Abstract

본 발명은 경쟁적 면역화학적 결정방법에 관한 것으로서, 본 발명에서는 분석물의 농도를 1단계 공정법으로 결정하며, 이러한 경우 분석물은 제1 및 제2특이적 결합성분과 함께 제3특이적 결합성분에 결합하기 위하여 경쟁하는데, 제1특이적 결합성분은 수-불용성 지지체에 결합되어 있으며, 제2특이적 결합성분은 신호-발생 표지를 구비하고 있다.

본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 다양한 항체 농도의 측정된 흡광도를 반로그적 표시로 나타낸 것이다.
제2도는 전형적인 항-HIV혈청변환(0193)의 시간경로를 도시한 것이다
제3도는 조사된 혈청변환 0193의 측정된 흡광도를 제1도의 검정곡선의 도움으로 정량화하여 도시한 것이다.

Claims (19)

  1. a) 분석물을, 수-불용성 고형상에 결합되어 있는 제1특이적결합성분, 신호-발생 표지를 구비하고 있는 제2특이적 결합성분 및 제3특이적 결합성분과 함께 배양하므로써, 분석물과 제1 및 제2특이적 결합성분이 제3 특이적 결합성분에 결합하기 위하여 경쟁하도록 하는 단계, b) 고형상에 결합된 신호-발생 표지를 결합되지 않은 분획으로부터 제3특이적 결합성분을 통하여 분리하는 단계, c) 표지의 결합된 분획에 의해 발생될 수 있는 신호를 측정하는 단계, 및 d) 단계 c)에서 밝혀진 값을 동일한 조건하에서 그래프화된 또는 이론적으로 산출된 표준곡선과 비교하므로써 분석물 농도를 결정하는 단계를 포함하는, 이질적 경쟁 결정법에 의해 분석물을 결정하기 위한 면역학적 방법.
  2. 제1항에 있어서, 사용된 면역학적 결정법이 1-단계 공정인 방법.
  3. 제1항 또는 제2항에 있어서, 항체 또는 항체 단편이 제1 및 제2특이적 결합성분으로서 사용되는 방법.
  4. 제3항에 있어서, 폴리클로날 항체 또는 적당한 항체 단편이 제1특이적 결합성분으로서 사용되는 방법.
  5. 제3항에 있어서, 항-HAV-특이적 항체 또는 상응하는 항체 단편이 제1특이적 결합성분으로서 사용되는 방법.
  6. 제3항에 있어서, 동일한 항-HAV-특이적 항체 또는 상응하는 항체 지지체가 제2특이적 결합성분으로서 사용되는 방법.
  7. 제6항에 있어서, 또 다른 항-HAV-특이적 항체 또는 상응하는 항체 지지체가 제2특이적 결합성분으로서 사용되는 방법.
  8. 제1항에 있어서, 효소가 신호-발생성분으로서 사용되는 방법.
  9. 제8항에 있어서, 양고추냉이 퍼옥시다제가 효소로서 사용되는 방법.
  10. 제7항에 있어서, 제1특이적 결합성분 및 제2특이적 결합성분이 공통 결합성분상의 상이한 결합 부위를 인식하는 방법.
  11. 제1항 또는 제2항에 있어서, 분석물이 항체인 방법.
  12. 제11항에 있어서, 분석물이 항-HAV-특이적 항체인 방법.
  13. 제12항에 있어서, 결정하고자 하는 분석물이 아부류 1gM 및/또는 1gG의 항-HAV-특이적 항체인 방법.
  14. 제1항에 있어서, 제3특이적 결합성분이 면역학적으로 관련있는 2개 이상의 상이한 결합부위를 가져야 하는 방법.
  15. 제14항에 있어서, 제3특이적 결합성분이 단지 최소한으로 정제된 형태로 사용되는 방법.
  16. 제14항에 있어서, 제3특이적 결합성분이 고분자인 방법.
  17. 제14항에 있어서, 제3특이적 결합성분이 바이러스 항원인 방법.
  18. 제14항에 있어서, 제3특이적 결합성분이 세포 배양중에 발생한 바이러스 항원인 방법.
