Abstract
본 발명은 신규 쿠마린 기반 티오페닐-피라졸릴티아졸 (coumarin-based thiophenyl-pyrazolylthiazole) 유도체 또는 이의 약학적으로 허용가능한 염, 및 이를 포함하는 티로시나제 (tyrosinase) 억제제에 관한 것이다.
Description
본 발명은 신규 쿠마린 기반 티오페닐-피라졸릴티아졸 (coumarin-based thiophenyl-pyrazolylthiazole) 유도체 또는 이의 약학적으로 허용가능한 염, 및 이를 포함하는 티로시나제 (tyrosinase) 억제제에 관한 것이다.
유기화학과 생물학은 의약품화학에서 가장 특권적인 조합으로 여겨진다. 다양한 치명적인 질병을 치료하기위한 의약품 개발에서 유기 분자의 중요성은 확실히 포괄적이다. 최근 과학자들은 피부를 희게하고 피부병을 치료하는 데 사용할 수있는 약물을 확인하고 합성하기 위해 열심히 노력해 왔다. 티로시나아제 (EC 1.14.18.1) 세균, 동물 등의 자연계에 널리 분포하는 멀티 구리 효소이다. 손상된 과일의 멜라닌 형성과 효소에 의한 갈변에 중요한 역할을 한다. 티로시나제는 멜라닌 형성 과정 동안 발생하는 2단계 반응의 촉매 역할을 한다. 히드록실화에 의해 모노페놀을 오르토디페놀(o-디페놀)로 변환한 다음 오르토디페놀이 산화되어 오르토퀴논(o-퀴논)으로 변환된다. 멜라닌 형성 과정에서 멜라닌 세포에 의해 생성되는 멜라닌은 인간의 피부, 머리카락 및 눈의 색소 침착을 일으키는 주요 염료이다. 피부에서 멜라닌의 형성은 태양에 해로운 자외선과 피부의 광발암에 대응하는 필수적인 광 보호 인자이다. 인간에서는 비정형의 멜라닌 손실과 색소 탈실, 또는 과도한 멜라닌 생성과 색소 침착 과잉은 심각한 심미적 및 피부 상태가 될 수 있다. 간반, 주근깨, 흑피증, 흑자, 달걀 흰자 및 파킨슨 병과 관련된 신경 퇴행성을 포함한 다양한 피부 질환은 멜라닌 색소 생산 증가로 인해 심각하고 정신적으로 고통스러운 문제를 일으킬 수 있다.
따라서, 티로시나제 억제제에 대하여 화장품 및 제약 산업에서 염료 탈실제로서 식품 및 농업 산업 모두에서 갈색화 방지제로 사용할 수 있는 효과적인 억제제를 개발하도록 장려해 왔다.
현대 의약품 화학에서 하이브리드 파마 코포어 기술은 두 개 이상의 의약품으로 중요한 비계를 결합하여 생물학적 활성 화합물을 설계하는 방법이 있다. 그러므로 신약과 의약품의 개발은 제약 업계와 학술 연구자들에게 도전적이고 필수적인 주제이다. 고품질의 치료제 후보를 개발하기 위해 세 개의 복소환 단위를 쿠마린 모티프와 결합했다.
쿠마린은 의학, 약리학, 화장품, 식품산업 등의 분야에서 그 다양한 생물활성에 의해 주목받고 있다. 피라졸린은 상피성장인자 수용체 티로신 키나아제 (EGFR-TK), 오로라 키나아제 (aurora kinase), COX-2 / B-Raf, 텔로 머라제 및 tubulin의 조립을 억제하는 것으로 보고된 잘 알려진 생물 활성 단위이다.
한편, 티아졸은 페니실린이나 비타민 B1의 일부이며, 그 유도체는 항균 [15], 항통합실조증, 항종양 및 세포독성, 항알레르기, 항염증, 항고혈압 및 진통 특성을 나타낸다. 또한, 티오펜은 진통 및 항염증, 항균, 항하이퍼텐시브 및 항종양 활성과 같은 다양한 용도의 중요한 복소환이다.
이러한 다양한 생물학적 활성에 비추어, 하나의 골격에서 다단계 합성을 사용하여 새로운 하이브리드 헤테로사이클 분자를 생성하기 위해 티로시나제 억제 및 세포독성을 조사했다. 분자 도킹과 동역학 연구는 실험 데이터와 양의 상관 관계를 보여주고 피부 질환의 신약 후보 개발에 사용될 수 있음을 시사했다.
