Abstract
본 발명은 트롤록스(Trolox)와 펩타이드가 화학적으로 결합된 구조를 갖는 트롤록스-펩타이드 결합체에 관한 것으로, 트롤록스-펩타이드 결합체는 5α-환원효소(5α-reductase)의 활성을 매우 효과적으로 억제할 뿐만 아니라, 모유두 세포 및 각질세포의 사멸을 억제하고 성장을 촉진함과 동시에 항산화 효과를 가지는바, 탈모를 방지 또는 예방함과 동시에 모발의 생성을 현저하게 촉진시킬 수 있다.
Description
본 발명은 트롤록스(Trolox)와 펩타이드가 화학적으로 결합된 구조를 갖는 트롤록스-펩타이드 결합체 및 그의 용도에 관한 것이다.
모낭은 포유동물의 피부에 존재하는 독특한 기관으로서, 원시 표피의 하부가 성장하여 보다 깊은 피부 층으로 신장된 기관이다. 모낭의 기부에는 소낭 또는 진피 유두 세포로서 알려진 세포의 플러그가 존재하며, 진피 유두 세포는 모낭의 정상적인 순환과 모간의 성장에 필수적이다. 모발 중에서도 특히 두피 위로 솟아있는 줄기인 모간은 케라틴 필라멘트와 필라멘트 응집 단백질로 충전된 단단하게 밀착된 상피 세포로 제조된 트레드 형상의 구조로 이루어져 있다.
인간의 모발의 주기는 모발을 성장시키는 성장기(anagen), 성장을 종료하고 모구부가 축소되는 시기인 퇴화기(catagen), 모유두가 활동을 멈추고 모발을 두피에 머무르게 하는 시기인 휴지기(talogen) 및 모유두가 활동을 시작하거나, 또는 새로운 모발을 발생시켜 오래된 모발의 탈모를 유도하는 시기인 발생기로 나뉜다. 상기 성장기는 모발이 성장하는 기간으로, 다시 모발이 모구로부터 모포로 나가려는 모발 생성단계와 딱딱한 케라틴이 모낭 안에서 만들어지는 단계로 세분화될 수 있으며, 이 시기에서 모발은 퇴행기가 될 때까지 지속적으로 성장을 계속한다. 상기 퇴화기는 성장이 끝나고, 모발의 형태를 유지하면서 대사과정이 느려지는 시기로, 이 시기에서는 케라틴이 만들어지지 않는다. 퇴화기에 해당하는 모발은 전체 모발 중 1%를 차지하며, 세포 분열은 정지해 있는 상태에 해당한다. 상기 휴지기는 모유두가 위축되고 모낭이 차츰 쪼그라들며 모근이 위쪽으로 밀려 올라가 빠지는 시기에 해당한다.
이와 같은 모발은 개체가 생명을 유지하는데 필수적인 기관은 아니지만, 건강의 상태를 나타내는 척도이자 외모를 결정짓는 신체의 중요한 일부분이다. 따라서 모발이 많은 일반인에게는 일상적인 탈모가 당연한 신체적 활동으로 여겨 지지만, 탈모가 진행 중인 사람들에게는 우울, 수치심, 사회적 고립 등으로 인해 정신건강과 삶의 질에 심각한 영향을 줄 수 있는 요인에 해당한다.
최근 탈모에 영향을 주는 요인으로는 기후, 빛 또는 열에 의한 노출 등의 환경적인 요인과 심한 스트레스, 질병, 출산, 호르몬 분비 및 변화, 약물의 복용, 영양상태 등의 내적인 요인이 있다. 그러나, 현재까지 탈모에 관한 명확한 원인은 밝혀져 있지 않으며, 최근 들어 식생활의 변화 또는 사회환경 등에 의한 스트레스의 증가 등으로 탈모로 고민하는 인구가 늘어나고 있고 그 연령 또한 낮아지고 있으며, 여성의 탈모 인구 역시 증가하고 있는 추세이다.
탈모 메커니즘에 관여하는 주요한 호르몬으로 5-알파 환원효소(5-α reductase)를 들 수 있다. 상기 5-알파 환원효소는 남성호르몬(androgen)의 일종인 테스토스테론(testosterone)을 DHT(dihydrotestosterone)로 변환시키고, 이렇게 생성된 DHT는 모발의 생장기를 짧게 하고, 휴지기를 길게 할 뿐만 아니라, 모낭 세포 사멸을 유도하는 세포신호전달에 관여하여 모낭 세포를 사멸시키는 것으로 보고되어 있다.
이에, 탈모의 원인을 치유하기 위하여, 머크사는 5-알파 환원효소의 활성을 억제함으로써 DHT의 생성을 억제하는 피나스테라이드를 개발하여 시판하고 있다(특허문헌 1). 상기 피나스테라이드는 복용이 편리하고, 효능이 매우 우수하지만, 이를 제조하는데 사용되는 시약이 고가이거나 독성이 존재하여 제조원가에 부담을 줄 수 있다. 특히, 특히 부수적으로 생성되는 생성물의 제거가 용이하지 않아 최종 생성물의 순도가 떨어질 수 있으며, 수분에 의해 활성이 저해되기 쉬운 시약이나 활성 유도체를 사용하여야 하기 때문에 대량 생산이 어렵다는 문제점이 존재한다. 이 외에도 각질 세포 또는 혈관 내피의 성장을 촉진시키거나, BMP에 속하는 단백질의 활성을 억제시킴으로써 모발 생성이 촉진되도록 하는 연구가 진행되고 있다(특허문헌 2). 그러나, 성장인자를 유효성분으로 포함하는 치료제는 효능이 매우 뛰어난 반면, 자연형의 성장인자를 얻기 위하여 추가 공정과 시간이 요구되고, 또한 정제 과정에서 대장균 유래의 오염원을 제거하기 위한 복잡한 정제 과정을 필요로 하게 될 뿐만 아니라, 분자량이 커 모발의 보호막을 쉽게 뛰어넘지 못한다는 등의 문제점이 존재한다.
따라서, 상기와 같은 문제점들을 해결하기 위하여 많은 연구가 진행되어 탈모를 방지 및 치료하기 위한 발모제들이 제시되었으나, 기존 발모제들은 탈모를 방지함과 동시에 발모를 촉진하는 기능을 갖지 못하기 때문에 실질적으로 탈모 치료 및 발모 촉진에 큰 효과를 나타내지 못하고 있는 실정이다.
본 발명은 탈모 억제 또는 발모 촉진 효과가 매우 우수한 트롤록스-펩타이드 결합체를 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위하여, 본 발명의 일 측면은 트롤록스(Trolox)와 펩타이드가 화학적으로 결합된 구조를 갖는 트롤록스-펩타이드 결합체를 제공하는 것이다.
아울러 본 발명의 다른 측면은 트롤록스-펩타이드 결합체를 유효성분으로 포함하는 약학적 조성물 및 화장료 조성물을 제공하는 것이다.
본 발명에 따르면, 트롤록스-펩타이드 결합체는 5α-환원효소(5α-reductase)의 활성을 매우 효과적으로 억제할 뿐만 아니라, 모유두 세포 및 각질세포의 사멸을 억제하고 성장을 촉진함과 동시에 항산화 효과를 가지는바, 탈모를 방지 또는 예방함과 동시에 모발의 생성을 현저하게 촉진시킬 수 있다.
다만, 본 발명의 효과는 상기에서 언급한 효과로 제한되지 아니하며, 언급되지 않은 또 다른 효과들은 하기의 기재로부터 당업자에게 명확히 이해될 수 있을 것이다.
도 1 내지 도 3은 본 발명의 서열번호 1의 펩타이드와 트롤록스의 결합체(서열 1의 결합체), 서열번호 2의 펩타이드와 트롤록스의 결합체(서열 2의 결합체) 및 서열번호 3의 펩타이드와 트롤록스의 결합체(서열 3의 결합체)에 대한 MASS 분석 데이터를 나타낸 것이다.

