Abstract
CIITA에 의해 전사 활성화를 억제하여 그 결과 MHC 클래스 Ⅱ 유전자 발현을 억제하는 화합물을 확인하는 방법이 기술되어 있다. 그러한 화합물은 면역 응답의 유도에 영향을 미칠 수 있다. 방법은 CIITA 활성화 및 상호작용 도메인을 독립적으로 사용한다. 방법은 아이소타이프-특이적 CIITA 단백질의 활성화 및 상호작용 도메인을 사용하고, 전사의 아이소타이프-특이적 억제제이고 자가면역계에 선택적으로 영향을 미치는데 유용한 화합물의 확인을 허여한다.

Claims (27)
- 화합물을 사용하여, CIITA에 전사 활성화 도메인을 함유하고 기능적 CIITA 상호작용 도메인을 함유하지 않은 폴리펩티드 전사-활성화 능력을 억제하는지의 여부를 결정하는 방법으로서, 전사의 억제가 상기 화합물이 역가있는 자가면역 질병의 치료제임을 지시하는 지표가 되는 것을 특징으로 하는 방법.
- 제1항에 있어서, a) DNA-결합 단백질에 융합된 상기 폴리펩티드를 함유하는 융합 단백질을 제공하는 단계; b) 상기 융합 단백질이 상기 조절 DNA 서열에 결합되였을 때, 상기 리포터 유전자의 전사에 적합한 시스템내에 리포터 유전자에 작동 가능하게 연결된 전사 조절 DNA 서열을 제공하는 단계; c) 상기 융합 단백질 및 상기 화합물을 상기 시스템내에 제공하는 단계; 및 d) 상기 화합물이 상기 리포터 유전자의 전사를 억제할 수 있는 능력을 측정하는 단계에 의해 전사 활성화의 억제를 측정하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 아이소타이프-특이적 CIITA 전사 활성화 도메인을 포함하고 기능적 CIITA 상호작용 도메인을 포함하지 않는 제2폴리펩티드의 전사-활성화 능력을 억제하는 상기 화합물의 능력을 측정하는 단계를 추가로 포함함으로써, 아이소타프-특이적 화합물을 확인하는 것을 특징으로 하는 방법.
- 기능적 CIITA 상호작용 도메인을 코드화하지 않으면서 CIITA 전사 활성화 도메인을 코드화하는 실질적으로 순수한 DNA.
- 제4항에 있어서, 상기 DNA가 제1도(서열번호:1)의 192-1149 누클레오티드 서열을 함유하거나, 이들의 변형체를 변이시키는 것을 특징으로 하는 DNA.
- 제4항에 있어서, 상기 DNA가 제1도의 DNA 서열의 192-1149 누클레오티드에 80% 이상 동일한 서열을 함유하는 것을 특징으로 하는 DNA.
- 제4항에 있어서, 상기 CIITA가 아이소타이프-특이적인 것을 특징으로 하는 DNA.
- CIITA 전사 활성화 도메인을 함유하고 기능적 CIITA 상호작용 도메인을 함유하지 않는 실질적으로 순수한 폴리펩티드.
- 제8항에 있어서, 제1도의 아미노산 서열의 아미노산 26-352와 실질적으로 동일한 아미노산 서열을 함유하는 것을 특징으로 하는 폴리펩티드.
- 제8항에 있어서, 상기 CIITA가 아이소타이프-특이적인 것을 특징으로 하는 폴리펩티드.
- 화합물을 사용하여, CIITA 상호 작용 도메인을 포함하고 기능적 CIITA 활성화 도메인을 함유하지 않는 폴리펩티드의 표적화 단백질의 결합능을 억제하는지의 여부를 결정하는 방법으로서, 결합의 억제가 상기 화합물이 자가면역 질병의 역가있는 치료제인 것을 지시하는 지표가 되는 것을 특징으로 하는 방법.