이러한 기술적 배경하에서, 본 출원의 발명자들은 신규하게 쿠마린 기반 티오페닐-피라졸릴티아졸 (coumarin-based thiophenyl-pyrazolylthiazole) 유도체를 합성하였고, 이러한 유도체는 새로운 클래스의 티로시나제 억제제로 설계 및 합성될 수 있음을 확인하고, 본 발명을 완성하였다.
본 발명은 신규 쿠마린 기반 티오페닐-피라졸릴티아졸 유도체 또는 이의 약학적으로 허용가능한 염을 제공하는 것이다.
본 발명의 다른 목적은 상기 쿠마린 기반 티오페닐-피라졸릴티아졸 유도체 또는 이의 약학적으로 허용가능한 염을 포함하는 티로시나제 활성 억제용 조성물을 제공하는 것이다.
상기와 같은 목적을 달성하기 위하여, 본 발명은 하기 화학식 1로 표시되는 화합물 또는 이의 약학적으로 허용가능한 염에 관한 것이다:

본 발명은 또한, 상기 화합물 또는 이의 약학적으로 허용가능한 염을 포함하는 티로시나제 (tyrosinase)의 활성 억제용 조성물에 관한 것이다.
새로운 티로시나제 억제제를 개발하기 위해 일련의 티오페닐-피라졸릴티아졸-쿠마린 하이브리드 유도체를 합성하고 생물학적으로 평가한 결과, 합성된 모든 화합물이 강력한 티로시나제 저해 활성을 가질 수 있음을 확인하였다.

멜라닌 형성과 과잉 색소 침착으로 잘 알려진 중요한 효소는 티로시나제이다. 티로시나제 억제제로서 티오페닐-피라졸릴티아졸-쿠마린 하이브리드의 새로운 시리즈를 확인하였다. 시험관내 티로시나제 저해의 결과는 모든 화합물이 강력한 티로시나제 저해 활성을 갖는다는 것을 나타내며, 특히 화합물 6g (IC 50 = 0.043 ± 0.006 iM)이 양성 대조군 (코지산, IC 50 = 18.521 ± 1.162 iM)과 비교하여 가장 활성이 높은 화합물로 확인되었다.
Lineweaver-Burk 플롯을 사용하여 역학 메커니즘을 분석하고 화합물 6g은 티로시나아제를 비 경쟁적으로 억제하여 효소 - 억제제 복합체를 형성했니다. 또한, 모든 화합물은 DPPH에 대해 매우 우수한 항산화 활성을 보였다. MTT 분석은 B16F10 흑색종 세포에 대한 모든 화합물의 세포 독성을 스크리닝하는 데 사용되었으며 세포에 독성 효과가 없었다. 화합물의 티로시나제와의 결합 친화도 또한 분자 도킹을 사용하여 조사되었고, 리간드는 양호한 결합 에너지 값을 나타냈다. 이러한 분자는 비독성 약리학 스캐폴드로서 피부의 색소 침착 및 관련 질환의 유망한 리드가 될 수 있다.

"치환 또는 비치환"은비치환될 수 있거나 또는 치환될 수있는 모 그룹(parent group)을 포함할 수있음을 의미하고, "치환된"은 1 이상의 치환기를 갖는 모그룹을, 치환기는 모기에 공유결합되거나 모그룹에융합된 화학적 부분을 의미한다.
구체적으로, 상기 화학식 1에서 상기 R은 수소, 4-C1-C6 알킬, 4-C1-C6 알콕시, 4-할로겐, 4-C1-C6 알콕시-C1-C6 알킬 또는 3-C1-C6 알콕시-4-C1-C6 알콕시페닐일 수 있다.
본 발명에 따른 구체적 실시예에서, 상기 화학식 1 중 상기 R은 수소, 4-Cl, 4-Br, 4-CH3, 4-OCH3, 4-OCH2-CH3 및 3-OCH3-4-OCH2Ph로 구성된 군에서 선택되는 하나 이상일 수 있다.

구체적으로, 2-아세틸 티오펜 (1)과 다양한 알데히드 (2a - 2h)를 순수한 에탄올에서 수산화 나트륨과 반응시켜 원하는 칼콘 (3a - 3h)을 수득하고, 칼콘 유도체 (3a - 3h)를 티오세미카르 바지드와 함께 에탄올 중 염기성 조건하 (KOH)로 환류시켜 피라졸 스캐폴드 (4a - 4h)를형성하였다. 마지막으로, 티오 아미드 유도체 (4a - 4h)와3- 브로모아세틸 쿠마린 (5)을환류 에탄올 중에서 반응시켜 비치환 페닐-티오페닐-4,5-디하이드로-1H - 피라졸릴-티아졸릴-2H-크로멘-2-온(6a - 6h)을 수득하였다.