이하, 본 발명을 상세히 설명한다.
1.
트롤록스-펩타이드 결합체
본 발명의 일 측면은 트롤록스(Trolox)-펩타이드 결합체를 제공한다.
본 발명의 상기 트롤록스는 6-하이드록시-2,5,7,8-테트라메틸크로만-2-카르복실산으로, 하기 화학식 1로 표현되는 구조를 갖는다.

본 발명의 상기 결합체는 트롤록스와 펩타이드가 화학적으로 결합된 구조를 갖는다.
상기와 같이 트롤록스에 펩타이드가 결합됨으로써, 물에 대한 용해도가 낮은 트롤록스의 물에 대한 용해도를 현저하게 높일 수 있을 뿐만 아니라, 5α-환원효소(5α-reductase)의 활성을 매우 효과적으로 억제하고, 모유두 세포 및 각질세포의 사멸을 억제하고 성장을 촉진하는바, 탈모를 방지 또는 예방함과 동시에 모발의 생성을 촉진하는 효과가 부여될 수 있다.
상기 펩타이드는 펩타이드 결합에 의하여 아미노산이 서로 결합되어 형성된 선형의 분자를 의미한다. 상기 펩타이드의 제작은 본 기술분야에 공지된 통상의 생물학적 또는 화학적 합성 방법에 의해 달성될 수 있는 것이고, 일 예로 고상 합성 기술(solid-phase synthesis techniques)과 같은 방법에 의해 달성될 수 있다.
상기 펩타이드는 5개 내지 30개, 바람직하게는 8개 내지 20개, 더욱 바람직하게는 10개 내지 15개의 아미노산으로 이루어진 것일 수 있다. 펩타이드를 이루는 아미노산의 개수가 5개 미만인 경우, 트롤록스의 물에 대한 용해도를 현저하게 높일 수 없고, 펩타이드를 이루는 아미노산의 개수가 30개 초과인 경우 펩타이드의 크기가 과도하게 크기 때문에 목적하는 약리 효과를 이루기 위한 흡수를 저하시키는 문제점이 발생할 수 있다.
상기 펩타이드는 친수성을 나타낼 수 있는 측쇄(side chain)를 가지는 아미노산의 비율이 50% 내지 100%, 예를 들면 80% 내지 100%일 수 있고, 상기 펩타이드는 소수성을 나타낼 수 있는 측쇄를 가지는 아미노산의 비율이 0% 내지 50%, 예를 들면 0% 내지 20%일 수 있다. 친수성을 나타낼 수 있는 측쇄를 가지는 아미노산의 비율이 상기 범위를 벗어나는 경우에는 트롤록스의 물에 대한 용해도가 현저하게 높아지는 효과가 발휘될 수 없다.
상기 친수성을 나타낼 수 있는 측쇄를 가지는 아미노산은 아르기닌(Arg), 히스티딘(His), 리신(Lys), 아스파라긴산(Asp), 글루탐산(Glu), 세린(Ser), 트레오닌(Thr), 아스파라긴(Asn), 글루타민(Gln), 시스테인(Cys), 셀레노시스테인(Sec), 글리신(Gly) 및 프롤린(Pro)으로 이루어진 군으로부터 선택되고, 상기 소수성을 나타낼 수 있는 곁사슬을 갖는 아미노산은 알라닌(Ala), 발린(Val), 이소루이신(Ile), 루이신(Leu), 메티오닌(Met), 페닐알라닌(Phe), 티로신(Tyr) 및 트립토판(Trp)으로 이루어진 군으로부터 선택되는 것일 수 있다.
상기 펩타이드는 펩타이드 결합에 의하여 적어도 50% 이상의 친수성을 나타낼 수 있는 측쇄를 가진 아미노산이 서로 결합되어 형성된 것으로, 수용성 펩타이드일 수 있다.
상기 펩타이드는 서열번호 1 내지 서열번호 3 중 어느 하나의 아미노산 서열을 포함한다.
상기 서열번호 1의 아미노산 서열로 구성된 펩타이드는 탈모 유발에 관여하는 유전자 DKK1, BMP4 및 TGF-β의 발현을 억제하여 탈모를 방지할 수 있다.
상기 서열번호 2의 아미노산 서열로 구성된 펩타이드는 모발의 주기를 권장하는 성장인자와 유사한 기능을 갖는 펩타이드로서, 상기 펩타이드에 의해 β-카테닌의 핵 내 전위가 유도되어 최종적으로 모세포가 새롭게 성장될 수 있도록 할 뿐만 아니라, 남성 호르몬으로 인한 디하이드로테스토스테론(dihydrotestosterone, DHT)의 기능을 저하시켜 탈모를 방지하는데 기여할 수 있다.
상기 서열번호 3의 아미노산 서열로 구성된 펩타이드는 신규 혈관 형성을 유도하여 건강하고 두꺼운 모발을 유지함으로써 탈모를 방지하는데 기여할 수 있다.
본 발명의 상기 트롤록스-펩타이드 결합체는 트롤록스와 펩타이드가 결합되어, 펩타이드가 가지는 발휘되는 고유의 탈모 방지 및 발모 촉진 기능을 유지할 뿐만 아니라, 나아가 트롤록스 또는 펩타이드를 단독으로 사용한 경우에 비하여 현저하게 향상된 탈모 방지 및 발모 촉진 효과를 가질 수 있다.
