Description
본 발명은 인간들과 같은 포유동물들에서 microRNA-27b (miR-27b)의 활성을 억제할 수 있는 새로운 화합물들 및 조성물들에 대한 것이다. 특히, 본 발명은 간질(epilepsy) 및 기억 장애들(memory disorders)을 포함하는, CNS 장애들을 치료하는 데 유용한 인간 인 비보(in vivo)에서 miR-27b의 활성을 조절할(modulating) 수 있는 안티센스 올리고뉴클레오티드 화합물들을 제공한다.
간질은 전 세계적으로 약 5천만 명의 사람들에게 영향을 미치는 반복적인(recurrent) 자발적(spontaneous) 발작들(seizures)로 특징되는 하는 심각한 만성 신경 장애(neurological disorder)이다.
현재 이용 가능한 항간질제들(anti-epileptic drugs)은 일반적으로 환자들의 삼 분의 이에서 발작들을 통제하지만, 근본적인 병태생리학(pathophysiology)에는 영향을 미치지 않는다. 간질 환자들의 나머지 삼분의 일은 약물 내성이 있거나 또는 현재 이용 가능한 약물들로부터 심각한 부작용들(side effects)로 고통받는다.

실시예 1: 세포 배양(culture)
안티센스 올리고뉴클레오타이드의 miRNA에 대한 효과들의 인 비트로(In vitro) 모델링은 보통 포유동물 세포주들에서 수행된다.
부착성(adherent) 래트(rat) 크롬친화세포종(pheochromocytoma) 세포주 PC-12 Adh (ECACC no. 88022401)은 ATCC (ATCC cat. no. CRL-1721.1™)로부터 구입되었고 2.5% 열-불활성화된 소 태아 혈청(heat-inactivated fetal bovine serum) (Sigma-Aldrich cat. no F4135-500 ml), 15% 열-불활성화된 말 혈청 (heat-inactivated horse serum) (Sigma-Aldrich cat. no. H1385-500ml 및 1% 페니실린(penicillin)/스트렙토마이신(streptomycin) (Sigma-Aldrich cat.no. P4333-100 ml) 으로 보충된 Ham's F-12K (Kaighn's) 배지(medium) (ThermoFischer Scientific cat.no. 21127022)에서 Corning® CellBIND® Surface cell culture flasks (Sigma-Aldrich cat.no. 실시예 2: 배양된 세포주들에서 루시페라제 리포터 분석들
간단하고 매우 민감한 접근법은 루시페라제와 같은, 리포터 유전자의 3' UTR에서 단일의 완벽 매치(match) miRNA 결합 부위를 나르는 miRNA 리포터 플라스미드의 구축(construction)을 수반한다. 이 방법은 miRNA 억제를 입증하고(validate) 또한 여러가지(different) antimiR 디자인들의 효능을 비교하기 위하여 배양된 세포들에서 광범위하게 사용되었다.
miR-27b 리포터는 듀얼-루시페라제 psiCHECK2 플라스미드 (Promega)에서 레닐라(Renilla) 루시페라제 유전자의 3' UTR 내로 인간 miR-27b에 대한 단일의 완벽-매치(single perfect-match) 타겟 부위에 해당하는 어닐링된(annealed) 올리고뉴클레오타이드들을 클로닝하여 생성되었다.
루시페라제 분석들을 위해, PC-12 adh 세포들이 트랜스펙션 전 날 웰 당 25,000 세포들의 밀도로 96-웰 Corning® CellBIND® 표면 세포 배양 마이크로웰 클레이트들(Surface cell culture microwell plates) (Sigma-Aldrich cat.no. CLS3330)에서 시딩(seeded)되었다. 세포들은 Opti-MEM™ I Reduced Serum Medium, GlutaMAX™ Supplement (ThermoFischer Scientific cat. no. 51985026)에서 0.5 μL/웰의 최종 농도(concentration)에서 리포펙타민(lipofectamine) 2000 (ThermoFischer Scientific cat. no. 11668-019)을 사용하여 트랜스펙트되었다. 17개의 안티센스 올리고뉴클레오타이드들의 라이브러리가 루시페라제 리포터 플라스미드 및 miR-27b 미믹으로 최종 농도들 0.2nM, 1 nM, 5 nM으로 각 antimiR-27b를 공-트랜스펙트하여 루시레아제(lucirease) 리포터 분석들을 사용하여 스크리닝되었다. 스크램블된(scrambled) 서열 올리고뉴클레오타이드, miRNA 매치 부위를 함유하지 않는 벡터 및 모크(mock) 트랜스펙션이 대조군들로 포함되었다. 모든 샘플들은 기술적 중복들(technical duplicates)로 실행(run)되었다. 4 시간 후 세포들은 Opti-MEM™ 배지에서 세척되었고 신선한 완전 세포 배양 배지 배지가 웰들에 첨가되었다.

