Abstract

본 발명은 아폽토시스 불록커라 알려진 bcl-2와 동종성을 갖는 신규한 아폽토시스 조절 유전자에 관한 것으로서 인간의 태아 간 조직으로부터 분리하여 fl-383d 클론으로 명명하였다. fl-383d는 총 734bp의 염기로 이루어졌으며, 한개의 오픈리딩 프레임을 가지고 있고, fl-383d 클론으로 형질전환된 383-E.coli는 한국종균협회에 KKCCM10061호로 기탁되어 있다. 아폽토시스의 분자적 기작이 거의 연구되지 않은 상황에서 아폽토시스 조절에 관하여 새로운 유전자의 발견은 아폽토시스 기작의 이해 및 이를 응용한 암의 진단시약, 암치료제의 개발 등에 유용하게 이용할 수 있다.

제1도는 램덤 씨퀀싱에 의해 결정된 fl-383d 클론의 염기서열과 컴퓨터 프로그램을 이용하여 상기 클론과 유사성을 갖는 유전자를 검색한 결과이며, 제2도는 fl-383d 클론의 전체 염기서열 및 이로부터 유추한 아미노산 서열을 나타낸 것이다, 제3도는 아미노산 수준에서 fl-383d 클론과 A1 유전자를 비교한 것이다.

  1. 하기의 염기배열 전체 또는 그 일부로 구성된 아폽토시스 조절 유전자.
  2. 제1항의 아폽토시스 조절 유전자 클론에 의해 형질전환된 E.coli(383-E.coli)(기탁번호 KCCM10061).