상기 서열번호 1 내지 서열번호 3의 아미노산은 상기 펩타이드가 트롤록스에 결합되어 물에 대한 수용성을 증가시키는데 영향을 미치지 않는 범위 내에서, 아미노산 잔기의 결실, 삽입, 치환 또는 이들의 조합에 의해서 상이한 서열을 가지는 아미노산의 변이체들 또는 단편들일 수 있으며, 경우에 따라서는 인산화(phosphorylation), 황화(sulfation), 아크릴화(acrylation), 당화(glycosylation), 메틸화(methylation), 파네실화(farnesylation) 등으로 변형될 수 있다. 또한 상기 서열번호 1 내지 서열번호 3의 아미노산은 상기 서열번호 1 내지 서열번호 3의 아미노산 서열을 포함하는 펩타이드와 실질적으로 동일한 아미노산 서열을 갖는 펩타이드 및 이의 변이체 또는 이의 활성 단편을 포함한다.

본 발명의 상기 조성물은 상기 "1. 트롤록스 - 펩타이드 결합체 " 항목에서 설명한 트롤록스-펩타이드 결합체를 유효성분으로 포함하여 탈모 방지 및 발모 촉진의 용도로 사용할 수 있고, 이에 대해서는 상기 "1. 트롤록스 - 펩타이드 결합체 " 항목의 설명을 원용하고, 이하에서는 탈모 방지 및 발모 촉진용 조성물의 특유한 구성에 대해서만 설명하도록 한다.
상기 "탈모 방지"는 모발이 빠지거나, 감소되는 것을 억제하는 것을 포함하는 의미이고, "발모 촉진"은 모발의 생장을 촉진하는 것을 의미한다.
상기 조성물은 약학적 조성물 또는 화장료 조성물의 형태로 제공될 수 있으나, 이에 제한되는 것은 아니다.

상기 약학적 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구투여(예를 들어, 근육 내, 정맥 내, 복강 내, 피하, 피내, 또는 국소에 적용)할 수 있으며, 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 시간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다.
본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에 있어서 '약학적으로 유효한 양'은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 상기 약학적 조성물은 개별 치료제로 투여하거나 다른 비만 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 동시에, 별도로, 또는 순차적으로 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.

상기 화장료 조성물은 본 기술분야에서 통상적으로 제조되는 임의의 제형으로 제조될 수 있고, 예를 들면, 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클린싱, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이 등으로 제형화될 수 있으나 이에 제한되는 것은 아니다.
상기 화장료 조성물은 유연 화장수, 영양 화장수, 영양 크림, 마사지 크림, 에센스, 아이 크림, 클렌징 크림, 클렌징 폼, 클렌징 워터, 팩, 스프레이, 파우더, 헤어토닉, 헤어크림, 헤어로션, 헤어샴푸, 헤어린스, 헤어컨디셔너, 헤어스프레이, 헤어 에어졸, 포마드, 젤 등과 같은 용액, 솔젤, 에멀젼, 오일, 왁스, 에어졸 등의 다양한 형태로 제조될 수 있으나, 이에 제한되는 것은 아니다.
상기 화장료 조성물의 제형의 형태가 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀루로오스 유도체, 폴리에틸렌글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
상기 화장료 조성물의 제형의 형태가 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토오스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 사용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진체가 포함될 수 있으나 이에 제한되는 것은 아니다.
상기 화장료 조성물의 제형의 형태가 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용될 수 있고, 예를 들면 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌글리콜 또는 소르비탄의 지방산 에스테르 등이 이용될 수 있다.