또한, 본 발명은 멜라닌 생성억제용 또는 항산화용 화장료 조성물일 수 있다.
상기 화장료 조성물은 총중량에 대하여 상기 화학식 I의 화합물 또는 이의 약학적으로 허용가능한 염을 0.001 내지 90중량%, 바람직하게는 0.01 내지 70중량%를 포함할 수 있다. 또한, 화장료 조성물은 상기 화학식 I의쿠마린 유도체 화합물 외에 비타민, 펩티드, 다당, 지질 등을 더 포함할 수있으며, 그 외통상 화장료에 배합되는 유지성분, 보습제, 계면활성제, 안료, 자외선 흡수제, 방부제, 살균제, 산화방지제, 식물추출물, pH 조정제, 알코올, 향료, 정제수 등을포함할 수 있으며, 이로 제한되지 않는다.
상기 화장료 조성물은 용액, 현탁액, 유탁액, 페이스트, 젤, 크림, 로션, 오일, 왁스, 파우더, 스프레이, 비누, 클렌징, 팩, 파운데이션, 메이컵베이스 및 모발화장료로 구성된 군으로부터 선택된 제형일 수 있으며, 이로 제한되지 않는다. 보다 상세하게는, 유연 화장수, 영양 화장수, 영양 크림, 맛사지 크림, 에센스, 아이크림, 헤어토닉, 샴푸, 린스, 클렌징 크림, 클렌징 포옴, 클렌징 워터, 팩, 스프레이 및 파우더의 제형으로 제조될 수 있다.
제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로 플루오로히드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진제를 포함할 수 있다.
제형이 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용되고, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌글리콜, 1,3-부틸글리콜, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다.
제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상의 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소 결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
제형이 계면활성제 함유 클렌징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성유, 라놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
실시예
1. 재료 및 방법
1H NMR 및 13C NMR 스펙트럼을 용매로서 DMSO-d6을 사용하여 400 및 100MHz 분광광도계 Bruker Avance III(독일)로 기록하였다. 화학적 이동(a)은 백만분의 1(ppm)로 보고되고 커플링 상수( J )는 Hz로 표시된다. FT-IR 스펙트럼은 Frontier IR 분광 광도계 (Perkin Elmer, 미국)를 사용하여 획득했다. Fisher Scientific (USA)의 MPA 160 장치를 사용하여 합성된 화합물의 융점 (보정되지 않음)을 결정했다. 모든 상업용 화학 물질 및 시약은 추가 정제없이 사용되었다. 반응의 진행은 박층 크로마토그래피(TLC) 방법에 의해 모니터링하였다.
2. 화합물의 합성 절차
2.1. 칼콘의 합성 순서 (3a-3h)
반응식 1에 나타낸 바와 같이, 2-아세틸 티오펜 (1,1 mmol)을 순수한 에탄올에 용해시키고 빙욕에서 5 분 동안 교반하였다. 이어서, 수산화나트륨(1.1 mmol)을 반응 플라스크에 조금씩 첨가하였다. 그 후, 다양한 방향족 알데히드 (2a-2h, 1mmol)를 반응 혼합물에 첨가하고 빙욕에서 24 시간 동안 교반 하였다. 반응 종료 후, 반응 혼합물을 부서진 얼음에 부어 염산으로 중화하였다. 생성된 침전물을 실온에서 교반한 후, 여과하고, 냉수/에탄올로 세척하고 건조시켜 원하는 칼콘 (3a -3h)을 얻었다.
2.2. 1-티오카르바모일피라졸 유도체의 합성 절차 (4a-4h)
이 단계에서, 1.5 mmol 티오세미카바지드와 순수 에탄올을 10 분간 혼합했다. 이어서, 1.1 mmol 수산화 칼륨을 첨가하고, 추가로 10 분 동안 교반한 후, 대응하는 칼콘(3a-3h, 1mmol)을 첨가하였다. 이어서, 반응이 완료될 때까지 환류하에 반응을 계속하였다 (TLC로 모니터링). 생성된 생성물을 깨진 얼음에 부어 염산으로 중화시키고 30 분 동안 교반 한 다음 여과하고 차가운 에탄올 로 세척하고 건조시켜 티오페닐 - 피라졸 - 카르보티오 아미드 유도체 ( 4a - 4h )을 얻었다.
