Abstract

En la presente invención se diseñaron y sintetizaron proteínas de fusión con actividad citocromo P450 encapsuladas en una nanocápside (nanopartículas cargadas con actividad citocromo P450) y que son inmunológicamente inertes y que son reconocidas por células de cáncer de mama. La metodología consistió en el encapsulamiento de moléculas de citocromo P450 dentro de cápsides virales o nanopartículas de biopolímeros. En un caso, el gen que codifica la enzima citocromo P450 se fusiona con la secuencia nucleotídica de una versión truncada de la proteína de andamiaje del bacteriófago P22. Se expresan las proteínas heterologamente y las cápsides conteniendo en su interior el citocromo P450 son purificadas. Las nanopartículas enzimáticas se cubren con polietilenglicol dendrítico bifuncional y finalmente se funcionalizarán con el péptido cíclico Arg-Gly-Asp-D-Phe-Cys u otro ligando para ser reconocidas por la integrina avjß3/5 u otro receptor de las células tumorales de mama. Esta invención lleva específicamente a células tumorales la actividad citocromo P450 que activa más eficientemente y en el sitio requerido los profármacos usados en quimioterapia.