상기 화장료 조성물의 제형이 계면-활성제 함유 클렌징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르 설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성유, 라놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
상기 화장료 조성물의 제형이 헤어샴푸인 경우에는 본 발명의 트롤록스-펩타이드 결합체에 증점제, 계면활성제, 점도 조절제, 보습제, pH 조절제, 방부제, 에센셜 오일 등과 같이 샴푸를 조성하기 위한 베이스 성분들이 혼합될 수 있다. 상기 증점제로는 CDE가 사용될 수 있고, 상기 계면활성제로는 음이온 계면활성제인 LES와, 양쪽성 계면활성제인 코코 베타인이 사용될 수 있고, 상기 점도조절제로는 폴리 쿼터가 사용될 수 있으며, 상기 보습제로는 글리세린이 사용될 수 있고, 상기 pH 조절제로는 구연산, 수산화나트륨이 사용될 수 있다. 상기 방부제로는 자몽 추출물 등이 사용될 수 있고, 이 외에도 시더우드, 페퍼민트, 로즈마리 등의 에센셜 오일과 실크 아미노산, 펜타올 또는 비타민 E가 첨가될 수 있다.
상기 화장료 조성물에 포함되는 성분은 유효 성분으로서 본 발명의 상기 트롤록스-펩타이드 결합체와 담체 성분 이외에 화장료 조성물에 통상적으로 사용되는 성분들, 예를 들면 항산화제, 안정화제, 용해화제, 비타민, 안료 및 향료와 같은 통상적인 보조제 등을 더 포함할 수 있으나, 이에 제한되는 것은 아니다.
이하, 본 발명을 실시예 및 실험예에 의하여 상세히 설명한다.
단, 하기 실시예 및 실험예는 본 발명을 구체적으로 예시하는 것이며, 본 발명의 내용이 하기 실시예 및 실험예에 의해 한정되지 아니한다.
[제조예 1]
펩타이드 합성
[1-1]
서열번호 2의 아미노산 서열을 포함하는 펩타이드 합성
700㎎의 클로로트리틸클로라이드 수지(chloro trityl chloride resin; CTL 수지, Nova biochem [0064] Cat No. 01-64-0021)를 반응 용기에 넣고, 상기 반응 용기에 10㎖의 메틸렌클로라이드(methylene chloride)를 넣고 3분 동안 교반하였다. 그런 다음, 용매를 제거하고 10㎖의 디메틸포름아마이드(dimethylformamide)를 넣고, 3분 동안 추가로 교반한 뒤 용매를 제거하였다. 상기 반응 용기에 10㎖의 디클로로메탄(dichloromethane)을 넣고, 200mmole의 Fmoc-Cys(trt)-OH (Bachem, Swiss)와 400mmole의 디이소프로필 에틸아민(disopropylethylamine)을 넣은 후 1시간 동안 교반하여 반응을 유도하였다. 상기 반응물을 세척한 뒤, 2:1의 비율로 혼합된 메탄올과 디이소프로필 에틸아민을 넣고 10분 동안 반응시키고, 1:1의 비율로 혼합된 디클로로메탄과 디메틸포름아마이드를 과량으로 넣어 다시 한번 세척하였다. 그런 다음, 용매를 제거하고 10㎖의 디메틸포름아마이드를 넣고 3분 동안 교반하고 용매를 제거하였다. 그런 다음, 탈보호 반응을 위하여, 10㎖의 탈보호 용액(20%의 피페리딘/디메틸포름아마이드)을 상기 반응 용기에 넣고 10분간 상온에서 교반한 후 용매를 제거하는 과정을 총 2회 수행하였다. 그 뒤, 용매를 제거하고 상기 반응물을 각각 3분 동안 디메틸포름아마이드로 2회, 메틸렌클로라이드로 2회, 디메틸포름아마이드로 1회 세척 하여 Cys(trt)-CTL 수지를 제조하였다.
새로운 반응 용기에 10㎖의 DMF 용액을 넣고 200mmole 의 Fmoc-His(trt)-OH(Bachem, Swiss), 200mmole의 HOBt(hydroxybenzotriazole) 및 200mmole의 Bop(benzotriazol-1-yloxytris(dimethylamino)phosphonium hexafluorophosphate)를 넣은 후 교반하였다. 상기 반응 용기에 400mmole의 디이소프로필 에틸아민을 2번에 걸쳐 나누어 넣은 후, 고체 상태의 반응물이 모두 용해될 때까지 최소 5분 이상 교반하였다. 그런 다음 상기 용액을 Cys(trt)-CTL 수지가 포함된 상기 반응 용기에 넣고, 1시간 동안 상온에서 교반한 뒤, 용매를 모두 제거하고 디메틸포름아마이드로 5분씩 3회 세척하고, 카이저 테스트(kaiser test, nihydrin test)를 통해 상기 반응물의 반응 정도를 점검하였다. 그런 다음, 상기 반응물을 상기 Cys(trt)-CTL 수지의 제조 과정에서 수행한 탈보호 반응을 수행하여 His(trt)-Cys(trt)-CTL 수지를 제조하였다. 상기 수지는 디메틸포름아마이드 및 메틸렌클로라이드로 충분히 세척하고 카이저 테스트를 통해 반응 정도를 점검하였다.
그런 다음, Fmoc-Cys(trt), Fmoc-Arg, Fmoc-Gln(trt), Fmoc-Val, Fmoc-Arg, Fmoc-Thr, Fmoc-Gln(trt) 및 Fmoc-Arg(pbf) 순서로 상기 His(trt)-Cys(trt)-CTL 수지와 연쇄반응을 수행하였다. 그 뒤, 탈보호 용액으로 10분씩 2번 반응시킨 후 잘 세척하여 Fmoc-보호기를 제거하였다. 상기 탈보호된 수지와 무수 아세트산, 디이소프로필 에틸아민 및 HOBt를 한 시간 동안 반응시켜 아세틸화 반응을 수행하였다. 그런 다음, 디메틸포름아마이드, 메틸렌클로라이드 및 메탄올로 각각 3번을 세척하고, 질소 공기를 천천히 흘려 건조시킨 뒤, P2O5하에서 진공으로 감압하여 완전히 건조시켰다.
상기 건조된 반응물에 30㎖의 탈루 용액(트리플루오로아세트산(trifluoroacetic acid, TFA) 95%, 증류수 2.5%, 티오아니졸(Thioanisole) 2.5%)을 넣은 후 상온에서 2시간 동안 교반하여 반응시켰다. 상기 반응이 완료된 수지를 필터를 이용하여 여과한 뒤, 소량의 트리플루오로아세트산으로 세척하고, 세척액을 모액과 혼합하였다. 상기 혼합 용액을 감압하여, 전체 부피가 절반 정도 남도록 증류한 뒤, 50㎖의 차가운 에테르를 가하여 침전을 유도하고, 원심분리기를 이용하여 침전물을 수득한 뒤 차가운 에테르를 이용하여 2번 세척하였다. 그런 다음, 모액을 제거하고 질소 하에서 충분히 건조하여 정제 전 NH2-Arg-Gln-Thr-Arg-Val-Gln-Arg-Cys-His-Cys-OH 펩타이딜 수지(서열번호 2)를 0.65 g 합성하였다(수율: 92.6%). 분자량 측정기를 이용하여 상기 서열번호 2의 아미노산 서열을 포함하는 펩타이드의 분자량을 측정한 결과, 그 분자량이 1287.1Da (이론값: 1286.5Da)에 해당함을 확인하였다.