결과들은 백그라운드 발광을 빼고(subtraction) 그 다음에 구성적(constitutive) 반딧불이(Firefly) 신호로 레닐라 신호를 정규화(normalizing)하여 분석되었다. 그 다음 두 기술 중복들(technical duplicates)의 평균이 빈 벡터(empty vector)로 정규화되고(normalized) 퍼센티지로 표현되었다. 결과들은 Graphpad Prism (version 9.0.2, GraphPad Software)에서 시각화되었다(visualized).
모든 17개 안티센스 올리고뉴클레오타이드들에 대한 반딧불이 루시페라제 활성에 대해 정규화된 레닐라 루시페라제 활성의 억제 해제 수준들이 도 1에 보여진다.
antimiR-27b 안티센스 올리고뉴클레오타이드들의 전체 라이브러리로부터 추가 분석들 및 IC50 측정들(determinations)을 위해 다섯 개의 가장 강력한 antimiR-27b 분자들이 선택되었다.
실시예 3: 배양된 세포주들에서 antimiR-27b 올리고뉴클레오타이드들에 대한 IC50의 측정
miR-27b를 억제하는 안티센스 올리고뉴클레오타이드들의 효능을 측정하기(determine) 위해, IC50 측정들이 수행되었다. 루시페라제 분석들이 실시예 2에 서술된 대로 수행되었다. 안티센스 올리고뉴클레오타이드들은 Xu et al (Oncotarget. 2017 Sep 19; 8(41): 70669-70684)로부터 miR-27b 안타고미르(antagomir)와 비교되었다. IC50 값들의 측정(determination)을 위하여, 2-배 희석(2-fold dilutions)에서 80 nM부터 0.0049 nM까지의 범위인 광범위한 antimiR-27b 농도들로 세포들이 트랜스펙트되었다. 레닐라 루시페라제 활성은 반딧불이 루시페라제 활성으로 정규화되었고 Graphpad Prism (version 9.0.2, GraphPad Software)에서 log(M)에 대해(against) 플롯팅되었다(plotted). 도즈-반응 곡선들은 3-파라미터 비-선형 피트(fit) 및 nM으로 계산된 IC50 값들을 사용하여 피트(fitted)되었다. 선택된 농도들 전체에 걸쳐 낮은 반응 때문에 antagomiR 대조군(control) 화합물에 대한 IC50 값을 측정하는(determine) 것은 가능하지 않았다.
도 2는 다섯 개의 antimiR-27b 올리고뉴클레오타이드들의 IC50 값들 및 도즈-반응 곡선들을 보여준다.
실시예 4: 배양된 U-87 Mg 세포들에서 IC50 측정(determination)
U-87 Mg 세포들에서 IC50 곡선들은 Lipofectamine 2000의 양이 웰 당 0.4 μL이었고 트랜스펙션들이 96-웰 Costar 블랙 플레이트들(black plates) (cat. no: 3603, Corning World, Corning, NY, USA)에서 수행되었다는 것을 제외하고는, 상기 실시예들에서 서술된 PC-12 Adh 세포들에서와 같이 수행되었다.
도 3은 다섯 개의 선택된 antimiR-27b 올리고뉴클레오타이드들 (seq id no's: 8, 12, 16, 19 및 20)의 IC50 값들 및 도즈 반응 곡선들을 보여준다.
