안정성이 확인된 피부 세포주를 대상으로, 장기산딸기 열수추출물을 농도별로 처리한 후, 세포 독성 테스트(MTT assay)를 통해 세포 사멸 정도를 측정하여 독성을 확인하였다. 세포 독성 테스트(MTT assay)는 원래 노란색을 띠는 MTT 포마잔이 살아있는 세포의 미토콘드리아에서 환원되어 보라색으로 그 색이 변화하게 되는 원리를 이용한 것이다. 세포 독성으로 인해 세포 사멸이 일어나는 정도에 따라 보라색으로 변하는 양이 줄어들기 때문에 흡광도 측정을 통해 쉽게 독성을 평가할 수 있다. 세포독성평가를 통해 장기산딸기 열수추출물 1 ~ 200ppm 처리 시, 95% 이상 세포생존율을 확인하였으며, 400ppm 농도로 추출물을 처리하였을 때 약 17% 가량 세포사멸되었음을 〈그림 1〉과 같이 확인하였다. 본 발명의 산딸기 바디 스크럽 조성물로 조성성분으로 포함되어 있는 산딸기 추출물은 200ppm 농도에서 추출한 추출물을 사용하였다.For the skin cell line whose stability was confirmed, the long-term raspberry hot water extract was treated by concentration, and then the degree of cell death was measured through a cytotoxicity test (MTT assay) to confirm toxicity. 타이로시나아제 활성저해시험을 통해 장기산딸기 열수추출물 1 ~ 400ppm 처리 시, 1 ~ 200ppm 농도에서 농도 의존적으로 미백 활성이 증가함을 확인하였으며, 400ppm 농도로 추출물을 처리하였을 때 약 21% 가량 미백 활성을 나타냄을 확인하였다. PPE (Porcine Pancreatic Elastase) Assay 시험을 통해 장기산딸기 열수추출물 1 ~ 400ppm 처리 시, 1 ~ 400ppm 농도에서 농도 의존적으로 주름개선 활성이 증가함을 확인하였으며, 400ppm 농도로 추출물을 처리하였을 때 약 52% 가량 주름개선 활성을 나타냄을 확인하였다.
