Abstract
본 발명은 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법에 관한 것으로서, 더욱 상세하게는 (a) 비행환경과 유사한 저산소 조건이 형성될 수 있는 동물실험용 챔버에 쥐를 투입하는 단계; (b) 상기 챔버에 간헐적으로 저산소 조건을 형성하여 상기 쥐를 간헐적 저산소 환경에 노출시키는 단계; (c) 상기 간헐적 저산소 환경에 노출된 쥐의 간으로부터 RNA를 수득하는 단계; (d) 상기 RNA를 NGS 장치를 이용하여 유전자의 발현변화를 측정하는 단계; 및 (e) 상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득하는 단계를 포함하는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법에 관한 것이다.
본 발명은 간헐적 저산소 환경에 노출된 경우 쥐 간의 유전자 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득할 수 있는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법을 제공할 수 있다.
또한 본 발명은 비행환경에서 발생하는 신체적, 유전적 변화 및 이를 통해 진행되는 질병에 관한 기초자료로 활용 가능한 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법을 제공할 수 있다.
본 발명은 간헐적 저산소 환경에 노출된 경우 쥐 간의 유전자 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득할 수 있는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법을 제공할 수 있다.
또한 본 발명은 비행환경에서 발생하는 신체적, 유전적 변화 및 이를 통해 진행되는 질병에 관한 기초자료로 활용 가능한 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법을 제공할 수 있다.
Description
본 발명은 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법에 관한 것으로서, 더욱 상세하게는 (a) 비행환경과 유사한 저산소 조건이 형성될 수 있는 동물실험용 챔버에 쥐를 투입하는 단계; (b) 상기 챔버에 간헐적으로 저산소 조건을 형성하여 상기 쥐를 간헐적 저산소 환경에 노출시키는 단계; (c) 상기 간헐적 저산소 환경에 노출된 쥐의 간으로부터 RNA를 수득하는 단계; (d) 상기 RNA를 NGS 장치를 이용하여 유전자의 발현변화를 측정하는 단계; 및 (e) 상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득하는 단계를 포함하는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법에 관한 것이다.
인간이 위치한 지상의 모든 생물은 지구상에 존재하게 된 때부터 대기환경에 적응하여 왔다.
대기환경에 변화가 발생하면 이는 모든 생물들에게 직접적인 영향을 미치게 되므로 생존을 위해서는 이에 적응할 필요가 있다.
한편 인간의 신체기능은 해수면과 유사한 고도에서는 별 영향을 받지 않지만 고공으로 상승하게 되면 그 기능에 장애를 일으키며 고도에 따라서는 치명적인 영향을 받게 된다.
문명의 발전과 생활의 다양화로 인하여 인간은 많은 스트레스를 받고 있으며, 특히 항공우주산업의 급속한 발달로 전투기 조종사를 포함한 공중근무자들은 저산소, 한랭, 소음, 급가속 등에 의하여 심한 신체적, 심리적 스트레스를 받게 된다.
그러나 비행환경에서 발생하는 신체적, 유전적 변화 및 이를 통해 진행되는 질병에 관한 연구는 전무한 실정이다.
따라서 이상환경에 장기간 노출되는 경우 발생하는 신체적, 유전적 변화를 분석할 수 있는 기반 기술 확보가 필요하다.
이와 관련하여 한국공개특허 제10-2008-0036579호, 한국등록특허 제10-1499710호, 한국공개특허 제10-2012-0089235호 등은 저산소 조건에 대한 식물의 내성을 증가시키는 방법, 벼의 비생물성 스트레스 노출 분석방법, 저산소 조건 하에 배양된 세포로부터의 조정 배지 등을 개시하고 있다.
본 발명은 상기 종래 기술의 문제점을 해결하기 위한 것으로서, 간헐적 저산소 환경에 노출된 경우 쥐 간의 유전자 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득할 수 있는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법을 제공하는데 그 목적이 있다.
