Abstract
Description
도 2는 OsASN1 유전자 넉아웃 변이체의 분리 및 특징을 나타낸 것으로, (A)는 OsASN1 유전자와 T-DNA 삽입 위치를 나타낸 그림으로, 박스와 선은 각각 코딩 부위 내의 엑손 및 인트론을 나타낸 것이다. osasn1-1(3D-02739) 와 osasn 1-2(3A-05359)에서 T-DNA는 첫번째 인트론과 4번째 인트론에 각각 삽입하였다. 수평 화살표는 T2자손의 지노타이핑에 사용한 프라이머를 나타낸 것이다. (B)는 OsASN1 RT-PCR 분석결과를 나타낸 것이다. 7일된 osasn1-1, osasn 1-2 및 격리된 야생형(WT)의 모종싹의 총 RNA를 OsASN1 특이 프라이머(Fw 및 Rv)를 사용하여 RT-PCR을 수행하였으며. OsUbi유전자를 내부 대조군으로 하여 증폭하였다. 도 6은 OsASN1 과발현 형질전환 식물체의 제작방법 및 특징을 나타낸 것으로, (A)는 옥수수유래 유비퀴틴 프로모터(pUbi) 및 nopaline synthase 터미네이터(Tnos)의 조절 하에서 OsASN1를 발현하는 벡터를 도식화한 것이다. OsASN1 특이 프라이머(OxF 및 OXR)를 사용하여 cDNA를 증폭시켰다. (B)는 야생형 및 OsASN1 과발현(OX) 형질전환 식물체의 모종 싹으로부터 RNA를 분리하여 OsASN1를 qRT-PCR 분석한 결과를 나타낸 것이다. (C)는 성숙 잎에서의 아스파라긴 및 글루타민 농도를 나타낸 것이고, (D)는 성숙 이삭에서의 아스파라긴 및 글루타민 농도를 나타낸 것이다. - 아스파라긴 합성효소 1을 코딩하는 유전자가 과발현되어 있는 형질전환 벼.
- 제1항에 있어서, 아스파라긴 합성효소 1을 코딩하는 유전자는 OsASN1 유전자인 것을 특징으로 하는 형질전환 벼.
- 제2항에 있어서, 상기 OsASN1 유전자는 서열번호 1의 염기서열로 표시되는 것을 특징으로 하는 형질전환 벼.
- 제1항에 있어서, 상기 아스파라긴 합성효소 1을 코딩하는 유전자는 옥수수 유비퀴틴 프로모터에 의해 발현조절되는 것을 특징으로 하는 형질전환 벼.
- 다음 단계를 포함하는 질소제한 조건에서 쌀의 생산방법:
(a) 질소 비료를 사용하지 않은 토양에서 제1항의 형질전환 벼를 재배하여, 쌀을 등숙시키는 단계; 및
(b) 상기 등숙된 쌀을 수확하는 단계.
