Abstract
본 발명은 NK 세포(natural killer cell) 활성 조절을 통한 비만의 예방 또는 치료 방법에 관한 것으로, 본 발명에 따른 비만의 예방 또는 치료 방법은 NK 세포의 활성 조절을 통해 체중 감소를 유도함으로써, 기존 비만 치료제와 차별화된 작용 기전을 갖는 비만 치료제를 연구 및 개발하는데 유용하게 사용될 수 있다.
Description
본 발명은 NK 세포(natural killer cell) 활성 조절을 통한 비만의 예방 또는 치료 방법에 관한 것이다.
2016년 기준으로 전세계의 비만 인구는 6억 5천만명(18세 이상 성인)으로 과체중까지 포함하면 19만명에 이르며, OECD 예측에 따르면 2030년까지 우리나라의 경우도 9%(450만명)까지 비만인구가 증가할 것으로 예상된다.
비만은 다른 대사질환과 다양한 암의 원인이 되며 다양한 질병으로의 이환에 따른 사망률을 증가시키기 때문에 비만 치료는 매우 중요한 과제 중 하나이다.
현재 주로 사용되고 있는 비만 치료제로는 식욕억제제가 있으나, 대부분의 식욕억제제는 심장질환, 고혈압, 부정맥과 같은 심혈관계 부작용과 우울증, 기억장애등의 정신적 부작용이 흔히 나타나며, 최근 점유율이 급증하고 있는 GLP-1 유사체(liraglutide, 삭센다)의 경우 매일 피하주사로 투여해야 하는 번거로움이 있어 삶의 질을 저하시키는 문제점이 있다.
이에 따라, 새로운 기전의 비만 치료제 개발이 절실히 요구되고 있는 실정이다.
한편, NK 세포(natural killer cell)는 선천적인 면역을 담당하는 혈액 속 백혈구의 일종으로 간과 골수에서 성숙하며, 자연 살해 세포라고도 한다.
바이러스에 감염된 세포나 암세포를 직접 공격해 없애는 것이 주기능으로, 바이러스에 감염된 세포나 암세포는 다른 세포와 달리 세포 표면에 특정 단백질(MHC Class I)이 적어지는 것 등의 이상이 생기는데, NK 세포는 이 이상을 감지해 바이러스에 감염된 세포나 암세포를 인지한다고 알려져 있다.
NK 세포가 바이러스에 감염된 세포나 암세포를 공격할 때 퍼포린(perforin)을 분비해 감염 세포나 암세포의 세포막에 구멍을 내고, 여기에 그랜자임(granzyme)을 내어 이 세포들을 사멸시킨다. 이 밖에 다른 면역세포의 증식을 유도하는 등의 역할을 하는 면역반응을 일으키는 물질인 케모카인(chemokine)과 사이토카인(cytokine)을 분비할 수도 있다.
특히, NK세포가 암세포를 공격해 암세포의 발생과 증식, 전이를 막는다는 것 외에도 암이 재발하는데 가장 중요한 역할을 하는 암 줄기세포를 효과적으로 제어할 수 있다는 것이 밝혀지며, 학계에서는 이 NK세포를 이용한 항암치료를 계속 연구하고 있다. 또한, 세포의 MHC class I 유전형이 같은 동종조혈줄기세포를 이식해 백혈병과 다발성골수종 등을 치료하려는 시도가 이루어지고 있으며, NK세포를 활성화시키는 사이토카인을 주입해 항암활성을 일으키는 연구도 진행 중이다.
일례로, 한국등록특허 제10-1693275호에는 IFN-γ 및 IL-32γ를 유효성분으로 포함하는 항암용 조성물이 개시되어 있고, 한국 등록특허 제10-1846464호에는 외래 미토콘드리아가 도입된 NK 세포를 사용한 항암용 조성물이 개시되어 있다.
그러나 현재까지 대사질환에서 NK 세포의 역할에 대한 연구는 미비한 편이며, 특히 NK 세포 활성조절을 통한 비만 치료 기전은 연구되어진 바 없다.