2.3. 1-티아졸릴-2-피라졸린 유도체 (6a-6h)의 합성 절차
티오페닐 - 피라졸 - 카르보티오 아미드 유도체 (4a - 4h , 1mmol)의 현탁액에 3- 브로모 아세틸 쿠마린 (5, 1mmol)을 첨가한 후 4-5 시간 동안 환류시켰다. 완료 후 (TLC에 의해), 반응 혼합물을 실온에서 냉각시키고, 침전물을 여과하고, 컬럼 크로마토 그래피 (n- 헥산, 에틸 아세테이트)로 정제하여 목적하는 표적 화합물 (6a - 6h)을 적당한 수확 비율(65-86%)로 수득하였다. 합성된 화합물의 물리적 및 분광학 데이터는 아래에 나와 있다.
3. 효능 검정
3.1. 항티로시나제 억제 검정
실험에 사용된 모든 화학 물질 (버섯 티로시나아제, L-DOPA ((3,4-dihydroxyphenylalanine) 및 kojic acid)은 미국 Sigma Chemicals에서 구입했다. 결과를 비교하기 위한 양성 대조군으로서 코디산을 사용하였다. IC 50 의 결과에는 총 8개의 다른 농도가 사용되었으며 GraphPad Prism 5의 비선형 회귀 분석을 사용하여 결과가 계산되었습니다. 티로시나제의 억제율은 다음 식을 사용하여 계산되었다.

3.2. 속도론적 분석 프로토콜
억제의 모드를 결정하기 위해 동역학 분석을 수행했다. 화합물 6g은 가장 강력한 활성에 기초하여 이들 시리즈로부터 선택되었다. 다른 농도의 기질 (L-DOPA (0.0625 ~ 2 mM))과 화합물 (0.00, 0.022, 0.043 및 0.086 iM)이 포함되어 있다. 최대 초기 속도는 30 초 간격에서 효소 첨가 후 5 분까지 흡광도의 초기 직선 부분에서 결정 되었다. 효소의 억제 유형은 기질 농도 1/[L-DOPA] mM-1의 역수가 아니라 속도의 역수(1/V)의 Lineweaver-Burk 플롯에 의해 분석되었다. EI 해리 상수 (Ki)는 억제제 농도에 대 한 1/V 2 차 플롯에서 계산되었다.
3.3. 자유 라디칼 포착 분석
라디칼 포착 활성 (DPPH) 분석은 이전에 공개 된 방법 [29]을 사용하여 수행되었다. 모든 화합물은 먼저 최종 희석을 위해 MeOH 용매를 사용하여 준비되었다. 모든 실험은 삼중으로 수행되었다.
3.4. 화합물의 세포 배양 및 처리
시험 화합물(6a - 6h)의 독성을 시험하는데 사용된 세포는 B16F10 흑색 종 세포주였다. 배양 및 처리 절차는 N. Hosseini Nasab, Y. Han, F. Hassan Shah, B.D. Vanjare, S. 4. 계산 방법론
4.1. 표적 단백질의 선택
버섯 티로시나제의 3차원(3D) 구조는 PDBID 2Y9X( www.rcsb.org/2y9x)가 있는 Protein Data Bank(PDB)에서 액세스되었다. 다음으로, 얻어진 단백질 구조는 공액 구배 알고리즘과 앰버 역장을 사용하여 에너지를 최소화하였다. 라마챈드런 그래프와 버섯 티로시나아제 값은 Molprobity 서버 (http://molprobity.biochem.duke.edu/)를 사용하여 점검하고 예측되었다.
4.2. 설계 리간드의 케모인포매틱스 분석
합성 리간드 (6a-6h)는 ACD/ChemSketch로 스케치되었으며 UCSF Chimera 1.10.1에 의해 더욱 최소화되었다. 반면 합성된 화합물 (6a-6h)의 생화학적 특성과 리핀 스키의 5 가지 법칙 (RO5)은 Molinspiration (http://www.molinspiration.com/) 및 Molsoft (http://www.molsoft.com/)을 사용하여 예측되었다.