또한 본 발명은 비행환경에서 발생하는 신체적, 유전적 변화 및 이를 통해 진행되는 질병에 관한 기초자료로 활용 가능한 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법을 제공하는 것을 목적으로 한다.
상기와 같은 목적을 달성하기 위하여 본 발명은 (a) 비행환경과 유사한 저산소 조건이 형성될 수 있는 동물실험용 챔버에 쥐를 투입하는 단계; (b) 상기 챔버에 간헐적으로 저산소 조건을 형성하여 상기 쥐를 간헐적 저산소 환경에 노출시키는 단계; (c) 상기 간헐적 저산소 환경에 노출된 쥐의 간으로부터 RNA를 수득하는 단계; (d) 상기 RNA를 NGS 장치를 이용하여 유전자의 발현변화를 측정하는 단계; 및 (e) 상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득하는 단계를 포함하는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법을 제공한다.
본 발명의 일 실시예에 있어서, 상기 (e) 단계 이후에, (f) 저산소 환경에 노출되지 않은 쥐의 간으로부터 RNA를 수득하고, 상기 RNA를 NGS 장치를 이용하여 유전자의 발현변화를 측정하며, 상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득하는 단계; 및 (g) 간헐적 저산소에 노출된 경우 및 저산소 환경에 노출되지 않은 경우의 유전자 발현변화를 비교하는 단계를 추가로 포함하는 것을 특징으로 한다.
본 발명의 일 실시예에 있어서, 상기 간헐적 저산소 환경은 8,000~30,000ft의 비행환경과 동일한 저산소 조건에서 오전 30분~2시간 및 오후 30분~2시간 노출시키는 것을 특징으로 한다.
본 발명의 일 실시예에 있어서, 상기 쥐는 만성부비동염이 유발된 쥐인 것을 특징으로 한다.
본 발명의 일 실시예에 있어서, 상기 정상 쥐의 경우 발현변화는 Hamp2 및 Ass1에서 높은 수치를 나타내는 것을 특징으로 한다.
본 발명의 일 실시예에 있어서, 상기 만성부비동염이 유발된 쥐의 경우 발현변화는 Mup17 및 Uox에서 높은 수치를 나타내는 것을 특징으로 한다.
본 발명의 일 실시예에 있어서, 상기 (c) 단계는 쥐의 간으로부터 mRNA를 정제, 파편화하고, 1차 cDNA 합성 및 2차 cDNA 합성을 순차적으로 진행한 후, PCR 증폭하는 과정을 포함하는 것을 특징으로 한다.
본 발명의 일 실시예에 있어서, 상기 단백질의 신호체계는 퍼옥시즘 증식 활성화 반응체 신호전달체계, 아디포사이토카인 신호전달체계 또는 인슐린 신호전달체계인 것을 특징으로 한다.
본 발명은 간헐적 저산소 환경에 노출된 경우 쥐 간의 유전자 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득할 수 있는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법을 제공할 수 있다.
또한 본 발명은 비행환경에서 발생하는 신체적, 유전적 변화 및 이를 통해 진행되는 질병에 관한 기초자료로 활용 가능한 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법을 제공할 수 있다.

본 발명에 사용되는 기술 용어 및 과학 용어는 다른 정의가 없다면 이 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 통상적으로 이해하고 있는 의미를 나타낸다.
본 발명은 (a) 비행환경과 유사한 저산소 조건이 형성될 수 있는 동물실험용 챔버에 쥐를 투입하는 단계; (b) 상기 챔버에 간헐적으로 저산소 조건을 형성하여 상기 쥐를 간헐적 저산소 환경에 노출시키는 단계; (c) 상기 간헐적 저산소 환경에 노출된 쥐의 간으로부터 RNA를 수득하는 단계; (d) 상기 RNA를 NGS 장치를 이용하여 유전자의 발현변화를 측정하는 단계; 및 (e) 상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득하는 단계를 포함하는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법에 관한 것이다.
상기 동물실험용 챔버는 이상환경과 유사한 조건을 형성할 수 있으며, 수면 80m 이하까지 그리고 고도 45,000ft까지의 환경과 유사한 조건을 만들 수 있다(도 1).