본 발명은 상기와 같은 문제점을 해결하기 위해 안출된 것으로, 본 발명의 목적은 NK 세포 활성조절을 통한 비만의 예방 또는 치료 방법을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 인간을 제외한 개체에 있어서, NK 세포(natural killer cell)의 활성을 증가시키는 단계를 포함하는 것을 특징으로 하는 비만의 예방 또는 치료 방법을 제공한다.
본 발명에 따른 비만의 예방 또는 치료 방법은 NK 세포의 활성 조절을 통해 체중 감소를 유도함으로써, 기존 비만 치료제와 차별화된 작용기전을 갖는 비만 치료제를 연구 및 개발하는데 유용하게 사용될 수 있다.
이하, 본 발명을 상세히 설명한다. 본 명세서 및 청구범위에 사용되는 용어나 단어는 통상적이거나 사전적인 의미로 한정해서 해석되어서는 아니되며, 발명자는 그 자신의 발명을 가장 최선의 방법으로 설명하기 위해 용어의 개념을 적절하게 정의할 수 있다는 원칙에 입각하여 본 발명의 기술적 사상에 부합하는 의미와 개념으로 해석되어야만 한다.
본 발명은 인간을 제외한 개체에 있어서, NK 세포(natural killer cell)의 활성을 증가시키는 단계를 포함하는 것을 특징으로 하는 비만의 예방 또는 치료 방법을 제공한다.
상기 비만은 과다한 체지방 축적으로 인해 건강의 위협을 주는 모든 상태를 의미할 수 있다.
상기 인간을 제외한 개체는 원숭이, 개, 고양이, 토끼, 모르모트, 랫트, 마우스, 소, 양, 돼지, 염소 등을 포함할 수 있다.
상기 NK 세포(natural killer cell)는 선천적인 면역을 담당하는 혈액 속 백혈구의 일종으로 간과 골수에서 성숙하며, 자연 살해 세포라고도 한다.
바이러스에 감염된 세포나 암세포를 직접 공격해 없애는 것이 주기능으로, 바이러스에 감염된 세포나 암세포는 다른 세포와 달리 세포 표면에 특정 단백질(MHC Class I)이 적어지는 것 등의 이상이 생기는데, NK 세포는 이 이상을 감지해 바이러스에 감염된 세포나 암세포를 인지한다고 알려져 있다.

상기 물질은 유기 또는 무기 화합물과 같은 단일 화합물, 펩타이드, 단백질, 핵산, 탄수화물 및 지질과 같은 생체 고분자 화합물 및 복수 화합물의 복합체 등을 포함할 수 있다.
또한, 상기 NK 세포(natural killer cell) 활성을 증가시키는 단계는 IL-12(Interleukin 12), IL-15(Interleukin 15,), IL-18(Interleukin 18), IFN-α(interferon alpha) 및 IFN-β(Interferon beta)로 이루어진 군으로부터 선택되는 어느 하나 이상의 사이토카인을 개체에 투여하여 이루어지는 것을 특징으로 할 수 있다.
상기 IL-12(Interleukin 12)는 세포 및 단구, 대식세포에서 생산되는 분자량이 약 70,000인 당단백질로, 정지기의 T 세포와 NK 세포에서 IFN-γ(Interferon gamma)생산의 유도, NK 세포의 활성 유도, 정지기 T세포의 렉틴 자극에 의한 세포증식능의 항진, 나이브세포에서 TH1세포로의 분화 촉진, 인터류킨3 존재하에서 조혈간세포의 증식 촉진 기능을 하며, 이식종양에 대한 생체 내 항종양 효과 등이 있는 것으로 알려져 있다.
상기 IL-15(Interleukin 15)는 상피세포에서 생성되는 분자량 14,000의 사이토카인으로 IL-2의 생물학적 활성의 대부분을 나타내며, NK 세포 분화에 필수적이다. 세포상해성 T 세포의 증식, 알로항원 특이적 세포상해성 T 세포 및 항원 비특이적 림포카인활성살생세포(LAK)의 시험관 내 작성을 촉진한다.
