Abstract

알부민 수용액을 음이온 교환제와 양이온 교환 그룹을 함유하는 폴리아크릴아미드형 담체인 양이온 교환제로 처리하고, 당해 용액을 열처리함을 포함하는 본 발명의 알부민 제제의 제조방법을 기술하였다. 폴리아크릴아미드형 담체를 양이온 교환제로 사용함으로써, 불순한 단백질로 인한 중합이 억제되고 고순도의 알부민 제제를 수득할 수 있게 된다.

Claims (15)

  1. 혈청 알부민을 함유하는 수용액을 음이온 교환체로 처리하는 단계(a), 단계(a)의 생성액을, 양이온 교환그룹을 함유하는 폴리아크릴아미드형 담체인 양이온 교환제로 처리하는 단계(b) 및 단계(b)의 생성액을 열처리하는 단계(c)를 포함하는 알부민 제제의 제조방법.
  2. 제1항에 있어서, 폴리아크릴아미드형 담체가 폴리(N-트리스[하이드록시메틸]메틸메타크릴아미드)인 방법.
  3. 제2항에 있어서, 양이온 교환제가 카복시메틸(CM)형 또는 설포프로필(SP)형 폴리(N-트리스[하이드록시메틸]메틸메타크릴아미드)인 방법.
  4. 제3항에 있어서, 양이온 교환제가 CM-트리스아크릴(Trisacryl) 또는 SP-트리스아크릴인 방법.
  5. 제1항에 있어서, 4 내지 8의 pH에서 양이온 교환제로 처리함을 포함하는 방법.
  6. 제5항에 있어서, 4.5 내지 6.5의 pH에서 양이온 교환제로 처리함을 포함하는 방법.
  7. 제6항에 있어서, 5.0 내지 6.0의 pH에서 양이온 교환제로 처리함을 포함하는 방법.
  8. 제1항에 있어서, 음이온 교환제가 강염기성 음이온 교환제인 방법.
  9. 제8항에 있어서, 음이온 교환제가 4급 아미노에틸(QAE)형 비닐 친수성 중합체 또는 4급 메틸암모늄(QMA)형 다공성 실리카겔인 방법.
  10. 제9항에 있어서, 음이온 교환제가 QAE-도오퍼얼(Toyopearl) 또는 QMA-세파로실(Sepharosil)인 방법.
  11. 제1항에 있어서, 3 내지 6의 pH에서 음이온 교환제로 처리함을 포함하는 방법.
  12. 제11항에 있어서, 4.5 내지 5.5의 pH에서 음이온 교환제로 처리함을 포함하는 방법.
  13. 제12항에 있어서, 5.1 내지 5.4의 pH에서 음이온 교환제로 처리함을 포함하는 방법.
  14. 제1항에 있어서, 50 내지 80℃에서 5 내지 20시간 동안 열처리함을 포함하는 방법.
  15. 알부민 중합체의 함량이 0.03중량% 미만인 알부민 제제.