상기 [1-1]의 합성 방법과 동일한 방법을 사용하여 서열번호 1 및 서열번호 3의 아미노산 서열을 포함하는 펩타이드를 합성하였다. 이때, Fmoc-Asp(OtBu), Fmoc-Ala, Fmoc-Pro, Fmoc-Arg (Pbf), Fmoc-Gly, Fmoc-Gly, Fmoc-Gly, Fmoc-His(Trt), Fmoc-Glu(OtBu), Fmoc-Ile, Fmoc-Leu 및 Fmoc-Glu(OtBu) 순서로 상기 CTL 수지와 연쇄반응을 수행하여 서열번호 1의 아미노산 서열을 포함하는 펩타이드를 합성하였으며, Fmoc-His(Trt), Fmoc-Thr(tBu), Fmoc-Trp, Fmoc-Gly, Fmoc-Gly, Fmoc-Lys(Boc), Fmoc-Lys(Boc), Fmoc-Ser(tBu), Fmoc-Lys(Boc) 및 Fmoc-Tyr(tBu) 순서로 상기 CTL 수지와 연쇄반응을 수행한 후 아세틸화하여 서열번호 3의 아미노산 서열을 포함하는 펩타이딜 수지를 합성하였다.

결합체의 제조
펩타이드 반응기에 1mole의 펩타이딜 수지와 10㎖의 1-메틸-2-피롤리돈(NMP)을 넣고, 270mg(2.0 equiv.)의 1-히드록시벤조트리아졸(1- hydroxybenzotriazole, 1-HOBt) 과 759 mg (2.0 equiv.)의 N,N,N',N'-테트라메일-O-(1H-벤조트리아졸-1-일)우로니움 헥사플루오로포스페이트(N,N,N′,N′-Tetramethyl-O-(1H-benzotriazol-1-yl)uronium hexafluorophosphate, HBTU)를 첨가하고 30분 동안 반응시켰다. 그런 다음, 상기 반응물에 388mg (3.0 equiv.)의 N,N'-디이소프로필에틸아민(N,N'-Diisopropylethylamine, DIEA)과 500mg (2.0 equiv.)의 트롤록스(trolox)를 첨가하여 상온에서 72시간 동안 반응시킨 뒤, 여과하였다. 그런 다음, 상기 여과물과 절단 용액(cleavage solution)을 상온에서 2시간 동안 반응시켜, 레진 및 보호기를 제거하였다. 마지막으로 10㎖의 디에틸에테르를 넣어 결정화시켜 트롤록스-펩타이드 결합체(즉, 서열번호 1, 서열번호 2, 또는 서열번호 3의 펩타이드와 트롤록스의 결합체)를 제조하였다. 트롤록스-펩타이드 결합체를 제조하기 위한 반응식은 하기 반응식 1과 같다.