4.3. PyRX를 이용한 리간드 설계 및 분자 도킹 시뮬레이션
도킹 실험에서는 버섯의 티로시나아제 구조를 분석하여 문헌 데이터로부터 활성 부위와 결합 포켓 아미노산을 확인했다. 설계된 화학 구조는 mol 형식으로 얻어지고 도킹 실험에서 추가 사용을 위해 PDB 형식으로 변환되었다. 도킹 실험은 활성 결합 부위 잔기에서 그리드 생성으로 구성되었으며 계산 된 도킹 에너지 값을 기반으로 스크리닝되었다. PyRx 스크리닝 도구는 버섯 티로시나아제에 대한 모든 리간드의 분자 도킹 실험을 수행하는데 사용되었다. 그리드 박스의 파라 메트릭 차원 값은 X = 2.6658, Y = 19.8796 및 Z = -46.7082로 조정되어 표적 단백질의 활성 영역에서 리간드의 입체 배치 위치를 보다 적절하게 얻었다. 단백질-리간드 도킹 복합체의 최상의 결합 형태 포즈를 달성하기 위해, 합성된 모든 리간드는 기본 망라성 값 = 8에서 버섯 티로시나제에 대해 개별적으로 도킹되었다. 마지막으로, 생성된 도킹 복합체는 최저 결합 에너지(Kcal/mol)값 및 구조 활성 상관 (SAR) 분석에 기초하여 평가되었다. 도킹된 모든 복합체의 그래픽 묘사는 Discovery Studio(2.1.0)에 의해 실현되었다.

반응식 1에 나타낸 바와 같이, 설계된 티오페닐-피라졸린-티아졸릴-크로메논 분자를 합성하기 위해 다단계 공정이 사용되었다. 합성은 2-아세틸 티오펜 (1)과 다양한 알데히드 (2a - 2h)를 순수한 에탄올에서 수산화 나트륨과 반응시켜 원하는 칼콘 (3a - 3h)을 얻기 시작했다. 이어서, 칼콘 유도체 (3a - 3h)를 티오세미카르 바지드와 함께 에탄올 중 염기성 조건하 (KOH)로 환류시켜 피라졸 스캐폴드 (4a - 4h)를 형성하였다. 마지막으로, 티오 아미드 유도체 (4a - 4h)와 3- 브로모아세틸 쿠마린 (5)을 환류 에탄올 중에서 반응시켜 비치환 페닐-티오페닐-4,5-디하이드로-1H - 피라졸릴-티아졸릴-2H-크로멘-2-온(6a - 6h)을 합리적인 수율 (65-86%)로 얻었다. 합성된 모든 화합물은 FT-IR, 1 H NMR 및 13 C NMR 분광법을 사용하여 구조적으로 확인되었다. 화합물 6d는 융점이 232-234°C의 노란색 비정질 고체로 생성되었다. 다른 작용기를 확인하기 위해 화합물의 진동 조사를 FT-IR 분광법을 사용하여 분석했다. 특징적인 흡수 밴드는 i 2903 (CH, 지방족 신축), 1716 (공액 C=O, 신축), 1606 (C=C, sp 2, 신축), 1546, 1485 (C=C, 방향환, 신축), 1383 (CH, 지방족, 굽힘), 1094 (CO, 신축성), 815, 751 (=CH, sp2, 면외 굽힘)에 나타났다. 1H NMR 분광 기술을 사용하여 양성자의 수와 유형을 결정하였다. 양성자 NMR 연구에서 세 개의 싱글렛 피크가 있었다. a 8.29 ppm의 고도로 차폐된 피크, a 2.28 ppm의 고도로 차폐된 피크 및 a 7.71 ppm의 다른 피크는 각각 H-23, H-33 및 H-20을 나타낸다 (도 1). 피라 졸린 고리의 CH2 및 CH 양성자를 포함하는 ABX 스핀 시스템은 a3.42 (dd, J = 16, 8Hz, 1H, H- 7A ), 4.07 (dd, J = 16, 12Hz , 1H, H- 7 B ), 5.66 (dd, J = 12, 8 Hz, 1H, H-8 x ). 4-메틸페닐 고리는 a 7.22 (d, J = 8 Hz, 2H, H-13 & 15), 7.38 (d, J = 8 Hz, 2H, H-12 & 16). 티오펜 유닛의 양성자는 a 7.19 (dd, J = 5, 3.6 Hz, 1H, H-3), 7.48 (dd, J = 3.5, 1 Hz, 1H, H-2)에서 두 개의 더블렛으로 식별되었고, a7.76-7.78에서 쿠마린링으로부터의 하나의 양성자와 합쳐진 하나의 다중선 (m, 2H, H-4 & H-28)으로 식별되었다. 쿠마린 고리의 나머지 3개의 양성자는 a 7.41-7.44 (m, 2H, H-29 & H-31) 및 7.60-7.64 (m, 1H, H-30)의 다중선으로 표현되었다. 