Abstract

본 발명은 인간 선호 코돈을 사용한 합성 인간 적혈구 성장촉진인자(EPO)유전자 및 이를 포함하는 발현벡터를 제공하며, 상기 발현벡터를 이용하여 동물세포내에서 EPO를 대량 생산하는 방법을 제공한다.

제1도는 인간에서 빈번하게 나타나는 코돈으로 설계된 EOP 59 유전자와 그 5′말단에 연결된 CD5 리더서열의 염기서열을 나타낸 것이고, 제2도는 EPO 60 유전자와 그 5′말단에 연결된 EPO 리더 서열의 염기서열을 나타낸 것이고, 제3도는 EOP 59 유전자와 EOP 60 유전자를 만드는데 이용된 프라이머 DNA의 염기서열을 나타낸 것임.

Claims (7)

  1. 하기의 각각의 아미노산에 대한 하기의 인간 선호 코돈이 전체 코돈의 70% 이상인 염기 서열로 구성된, 인간 적혈구 성장촉진 인자(EPO)를 코드화하는 합성 EPO 유전자:
  2. 제1항에 있어서, 하기의 염기성을 가지는 합성 EPO 유전자:
  3. 제1항에 있어서, 하기의 염기서열을 가지는 합성 EPO 유전자:
  4. 제1항 내지 3항 중 어느 한 항의 유전자를 포함하는 발현벡터.
  5. 제3항에 있어서, EPO 유전자 앞에 EPO 리더 서열, EPO 이외의 세포외 분비단백질 리더 서열 또는 타입-I 세포막 표지 단백질의 리더 서열을 포함하는 발현 벡터.
  6. 제4항에 있어서, 발현벡터 pMI-EPO 59 또는 pMI-EPO 60.
  7. 제4항 내지 제6항 중 어느 한 항의 발현 벡터에 의해 형질전환된 동물세포를 배양함을 포함하는, 인간 적혈구 성장 촉진인자의 제조방법.