실시예 5는 배양된 PC-12 Adh 세포주에서 miR-27b 타겟 mRNA 억제 해제(derepression)를 보여준다.
miRNA들은 그것들의 타겟 mRNA들의 수준들을 부정적으로(negatively) 조절하므로, antimiR 올리고뉴클레오타이드들에 의한 miR-27b 억제의 기능적 효과들은 타겟 mRNA들의 그 다음의(subsequent) 상향 조절에 의해 측정될 수 있다. miR-27b의 주요(principal) 타겟은 전사 인자 Nrf2이다; Hmox1 및 Nqo1과 같은 항산화(antioxidant) 및 해독(detoxifying) 인자들의 상향 조절을 담당한다(responsible). 이들 세 마커들에서 상향 조절은 Nrf2 수준들에 대한 기능적 효과를 단지 의미할(signify) 뿐 아니라 Nrf2에 의해 조절되는 다운-스트림 분자적 경로들의 활성화를 보여준다.
루시페라제 리포터가 사용되지 않았고 웰 당 6 μL Lipofectamine2000을 사용하여, 3x105 세포들/웰에서 12-웰 CellBind 플레이트들(plates) (cat. no: CLS3336, Corning World, Corning, NY, USA)에서 세포들이 시딩되었다는 것을 제외하고 상기 실시예들에 서술된 것과 같이 PC-12 Adh 세포들이 트랜스펙트되었다. 직접 현미경 검사(microscopy)로 검사하여 트랜스펙션 효율을 확인하기 위해 별개의 웰에서 FAM-표지된 올리고뉴클레오타이드들이 트랜스펙트되었다. 트랜스펙션 사십-팔 시간 후, 제조업자의 지침에 따라 miRNeasy 미니 키트 (cat. no: 217004, Qiagen, Hilden, Germany)을 사용하여 RNA 추출(extraction)이 수행되었다. RNA 는 추가 분석까지 -80℃에서 보관되었다. 랜덤 헥사머(hexamer) 프라이머 (cat. no: SO142, Thermo Fischer Scientific, Waltham, MA, USA)를 사용하여 그리고 ezDNase™ (cat. no: 11766051, Thermo Fischer Scientific, Waltham, MA, USA)에 의해 gDNA 제거(removal)을 포함하는, 제조업자의 지침에 따라 Superscript IV 역전사효소(reverse transcriptase) (cat. no: 18090010, Thermo Fischer Scientific, Waltham, MA, USA)를 사용하여 역전사가 수행되었다. qPCR이 제조업자의 지침들에 따라 통합된(Integrated) DNA Technologies (Newark, NJ, USA) 및 TaqMan™ Universal Master Mix II, no UNG (cat. no: 4440040, Thermo Fischer Scientific, Waltham, MA, USA)에 의해 합성된 Taqman 분석들 (표 2)를 사용하여 QuantStudio 6 Flex (Applied Biosystems, Waltham, MA, USA)에서 수행되었다. 모든 qPCR 분석들은 엑손-스패닝(exon-spanning)으로 설계되었으며 특이성은 프라이머들의 블라스트(blast)에 의해 확인되었고 프라이머들의 효율은 다섯-배 희석(five-fold dilution) 시리즈를 사용하여 테스트되었다. 도 4에서 막대(bar) 다이어그램은 miR-27b 타겟 유전자 억제 해제 (Nrf2, Hmox1 and Nqo1)에 대한 antimiRs (Seq ID 8, 12, 16, 19 및 20 )의 효과를 보여준다. 이 실험에서 사용된 Seq ID 20으로 정의된 올리고뉴클레오타이드는 그것이 Seq ID 22에 대응하도록 만들기 위해 하나의 구아닌이 이노신 치환되었다(substituted).

세포들이 콜라겐 (Sigma-Aldrich cat. no. C8919)으로 전처리된 투명한 바닥의(clear-bottom), 흰색 96-웰 플레이트들 (cat.no 3610, Corning)에서 시딩되었고 IC50 실험을 위해 세 개의 기술적 복제들(technical replicates)이 사용되었고 백그라운드 빼기(substraction)를 수행하지 않았다는 점을 제외하고, 실시예들 2 및 3에서 서술된 대로 트랜스펙션(transfection) 및 루시페라제 분석이 수행되었다. 도즈-반응 실험 및 IC50 실험의 결과들이 각각 도 5 A 및 B에 보여진다.
구현예들(Embodiments)