생성된 3종의 트록록스-펩타이드 결합체에 대한 MASS 분석 데이터를 각각 도 1 내지 도 3에 나타내었다. 상기 MASS 분석 데이터에서, 각각 분자량 1550.7 Da, 1487.62 Da, 및 1423.6 Da에 해당하는 서열 1 내지 서열 3의 트롤록스-펩타이드 결합체가 합성되었음을 명확하게 확인할 수 있다.
[실시예 1]
증식 촉진 효과 확인
HFDPC(human hair follicle dermal papilla cell) 및 인간 각질형성세포(keratinocyte)인 HaCaT를 96웰 플레이트의 각 웰에 2×103 개씩 분주하고 밤새(overnight) 배양하였다. 상기 배양된 세포들의 배양액을 혈청이 포함되지 않은 배지(serum free media)로 교체한 뒤, 0.5μM, 5μM 또는 50μM의 트롤록스-펩타이드 결합체들을 각각 처리하고, 3일 동안 추가로 배양하였다. 그런 다음, 세포의 증식을 확인하기 위하여 4mg/ml의 MTT(3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide)를 웰에 각각 100㎕씩 넣고, 4시간 동안 반응시켜 생성된 포마잔을 DMSO를 처리하여 녹인 뒤, 마이크로플레이트 리더기를 사용하여 560nm에서 흡광도를 측정하였다. 이때, 양성 대조군으로는 HFDPC 및 HaCaT 세포의 증식을 촉진하는 것으로 알려져 있는 성장인자인 1μM의 IGF-1 또는 EGF를 사용하였고, 비교군으로는 본 발명의 트롤록스-펩타이드 결합체와 동일한 함량의 트롤록스 및 서열번호 1 내지 서열번호 3의 아미노산 서열을 갖는 펩타이드 단독 화합물을 사용하였다.
그 결과, 도 4 및 도 5에 도시된 바와 같이, HFDPC 및 HaCaT 세포에 트롤록스-펩타이드 결합체들을 처리한 경우에, 모두 그 세포 증식이 증가되는 것으로 확인되었고, 이러한 세포 증식이 증가되는 효과는 처리되는 트롤록스-펩타이드 결합체의 농도가 커질수록 더욱 증가되는 것으로 확인되었다. 특히, HaCaT에 50μM의 트롤록스-펩타이드(서열번호 2) 결합체를 처리한 경우에는 양성 대조군인 EGF 만큼, 세포 증식이 현저하게 증가되는 것으로 확인되었다. 이와 대조적으로 비교군으로 사용된 트롤록스 화합물은 세포 증식 효과가 거의 나타나지 않았고, 서열번호 1 내지 서열번호 3의 아미노산 서열을 갖는 펩타이드 화합물들은 세포 증식 효과가 미약하거나, 동일한 함량의 트롤록스-펩타이드 결합체보다 세포 증식 효과가 낮은 것으로 나타났다.
상기와 같은 결과로부터, 트롤록스-펩타이드 결합체를 처리하는 경우 HFDPC 및 HaCaT 세포의 증식을 촉진함으로써 모발 성장 촉진 효과가 발휘될 수 있음을 알 수 있다.
[실시예 2]
세포 내(intracellular) ROS(reactive oxygen species) 저해 효과 확인
HFDPC를 6웰 플레이트의 각 웰에 3×105 개씩 분주하고 밤새 배양하였다. 상기 배양된 세포에 50mJ의 UVB를 조사하고, 상기 세포들의 배양액을 혈청이 포함되지 않은 배지로 교체한 뒤, 5μM 또는 50μM의 트롤록스-펩타이드 결합체들을 각각 처리하고, 24시간 동안 추가로 배양하였다. 그런 다음, 세포 내 ROS의 농도를 측정하기 위하여 DCFH-DA(dichlorofluorescin diacetate)를 처리하고, 30분 동안 반응시킨 뒤, 세포를 회수하여 FACS를 이용하여 평균 FL1 값의 변화를 확인하였다. 이때, 양성 대조군으로 세포 내 ROS 저해 효과가 있다고 알려진 5mM의 NAC(N-acetyl-L-cysteine)를 사용하였다.
그 결과, 도 6 내지 도 8에 도시된 바와 같이, 트롤록스-펩타이드 결합체들을 처리한 경우, 양성 대조군과 유사한 정도의 수준으로 UVB 조사에 의해 증가된 세포 내 ROS가 저해되는 것으로 확인되었다.
상기와 같은 결과로부터, 트롤록스-펩타이드 결합체를 처리하는 경우 세포 내 ROS가 저해되어, 모유두 세포 내 산화적 스트레스를 현저하게 감소시킴으로써 탈모 방지 효과가 발휘될 수 있음을 알 수 있다.

