Abstract

본 발명은 한국형 C형 간염바이러스 Core22 유전자 (KHCV LBC1의 346번째에서 915번째 염기까지의 유전자)를 곤충세포에서 발현시키기 위한 발현벡터, 이 발현벡터로 형질전화된 숙주세포 및 이를 이용하여 한국형 C형 감염바이러스의 Core22핵 단백질을 제조하는 방법에 관한 것이다.

제1도는 한국형의 C형 간염바이러스의 Core22핵 단백질의 뉴클레오티드 서열 및 아미노산 서열을 나타낸 것이고,
제2도는 한국형 C형 간염바이러스의 Core22핵 단백질 유전자를 베큘로 바이러스 발현베터 pAC360에 접합시키기 위한, 아미노 및 카복시 말단 프라이머를 나타낸 것이고,
제3도는 한국형 C형 간염바이러스의 Core22핵 단백질 유전자의 접합전략 및 발현벡터의 제조 과정을 나타낸 것이고,
제4도는 Core22 유전자를 함유하는 재조합 베큘로바이러스를 곤충세포에 감염시켜 핵단백질의 발현을 유도시킨 후 웨스턴 블롯팅에 의해 확인한 결과이다.

Claims (7)

  1. 한국형 C형 간염 바이러스의 Core22 유전자를 포함하는 재조합 베큘로바이러스 발현 벡터.
  2. 제1항에 있어서, 야생형 베큘로바이러스 DNA의 폴리히드린 유전자를 상기 Core22 유전자로 치환시킨 발현 벡터.
  3. 제2항에 있어서, Core22 단백질 유전자를 포함하는 재조합 베큘로바이러스 클로닝 벡터를 야생형 베큘로바이러스 DNA와 함께 곤충세포에 공형질감염시켜 제조한 발현 벡터.
  4. 제3항에 있어서, 상기 재조합 베큘로바이러스 클로닝 벡터가 pVC360 Core22인 발현 벡터.
  5. 제1항 내지 제4한 중 어느 한 항의 발현 벡터를 포함하는 재조합 베큘로바이러스.
  6. 제5항에 있어서, BVLUC-Core22(ATCC VR 2419)인 재조합 베큘로바이러스.
  7. 제5항 또는 제6항의 재조합 베큘로바이러스로 곤충세포를 감염시켜 배양함을 포함하는, 한국형 C형 간염 바이러스의 Core22핵 단백질의 제조방법.
