Recombinant nucleotidevolgorde welke codeert voor een gen
De onderhavige uitvinding heeft betrekking op een recombinant nucleotidevolgorde welke codeert voor een gen.
Een algemeen probleem in de biotechnologie is het op een gewenste tijd en, in een meercellig organisme, op de ge-5 wenste plaats tot expressie brengen van het gen. Voor het tot expressie brengen van het gen staat het onder controle van een promotervolgorde, bij voorkeur onder controle van een in-duceerbare promoter.
Een probleem is dat er in de praktijk vaak ook on-10 controleerbare, lage expressie ("leaky" expressie) optreedt van het gen wanneer de promoter niet is geïnduceerd.
De onderhavige uitvinding beoogt dit probleem te verminderen en volgens voorkeursuitvoeringen in wezen uit te sluiten.
15 Daartoe wordt de recombinant nucleotidevolgorde vol gens de uitvinding gekenmerkt doordat deze codeert voor een gen dat een tussen twee exonen gelegen intron bevat, welk intron een eerste, tweede en derde volgorde bevat, de eerste en derde volgorde herkenningsvolgorden zijn voor een recombina-20 tiesysteem dat recombinatie mogelijk maakt, en de tweede volgorde ten minste een eerste verstorende volgorde omvat gekozen uit de groep bestaande uit i) een transcriptie-terminatievolgorde voor transcriptie, en ii) een vreemd exon dat optioneel een tweede verstorende volgorde bevat gekozen 25 uit a) een transcriptie-terminatievolgorde voor transcriptie, b) een stop-codon voor translatie, en c) een leesraam-verschuivende volgorde, dan wel de complementaire volgorde daarvan.
Een dergelijk construct heeft als voordeel dat zo 30 het gen ongewenst wordt afgelezen, dit door één of meer van de hier genoemde maatregelen niet leidt tot een functionele ribonucleotidevolgorde en in het bijzonder een functioneel eiwitproduct van het gen. Met de recombinant nucleotidevolgorde volgens de uitvinding is het mogelijk om een eukaryoot 35 organisme of cel op een gewenst moment permanent te veranderen.

Volgens de uitvinding kan de recombinant nucleotide-volgorde een RNA of DNA volgorde zijn. De wijze waarop de volgorde is verkregen, bijvoorbeeld middels synthese of genetische manipulatie, doet niet ter zake. Ook de complementaire 5 volgorde wordt volgens de uitvinding omvat, aangezien daar een sensevolgorde van kan worden gemaakt met de volgens de uitvinding te bereiken voordelen.
Het gen kan niet alleen een eukaryoot gen zijn, doch ook een prokaryoot gen.
10 In de onderhavige uitvinding wordt onder een vreemd exon verstaan een exon dat niet of, zo dit toch het geval is, niet op die plaats in het gen dat codeert voor een functionele ribonucleotidevolgorde of functioneel eiwit wordt aangetroffen. Het vreemde exon wordt volgens de uitvinding middels 15 algemeen bekende recombinatietechnieken in het gen ingébracht. Niet ter zake kundigen wordt verwezen naar Sambrook et al. Molecular cloning, a laboratory manual, 2e druk, Cold Spring Harbor, New York 1989.
Bij voorkeur staat het gen onder controle van een 20 weefsel-specifieke promoter. Het intron kan zich in een regulerende volgorde, zoals een promotervolgorde, van het gen bevinden, doch bij voorkeur in een door transcriptie afgelezen deel van het gen, en met de meeste voorkeur in het open leesraam.
25 Dit maakt de recombinant nucleotidevolgorde volgens de uitvinding bij uitstek geschikt voor toepassing in eukaryote organismen, zoals zoogdieren en in het bijzonder de mens, waarbij voor diverse toepassingen, zoals medische toepassingen, een uitermate hoog niveau van controle over het (inge-30 brachte) gen vereist is.
Bij voorkeur is het recombinatiesysteem een recombi-natiesysteem dat ten minste twee door verschillende genen gecodeerde eiwitten omvat, waarbij ten minste een van de verschillende genen onder controle staat van een extern stimu-35 leerbare promoter.
Hierdoor wordt bevorderd dat "leaky" expressie van het recombinatiesysteem niet leidt tot excisie van het intron en daarmee, in geval van "leaky" expressie van het gen of on- * Ü l ( o o ^ 3 der invloed van de eerder genoemde weefsel-specifieke promoter, tot expressie van het gen. De (ten minste) twee eiwitten kunnen de subeenheden zijn van één eiwit dat het gehele re-combinatie-systeem is.
5 Bij voorkeur bevat de recombinant nucleotidevolgorde de recombinant nucleotidevolgorde tevens een tweede gen, of complementaire volgorde daarvan, coderend voor een eiwit van het recombinatiesysteem of een subeenheid daarvan.
