용성 성숙 hIL-β 및 변경된 생물학적 활성을 갖는 유도체를 이 콜라이에서 고도로 발현시키는 플라즈미드 및 그 방법

제1도는 플라즈미드 PDP 506의 제조공정도이고,
제2도는 Pro hIL-1β를 암호화하는 DNA를 함유하는 1.6Kb 절편을 나타내며.

Claims (31)

  1. 실질적으로 플라즈미드 pUC 8 및 성숙(nature) hlL-1β의 아미노산 서열 5 내지 153을 암호화하는 DNA로 이루어지고, pUC 8의 lac 프로모터 하부에 있는 ATG/Met 해독 출발 코돈이 위치 5의 TCA Ser코돈으로부터 아미노산 코돈 4 내지 18개의 간격을 유지하는, 성숙 hlL-1β의 아미노산 서열 5 내지 153을 함유하는 용성 단백질을 이.콜라이(E.coli)에서 고도로 발현시킬 수 있는 플라즈미드.
  2. 제1항에 있어서, 성숙 hlL-1β의 아미노산 서열 5 내지 153을 암호화하는 DNA절편이 pUC 8의 Pst I부위에 삽입된 플라즈미드.
  3. 제2항에 있어서, ATG/Met 해독 출발 코돈이 TCA/Ser 코돈으로부터 4 내지 8개 코돈의 간격을 유지하는 플라즈미드.
  4. 제3항에 있어서, ATG/Met 해독 출발 코돈이 TCA/Ser코돈으로부터 서열 Thr Asn Ala Pro Val Arg을 암호화하는 6개 코돈의 간격을 유지하는 플라즈미드.
  5. 제4항에 있어서, pDP 516, 즉 ATCC 부여번호 67303인 플라즈미드.
  6. 제3항에 있어서, ATG Met/출발 코돈이 TCA/Ser 코돈으로부터 서열 Thr Pro Val Arg을 암호화하는 4개 코돈의 간격을 유지하는 플라즈미드.
  7. 제6항에 있어서, pDP 516-22A, 즉 ATCC 부여번호 67304인 플라즈미드.
  8. 제3항에 있어서, ATG Met/출발 코돈이 TCA/Ser코돈으로부터 서열 Thr Val Arg을 암호화하는 4개 코돈의 간격을 유지하는 플라즈미드.
  9. 제8항에 일어서, pDP 516-18, 즉 ATCC 부여번호 67305인 플라즈미드.
  10. 제2항에 있어서, ATG/Met 해독 출발 코돈이 TCA/Ser코돈으로부터 서열 Thr Met Ile Thr Asn Ser Arg Gly Ser Val Asp Leu His Asp Ala Pro Val Arg을 암호화하는 18개 코돈의 간격을 유지하는 플라즈미드.
  11. 제9항에 있어서, pDP 506, 즉 ATCC 부여번호 67302인 플라즈미드.
  12. 제1항에 있어서, ATG/Met 해독 출발 코돈이 TCA/Ser 코돈으르 4개 코돈의 간격을 유지하는 플라즈미드.
  13. 제12항에 있어서, 4개 코돈이 Thr Met Gly Arg을 암호화하는 플라즈미드.
  14. 제12항에 있어서, 4개 코돈이 Thr Met Pro Arg을 암호화하는 플라즈미드.
  15. 제12항에 있어서, 4개 코돈이 Thr Met Leu Arg을 암호화하는 플라즈미드.
  16. 제12항에 있어서, 4개 코돈이 Thr Met Thr Arg을 암호화하는 플라즈미드.

  24. 위치 1 내지 4의 1개 이상에서 아미노산 치환체를 갖는 성숙 hIL-1β의 유도체.
  25. 제24항에 있어서, 위치 1 내지 2의 어느 1개 또는 2개에서만 아미노산 치환체를 갖는 유도체.
  26. 제25항에 있어서, 위치 1에서 아미노산 치환체를 갖는 유도체.
  27. 제26항에 있어서, 치환체가 Ala 대신 Thr인 유도체.
  28. 제25항에 있어서, 위치 1 및 2에서 2개의 아미노산 치환체를 갖는 유도체.
  29. 제28항에 있어서, 치환체가 Ala 및 Pro대신 각각 Thr 및 Met인 유도체.
  30. 제24항에 있어서, 위치 1 내지 3에서 3개의 아미노산 치환체를 갖는 유도체.