5α-환원효소(5α-reductase)에 의해 테스토스테론(testosterone)으로부터 생성된 DHT(dihydrotestosterone)는 탈모를 유발하는 단백질인 DKK1의 발현이 증가되도록 유도하는 것으로 알려져 있다. 이에, 상기 트롤록스-cg-Wint(서열번호 2) 결합체에 의해 DKK1의 발현이 억제되는지 확인하였다.
HFDPC를 6웰 플레이트의 각 웰에 3×105 개씩 분주하고 밤새 배양하였다. 상기 배양된 세포들의 배양액을 혈청이 포함되지 않은 배지로 교체한 뒤, 5μM 또는 50μM의 트롤록스-펩타이드(서열번호 2) 결합체, 5α-환원효소가 포함되어 있는 마우스 간 추출물 및 테스토스테론을 처리하고 24시간 동안 추가로 배양하였다. 그런 다음, DKK-1 단백질에 특이적인 항체(santacruz biotechnology, 미국)로 웨스턴 블롯을 수행하여 단백질 발현 양상을 확인하였다.
그 결과, 도 9에 도시된 바와 같이, 테스토스테론과 마우스 간 추출물의 처리에 의하여 DKK1 단백질의 발현이 증가되었고, 트롤록스-펩타이드(서열번호 2) 결합체가 처리된 경우, 상기와 같이 증가된 DKK1 단백질의 발현이 감소된 것으로 확인되었다.
상기와 같은 결과로부터, 트롤록스-펩타이드 결합체는 5α-환원효소의 활성을 억제시켜 DHT의 생성이 억제되도록 함으로써, 궁극적으로 탈모를 유발하는 DKK1 단백질의 발현이 매우 효과적으로 억제되도록 할 수 있음을 알 수 있다.
[실시예 4]
세포 사멸 관련 단백질 발현 변화 확인
HFDPC를 6웰 플레이트의 각 웰에 3×105 개씩 분주하고 밤새 배양하였다. 상기 배양된 세포들의 배양액을 혈청이 포함되지 않은 배지로 교체한 뒤, 50μM의 트롤록스, 50μM의 트롤록스-펩타이드(서열번호 2) 결합체 또는 50μM의 트롤록스-펩타이드(서열번호 3) 결합체를 각각 처리하고, 24시간 동안 추가로 배양하였다. 그런 다음, Bcl-2 및 Bax 단백질에 특이적인 항체(santacruz biotechnology, 미국)로 웨스턴 블롯을 수행하여 단백질 발현 변화를 확인하였다.
그 결과, 도 10에 도시된 바와 같이, 트롤록스만을 처리한 경우에는 Bcl-2의 발현에 변화가 없고, Bax의 발현이 증가된 것으로 확인되었다. 반면, 트롤록스-펩타이드(서열번호 2) 결합체 및 트롤록스-펩타이드(서열번호 3) 결합체를 처리한 경우에는 Bcl-2의 발현이 증가되고, Bax의 발현이 현저하게 감소되는 것으로 확인되었다.
상기와 같은 결과로부터, 트롤록스 단독으로 사용되는 경우에는 세포 사멸이 유도될 수 없으나, 상기 트롤록스에 펩타이드가 결합되는 경우에는 세포 사멸과 관련된 단백질의 발현 변화를 유도함으로써 세포 사멸을 유도할 수 있음을 알 수 있다.
[실시예 5]
MSX2 유전자의 발현 증가 여부 확인
HHGMC(human hair germinal matrix cell)에서 상기 트롤록스-펩타이드 결합체에 의해 모낭 발달, 모발 주기의 제어 및 모간의 분화에 관여하는 MSX2 유전자의 발현이 변화되는지 확인하였다.
HHGMC를 6웰 플레이트의 각 웰에 3×105 개씩 분주하고 밤새 배양하였다. 상기 배양된 세포들의 배양액을 0.5%의 우태아혈청이 포함된 배지로 교체한 뒤, 5μM 또는 50μM의 트롤록스-펩타이드(서열번호 1) 결합체, 트롤록스-펩타이드(서열번호 2) 결합체 또는 트롤록스-펩타이드(서열번호 3) 결합체를 각각 처리하고, 24시간 동안 추가로 배양하였다. 그런 다음, 상기 세포로부터 전체 RNA를 추출하고, cDNA 합성 키트(Intron, 한국)를 이용하여 정량한 뒤 동일 양의 전체 RNA로부터 cDNA를 합성하였다. 상기 합성된 cDNA를 주형으로 하기 표 2의 프라이머를 이용하여 MSX2 유전자에 대한 PCR 반응을 수행하였다. 그 뒤, 상기 PCR 산물을 5%(w/v) 아가로오스 겔에서 전기영동시켜, MSX2 유전자의 발현 정도를 비교하였다.

그 결과, 도 11에 도시된 바와 같이, 트롤록스-펩타이드 결합체를 처리하는 경우에 MSX2 유전자의 발현이 증가되는 것으로 확인되었다.상기와 같은 결과로부터, 상기 트롤록스-펩타이드 결합체에 의해 모낭 발달, 모발 주기의 제어 및 모간의 분화에 관여하는 MSX2 유전자의 발현이 증가되어, 탈모를 방지할 수 있음을 알 수 있다.
[실시예 6]
Keratin-14 유전자의 발현 증가 확인
HaCaT 세포를 6웰 플레이트의 각 웰에 5×105 개씩 분주하고 밤새 배양하였다. 상기 배양된 세포들의 배양액을 혈청이 포함되지 않은 배지로 교체한 뒤, 5μM 또는 50μM의 트롤록스-펩타이드(서열번호 1) 결합체, 트롤록스-펩타이드(서열번호 2) 결합체 또는 트롤록스-펩타이드(서열번호 3) 결합체를 각각 처리하고, 24시간 동안 추가로 배양하였다. 그런 다음, 상기 [실시예 5]와 동일한 과정으로 Keratin-14 유전자의 발현 정도를 비교하였다.

그 결과, 도 12에 도시된 바와 같이, 트롤록스-펩타이드 결합체를 처리하는 경우에 Keratin-14 유전자의 발현이 증가되는 것으로 확인되었다. 이러한 Keratin-14 유전자의 발현이 증가되는 효과는 처리되는 트롤록스-펩타이드 결합체의 농도가 커질수록 더욱 증가되는 것으로 확인되었다.상기와 같은 결과로부터, 상기 트롤록스-펩타이드 결합체에 의해 keratin-14 유전자의 발현이 증가되어, 발모를 촉진할 수 있음을 알 수 있다.
[실시예 7]
세포신호전달 효과 확인
모낭 주기를 조절하는 신호전달은 Wnt, Shh 또는 BMP 등이 존재하는 것으로 보고되어 있다. 따라서, 상기 트롤록스-펩타이드 결합체들이 어떠한 세포신호전달에 관여함으로써, 탈모 억제(또는 방지) 또는 발모 촉진 효과를 발휘하는지 확인하였다.
HFDPC를 6웰 플레이트의 각 웰에 3×105 개씩 분주하고 밤새 배양하였다. 상기 배양된 세포들의 배양액을 혈청이 포함되지 않은 배지로 교체한 뒤 아래의 실험을 수행하였다.

