Abstract

La présente invention concerne un procédé d'extraction acide à faible température et depurification de collagène marin natif type 1 soluble à partir des écailles de sardine (sardinepilchardus) fraiches ou congelées, caractérisée en ce qu'il comprend par les étapes de 1)lavage à l'eau des écailles, 2) séchage des écailles 3) traitement alcalin pour éliminer lesprotéines non collagénique, 4) traitement acide pour éliminer les minéraux, 5)extraction àfroid à ph acide pour solubiliser les fibrilles de collagène natif, 6)ultracentrifugation pouréliminer les gros agrégats, 7) précipitation du collagène par adjonction d'un sel neutre 8)remise en suspension du collagène précipité et 9) dialyse au moyen d'un sac de dialyse. Ceprocédé permet dans un délai raisonnable de produire un collagène avec une puretémaximale en supprimant la quasi-totalité des impuretés non collagénique et des minéraux,en solubilisant la totalité du collagène et en limitant sa dénaturation. En plus de l'obtentionde collagène marin natif type 1avec un rendement considérable, ce procédé optimisé par laméthode de réponse de surfacersm permet de réduire la durée du traitement alcalin etacide, de diminuer considérablement la durée de l'extraction acide. Il permet égalementd'amoindrir la quantité de réactifs utilisés en l'occurrence, la soude, l'acidedéminéralisateur et l'acide extracteur. Le procédé résout le problème particulier de lagestion des produits chimiques, de manière plus durable et plus économique. Le collagènenatif de haute qualité ainsi obtenu est propice à des applications en alimentaire, encosmetfood et en biomédical.The present invention relates to a process for low temperature acid extraction and purification of soluble native marine collagen type 1 from sardine scales (sardinepilchardus) fresh or frozen, characterized in that it comprises by the steps of 1) washing at scaling water, 2) drying of scales 3) alkaline treatment to remove non-collagenic proteins, 4) acid treatment to remove minerals, 5) acid ph-cold extraction to solubilize native collagen fibrils, 6) ultracentrifugation to eliminate aggregates, 7) collagen precipitation by addition of a neutral salt 8) resuspension of precipitated collagen and 9) dialysis by means of a dialysis bag. This method allows within a reasonable time to produce a collagen with a pureteamximal eliminating almost all non-collagenic impurities and minerals, by solubilizing all the collagen and limiting its denaturation. In addition to obtaining a type 1 native marine collagen with a considerable yield, this method optimized by the surfacersm response method makes it possible to reduce the duration of the alkaline and acid treatment, considerably reducing the duration of the acid extraction. It also reduces the amount of reagents used in this case, sodium hydroxide, acemineminaliser and extractor acid. The process solves the particular problem of managing chemicals in a more sustainable and economical way. The resulting high quality collagen is suitable for food, encosmetfood and biomedical applications.
