Abstract

본 발명은 인체 락토페린을 포유동물로부터 생산하기 위한 발현 벡터, 상기 발현벡터로 형질전환된 포유동물 및 상기 포유동물로부터 인체 락토페린을 생산하는 방법에 관한 것으로, 소의 베타-카제인 유전자, 하이브리드 인트론 및 인체 락토페린 유전자를 포함하는 발현벡터를 포유동물의 수정란에 주입하여 형질전환된 동물을 탄생시킨 후, 동물을 사육하여 유즙으로부터 인체 락토페린을 분리하는 본 발명의 방법에 의하면 높은 효율로 인체 락토페린을 생산할 수 있다.

본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 플라스미드 pBL1의 제작과정 및 구조를 나타낸 것이고,
제2도는 생쥐의 조직에서 인체 락토페린의 발현여부를 확인한 결과를 나타낸 것이다.

Claims (8)

  1. 소의 베타-카제인 유전자, 하이브리드 인트론 및 인체 락토페린 유전자를 포함하는, 포유동물에서 인체 락토페린을 생산하기 위한 발현벡터.
  2. 제1항에 있어서, 상기 하이브리드 인트론이 소 베타-카제인의 인트론 1과 토끼 베타-글로빈 유전자의 인트론 2를 결합한 것임을 특징으로 하는 발현벡터.
  3. 제2항에 있어서, 상기 하이브리드 인트론이 베타-카제인의 분비 신호 앞에 위치함을 특징으로 하는 발현벡터.
  4. 제1항에 있어서, 인체 락토페린이 유선에서만 특이적으로 발현되도록 소 베타-카제인 5'-인접(flanking) 서열을 포함하는 발현벡터.
  5. 제1항에 있어서, 2kb의 소 베타-카제인 5'-인접(flanking) 서열, 베타-카제인의 엑손 1, 하이브리드 인트론, 베타-카제인의 분비 신호, hLF의 cDNA로부터 얻은 DNA 서열, hLF의 번역 종결 서열, 베타-카제인의 엑손 8 및 엑손 9, 및 SV40 바이러스의 폴리아데닌 첨가신호를 순서대로 포함하는 발현벡터.
  6. 제5항에 있어서, 플라스미드 pBL1(KCTC 8630P).
  7. 제1항 내지 제6항 중 어느 한 항에 발현벡터로 형질전환된 포유동물.
  8. 제1항 내지 6항 중 어느 한 항에 발현벡터를 포유동물의 수정란에 주입하여 형질전환된 동물을 탄생시킨 후, 동물을 사육하여 유즙으로부터 인체 락토페린을 분리함을 포함하는, 인체 락토페린의 생산 방법.
