Abstract

Se proveen moléculas del tipo avidina modificadas de union a biotin en donde el residuo de tirosina esencial en el sitio de union a biotin se modifica de tal manera que su pKa disminuye en comparacion con el pKa del residuo de tirosina no modificado en la molécula del tipo avidina no modificada correspondiente; las moléculas del tipo avidina incluyen: (i) avidina de clara de huevo nativa; (ii) avidina recombinante; (iii) formas desglicosiladas de avidina; (iv) estreptoavidina bacteriana; (v) estreptoavidina recombinante; (vi) estreptoavidina truncada; y (vii) derivados de (i)-(vi) que se modifican en sitios diferentes al residuo de tirosina esencial; la modificacion se logra mediante la substitucion en una o ambas posiciones otro al radical hidroxi del residuo de tirosina por radicales tales como nitro, halogeno, azo y amino; las moléculas del tipo avidina modificadas pueden usarse en todas las aplicaciones de la tecnología de avidina-biotin.
