Abstract

Blood serum is used for many types of analyses and may provide useful insights into a state of health of a person of whom the blood serum belongs. Blood serum needs to be separated from blood, which in turn has to be collected from a person. A device is provided in which a strip of lateral flow paper may be placed. When a sample of blood is provided on a receiving section of the strip of lateral flow paper, due to capillary action, the blood is transported through the lateral flow paper. While being transported, the lateral flow paper acts a sieve in which amongst others red blood cells are trapped. Blood serum, and many constituents it holds, is not sieved and is therefor separated from the blood. The blood serum on the lateral flow paper may be used for analysis when dissolved in an extraction liquid.

Description

Title: Device for collecting a sample of blood.
TECHNICAL FIELD
The present invention relates to a device for collecting blood samples.
BACKGROUND
Examples of conventional ways of collecting a sample of blood are inserting a needle into a vein, and finger puncture or heel puncture sampling, wherein the skin is pierced with a lancet, and a small sample of blood may be collected in a capillary tube or on a piece of paper.
When collecting blood by inserting a needle into a vein, an unnecessarily large sample of blood is collected into a tube, which has to be stored under precise conditions and eventually has to be disposed of properly. A sample collected from a finger prick or heel prick sampling on paper allows for a smaller amount of analysis options, as the blood dries after collection.
From a collected blood sample, obtained by venepuncture, blood serum may be separated by allowing blood to clot and subsequently separated from the serum by centrifuging the blood. The blood serum then needs to be stored under specific conditions in order to prevent deterioration.
SUMMARY
It is preferred to provide a device for collecting small blood samples. Furthermore, it is preferred to provide a method for separating blood cells from serum, and for preserving the blood serum to be used lateron for analysis without the need for precisely conditioned storage.
A first aspect provides a device for collecting a blood sample, comprising an elongated frame, arranged to hold a strip of lateral flow paper. The device comprises: a first frame part, comprising an opening arranged to expose a receiving section of the strip of lateral flow paper such that the sample of blood can be provided directly to the receiving section through the opening; a second frame part, arranged to be connected to the first frame part to support the strip of lateral flow paper in the device between the first frame part and the second frame part. At least one of the first frame part and the second frame part comprises one or more support protrusions arranged to support the strip of lateral flow paper; and the first frame part and the second frame part are releasably connected, such that the strip of lateral flow paper may be removed from the device.
Lateral flow paper is paper comprising fibres oriented in a particular direction, parallel to one another. Such paper may comprise cellulose fibres constituting the paper and oriented glass fibres which glass fibres are provided parallel to one another. Such paper is used in pregnancy tests to allow liquid matter to flow towards markers that react with certain constituents resolved in the liquid.
In an embodiment of the device for collecting a blood sample, the second frame part is hingedly connected to the first frame part. The first frame part may comprise a scraping protrusion adjacent to the opening, arranged to increase the ease with which the sample of blood may be collected from a finger in which a small hole has been punctured.
A second aspect provides a method for separating blood serum from a sample of blood comprising providing the sample of blood to a receiving section a strip of lateral flow paper, wherein the lateral flow paper is arranged to transport the sample of blood from the receiving section through a separation section, wherein in the separation section blood cells are separated from the blood serum.
Lateral flow paper may be fragile and it’s effectiveness in separating blood serum from a sample of blood may be at least partially comprised when handled incorrectly. The device for collecting a blood sample is arranged to protect the lateral flow paper from incorrect handling, and to increase easy with which the sample of blood may be collected, by substantially enclosing the lateral flow paper. Incorrect handling may be applying to much pressure to the lateral flow paper, touching the paper with dirty or greasy fingers, accidentally tearing the paper, or any other form of handling which may comprise the working of the lateral flow paper.
The receiving section of the lateral flow paper may be provided with a larger thickness than the separating section to allow a larger sample of blood to be collected. The larger thickness may be achieved by folding part of the strip of lateral flow paper double at the receiving section.

DETAILED DESCRIPTION OF THE EMBODIMENTS
Fig. 1A shows an embodiment of a device 100 for collecting a sample of blood, comprising an elongated frame arranged to hold a strip of lateral flow paper. The elongated frame comprises a first frame part 110 and a second frame part 120. The first frame part 110 comprises a receiving opening 111, arranged to expose a receiving section of the strip of lateral flow paper. The receiving section is arranged to receive the sample of blood.