탄소의 수와 유형도 13 C NMR 분광법을 사용하여 결정되었다. 분자 내에 몇 개의 대칭 탄소가 있기 때문에 총 24개의 탄소 공명이 관찰되었다. CH3, CH2 및 CH의 세 가지 지방족 신호는 a21.18 (CH3 ) , 44.22 (C-7), 64.63 (C-8)에 나타났다. 2개의 메틴 탄소는 a 111.55 (C-20) 및 129.83 (C-23)에서 검출되었다. 또한, 티오펜 단위의 4 개의 신호는 a 125.30 (C-4), 128.58 (C-3), 129.09 (C-2) 및 138.89 (C-1)에서 관찰되었다. 4-메틸페닐링의 두 대칭 공명은 a 127.45 (2C , C-12 & C-16) 및 129.62 (2C, C-13 & C-15)에서 발견되었다. 쿠마린 고리의 카르보닐 탄소 신호는 a163.78(C-27)에 나타나고 나머지 4개의 방향족 신호는 a116.41(C-31), 130.33(C-29), 132.19(C-28) 및 134.43 에서 확인되었다. (C-30). a119.56 (C-24), 120.94 (C-22), 137.42 (C-11), 138.81 (C-14), 144.29 (C-19), 149.77 (C-25), 152.77(C-6) 및 159.10 (C-17)에서 8개의 4차 신호가 지정되었다. (도 1).

실시예 2. 생물학적 평가
2.1. 항티로시나제 활성과 구조 활성 상관관계
신규 합성 화합물 (6a - 6h)을 통해 티로시나아제 효소에 대한 억제 활성을 측정 하였다. 결과를 표 1에 도시하였다. 참조 약물의 kojic acid (IC 50 = 18.521 ± 1.162 iM)과 비교할 때, 이들 화합물은 모두이 효소에 대해 0.043 ± 0.006 ~ 4.508 ± 0.348 iM 범위의 매우 강력한 억제 활성을 나타냈다. 분자의 전반적인 참여는 저해 활성에 영향을 미치지만 상응하는 표적 화합물의 다른 치환기는 제한된 구조 활성 상관 (SAR)을 나타낼 수 있다. 아릴 엔티티는 이러한 화합물의 유일한 가변 부분이기 때문에, 이 부분의 다양한 치환이 억제 가능성에 영향을 미친다. SAR 결과를 다음과 같이 요약했다. (1) 아릴환에 치환기를 갖지 않는 화합물 6h는 효소와의 강력한 소수성 상호작용이 없기 때문에, 중간 정도의 활성을 나타낸다. (2) 수소 원자 1 개를 할로겐 원자 (-F, -Cl, -Br)로 치환함으로써 저해 효과가 변화한다 (6a - 6c). 표 1에 나타낸 바와 같이, 화합물 6a (IC 50 = 1.946 ± 0.751 iM), 6b (IC 50 = 2.028 ± 0.035 iM) 및 6c (IC 50 = 0.809 ± 0.054 iM )는 각각 플루오로, 클로로 및 브로모 치환기를 포함한다. 화합물 6c는 활성을 크게 증가시킨다. 이는 할로겐 원자의 크기가 커짐에 따라 효소와의 상호작용이 개선되고 억제 활성이 증가한다는 것을 시사한다. (3) 화합물 6d (IC 50 = 4.071 ± 0.136 iM), 6e (IC 50 = 0.278 ± 0.023 iM) 및 6f (IC 50 = 4.508 ± 0.348 iM) 에 1 개의 전자 증가로 표준 약제인 kojic acid 나은 활성을 나타내고, 식별 가능한 패턴은 없다. 화합물 6e는 파라 위치에서 강한 전자 공여 그룹(-OCH3)으로 인해 두 번째로 가장 효과적인 억제제인 반면, 화합물 6f는 부피가 작은 극성 그룹(-OCH2CH3)을 가지며 최종 강력한 억제제이다. (4) 놀랍게도 화합물 6g (IC 50 = 0.043 ± 0.006 iM)에 벤질옥시기와 메톡시기와 같은 두 개의 다른 그룹을 도입하면 가장 우수한 티로시나아제 억제제로서 시너지 효과로 억제 활성이 증가한다. 이것은 화합물 6g이 효소의 활성 부위와의 적절한 상호 작용을 향상시키고 더 많은 수소 결합을 생성한다는 것을 의미한다. 전자 공여기 및 아릴 고리에 두 개의 다른 그룹을 가진 화합물이 티로시나제 효소의 강력한 억제제 역할을 하는 것으로 관찰되었다.

