Abstract

La invención proporciona el descubrimiento y caracterización de una proteína de arterivirus novedosa (nsp2TF), cuya expresión es dependiente del desplazamiento de estructura ribosomal -2 en un sitio ubicado en la región de codificación nsp2. La región de codificación para el dominio TF único de nsp2TF se sobrepone a la parte de ORF1 que codifica la región de transmembrana de nsp2 en artervirus, incluyendo PRRSV, LDV y SRFV. Las mutaciones que afectan la expresión de nsp2TF deterioran la replicación de PRRSV y dan por resultado un fenotipo de placa más pequeño. En la presente se proporcionan arterivirus que exhiben traducción reducida de nsp2TF y/o traducción alterada de uno o más productos corriente abajo, arterivirus en los cuales la función de nsp2TF se reduce y/o está ausente, y vacunas que comprenden los arterivirus. En la presente también se proporcionan métodos de diagnóstico, métodos para identificar compuestos que inhiben el desplazamiento de estructura -2, y herramientas de expresión génica para sistema eucarióticos utilizando el desplazamiento de estructura -2.