Blood serum provided in the sample of blood is transported through the lateral flow paper, during which the blood serum is separated from the sample of blood in a separation section of the strip of lateral flow paper as red blood cells are left behind during transportation. The separation section is provided adjacent to the receiving section of the strip of lateral flow paper, and both the receiving section and the separation section may be comprised by a one and the same strip of lateral flow paper.
The second frame part 120 is arranged to be connected to the first frame part 110 to support the strip of lateral flow paper in the device 100. The first frame part 110 and the second frame 120 may be releasably connected, such that the strip of lateral flow paper may be removed from the device 100, for example after the blood serum is separated from the sample of blood.
In the embodiment as shown in Fig. 1A, the first frame part 110 comprises laterally elongated protrusions at the receiving opening 111 as a first set of support protrusions 131. The second frame part 120 comprises laterally elongated protrusions as a second set of support protrusion 132. The first set of support protrusions 131 and the second set of support protrusions 132 are arranged to support at least a substantial part of the receiving section of the strip of lateral flow paper.
In the embodiment of the device 100 as shown in Fig. 1A, the first frame part 110 further comprises pins protruding from the first frame part 110 as a third set of support protrusions 133. The second frame part 120 further comprises pins protruding from the second frame part 120 as a fourth set of support protrusions 134. The pins comprised by the third set of support protrusions 133 and the fourth set of support protrusions 134 are arranged to support the strip of lateral flow paper with a minimal or at least very small contact area to optimize flow of the sample of blood through the lateral flow paper. The pins may therefor be arranged with a rounded of end, or as at least partially tapered pins, tapered away from the frame part from which the pins protrude, as to minimize the contact area between protrusions and the strip of lateral flow paper. A minimal contact area is to be understood as a contact area that is as small as possible to manufacture with, for example, an injection moulding system or, alternatively or additionally, is of such size that it has no or a negligible effect on a flow of blood serum through the strip of lateral flow paper.
In an embodiment of the device 100, at least part of the support protrusions comprised by the first frame part 110 and at least part of the support protrusions comprised by the second frame part 120 are aligned. The alignment is such that the strip of lateral flow paper is at both sides of the paper supported at the same area, which minimises a negative impact contact between the lateral flow paper and the support protrusions may have on the flow of blood serum through the strip of lateral flow paper.
The first set of support protrusions 131, second set of support protrusions 132, third set of support protrusions 133, and the fourth set of support protrusions 134 may comprises any number of protrusions, including only one protrusion, and the protrusions may have any shape. The protrusions may all have the same shape or different shapes. In a closed position of the device 100, the pins of the third set of support protrusions 133 are preferably provided substantially opposite to the pins of the fourth set of support protrusions 134.
Embodiments of the device 100 are envisioned comprising none, one or more of the first set of support protrusions 131, second set of support protrusions 132, third set of support protrusions 133, and the fourth set of support protrusions 134, in any combination thereof.
The first frame 110 in the embodiment of the device 100 as shown in Fig. 1A comprises the receiving opening 111, arranged to expose the receiving section of the strip of lateral flow paper such that the receiving section may receive the sample of blood. The receiving opening 111 is provided parallel to the lateral flow paper.
In a further embodiment of the device 100, the second frame part 120 comprises one or more through holes 121, arranged to expose one or more sections of the strip of lateral flow paper. The exposure is arranged to provide a fluid connection between the strip of lateral flow paper and ambient air, which allows the sample of blood and the separated blood serum to dry more quickly.
In the embodiment of the device 100 as shown in Fig. 1A, the first frame part 110 and the second frame part 120 are arranged to be hingedly connected. In the embodiment, a hinge line 101 is provided at the same side of the device 100 as the receiving opening 111. In an alternative embodiment of the device 100, the hinge line 101 is provided at a side opposite to the side at which the receiving opening 111 is provided. In an embodiment of the device 100 where the first frame part 110 and the second frame part 120 are hingedly connected, a first of the first frame part 110 and the second frame part 120 comprises one or more hinge protrusions. The hinge protrusions are arranged to engage with one or more hinge indentations provided by a second of the first frame part 110 and the second frame part 120. The hinge line 101 as a rotation axis over which the hinging movement may be performed is then parallel to a direction of protrusion of the one or more hinge protrusions.
To releasably connect the first frame part 110 and the second frame part 120, a first of the first frame part 110 and the second frame part 120 may be provided with one or more connection protrusions 141, arranged to mate with one or more connection indentations 142 provided by a second of the first frame part 110 and the second frame part 120. The fit between the connection protrusions 141 and the connection indentations 142 may be chosen such that the connection may be released. A separate tool may be provided with the device 100 arranged to help release the connection between the first frame part 110 and the second frame part 120.