상기 간헐적 저산소 환경은 8,000~30,000ft의 비행환경과 동일한 저산소 조건에서 오전 30분~2시간 및 오후 30분~2시간 노출시킬 수 있다.
본 발명은 상기 (e) 단계 이후에, (f) 저산소 환경에 노출되지 않은 쥐의 간으로부터 RNA를 수득하고, 상기 RNA를 NGS 장치를 이용하여 유전자의 발현변화를 측정하며, 상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득하는 단계; 및 (g) 간헐적 저산소에 노출된 경우 및 저산소 환경에 노출되지 않은 경우의 유전자 발현변화를 비교하는 단계를 추가로 포함할 수 있다.
상기 쥐는 정상 쥐 또는 만성부비동염이 유발된 쥐인 것을 특징으로 한다.
상기 유전자 신호체계 및 단백질 대사는 퍼옥시즘 증식 활성화 반응체 신호전달체계(ME1, SCD1 등), 아디포사이토카인 신호전달체계(PPARA, G6PC 등), 인슐린 신호전달체계(SREBF1, SOCS2 등), 당단백질 대사(Glut1, Gclc, Gclm 등) 등이 해당된다.
상기 간헐적 저산소에 노출된 정상 쥐의 경우 발현변화는 Hamp2 및 Ass1에서 높은 수치를 나타낸다.
상기 간헐적 저산소에 노출된 만성부비동염이 유발된 쥐의 경우 발현변화는 Mup17 및 Uox에서 높은 수치를 나타낸다.
이하 실시예 및 비교예를 통해 본 발명을 상세히 설명한다. 하기 실시예는 본 발명의 실시를 위하여 예시된 것일 뿐, 본 발명의 내용이 하기 실시예에 의하여 한정되는 것은 아니다.
(실시예 1) 시료의 전처리
도 2는 본 발명에 사용된 시료의 전처리 과정을 나타낸다.
쥐의 간으로부터 mRNA를 정제, 파편화하고, 1차 cDNA 합성, 2차 cDNA 합성 등을 순차적으로 진행한 후, PCR 증폭하였다.
Validate Library를 위해 Agilent DNA 1000 kit을 사용하였으며, Normalize and Pool Libraries를 수행하였다.
도 3은 NGS 실험에 적합한 RNA quality check을 나타낸다.
Co는 정상 쥐, Cs는 만성부비동염이 유발된 쥐, CoH는 간헐적 저산소에 노출된 정상 쥐, CsH는 간헐적 저산소에 노출된 만성부비동염이 유발된 쥐를 나타낸다.
표 1은 Bioanalyzer 2100 Bioanlyzer Desktop system으로 분석한 각 시료의 Library construction 결과를 나타낸다.
혼합한 시료들은 각 16 well plate에 각 시료마다 3번 정도 반복 실험을 할 수 있도록 한 웰에 1㎕를 주입하고 DNA 1000 bioanalyzer labchip에 1㎕를 주입하여 라이브러리 크기 분포 및 농도를 구하고, 10㎕ 부피로 하여 Sequencing에 이용하여 분석 자료를 얻었다.
이 실험에 사용한 장비는 Agilent 사에서 구매한 2100 Bioanalyzer를 이용하였고, Illumina사의 NextSeq 500 Sequencing System을 이용하였다.

(실시예 2) 유전자 발현변화
도 4는 간헐적 저산소에 노출된 정상 쥐의 유전자 발현변화를 나타낸다.
동물실험용 챔버에 정상 쥐를 투입하여 25,000ft의 비행환경과 동일한 저산소 조건에서 오전 1시간 및 오후 1시간 노출시켰다.
노출된 정상 쥐의 간에서 NGS 분석장비를 이용하여 유전자 발현변화의 피크 패턴을 관찰하였으며, Hamp2, Ass1 순으로 높은 발현변화를 볼 수 있었다.