Aldus wordt een enkele recombinant nucleotidevolgor- 10 de verschaft welke zowel het onder gecontroleerde omstandigheden tot expressie te brengen gen bevat alsmede de middelen voor excisie van het intron. Een dergelijk recombinant nucleotidevolgorde kan zeer goed worden gebruikt als vector voor genetische manipulatiedoeleinden. De recombinant nucleotide- 15 volgorde bevat bij voorkeur de volledige genetische code voor het recombinatiesysteem.
Bij voorkeur zijn de eerste en derde volgorden door een recombinase erkende volgorden.
Recombinase is zeer geschikt als recombinatiesys- 20 teem.
Tenslotte heeft de uitvinding betrekking op een werkwijze voor het genetisch modificeren van een eukaryote cel, hierdoor gekenmerkt, dat een recombinant nucleotidevolgorde wordt toegepast volgens de uitvinding.
25 Derhalve codeert het tweede gen voor de recombinase, of een subeenheid daarvan.
De uitvinding zal thans worden toegelicht aan de hand van het volgende voorbeeld.
VOORBEELD
30 Constructie van een vector voor spatieel-temporeel gecontroleerde expressie van een gen.
Een mogelijke vector volgens de uitvinding, aangeduid als Loxmid, is opgebouwd uit bestaande genetische vectoren en in de literatuur beschreven genetische elementen. In 35 een vector, pUC18 of pUC19, wordt een aantal additionele, zeldzame herkenningsplaatsen voor restrictie-endonucleasen geïntroduceerd. Deze plaatsen zijn van belang voor latere kloneringstappen Met deze gemodificeerde vector(en) worden ^ W £__ 4 twee "ouderlijke" vectoren gemaakt, de een pDAD genaamd met de coderende sequentie voor een recombinase of een subeenheid daarvan (bijvoorbeeld Cre recombinase) onder de controle van een induceerbare promoter; bijvoorbeeld een met tetracycline 5 induceerbare, of met een respons element voor vitamine A, vitamine D of een ander steroid-hormoon. Het Cre gen en haar induceerbare promoter wordt geflankeerd door restrictieplaat-sen die de klonering in de uiteindelijke Loxmid vergemakkelijken. De andere "ouderlijke" vector (pMOM genaamd) wordt 10 zodanig geconstrueerd dat deze een gen bevat bestaande uit de coderende sequentie voor het gen van interesse, met daarin een al dan niet natuurlijk intron, onder de controle van een weefselspecifieke promoter; het intron zal, tussen twee her-kenningsplaatsen voor het recombinase dat in de andere vector 15 is gekloneerd, bijvoorbeeld een transcriptie-terminatie-volgorde bevatten voor transcriptie, of een stopcodon voor translatie, een leesraamverschuivende nucleotiden-volgorde, of een combinatie daarvan. Een derde vector wordt geconstrueerd (de Loxmid, ofwel lxCHILD) met daarin specifieke klone-20 ringplaatsen waarin enerzijds de coderende sequentie voor een recombinase onder de controle van een induceerbare promoter wordt geplaatst, en anderzijds het gewenste gen onder de controle van de weefselspecifieke promoter.
Alle kloneringstappen worden uitgevoerd met behulp 25 van klassieke moleculair-biologische technieken, ondersteund door computer-assisted vector design, zoals bij de gewone ter zake kundige welbekend.
De resulterende vector (Loxmid genaamd) kan worden gebruikt om in een gespecificeerd weefsel of celtype genex-30 pressie te induceren op de door de onderzoeker gewenste tijd. Zo kan in het Loxmid Cre worden gekloneerd onder de controle van een promoter die transcriptie aandrijft onder invloed van vitamine A, en kan Fas ligand worden gekloneerd onder de invloed van de keratinocyt-specifieke K14 promoter. Fas ligand 35 is door de aanwezigheid van transcriptie- en/of translatie-terminerende sequenties in het intron niet actief; deze sequenties liggen tussen herkenningsplaatsen voor het Cre recombinase .

De promoter voor Cre bindt aan de nucleaire retinoid acid binding receptor, dat op zijn beurt weer wordt geactiveerd door vitamine A. Wanneer nu het Loxmid wordt ingebracht in gekweekte, prolifererende epidermale huidcellen (zogenaam-5 de keratinocyten) zal het Cre recombinase alleen maar tot expressie komen wanneer aan het medium vitamine A wordt toegevoegd. Tengevolge van Cre expressie zullen de terminator sequenties worden verwijderd uit het intron van Fas ligand, hetgeen weer tot gevolg heeft dat transcriptie en vervolgens 10 translatie van het intacte Fas ligand plaatsvindt onder invloed van de K14 promoter die endogeen actief is in keratinocyten. Wanneer actief Fas ligand wordt aangemaakt tengevolge van vitamine A toediening in de kweek, zal er massaal geprogrammeerde celdood (apoptose) optreden in keratinocyten. Fas 15 ligand induceert namelijk via haar receptor Fas, die in keratinocyten tot expressie komt, een gecoördineerde cascade van biochemische reactie die kunnen leiden tot celdood, en zal in een kweekmodel, zoals hier beschreven, aanleiding geven tot geprogrammeerde celdood ("apoptose") van keratinocyten.