2.2. 동역학 분석
억제의 종류와 티로시나제 억제 상수를 평가하기 위해 가장 효과적인 화합물 (6g)를 선택했다. 유리 효소 및 효소-기질 복합체를 억제하는 화합물의 능력은 각각 EI 및 ESI에 의해 결정되었다. 다양한 억제제 농도가 있는 상태에서 1/V 대 기질 L-DOPA 1/[S]의 Lineweaver-Burk 플롯을 사용하는 효소의 동역학 연구는 도 2A와 같이 일련의 직선을 생성했다. 화합물 6g의 효과는 화합물이 두 번째 사분면 내부에서 교차하는 것을 확인했다. 분석에서 Vmax는 새로운 억제제 용량으로 감소했지만 Km은 동일하게 유지됨을 확인했다. 이 사실에 따르면 화합물 6g는 비경쟁적으로 tyrosinase를 억제하여 효소-억제제 복합체를 형성한다. 억제제 농도 대 기울기의 2차 플롯은 효소 억제제 해리 상수(Ki)를 보여주었다(도 2B). 표 2는 동역학 결과를 제공한다.

2.3. 자유 라디칼 소거 활성
이 실험에서 새로 합성된 모든 화합물 (6a - 6h)을 항산화 활성을 위한 DPPH에 대해 스크리닝했다. 첫째, 모든 화합물을 고농도 (500 μg/mL)로 조사하였다. 활성을 확인한 후 최종 실험 분석을 위해 5 가지 다른 농도 (500, 400, 300, 200 및 100 ig/mL)를 선택했다. 비타민 C (아스코르브 산)를 양성 대조군으로 사용하고 결과를 비교했다 (도 3). 그래프에서 알 수 있듯이, 모든 표적 화합물은 모든 시험 농도에서 매우 우수한 활성을 보였다. 처음 4 가지 농도에서 모든 화합물과 양성 약물은 80% 이상의 활동을 보였다. 100 g/mL 농도의 화합물 6b 만이 다른 것에 비해 50% 감소를 보였다. 그러나, 양성 약물과 비교하여 화합물은 모두 매우 높은 항산화력을 가지고 있다.
2.4. 세포 생존율
B16F10 세포주를 사용하여 세포 증식에 대한 8개의 합성 화합물(6a - 6h)의 효과를 조사하였다. Microculture Tetrazolium Assay를 사용하여 이러한 화합물의 독성을 측정했다. 성장 배지에 포함된 적절한 수의 세포를 먼저 96-웰 ELISA 플레이트에 이식하고 인큐베이터에 24시간 동안 두었다. 세포가 얻은 밀도에 도달 한 후, 이러한 화합물과 대조군 (엘로티닙)을 다양한 농도 (0, 1, 2, 4, 6, 10, 20, 30 및 40 iM)의 세포로 처리하였다. 이 방법으로 얻은 세포 생존율의 결과를 표 3에 나타내었다. 이 화합물들 중 6a - 6d는 1-6 iM에서 영향을 미치지 않았으며, 이러한 농도를 초과하면 세포 집단이 약 20 % 감소했다 (세포생존율 ~ 80 %). 화합물 6e는 2 iM까지 비독성이었지만, 화합물의 농도가 증가함에 따라 세포 증식은 최대 25% 감소하였다. 대조군은 항증식 효과를 나타내는 세포 생존율의 용량 의존적 감소를 나타내었다. 6f - 6h는 세포의 생존율을 약 27% 감소시켰지만, 6f - 6g는 1 μM 농도에서 세포에 영향을 주지 않았다. 이러한 화합물에 의해 영향을 받은 전반적인 세포 생존율은 25-27%이었다. 이러한 생존율의 결과를 고려하면, 이들 화합물은 적당한 독성을 가지며, 다양한 생물학적 활동에 더욱 이용될 수 있다고 생각한다.

