30 - 40 s duurt, — het 2 tot 3 keer wassen van de rijstzaden met steriel water en vervolgens — het steriliseren van de rijstzaden met een 5 -10 gew.% NaClO-oplossing gedurende 5- 15 min, — het wassen van de rijstzaden met steriel water gedurende 3 - 5 maal, en — ten slotte het drogen van de oppervlakken van de zaden met steriel filtreerpapier.
3. De werkwijze voor het induceren van rijst ter verbetering van de tolerantie voor zout volgens conclusie 1, waarbij de omgeving waar zoutstress heerst als volgt is: een zoutstressoplossing als irrigatiewater is een gemengde oplossing van 5 - hydroxy - L - norvaline en NaCl, waarbij de 5 - hydroxy - L - norvaline een concentratie heeft van O - 400 mg / L, en de NaCl een concentratie heeft van 3 - 10wt.%o.
4. De werkwijze voor het induceren van rijst ter verbetering van de tolerantie voor zout volgens conclusie 1, waarbij in stap 1 de oppervlakken van de rijstzaden worden gedesinfecteerd en vervolgens de 5 - hydroxy - L - norvaline wordt opgelost in een 5 wt.%o NaCl - oplossing om een 5 - hydroxy - L - norvaline-oplossing te bereiden met een concentratie van 200 mg/l..
5. De werkwijze voor het induceren van rijst ter verbetering van de tolerantie voor zout volgens conclusie 1, waarbij een werkwijze voor het inspecteren van de kiemkracht en de groeisnelheid van de rijstzaden in stap 2 de volgende stappen omvat: — het plaatsen van de in stap 2 behandelde rijstzaden in kiemdozen voor een kiemproef,, — het opvullen van twee lagen steriel filtreerpapier in elk van de kiemdozen, waarin telkens 100 zaden worden gelegd — het in elke kiemdoos toevoegen van de gemengde oplossing van de 5 - hydroxy - L - norvaline en de NaCl met dezelfde concentratie als in stap 1 om het filtreerpapier vochtig te kunnen houden, — het afdekken van elke kiemdoos, — het plaatsen van elke kiemdoos in een omgeving met een temperatuur van 20 - 28°C om in het donker te kweken gedurende 2 - 3 dagen en vervolgens in het licht waarbij wordt gekweekt gedurende 2 — 3 dagen met een lichtintensiteit van 4000 - 10000 Ix, en het tellen van de kieming na kweken in het licht gedurende 2 - 3 dagen, — het selecteren van 30 kiemplanten met een constante groei voor een kiemplantgroeiproef in drievoud; en — het overbrengen van de ontkiemde zaailingen in een PCR-plaat met 96 putjes, waarvan de bodem is afgeknot, — het in elk putje plaatsen van een zaailing, — het kweken van de zaailingen in een kunstmatige klimaatincubator onder kweekomstandigheden waarbij de temperatuur 25 - 28°C bedraagt, de lichtintensiteit 3000 - 10000 Ix bedraagt, de vochtigheidsgraad 60 - 80% bedraagt, en de tijdverhouding tussen kweken in het licht en in het donker 16 - 8 uur bedraagt, en vervolgens — het onder het vloeistofoppervlak planten in de gemengde oplossing van 5-hydroxy-L- norvaline en NaCl in dezelfde concentratie als die in stap 1, — het eens in de 3 dagen verversen van de beplantingsoplossing, en — het meten van de planthoogte, de wortellengte en het verse gewicht van elke zaailing na 2 weken.
6. De werkwijze voor het induceren van rijst ter verbetering van de tolerantie voor zout volgens conclusie 5, waarbij bij het tellen van de kiemkracht een zaailing met een som van de wortellengte en de planthoogte van meer dan 0,5 cm als gekiemd wordt beschouwd.