An embodiment of the device 100 comprises the strip of lateral flow paper, a thickness of the receiving section of the strip of lateral flow paper has a larger thickness than the separation section of the strip of lateral flow paper. A larger thickness of the receiving section of the strip of lateral flow paper allows a larger sample of blood to be collected by the device 100. Only a certain amount of blood can be absorbed by the strip of lateral flow paper with a certain thickness before the blood leaks through the paper. Increasing the thickness increases the amount of blood that may be provided before the bloods leaks through the paper, and allows more blood serum to be extracted since more blood comprises more blood serum.
To accommodate the different thicknesses of the strip of lateral flow paper, the first set of support protrusions 131 and the second set of support protrusions 132 may be arranged with a smaller protruding length than the third set of support protrusions 133 and the fourth set of support protrusions 134.
Fig. IB shows the device 100 from Fig. 1A in a closed position, in which the device 100 is arranged to constrain the strip of lateral flow paper between the first frame part 110 and the second frame part 120.
In the device 100 as shown in Fig. IB, the first frame part 110 further comprises a scraping protrusion 114, protruding outwards away from the first frame part 110. The scraping protrusion 114 is provided adjacent to the receiving opening 111, and is arranged to collect at least part of the blood sample. The scraping protrusion 114 is arranged to scrape blood towards the receiving opening 111, such that the scraped blood may be provided to the receiving section of the strip of lateral flow paper. When the blood is to be collected from a finger, the scraping protrusion 114 may be provided in a shape substantially corresponding to at least part of a shape of a tip of the finger.
In an embodiment of the device 100, the first frame part 110 further comprises an optional viewing opening 112, arranged to expose a pre-determined part of the strip of lateral flow paper. The exposure allows the pre-determined part of the strip of lateral flow paper to be viewable by a user of the device 100. As a certain pre-determined volume of blood is received by the receiving section of the lateral flow paper, the predetermined part of the strip of lateral flow paper will be coloured by virtue of red blood cells present in the received blood. The colouring of the predetermined part of the strip of lateral flow paper may indicate to the user that a sufficient amount of blood has been received by the device 100.
Fig. 2A shows a piano 200 for an embodiment of the device 100 for collecting a blood sample. Dashed lines indicate folding lines over which the piano 200 may be folded to form the device 100 as shown in Fig. 2B. Dashdotted lines indicate weakened lines over which the device 100 may be torn. The piano 200 comprises a first section 211 and a second section 212, which are arranged to be folded into the first frame part 110 as shown in Fig. 2B. The piano 200 further comprises a third section 213, a fourth section 214 and a fifth section 215, which are arranged to be folded into the second frame part 120 as shown in Fig. 2B.
The piano 200 is preferably one and the same piece of material, in which the material may be paper, cardboard, any other material, or any combination thereof. The folding lines, shown as dashed line in Fig. 2A, may be provided as weakened sections to increase ease of folding. After folding the piano 200 into the shape as shown in Fig. 2B, the first section 211 and the second section 212 may be connected. Similarly, any combination of the third section 213, the fourth section 214 and the fifth section 215 may be connected in order to maintain the shape of the device 100 as shown in Fig. 2B after folding. The connection may be applied using for example glue, tape, a clamping device such as a paper clip, any other connection method or any combination thereof.
The first section 211 and the section 212 comprise the receiving opening 111 and the viewing opening 112. The third section 213 comprises the fourth set of protrusions 134, which may be provided by embossing areas of the third section 213 adjacent to the protrusions of the fourth set of protrusions 134. The fourth section 214 comprises the second set of protrusions 132. The protrusions may be formed by removing a layer of the material of the piano 200 or by depressing areas adjacent to the intended locations of the protrusions. In Figure 2 B, the rectangles adjacent to the protrusions of the fourth set of protrusions 134 indicate depressed or compressed areas of material.
The device 100 as shown in Fig. 2B comprises the first frame part 110 and the second frame part 120, which are arranged to be folded towards each other over hinge line 101. The device 100 as shown in Fig. 2B is shown in an open position, arranged to be provided with the strip of lateral flow paper. To constrain the strip of lateral flow paper, the first frame part 110 and the second frame part 120 may be hinged into a closed position over the hinge line 101. In the closed position the strip of lateral flow paper is supported between the first frame part 110 and the second frame part 120. To connect the first frame part 110 and the second frame part 120 in the closed position, glue, tape, a clamping device such as a paperclip, any other connection method or a combination thereof may be used.