NK 세포외에 항암면역 반응을 억제하는 조절 T세포(Treg)를 증폭할 수 있는 IL 2와 달리, IL-15의 경우 NK 세포에 선택적으로 작용하며 비교적 낮은 농도에서 NK 세포를 활성화 할 수 있다.
상기 IL-18(Interleukin 18)는 1995년에 쿠퍼세포가 생산하는 사이토카인으로서 클로닝된 인터류킨으로, 분자량이 24,000인 IL-18의 전구체는 ICE에 의한 프로세싱을 받아 분자량이 18,000인 활성형이 되어 세포 밖으로 분비된다.
생물학적 활성으로, T 세포나 NK 세포에 있어서 IFN-γ(Interferon gamma)생산을 유도하며, NK세포의 활성을 증강한다. 또한, IFN-γ(Interferon gamma)생산 유도에 있어서는 IL-18와 IL-12는 강한 상승작용을 나타내는 것으로 알려져 있다.
상기 IFN-α(interferon alpha)는 사람의 백혈구가 바이러스에 감염될 때 특이적으로 유발되기 때문에 백혈구 인터페론이라고 하며, 핵이 없는 세포들이 바이러스에 대한 침입에 대항하도록 유도하고, I형 MHC의 발현을 증가시키며, NK 세포를 활성화시킨다.
상기 IFN-β(Interferon beta)는 사람의 섬유아세포에 2중나선 RNA를 첨가할 때 특이적으로 유발되어 섬유아세포인터페론이라고도 하며, 핵이 없는 세포들이 바이러스에 대한 침입에 대항하도록 유도하고, I형 MHC의 발현을 증가시키며, NK 세포를 활성화시킨다.
또한, 상기 NK 세포(natural killer cell) 활성을 증가시키는 단계는 리포폴리사카라이드(LPS) 또는 폴리 I:C(Polyinosinic:polycytidylic acid)를 개체에 투여하여 이루어지는 것을 특징으로 할 수 있다.
상기 투여는 임의의 적절한 방법으로 개체에게 소정의 NK 세포의 활성을 증가시키는 물질 제공하는 것을 의미한다.
예를 들면, 경구, 직장 또는 정맥, 근육, 피하, 자궁 내 경막 또는 뇌혈관 내(intracerebroventricular) 주사에 의해 투여될 수 있다.
상기 NK 세포(natural killer cell)는 지방조직 내의 NK 세포(natural killer cell)인 것을 특징으로 할 수 있다.
조직학적인 관점에서 지방조직은 크게 지방세포(성숙한 지방세포)와 지방세포 외 다양한 면역세포들로 이루어진 두 개의 독립체로 구성된다. 이 독립체는 다양한 상호작용을 통해 지방조직에서 지질대사, 지방조직의 유지 그리고 사멸화된 세포를 제거하는 일을 담당한다.
지방조직에는 백색 지방조직(white adipose tissue)과 갈색 지방조직(brown adipose tissue)이 존재하는데, 백색 지방조직은 주로 에너지를 저장하며 갈색 지방조직은 열을 생성하는 역할을 담당하고 있다.
갈색 지방조직은 백색 지방조직과 달리 세포 밖에 혈관과 교감신경이 매우 치밀하게 분포하고 있고, 세포 내에는 작은 지방 방울이 촘촘하게 분포할 뿐만 아니라 미토콘드리아가 많이 분포되어 있으며, 미토콘드리아 내막에 존재하는 Ucp1(uncoupling protein 1)에 의해 ATP 대신 열을 생성할 수 있는 특징이 있다.
