Claims (10)

  1. 시험 샘플중에서 특정 항원에 대한 항체를 검출하기 위하여 검출대상 항체에 특이적인 제1항원을 고체상에 고정화시키고, 제1항원에 시험 샘플중의 검출대상 항체를 결합시켜 고정화시키며, 고정화된 항체에 표지된 제2항원을 추가 결합시키고, 상기 제1항원 및 제2항원은 동종 공급원으로부터 유도되는 샌드위치 면역 검정법을 수행하는 방법에 있어서, 제2항원을 제1항원의 공급원에 대해 이종인 공급원으로부터 유도됨을 특징으로 하는 개선된 방법.
  2. (a) 검출대상 항체에 특이적인 재조합-유도된 제1항원을 고체상에 고정화시키고, (b) 단계(a)에서 생성된 고체상에 항원-특이적 항체를 함유하거나 함유하고 있는 것으로 기대되는 수성상 시험 샘플을 접촉시키며, (c) 단계(b)에서 생성된 고체상에 재조합-유도된 제1항원의 공급원에 대해 이종인 공급원으로부터 유도되고 표지를 부착시킨 재조합-유도된 제2항원을 함유한 수성상을 접촉시키고, (d) 수성상을 고체상으로 부터 분리시키며, (e) 고체상 또는 액체상에서 표지의 존재를 측정하여 시험 샘플중의 항체를 검출 및/또는 역가하는 단계를 포함함을 특징으로 하여, 시험 샘플중의 항원-특이적 항체를 검정하는 방법.
  3. 제 2 항에 있어서, 표지가 효소 또는 방사성 동위원소인 방법.
  4. 제 2 항에 있어서, 재조합-유도된 제1및 제2항원이 둘다 동시에, HTLV Ⅲ의 p24항원, HTLV Ⅲ의 p41항원, HTLV Ⅲ의 gp120항원, HBsAg, HBcAg, HBeAg, 또는 항원성 단편 또는 이의 에피토프를 포함하는 그룹중 하나이며, 단, 재조합-유도된 제1 및 제2항원이 시험 샘플중의 항원-특이적 항체와 가교결합 하기에 충분한 정도의 공통된 항원 결정기를 갖는 방법.
  5. 제 2 항에 있어서, 단계(b) 및 (c)를 동시에 수행하는 방법.
  6. 제 2 항에 있어서, 고체상이 폴리스티렌 비드인 방법.
  7. (a) 폴리스티렌 비드를 효모로부터 정제된 p41항원으로 피복시키고, (b) 피복된 비드에 인간 혈청 샘플을 가하며, (c) 약 40℃에서 약2시간 동안 배양하고, (d) 비드를 탈이온수로 세척하며, (e) 비드에 정제및 표지된 p41항원 또는 정제 및 표지된 단편 또는 이의 에피토프를 가하고, 이때 p41항원 또는 단편 또는 이의 에피토프는 비드상의 p41항원을 생성시키기 위해 사용된 공급원과 이종인 유기체로부터 유도되며, 단편 또는 에피토프는 단계(a)의 항원과 공통된 항원 결정기를 충분히 가짐으로써 인간 혈청중의 p41에 대한 항체와 가교결합을 할 수 있으며, (f) 약 40℃에서 약 1시간 동안 배양시키고, (g) 비드를 탈이온수로 세척하고, (h) 반응하지 않은 시약을 비드로부터 분리시키며, (i) 비드상에 존재하는 표지물을 측정하는 단계를 포함함을 특징으로 하여, p41(HTLV Ⅲ)인간 혈청중의 검정하는 방법.
  8. 제 7 항에 있어서, 단계(a)의 p41항원이 효모에서 생성되고, 단계(e)의 표지된 p41항원이 이 콜라이에서 생성되는 방법.
  9. 제 7 항에 있어서, 단계(e)의 p41항원이 마우스세포에서 생성되는 방법.
  10. 제 7 항에 있어서, 표지물이125I인 방법.
