섬유아세포 성장 인자에 대해 친화성을 갖는 올리고사카라이드 및 이의 제조방법

제1도는 bFGF에 대해 고친화력을 갖는 헤파란 설페이트 샘플의 bFGF-세파로즈 칼럼(Sepharose column)으로부터의 용출 패턴을 도시하며, 여기서는 소의 대동맥에서 유래된 헤파란 설페이트이며 ○는 마우스의 EHS 종양에서 유래된 헤파란 설페이트이고 ;
제2도는 bFGF에 대해 친화력을 갖는 올리고사카라이드 샘플의 bFGF-.세파로즈 칼럼으로부터의 용출 패턴을 도시하며 ;
제3도는 FGF에 대해 친화력을 갖는 올리고사카라이드 샘플의 세파덱스(Sephadex) G-50 칼럼으로부터의 용출 패턴을 도시하고 ;
제4도는 〔3H〕티미딘을 DNA내로 혼입시킨데 대한, FGF에 대해 친화력을 갖는 올리고사카라이드 샘플의 영향을 도시한 그래프이며, 여기서, ○는 aFGF이고, ●는 aFGF+트립신 처리를, △는 aFGF+FGF=에 대해 친화력을 갖는 올리고사카라이드+트립신 처리를 ▲는 aFGF+FGF에 대해 친화력을 갖지 않는 올리고사카라이드+트립신 처리를 나타낸다.

Claims (9)

  1. 주요 디사카라이드 단위가 L-이두론산 2-설페이트 및 N-설포-D-글루코사민을 포함하는 8 내지 18개의 모노사카라이드 잔기로 이루어진, 섬유아세포 성장 인자에 대한 친화성을 갖는 올리고사카라이드.
  2. 헤파란 설페이트를 헤파리티나제 I로 분해시킴을 특징으로 하여, 주요 디사카라이드 단위가 L-이두론산 2-설페이트 및 N-설포-D-글루코사민을 포함하는 8 내지 18개의 모노사카라이드 잔기로 이루어진, 섬유아세포 성장 인자에 대한 친화성을 갖는 올리고사카라이드를 제조하는 방법.
  3. 제2항에 있어서, 생성된 분해물을 콘드로이틴설페이트의 존재하에 산성 또는 염기성 섬유아세포 성장 인자-결합 담체와 함께 배양시킨 다음, 상기 담체로부터 결합 분해물을 탈착시키는 방법.
  4. 제2항에 있어서, 헤파란 설페이트가 소, 돼지 또는 양의 소장 또는 대동맥 및 집에서 기르는 수탉의 볏으로부터 수득되는 방법.
  5. 제3항에 있어서, 상기 담체가 아가로즈 겔인 방법.
  6. 헤파란 설페이트를 산성 또는 염기성 섬유아세포 성장 인자-결합 담체와 함께 배양시키고 이 담체에 결합된 헤파란설페이트의 분획을 분리시켜서 분리된 헤파란 설페이트 분획을 헤파리티나제 I로 분해시킴을 특징으로 하여, 주요 디사카라이드 단위가 L-이두론산 2-설페이트 및 N-설포-D-글루코사민을 포함하는 8 내지 18개의 모노사카라이드 잔기로 이루어진, 섬유아세포 성장 인자에 대한 친화성을 갖는 올리고사카라이드를 제조하는 방법.
  7. 제6항에 있어서, 생성된 분해물을 콘드로이틴설페이트의 존재하에 산성 또는 염기성 섬유아세포 성장 인자-결합 담체와 함께 배양시킨 다음, 상기 담체로 부터 결합분해물을 탈착시키는 방법.
  8. 주요 디사카라이드 단위가 L-이두론산 2-설페이트 및 N-설포-D-글루코사민을 포함하는 8 내지 18개의 모노사카라이드 잔기로 이루어진, 섬유아세포 성장 인자에 대한 친화성을 갖는 올리고사카라이드를 2개 이상 포함하는 조성물.
  9. 섬유아세포 성장 인자 및 1개 이상의 주요 디사카라이드 단위가 L-이두론산 2-설페이트 및 N-설페이트 및 N-설포-D-글루코사민을 포함하는 8 내지 18개의 모노사카라이드 잔기로 이루어진 섬유아세포 성장 인자에 대한 친화성을 갖는 올리고사카라이드를 약제학적으로 허용되는 담체와 함께 포함하는 약제학적 조성물.
