Abstract

본 발명은 형질전환된 세포상에 악성 포유동물 세포의 형질전환된 표현형을 부여하는 유전 억제 요소, 상기 유전 억제 요소를 확인 및 수득하는 방법, 상기 유전 억제 요소에 상응하는 유전자를 단리 및 확인시키는 방법, 및 상기 유전 억제 요소를 사용하는 방법에 관한 것이다. 또한, 본 발명은 본 발명의 GSE에 상응하는 유전자에 관한 것이다.

Claims (20)

  1. 포유동물 세포에서 형질전환된 표현형의 확정 및 유지와 관련된 유전 억제 요소를 단리시키는 방법으로서, (a) 세포의 전체 mRNA로부터 제조된 불규칙하게 단편화된 cDNA를 합성하여 DNA 단편을 생성시키는 단계; (b) DNA 단편을 발현 벡터에 이동시켜, 발현 벡터가 형질전환된 표현형을 발현시킬 수 있는 생존 세포에서 DNA 단편을 발현시킬 수 있는 유전 억제 요소 라이브러리를 생성시키는 단계; (c) 세포에 상기 유전 억제 요소 라이브러리를 도입시켜 생존 세포를 유전적으로 변형시키는 단계; (d) 형질전환된 세포가 확인될 수 있는 조건하에 세포를 선별하여, 세포상에 형질전환된 표현형을 부여하는 유전 억제 요소를 함유하는 유전적으로 변형된 생존 세포를 단리시키거나 인리치(enrich)시키는 단계; (e) 살아남은 유전적으로 변형된 세포로부터 형질전환된 표현형을 부여하는 유전 억제 요소를 수득하는 단계를 포함하는, 포유동물 세포에서 형질전환된 표현형의 확정 및 유지와 관련된 유전 억제 요소를 단리시키는 방법.
  2. 제1항의 방법에 의해 확인된 유전 억제 요소.
  3. 제2항에 있어서, 펩티드를 코우드화하는 센스-배향된 유전 억제 요소인 것을 특징으로 하는 유전 억제 요소.
  4. 제2항에 있어서, 앤티센스 RNA를 코우드화하는 앤티센스-배양된 유전 억제 요소인 것을 특징으로 하는 유전 억제 요소.
  5. 제3항의 방법에 따라 제조된 GSE에 의해 코우드화된 아미노산 서열의 6개의 아미노산 내지 전체 아미노산 서열에 상응하는 아미노산 서열을 갖는 합성 펩티드.
  6. 제4항에 의해 제조된 GSE에 의해 코우드화된 앤티센스 RNA의 뉴클레오티드 서열의 12개의 뉴클레오티드 내지 전체 뉴클레오티드 서열에 상응하는 뉴클레오티드 서열을 갖는 합성 올리고뉴클레오티드.
  7. 제2항의 GSE 또는 그것의 보체의 뉴클레오티드 서열과 상동인 클로닝된 유전자.
  8. 제7항에 있어서, 상기 GSE가 GSE Tr6, Tr19 및 Tr22로 이루어진 그룹으로부터 선택되는 것을 특징으로 하는 클로닝된 유전자.
  9. 제2항에 따라 GSE를 발현시키는 포유동물 세포.
  10. 제3항에 따라 GSE를 발현시키는 포유동물 세포.
  11. 제4항에 따라 GSE를 발현시키는 포유동물 세포.
  12. 제7항의 유전자에 의해 코우드화된 단백질 또는 제7항의 유전자의 일부에 의해 코오드화된 펩티드에 대해 배양된 항체.
  13. 제8항의 유전자에 의해 코우드화된 단백질 또는 제8항의 유전자의 일부에 의해 코오드화된 펩티드에 대해 배양된 항체.
  14. 진단적 검정 방법으로서, (a) 동물의 암세포로부터 전령 RNA를 포함하는 세포성 RNA를 단리시키는 단계; (b) 동물의 암세포로부터 제7항의 유전자에 상응하는 mRNA의 발현 수준을 측정하는 단계; 및 (c) 상기 단계(b)에서 특정된 상기 mRNA의 발현 수준이, 유전자가 동물의 암세포내에서 수준이상-발현 또는 수준이하-발현됨을 나타내는지의 여부를 결정하는 단계를 포함하는 진단적 검정 방법.
  15. 진단적 검정 방법으로서, (a) 동물의 암세포로부터 세포성 단백질을 단리시키는 단계; (b) 동물의 암세포내에서 제7항의 유전자에 상응하는 단백질의 양을 측정하는 단계; 및 (c) 상기 단계(b)에서 측정된 상기 단백질의 양이, 유전자가 동물의 암세포내에서 수준이상-발현 또는 수준이하-발현됨을 나타내는지의 여부를 결정하는 단계를 포함하는 진단적 검정 방법.
  16. 진단적 검정 방법으로서, (a) 제7항의 유전자에 상응하는 단백질의 특성은 효소적 활성이므로, 동물의 암세포내에서 효소적 활성량을 측정하는 단계; 및 (c) 상기 단계(b)에서 측정된 효소적 활성량이, 유전자가 동물의 암세포내에서 수준이상-발현 또는 수준이하-발현됨을 나타내는지의 여부를 결정하는 단계를 포함하는 진단적 검정 방법.
  17. 제14항, 15항 또는 16항에 따라 측정된 유전자의 발현 수준을 정량하는 단계를 포함하여, 동물의 악성종양을 특정화시키는 진단적 검정 방법.
  18. 제14항, 15항 또는 16항에 있어서, 상기 유전자가 SAHH, 1bb1, Tr6, Tr19 및 Tr22로 이루어진 그룹으로부터 선택된 GSE의 뉴클레오티드 서열 또는 그것의 보체와 상동인 것을 특징으로 하는 방법.
  19. 암세포내에서 제7항의 완전한 유전자를 발현시키는 단계를 포함하는, 동물내 악성 암세포의 성장을 저해하는 방법.
  20. 제19항에 있어서, 상기 GSE가 SAHH, P120 및 GSE Tr6, Tr19 및 Tr22와 상동인 유전자로 이루어진 그룹으로부터 선택되는 것을 특징으로 하는 방법.