BMP(bone morphogenetic protein) 2 세포신호전달 억제 효과
상기 HFDPC 세포에 50ng/㎕의 BMP-2와 5μM 또는 50μM의 트롤록스-펩타이드(서열번호 1) 결합체를 처리하고, 15분 동안 추가로 배양하였다. 그런 다음, BMP2 세포신호전달에 의해 발현이 조절되는 하위 단백질인 P-smad1/5/8 단백질에 특이적인 항체(cell signaling, 미국)로 웨스턴 블롯을 수행하여 단백질 발현 변화를 확인하였다.
그 결과, 도 13에 도시된 바와 같이, 50μM의 트롤록스-펩타이드(서열번호 1) 결합체를 처리한 경우 BMP2의 처리에 의해 증가된 P-smad 1/5/8의 단백질 발현 수준이 감소된 것으로 확인되었다.
상기와 같은 결과로부터, 상기 트롤록스-펩타이드(서열번호 1) 결합체에 의해 탈모를 유도하는 BMP2 세포신호전달을 억제되어, 효과적으로 탈모를 억제 또는 예방할 수 있음을 알 수 있다.

트롤록스-펩타이드 결합체의 세포신호전달 효과
상기 HFDPC 세포에 5μM 또는 50μM의 트롤록스-펩타이드(서열번호 2) 결합체를 처리하고, 30분 동안 추가로 배양하였다. 그런 다음, 상기 세포를 회수하고 핵에 존재하는 단백질을 분획하여 추출한 뒤, β-catenin 단백질에 특이적인 항체(santacruz, 미국)로 웨스턴 블롯을 수행하여 단백질 발현 변화를 확인하였다.
그 결과, 도 14에 도시된 바와 같이, 트롤록스-펩타이드(서열번호 2) 결합체를 처리한 경우 핵 내 존재하는 β-catenin의 양이 현저하게 증가되는 것으로 확인되었다.
상기와 같은 결과로부터, 트롤록스-펩타이드(서열번호 2) 결합체에 의해 Wnt 세포신호전달을 활성화 됨으로써 발모 촉진 효과가 발휘됨을 알 수 있다.

트롤록스-펩타이드(서열번호 3) 결합체의 세포신호전달 효과
상기 HFDPC 세포에 5μM 또는 50μM의 트롤록스-펩타이드(서열번호 3) 결합체를 처리하고, 15분 동안 추가로 배양하였다. 그런 다음, 상기 세포를 회수하고 핵에 존재하는 단백질을 분획하여 추출한 뒤, p-Erk1/2 및 p-Akt 단백질에 특이적인 항체(cell signaling, 미국)로 웨스턴 블롯을 수행하여 단백질 발현 변화를 확인하였다.
그 결과, 도 15에 도시된 바와 같이, 트롤록스-펩타이드(서열번호 3) 결합체를 처리한 경우 Erk1/2 및 Akt의 인산화가 현저하게 증가되는 것으로 확인되었다. 이러한 인산화 증가 효과는 처리되는 트롤록스-펩타이드 결합체의 농도가 커질수록 더욱 증가되는 것으로 확인되었다.
상기와 같은 결과로부터, 트롤록스-펩타이드(서열번호 3) 결합체는 Erk1/2 및 Akt의 인산화와 관련되어 있는 KGF 세포신호전달을 활성화시킴으로써 발모 촉진이 효과적으로 유도될 수 있도록 하는 것을 알 수 있다.
이상에서 본 발명은 기재된 실시예에 대해서만 상세히 설명되었지만 본 발명의 기술사상 범위 내에서 다양한 변형 및 수정이 가능함은 당업자에게 있어서 명백한 것이며, 이러한 변형 및 수정이 첨부된 청구범위에 속함은 당연한 것이다.

- 트롤록스(Trolox)와 펩타이드가 화학적으로 결합된 구조를 갖는 트롤록스-펩타이드 결합체.
- 청구항 1에 있어서,
상기 펩타이드는 펩타이드 결합에 의해 아미노산이 서로 결합되어 형성된 선형의 분자인 것인, 트롤록스-펩타이드 결합체. - 청구항 1에 있어서,
상기 펩타이드는 5개 내지 30개의 아미노산 서열로 이루어지는 것인, 트롤록스-펩타이드 결합체. - 청구항 1에 있어서,
상기 펩타이드는 친수성을 나타낼 수 있는 측쇄(side chain)를 포함하는 아미노산의 비율이 50% 내지 100% 이고, 및
상기 펩타이드는 소수성을 나타낼 수 있는 측쇄를 포함하는 아미노산의 비율이 0% 내지 50%인 것인, 트롤록스-펩타이드 결합체. - 청구항 4에 있어서,
상기 친수성을 나타낼 수 있는 측쇄를 포함하는 아미노산은 아르기닌(Arg), 히스티딘(His), 리신(Lys), 아스파라긴산(Asp), 글루탐산(Glu), 세린(Ser), 트레오닌(Thr), 아스파라긴(Asn), 글루타민(Gln), 시스테인(Cys), 셀레노시스테인(Sec), 글리신(Gly) 및 프롤린(Pro)으로 이루어진 군으로부터 선택되고,
상기 소수성을 나타낼 수 있는 측쇄를 포함하는 아미노산은 알라닌(Ala), 발린(Val), 이소루이신(Ile), 루이신(Leu), 메티오닌(Met), 페닐알라닌(Phe), 티로신(Tyr) 및 트립토판(Trp)으로 이루어진 군으로부터 선택되는 것인, 트롤록스-펩타이드 결합체. - 청구항 1에 있어서,
