실시예
인간 유도 만능 줄기 세포(iPSC)의 CISH 유전자는 CRISPR/Cas9 시스템을 사용하여 파괴되었고 CISH-/- iPSC로부터의 NK 세포는 2 단계 생체외 분화 프로토콜을 사용하여 유도되었다. 조혈 전구 세포로의 분화의 제1 단계는 WT 또는 CISH-/- iPSC를 사용한 정상(80% 초과 CD34+ 세포) 이었다. iPSC에서 CISH의 결실은 생체외 NK 세포 분화의 제2 단계를 지연시켰다(도 1 및 도 2). 특히, NK 세포 분화는 전형적으로 WT iPSC를 사용한 4주 후 90% 초과 NK 세포로 완전히 완료되는 반면, CISH-/- iPSC-유도 세포는, 5주에는 80% 초과 NK 세포였지만, 4주에는 10% CD45+CD56+ NK 세포만 생성했다. 이 시간 이후, CISH-/- iPSC-유도 NK 세포는 표현형적으로 성숙되었고 CD94, CD16, NKG2D, NKp44, NKp46과 같은 전형적인 NK 표지 마커(surface maker) 발현을 보였다.

생성된 CISH-/- 인간 iPSC-NK 세포는 IL-2/IL-15 자극에 과민 반응 및 저농도의 IL-2(5 U/ml) 및 IL-15(5 ng/ml)로 확장 및 항종양 기능을 유지하는 능력을 나타냈다(도 3a 내지 3b). CISH -/- IPSC-NK 세포는 생체외에서 3주를 초과하여 저농도의 IL-2(5 U/ml) 및 IL-15(5 ng/ml)로 확장 및 세포 독성 기능을 유지할 수 있다.

Claims (20)