Fig. 2C shows an embodiment of the device 100 based on the piano 200 of Fig. 2A in the closed position, in which the strip of lateral flow paper may be substantially enclosed by the device 100. At least part of the receiving section of the strip of lateral flow paper is exposed via the receiving opening 111.
The enclosed strip of lateral flow paper is provided on the second set of protrusions 132 and fourth set of protrusions 134 comprised by the second frame part 120, provided underneath the first frame part 110 in Fig. 2C. The receiving opening 111 exposes at least part of the receiving section of the strip of lateral flow paper, and the viewing opening 112 exposes a predetermined part of the strip of lateral flow paper.
Fig. 2D shows the device 100 provided with a strip of lateral flow paper 300, on which on a receiving section 301 a sample of blood is provided. As the blood serum is transported through the strip of lateral flow paper 300, in Fig. 2D from left to right, red blood cells, in Fig. 2D shown as a shaded section, are left behind. Part of the separation section 302 of the strip of lateral flow paper is visible through the viewing opening 112, making also the progress of the separation process visible.
Part of the device 100 may be removed from the rest over the device by breaking off the device at the weakened section 150. This allows the strip of lateral flow paper 300 to be removed from the device 100 for further processing.
In the embodiments of the device 100 as shown in Figs. 2A, 2B, 2C and 2D, the viewing opening 112 is arranged to expose a pre-determined part of the strip of lateral flow paper, such that at least part of the predetermined part of the strip of lateral flow paper may be viewable by the user of the device 100. The viewability of the pre-determined part of the strip of lateral flow paper may provide an indication to the user of the volume of the sample of blood that has been provided to the receiving section of the strip of lateral flow paper.
The first frame part 110 may be provided with one or more markings to further indicate the volume of blood which has been provided to the receiving section. The markings may indicate a specific volume, whether a sufficient amount of blood has been provided, or a combination thereof. The exposure of the pre-determined part of the strip of lateral flow paper further decreases the contact area between the device 100 and the strip of lateral flow paper, which may increase the flow of blood through the strip of lateral flow paper.
In an alternative embodiment of the device 100, the receiving opening 111 is provided substantially perpendicular to the strip of lateral flow paper, such that at least part of the receiving section of the strip of lateral flow paper may protrude from the device 100. In yet another embodiment of the device 100, the device comprises a lid arranged to substantially seal off or at least substantially close the receiving opening
111.
The larger thickness of the receiving section of the strip of lateral flow paper may be provided in the production process of the strip of lateral flow paper. Alternatively, a strip of substantially uniform thickness may be provided in the device 100, of which part of the strip is provided over the hinge line between the first frame part 110 and the second frame part 120 when the first frame part 110 and the second frame part 120 are in the open position. When hinging the first frame part 110 relative to the second frame part 120, the part of the strip which is provided over the hinge line will be folded double, and herewith provided the increased thickness of the strip of lateral flow paper. Alternatively, the strip of lateral flow paper may be folded before being placed in the device 100.
In the embodiment of Figure 2 B, a first set of support protrusions is omitted compared to the embodiment shown by Figure 1 A. When the strip of lateral flow paper is to be folded double, the second set of support protrusions 132 may be arranged to clamp the strip to the first frame part 110 and in particular the part of the first frame part 110 surrounding the receiving opening 111 such that a substantial part of the folded part is pressed onto the strip to provided a fluid connection between the folded part and the strip of lateral flow paper. In another embodiment similar to that of Figure 2 B, the first set of support protrusions may be provided, in any way.
At least a part of at least one of the first frame part 110 and the second frame part 120 may be provided at least partially transparent, such that a pre-determined part of the strip of lateral flow paper provided in the device 100 may be viewable by the user of the device. In such an embodiment, the user may view the progression of the separation of the blood serum from the sample of blood, view the amount of blood provided to the strip of lateral flow paper, or a combination thereof.
An embodiment of the device is also envisioned wherein the device is arranged to hold a different shape of lateral flow paper, such as a square, rectangle, circle, or any other shape.
In an embodiment of the strip of lateral flow paper, the receiving section and the separation section are comprised by two individual strips of lateral flow paper which when provided in the device are arranged in a fluid connection such that at least part of the sample of blood may flow from the receiving section to the separation section.
In summary, blood serum is used for many types of analyses and may provide useful insights into a state of health of a person of whom the blood serum belongs.