베이지 지방세포(beige adipocyte)는 피하지방조직에 존재하는 지방세포로 평상시에는 백색지방의 역할을 하나, 추위나 교감신경계 자극에 의해 갈색지방의 성질을 가지게 되는 지방세포이다. 비만인 경우 활성화가 감소하는 것으로 알려져 있으며, 사람의 갈색 지방세포는 베이지 지방세포와 성질이 매우 유사함이 밝혀지고 있다.
상기 백색 지방조직 내의 NK 세포(natural killer cell) 활성이 증가될 경우, 백색 지방조직 내 지방세포의 에너지 저장이 감소하여 체중 감소 효과가 유도될 수 있다.
상기 갈색 지방조직 내의 NK 세포(natural killer cell) 활성이 증가될 경우, 갈색 지방조직 내 지방세포의 활성이 증가되어 열생성을 증가시킴으로써 체중 감소 효과가 유도될 수 있다.
상기 갈색 지방조직 내의 NK 세포(natural killer cell) 활성 증가는 갈색 지방 세포내의 UCP-1(uncoupling protein-1)의 발현을 증가시켜 열생성을 증가시킬 수 있다.
또한, 본 발명은 인비트로(in vitro) 상에서 백색 지방세포 및 갈색 지방세포와 NK 세포(natural killer cell)를 공동배양 하는 단계; 및
상기 NK 세포(natural killer cell)의 활성을 증가시키는 단계를 포함하는 것을 특징으로 지방세포의 활성화 방법을 제공한다.
상기 NK 세포(natural killer cell) 활성을 증가시키는 단계는 NK 세포의 활성을 증가시키는 모든 물질을 개체에 투여하여 이루어질 수 있다.
상기 물질은 유기 또는 무기 화합물과 같은 단일 화합물, 펩타이드, 단백질, 핵산, 탄수화물 및 지질과 같은 생체 고분자 화합물 및 복수 화합물의 복합체 등을 포함할 수 있다.
상기 NK 세포(natural killer cell)의 활성을 증가시키는 단계는 IL-12(Interleukin 12), IL-15(Interleukin 15,), IL-18(Interleukin 18), IFN-α(interferon alpha) 및 IFN-β(Interferon beta)로 이루어진 군으로부터 선택되는 어느 하나 이상의 사이토카인을 투여하여 이루어지는 것을 특징으로 할 수 있다.
이하, 본 발명을 구체적으로 설명하기 위해 실시예 및 실험예를 들어 상세하게 설명하기로 한다. 그러나 본 발명에 따른 실시예들은 여러 가지 다른 형태로 변형될 수 있으며, 본 발명의 범위가 아래에서 상술하는 실시예들에 한정되는 것으로 해석되어서는 안 된다. 본 발명의 실시예들은 당업계에서 평균적인 지식을 가진 자에게 본 발명을 보다 완전하게 설명하기 위해서 제공되는 것이다.
<실험예 1> NK 세포 활성화에 따른 체중 감소 효과 확인
(1) 실험 방법
8주령 C57BL6/J 수컷 마우스를 사용하여, 60% 고지방식이(D12492,research diet) 사료를 8주간 섭취시켜 비만을 유도하였다.
비만 유도된 수컷 마우스 5마리 각각에 10ug/7mL 농도의 인터루킨-15(IL-15)을 300ul씩, 주 2회, 2주간 투여하였다.
대조군으로, 비만 유도된 수컷 마우스 5마리 각각에 PBS(Phosphate-Buffered Saline)을 동일한 조건으로 투여하였으며, 투여 종료 후, 2주간 체중 변화를 관찰하였다.
(2) 실험 결과
상기 실험의 결과를 표 1에 나타내었다.
나타낸 바와 같이, 투여 종료된 시점을 기준으로 2주 뒤인 4주 차에 실험군에서는 체중이 감소된 결과가 나타났으나, 대조군에서는 체중이 증가된 결과가 나타났다.
이를 통해, IL-15 투여로 활성화된 NK 세포는 비만 유도된 쥐에서 체중 감소 효과를 나타내어, 비만의 예방 또는 치료에 사용될 수 있음을 확인할 수 있었다.
