Abstract
본 발명은 항체- 또는 펩티드-약물 콘쥬게이트 화합물에 관한 것으로, 여기서 하나 이상의 크립토파이신 유도체 (매크로시클릭 뎁시펩티드)는 자가-희생 링커에 의해 공유적으로 부착되고, 상기 자가-희생 링커는 하나 이상의 종양-관련 항원 또는 세포-표면 수용체에 결합한다. 상기 링커는 디케토피페라진을 형성할 수 있는 프로테아제 및 디펩티드 단위에 대한 분열 사이트를 포함한다. 이들 화합물은 암, 다른 질병 및 질환, 예를 들여 면역 또는 감염성 질병의 진단 또는 치료 방법에 유용할 수 있다.
Description
본 발명은 의약화학에 관한 것으로, 특히 항암 활성을 갖는 화합물, 보다 구체적으로 화학요법적 매크로시클릭(macrocyclic) 뎁시펩티드(depsipeptide) 약물 또는 톡신과 콘쥬게이트된 항체에 관한 것이다. 본 발명은 또한, 암, 자가면역 질환 또는 감염성 질환에 의해 영향받은 포유류 또는 대상의 인비트로 (in vitro), 인 사이투(in situ), 및 인비보 ( in vivo) 진단 또는 치료에 사용되는 용도의 상기 화합물에 관한 것이다.
항체 치료법은 암, 면역학적 및 맥관기원(angiogenic) 질병을 갖는 환자의 표적 치료를 위해 수립되어 왔다. 이러한 목적으로, 치료적 모노클론 항체(TMAs)가 개발되어 왔다(Firer M. A. J. Hematol . Oncol . 2012, 5, 70; Mullard, A. Nature Rev. Drug Discov . 2013, 12, 329). 그러나, 그들 자체에 사용되었을 때 임상전 연구에서 전도 유망했던 많은 TMAs는 불충분한 암 세포 독성 때문에 임상에서는 실패하였다.
모노클로날 항체 치료법은 암, 면역학적 및 맥관기원 질병을 갖는 환자의 표적 치료를 위하여 수립되어 왔다. 성공적인 항체 치료법의 예는 HERCEPTIN® (트라스투주맵)이며, 이는 인간 표피 성장 인자 수용체 2 단백질의 세포외 도메인, HER2 (ErbB2)에 대하여 높은 친화도를 가지고 선택적으로 결합하는 재조합 DNA-유래 인간화된 모노클로날 항체이다(US 5,821,337; US 6,054,297; US 6,407,213; US 6,639,055; Coussens L, et al (1985) Science 230:1132-9; Slamon D J, et al (1989) Science 244:707-12). HERCEPTIN은 항암치료 전에 광범위하게 받는 ErbB2-과다 발현하는 유방암 환자 치료에서 돌파구이지만, 이러한 집단에서 환자 대다수는 HERCEPTIN 치료에 대해 실패하거나 또는 좋지 못한 반응만을 나타낸다.
그러한 문제를 극복하기 위하여, 항체-약물 콘쥬게이트(ADCs) 분야에서 지수적 성장이 있어왔으며, 여기서 모노클로날 항체 (mAb, e.g. 트라스투주맵)은 높은 세포독성 약물에 연결된다. ADCs은 암 치료법에서 점점 더 중요해진다. 3개 ADCs는 US FDA에서 승인되었다:Mylotarg® (겜투주맵 오조가미신, Wyeth Pharmaceuticals), Adcetris® (브렌툭시맵 베도틴, Seattle Genetics), 및 Kadcyla® (아도-트라스투주맵 엠탄신, Roche), 반면에 35 ADCs은 현재 임상 시험 중에 있다. 종양 선택성은 mAb에 의해 나타나는 반면에 이 링커는 생리학적 조건 하에서 약물의 방출, 안정성 및 전체 용해성에 책임이 있다.
Mylotarg®은, 칼리키아마이신(calicheamicin)에 연결된 hu CD33 항체로 구성되며, 급성 골수성 백혈병의 치료를 위해 2000년에 승인되었으나, 2010년에 취소되었다. Adcetris®(브렌툭시맵 베도틴, Seattle Genetics, 이하 화학식 1)는 2011년에 승인되었고, 전통적인 호지킨 림프종 (HL) 및 전신역형성대세포림프종 (systemic anaplastic large cell lymphoma, sALCL)에서 나타나는, CD30에 대한 항체에 연결된 모노메틸 아우리스타틴 E (MMAE)으로 구성되며, Kadcyla® (아도-트라스투주맵 엠탄신, Roche, 이하 화학식 2)는 난치성 인간 표피 성장 인자 수용체 2 (HER2) 양성 전이 또는 국소적으로 진행된 유방암의 치료를 위해 2013년에 승인되었고, 세포 독성제 메르탄신(DM1)에 연결된 모노클로날 항체 트라스투주맵 (Herceptin)을 포함한다.
아도-트라스투주맵 엠탄신 (Kadcyla®) 및 브렌툭시맵 베도틴 (Adcetris®)은 US FDA 및 EMA(European Medicine Agency)에 의해 현재 승인된 단 2개의 ADCs이다.

