본 발명은 이. 콜리(E, coli)에서 단백질을 낮은 내지 중간 정도로 발현시키기에 유용한 DNA 서열에 관한 것이다. 서열은 트랜스포슨 Tn10의 tet A 유전자의 억제체 결합 부위의 변형을 기초로 한다.

Claims (11)

  1. 억제제 결합부위, 리보솜 결합부위의 5' 영역 및 변역 출발 코돈을 가진 Tn 10 tet A 유전자의 부분을 포함하는 DNA 서열에 있어서, 교체 뉴클레오타이드 서열에 의한 억제체 결합 부위의 적어도 부분적인 치환을 포함함을 개선점으로 하는 DNA 서열.
  2. 제1항에 있어서, 교체 뉴클레오타이드 서열은 적어도 하나의 제한 부위를 포함하는 서열.
  3. 제1항에 있어서, 단백질에 관한 암호 영역을 포함하는 서열.
  4. 제3항에 있어서, 단백질이 트랜스 포슨 Tn 10의 tet A 유전자, 또는 에스, 아우레우스(S. aureus)의 tet K 유전자에 의해 암호화된 테트라사이클린 유출 펌프인 서열.
  5. 제4항에 있어서, Sal I 제한부위, ATG 출발 코든, 및 tet A 암호영역 또는 tet K 암호영역을 포함하는 서열.
  6. 제5항에 있어서, 제1B도에 나타낸 바와같이, pCBSal에 존재하거나, 또는 제1B도에 나타낸 바와 같이 pGG57에 존재하는 서열.
  7. 제1-6항 중 어느 한 항의 서열을 포함하는 벡터.
  8. 제7항에 있어서, 플라스미드인 벡터.