20 Wanneer ditzelfde Loxmid wordt ingebracht in de kiemlijn van muizen (om transgene muizen te creëren) kan in theorie de geprogrammeerde celdood in gang gezet worden in de basale of kiemlaag van de muizenepidermis (aannemende dat de gekozen genetische elementen actief zijn in de muis) wanneer 25 vitamine A wordt toegediend. Indien vitamine A systematisch wordt toegediend, zal apoptose nog steeds slechts plaatsvinden in de kiemlaag, aangezien dat de enige laag is waar de K14 promoter actief is. Hoewel Fas ligand dan slechts in een aantal cellen tot expressie komt, zal het toch leiden tot een 30 mogelijk levensbedreigende situatie omdat de muis de opperhuid zal verliezen, hetgeen leidt tot uitdroging en infecties. In mensen komt soms een vergelijkbare aandoening voor die bekend staat als toxische epidermale necrolyse. Deze ziekte is geassocieerd met geneesmiddelenallergie. Derhalve 35 kan de transgene muis voor deze menselijke ziekte model staan. Wanneer echter vitamine A plaatselijk, dat wil zeggen op de huid, wordt toegediend, zal slechts op de plaats van toediening apoptose plaatsvinden. Daarmee is de transgene - 1017852 6 muis een model bij onderzoek naar bijvoorbeeld therapie voor brandwonden. Uiteraard kunnen ook andere genen dan Fas ligand gebruikt worden in het Loxmid, en het Loxmid, afhankelijk van het modelorganisme, aangepast worden. Zo kan het worden ge-5 bruikt in basaal wetenschappelijk onderzoek naar genfunctie en genregulatie in celkweek, of als therapeuticum in gentherapie .

Claims (8)
1. Recombinant nucleotidevolgorde welke codeert voor een gen dat een tussen twee exonen gelegen intron bevat, welk intron een eerste, tweede en derde volgorde bevat, de eerste en derde volgorde herkenningsvolgorden zijn voor een recombi- 5 natiesysteem dat recombinatie mogelijk maakt, en de tweede volgorde ten minste een eerste verstorende volgorde omvat gekozen uit de groep bestaande uit i) een transcriptie-terminatievolgorde voor transcriptie, en ii) een vreemd exon dat optioneel een tweede verstorende volgorde bevat gekozen 10 uit a) een transcriptie-terminatievolgorde voor transcriptie, b) een stop-codon voor translatie, en c) een leesraam-verschuivende volgorde, dan wel de complementaire volgorde daarvan.
2. Recombinant nucleotidevolgorde volgens conclusie 15 1, met het kenmerk, dat het gen onder controle staat van een weefsel-specifieke promoter.
3. Recombinant nucleotidevolgorde volgens conclusie 1 of 2, met het kenmerk, dat het recombinatiesysteem een re-combinatiesysteem is dat ten minste twee door verschillende 20 genen gecodeerde eiwitten omvat, waarbij ten minste een van de verschillende genen onder controle staat van een extern stimuleerbare promoter.
4. Recombinant nucleotidevolgorde volgens een der voorgaande conclusies, met het kenmerk, dat de recombinant 25 nucleotidevolgorde tevens een tweede gen bevat, of complementaire volgorde daarvan, coderend voor een eiwit van het recombinatiesysteem of een subeenheid daarvan.
5. Recombinant nucleotidevolgorde volgens een der voorgaande conclusies, met het kenmerk, dat de recombinant 30 nucleotidevolgorde voor het volledige recombinatiesysteem codeert .
6. Recombinant nucleotidevolgorde volgens een der voorgaande conclusies, met het kenmerk, dat de eerste en derde volgorden door een recombinase herkende volgorden zijn.
7. Recombinant nucleotidevolgorde volgens conclusie ' ' - - ' £ o 6, met het kenmerk, dat het tweede gen codeert voor de recom-binase, of een subeenheid daarvan.
8. Werkwijze voor het genetisch modificeren van een eukaryote cel, met het kenmerk, dat een recombinant nucleoti-5 devolgorde wordt toegepast volgens een van de conclusies 1-7. ~j O '/