- 제11항에 있어서, 화합물을 사용하여 상기 폴리펩티드의 전사 중재 능력을 억제하는지의 여부를 결정하는 것을 포함하는 방법으로서, 전사의 억제가 상기 화합물의 자가면역 질병의 역가있는 치료제인 것을 지시하는 지표가 되는 것을 특징으로 하는 방법.
- 제12항에 있어서, a) DNA-결합 단백질에 융합된 상기 폴리펩티드를 포함하는 제1융합 단백질을 제공하는 단계; b) 리포터 유전자에 작동 가능하게 연결되도록 DNA 서열이 전사 조절 서열을 상기 리포터 유전자를 전사시키기에 적합한 시스템내에 제공하는 단계; c) CIITA 상호작용 도메인의 표적화 단백질에 융합된 전사 활성화 도메인을 포함하는 것을 특징으로 하는 제2융합 단백질을 제공하는 단계; d) 상기 제1 및 제2융합 단백질 및 상기 화합물을 시스템내로 제공하는 단계; 및 e) 상기 화합물이 상기 리포터 유전자의 전사를 억제하는 능력을 측정하는 단계 억제를 측정하는 것을 특징으로 하는 방법.
- 제11항에 있어서, 화합물을 사용하여 아이소타이프-특이적 CIITA 상호작용 도메인을 포함하고 기능적 CIITA 활성화 도메인을 포함하지 않는 제2폴리펩티드의 표적화 단백질 결합능 억제를 측정하는 것을 추가로 포함하고, 아이소타이프-특이적 화합물을 확인하는 것을 특징으로 하는 방법.
- 제14항에 있어서, 상기 제2폴리펩티드가 클론 13 CIITA 상호작용 도메인을 함유하는 것을 특징으로 하는 방법.
- 기능적 CIITA 전사 활성화 도메인을 코드화하지 않으면서 CIITA 상호작용 도메인을 코드화하는 실질적으로 순수한 DNA.
- 제16항에 있어서, 상기 DNA가 제1도의 누클레오티드 1016-3509의 누클레오티드 서열을 가지거나 이들의 변종을 변이시키는 것을 특징으로 하는 DNA.
- 제16항에 있어서, 상기 DNA가 제1도의 DNA 서열의 누클레오티드 1016 내지 3509에 80% 이상 동일한 서열을 함유하는 것을 특징으로 하는 DNA.
- 제16항에 있어서, 상기 CIITA가 아이소타이프-특이적인 것을 특징으로 하는 DNA.
- 제19항에 있어서, 상기 CIITA가 클론 13 CIITA인 것을 특징으로 하는 DNA.
- CIITA 상호 작용 도메인을 함유하고 기능적 CIITA 전사 활성화 도메인을 함유하지 않는 실질적으로 순수한 폴리펩티드.
- 제21항에 있어서, 제1도의 아미노산 서열의 아미노산 301 내지 1130와 실질적으로 동일한 아미노산 서열을 함유하는 것을 특징으로 하는 폴리펩티드.
- 제21항에 있어서, 상기 CIITA가 아이소타이프 특이적인 것을 특징으로 하는 폴리펩티드.
- 제23항에 있어서, 상기 CIITA가 클론 13 CIITA인 것을 특징으로 하는 폴리펩티드.
- 화합물을 사용하여 전사 활성화 도메인에 융합된 CIITA 상호작용 도메인을 함유하고 기능적 CIITA 전사 활성화 도메인을 함유하지 않는 폴리펩티드의 전사-중재능을 억제하는지의 여부를 결정하는 방법으로서, 전사의 억제가 상기 화합물이 자가면역 질병의 역가있는 치료제인 것을 지시하는 지표가 되는 것을 특징으로 하는 방법.
- 제25항에 있어서, B림프구내에 상기 폴리펩티드를 제공하고 MHC 클래스 II 유전자의 발현을 검정하는 것을 포함하는 것을 특징으로 하는 방법.
