Abstract

La presente invención se relaciona con un dispositivo para la unión eficiente de ácidos nucleicos en una micromatriz, que comprende una zona de reacción que comprende una micromatriz, y una zona de captura que comprende un sustrato de membrana porosa, en donde la zona de captura es capaz de capturar específicamente hebras sentido de moléculas diana, mientras que hebras antisentido complementarias son capturadas en la zona de reacción, o viceversa. La invención contempla además unidades reguladoras de temperatura en el dispositivo que permiten llevar o mantener la captura a una temperatura que no permite la hibridación o unión de ácidos nucleicos a las moléculas de captura o a una temperatura adecuada para la hibridación de ácidos nucleicos. La invención también se relaciona con un método de unir eficientemente ácidos nucleicos sobre una micromatriz, que comprende introducir un medio que contiene una o más moléculas diana en forma desnaturalizada en una zona de captura de un dispositivo de la presente invención, llevar a cabo una reacción de interacción entre las moléculas diana y moléculas de captura inmovilizadas en la zona de captura, y transportar hebras de moléculas diana no unidas a una zona de reacción que comprende una micromatriz, permitiendo así una interacción entre las hebras de moléculas diana y moléculas de captura inmovilizadas sobre la micromatriz. En un aspecto adicional la invención se relaciona con el uso del dispositivo para enriquecer moléculas diana, seleccionando específicamente moléculas diana, para análisis de expresión, hibridación genómica comparativa, la detección de SNPs, o para la secuenciación basada en selección genómica basada en micromatriz.