NGS 유전자 분석결과를 다시 Excel로 통계 처리함으로써 한꺼번에 높고 낮은 발현변화를 나타내는 다양한 유전자를 확인할 수 있었다.
도 5는 간헐적 저산소에 노출된 만성부비동염이 유발된 쥐의 유전자 발현변화를 나타낸다.
동물실험용 챔버에 만성부비동염이 유발된 쥐를 투입하여 25,000ft의 비행환경과 동일한 저산소 조건에서 오전 1시간 및 오후 1시간 노출시켰다.

기존에는 다양한 유전자, 단백질들의 신호체계와 대사 등 Real-time RT PCR를 수십 번 또는 수백 번 실험하여 자료를 얻지만, 본 발명은 한 번에 많은 유전자 발현변화 결과를 얻으므로 비행환경에 장기간 노출되는 경우 발생하는 유전자 변화 및 질환연구에 많은 도움이 될 수 있다.
상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득할 수 있으며, 이로부터 간헐적 저산소에 노출되는 경우 신체적, 유전적 상태의 변화를 확인할 수 있다.

한편 저산소 환경에 노출되지 않은 쥐의 간으로부터 RNA를 수득하고, 상기 RNA를 NGS 장치를 이용하여 유전자의 발현변화를 측정하며, 상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득할 수 있다.
간헐적 저산소에 노출된 경우 및 저산소 환경에 노출되지 않은 경우의 유전자 발현변화를 비교함으로써 간헐적 저산소가 신체적, 유전적 변화에 미치는 영향을 확인할 수 있으며, 이를 통해 발생 가능한 질환에 관한 연구를 할 수 있을 뿐 아니라 발생 가능한 질병을 미리 예방할 수도 있다.
정상 쥐, 만성부비동염이 유발된 쥐, 간헐적 저산소에 노출된 정상 쥐 및 간헐적 저산소에 노출된 만성부비동염이 유발된 쥐는 서로 상이한 유전자, 신호체계 및 대사를 나타낸다.
특정질환을 가지는 개체에서 간헐적 저산소에 노출되는 경우 개체에서 발현되는 신체적, 유전적 변화는 질환이 없는 개체에서 간헐적 저산소에 노출되는 경우와 상이하며, 이를 통해 특정질환을 가진 개체가 비행환경에 노출되는 경우 질환의 변화를 고찰할 수 있으며, 간헐적 저산소에 노출된 개체의 유전자들이 질환에 관여하는 메커니즘도 분석할 수 있다.
Claims (8)
- (a) 비행환경과 유사한 저산소 조건이 형성될 수 있는 동물실험용 챔버에 쥐를 투입하는 단계;
(b) 상기 챔버에 간헐적으로 저산소 조건을 형성하여 상기 쥐를 간헐적 저산소 환경에 노출시키는 단계;
(c) 상기 간헐적 저산소 환경에 노출된 쥐의 간으로부터 RNA를 수득하는 단계;
(d) 상기 RNA를 NGS 장치를 이용하여 유전자의 발현변화를 측정하는 단계; 및
(e) 상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득하는 단계를 포함하는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법.
- 제1항에 있어서,
상기 (e) 단계 이후에, (f) 저산소 환경에 노출되지 않은 쥐의 간으로부터 RNA를 수득하고, 상기 RNA를 NGS 장치를 이용하여 유전자의 발현변화를 측정하며, 상기 유전자의 발현변화로부터 유전자, 단백질의 신호체계 및 대사의 상관관계를 수득하는 단계; 및
(g) 간헐적 저산소에 노출된 경우 및 저산소 환경에 노출되지 않은 경우의 유전자 발현변화를 비교하는 단계를 추가로 포함하는 것을 특징으로 하는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법.
- 제1항에 있어서,
상기 간헐적 저산소 환경은 8,000~30,000ft의 비행환경과 동일한 저산소 조건에서 오전 30분~2시간 및 오후 30분~2시간 노출시키는 것을 특징으로 하는 간헐적 저산소에 노출된 쥐 간의 유전자 분석방법.

- 제1항에 있어서,
