Abstract

본 발명은 네오마이신 내성유전자, 디하이드로폴레이트 리덕타제(Dihydrofolate reductase ; DHFR) 유전자 및 MMTV LTR(mouse mammary tumor virus long terminal repeata)르로모터를 함유하며, 대장균 및 동물세포 내에서 복제 및 발현이 가능한 재조합 플라스미드 벡터에 관한 것으로서 궁극적으로는 상기 플라스미드 벡터를 이용하여 동물세포내에서 원하는 특정 단백질을 좀더 효율적으로 제조하는 것을 목적으로 한다.

본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 플라스미드 pCMVNEO외 pMSG로부터 제조된 복합 플라스미드 pCMSV1의 제작도이다.
제2도는 플라스미드 pCMSV1에 DHFR 유전자를 도입시킨 플라스미드 pNOHA31의 제작도이다.
제3도는 최종 발현용 벡터인 플라스미드 pNOHA31에 포함되어 있는 유전자 및 제한효소의 상세한 지도이다.

Claims (5)

  1. 네오마이신 내성유전자, 디하이드로폴레이트 리덕타제(Dihydrofolate reductase ; DHFR) 유전자 및 MMTV LTR(mouse mammary tumor virus long terminal repeata) 프로모터를 함유하는 재조합 플라스미드 pNOHA31.
  2. 제1항의 재조합 플라스미드 pNOHA31로 형질전환된 숙주세포주.
  3. 제2항에있어서, 대장균 또는 동물세포주임을 특징으로 하는 형질전환된 숙주세포주.
  4. a) 제1항에 따른 재조합 플라스미드 pNOHA31을 제조하고; b) 상기 플라스미드에 외재에 목적단백질 유전자를 삽입시키고; c) 이를 이용하여 동물세포주를 형질전환시킨 후; d) 이 동물세포주를 발현에 적합한 조건하에 배양하고 발현된 단백질을 회수함을 특징으로 하는 목적단백질을 재조하는 방법.
  5. 제4항에 있어서, 목적단백질이 에리트로포이에틴(Erythropoietin ; EPO)임을 특징으로 하는 방법.
