Abstract

본 발명은 안티센스 방향으로 가광견병 바이러스 서열, 특히 PRV의 gII 부위로 부터의 서열을 함유하는 폴리뉴클레오티드, 그의 제조방법 및 그의 용도에 관한 것이다.

가광견병 바이러스 폴리뉴클레오티드 및 바이러스-내성 진핵세포의 제조를 위한 그의 용도

제1도는 PRV의 gII 부위를 포함하는 PRV의 제한지도의 한 부분 및 예로써 인용된 안티센스 작제물의 위치를 나타낸다.
제2도는 벡터의 SV40초기 프로모터에 대해 안티센스 방향으로 180bp길이의 PRV-gII단편(서열범위 879-1058)을 갖는 pBEH의 벡터지도를 나타낸다.
제3도는 진핵성 발현벡터의 프로모터에 대해 안티센스 방향으로, 진핵세포내에서 PRV에 대한 고도의 내성을 야기시키는 PRV gII 부위에서 유래한 180bp 길이의 DNA단편(서열범위 879-1058)의 뉴클레오티드 염기서열을 나타낸다.

Claims (18)

  1. 프로모터 서열에 대해 안티센스 방향으로 위치하는 PRV 유전자 서열을 함유함을 특징으로 하는 안티센스 DNA 폴리뉴클레오티드.
  2. 프로모터 서열에 대해 안티센스 방향으로 위치하는 것으로 PRV 의 gII 부위로부터 유래한 유전자 서열을 함유함을 특징으로 하는 안티센스 DNA 폴리뉴클레오티드.
  3. 프로모터 서열에 대해 안티센스 방향으로 위치하는 것으로, PRV의 gII 부위의 뉴클레오티드 서열 879-1058에 상응하는 180bp 크기의 부위를 함유함을 특징으로 하는 안티센스 DNA 폴리뉴클레오티드.
  4. PRV의 유전자 서열을 함유함을 특징으로 하는 안티센스 RNA 폴리뉴클레오티드.
  5. PRV의 gII 부위로 부터 유리한 유전자 서열을 함유함을 특징으로 하는 안티센스 RNA 폴리뉴클레오티드.
  6. PRV의 gII 부위d의 뉴클레오티드 서열 879-1058에 상응하는 180bp 크기의 부위를 함유함을 특징으로 하는 안티센스 RNA 폴리 뉴클레오티드.
  7. PRV의 바이러스성 RNA에 대해 상보적인 PRV의 유전자 서열을 함유함을 특징으로 하는 안티센스 올리고뉴클레오티드.
  8. PRV의 바이러스성 gII RNA에 대해 상보적인 PRV gII부위의 유전자 서열을 함유함을 특징으로 하는 안티센스 올리고뉴클레오티드.
  9. 바이러스성 gII RNA에 대해 상보적인, PRV의 gII부위로 부터 유래한 180bp 크기의 부위(879-1058)로부터의 유전자 서열을 함유함을 특징으로 하는 안티센스 올리고뉴클레오티드.

  14. 제1항 내지 6항 및 제10항 내지 12항중의 어느 하나에 따르는 안티센스 폴리뉴클레오티드를 세포내에 함유하는 돼지.
  15. 제7항 내지 9항중의 어느 하나에 따르는 안티센스 올리고 뉴클레오티드를 세포내에 함유하는 돼지.
  16. 제1항 내지 6항 및 제10항 내지 12항중의 어느 하나에 따르는 폴리뉴클레오티드를 바이러스-감수성 세포내에 이입시키고, 바이러스성 RNA에 대해 상보적인 RNA를 형성시킴으로써 PRV의 바이러스성 복제를 방지함을 특징으로 하여 PRV-내성세포를 제조하는 방법.
  17. 제7항 내지 9항중의 어느 하나에 따르는 안티센스 올리고 뉴클레오티드를 동물에게 투여함을 특징으로 하여 PRV 감염을 억제 시키는 방법.
  18. 형질 변형된 PRV-내성 동물의 생산에 적합한 세포내에 이입시키기 위한, 제1항 내지 6항 및 제10항 내지 12항중의 어느 하나에 따르는 폴리뉴클레오티드의 용도.