실시예 3. 인실리코 분석
3.1. 설계 리간드의 케모 인포매틱스 분석
합성 화합물 (6a-6h)은 RO5 분석에 의해 검증되었다. RO5의 표준 매개 변수는 분자량과 log P 값이 각각 500(g/mol)과 5 미만이어야 함을 나타낸다. 또한, 화합물은 각각 10개 이하의 HBA 및 5개의 HBD를 가져야한다. 수소 결합 능력은 약물 투과성의 중요한 파라미터로 간주되어 왔다. 합성된 리간드가 <10 HBA 및 <5 HBD 값을 갖고 표준값에 필적한다는 것을 정당화하였다. 대부분의 화합물의 log P 값은 표준 log P값과 동등했다. 모든 화합물의 분자량 (g/mol)이 기준값을 초과했다 (표 4). 그러나, 일부 기존 약물 중 RO5 위반에서 벗어나는 사용 가능한 데이터가 있다. 극성 표면적 (PSA Polar surface area)도 장관 흡수, 생체 이용률, 혈액 뇌 장벽의 투과와 같은 약물 흡수를 특성화하는 훌륭한 설명자로 간주된다. 예측 결과는 모든 화합물이 140 A2 미만의 값을 가지며 양호한 납과 같은 거동을 나타내는 것을 보여주었다. 약물 유사성 점수는 분자가 알려진 약물과 유사한지 여부를 결정하는 화합물의 다양한 분자 특성과 구조적 특성의 복잡한 균형의 혼합물이다. 중요 특성은 소수성, 전자 분포, 수소 결합 특성, 분자 크기 및 유연성, 물론 생체 내 분자의 거동에 영향을 미치는 다양한 파마코포어 기능의 존재이며 생체 이용률, 수송 특성, 단백질 에 대한 친화력, 반응성, 독성, 대사 안정성이다. 모든 합성 화합물 (6a-6h)이 양호한 약물 점수값을 보였다는 것을 보여주었다 (표 4).

3.2. 분자 도킹 및 결합 에너지 분석
합성 리간드 (6a ~ 6h)의 도킹 복합체를 최소 에너지 값과 리간드 상호 작용 패턴을 기반으로 분석했다. 결과는 모든 리간드가 양호한 결합 에너지 값을 나타내었고 표적 단백질의 활성 부위 내에서 그들의 상호 작용을 나타냈다는 것을 보여주었다 (표 5). 결합 에너지 분석은 대부분의 리간드가 양호한 결합 에너지 값을 나타내며 모든 합성 화합물 간에 큰 에너지 차이가 관찰되지 않음을 보여주었다. 통상 Autodock의 표준 오차는 2.5 Kcal/mol인 것으로 나타났다. 그러나 모든 도킹 복합체에서 예측된 에너지 값의 차이는 표준 에너지 값보다 작았다. 합성된 모든 화합물의 기본적인 핵은 유사했지만, 따라서 대부분의 리간드는 뛰어난 효율적인 에너지 값을 가지며 큰 에너지 변동에 차이가 없었다.

3.3. 표적 단백질에 대한 리간드의 결합 분석
도 4에 나타낸 바와 같이, 리간드 - 단백질 결합 분석은 6g이 표적 단백질의 활성 결합 포켓에 갇혀 있음을 나타냈다. 6g 수용체 도킹 복합체의 도킹 결과는 도킹 복합체에서 3개의 수소 결합이 관찰됨을 보여주었다. 6g의 수소 원자는 결합 길이 2.73A에서 His85에 대한 수소 결합을 형성했다. 또한, 6g의 다른 산소 원자는 각각 2.43A 및 2.70A의 결합 길이를 갖는 Asn81과 수소 결합을 형성했다. 모든 수소 결합 거리는 3A 미만이었고 도킹 복합체에 대해 양호한 안정성을 보였다.
요약하면, 몇 가지 새로운 thiophenyl-pyrazolylthiazole-coumarin 화합물을 설계 및 합성하고 티로시나제 억제제에 대해 평가했다. 그 결과 모든 화합물이 kojic acid(양성대조군)보다 버섯 티로시나제 효소에 대한 억제 효과가 더 큰 것으로 나타났다. 화합물 6g는 시리즈 전반에 걸쳐 상당히 효율적인 것으로 보였고 표적 효소와 경쟁적이지 않은 것으로 밝혀졌다. 또한, 합성된 화합물의 세포 생존 가능성을 평가하기 위해 다양한 농도의 B16F10에 대해 MTT 분석을 수행한 결과 세포 생존에 대한 독성 효과가 나타나지 않았다. 표적 단백질의 활성 부위와 리간드의 결합 친화도를 조사하기 위해 분자 도킹 연구를 수행했으며 모든 화합물은 버섯 티로시나제와의 허용 가능한 도킹 결합 친화도 및 양호한 도킹 점수를 입증했다. 화합물, 특히 6g은 티로시나제 효소 억제제 개발에 있어 구조적 주형으로 활용될 수 있다.
이상으로 본 발명의 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시예일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다.