크립토파이신의 놀라운 활성은 일부 환자들에게서는 늦게 인식되었다. Endocyte사는 분열가능한 디설파이드 링커를 장착한 크립토파이신 폴레이트 콘쥬게이트(화학식 3)을 특허받았다. 그러한 폴레이트 콘쥬게이트는 폴레이트 수용체를 과발현하는 암 세포를 표적으로 한다 (US2009/002993).

Sanofi-Aventis는 아주 최근에 크립토파이신 항체-약물 콘쥬게이트 (ADCs)에 대한 데이터를 개시하는 2개 특허를 공개하였다(US2011/001052; FR2947269). 변형된 아릴 치환기를 갖는 크립토파이신 유도체는 주로 적당한 기능성을 갖는 크립토파이신을 얻도록 선택되었다(화학식 4).

이들 중 다른 링커 타입, 분열가능한 디설파이드-, PEG-, 티오에테르- 및 티아졸 링커가 사용되었다. 모든 경우에, 상기 링커는 hu2H11에 연결되었고, 이는 EphA2 수용체를 표적으로 하는 모노클로날 항체이다. 2개 크립토파이신 ADCs에 대한 IC50의 값은 또한 인간 유방암 세포-주 MDA-MB-231에 대해 보고되었다. 화학식 4로 나타난 ADC는 입체적으로 가려진 티오에테르 링커를 포함하며, IC50값이 0.710nM을 나타낸다. 또 다른 ADC는 입체적으로 가려진 디설파이드 링커(IC 50: 0.150nM)를 포함한다. 치환된 크립토파이신은 약물 저항성 세포에 대해 세포독성 활성을 느슨하게 하는 것으로 보고되었다.
대조적으로, 콘쥬게이트되지 않은 크립토파이신(화학식 5)는 다중-약물 저항(MDR) 암 세포에 대하여 조차 매우 높은 세포독성을 갖는 것으로 보고된다. 크립토파이신-52은 다중-약물 저항(MDR) 종양 세포주에 대하여 매우 높은 활성을 나타낸다. 많은 MDR 종양 세포는 P 글리코단백질(P-gp)를 발현하고, 이는 유출 펌프로서 작동한다. P-gp 저항성 약물을 합리적으로 디자인하는 것은 여전히 어렵기 때문에 이러한 활성은 크립토파이신의 또 다른 귀중한 특징이다.
첫번째로 대표적인 것은 1990년에 시아노박테리아 Nostoc sp.로부터 분리되었다. 이들의 생물 작용(bioactivity)은 단백질 튜블린과 이들의 상호작용을 기초로 한다. 크립토파이신은 마이크로튜블 동력학(microtubule dynamics)의 억제로 인하여 아포프토시스를 유도하는 것으로 밝혀졌다. 결과적으로, 크립토파이신 유사체는 잠재적 항종양제인 것으로 생각된다.
크립토파이신-52 (LY355703)은 임상 I 연구를 통과하였으나, 이어지는 임상 II 단계 연구는 인비보 효능의 결핍, 높은 신경독성 및 선택된 용량에서 용량-제한 독성 때문에 중단되었다.

단지 몇가지 ADCs만이 현재 암 치료법에 대해 승인되었다. 보다 높은 다양성의 ADCs가 약물 저항성과 협력하기 위해서 필요하다.
현재까지 어떠한 크립토파이신-계 ADC가 승인되지 않았고, 어떠한 크립토파이신-55-글리시네이트도 항체에 대한 콘주게이션을 설명하고 있지 않다.
모든 설명된 크립토파이신 콘쥬게이트는 긴 합성 경로에 의해서만 접근 가능하다(예를 들어 W02011/001052 참조). 그러나, Sewald 및 공동 연구자들은, 짧으면서도 효율적인 합성 경로를 설명하였다 (Nat. Prod. Rep. 2013, 30, 924-940).
질병, 특히 암, 면역학적 질병, 감염성 질병의 치료에서 안전한 사용을 위하여 약물 콘쥬게이트, 특히 항체-약물 콘쥬게이트(또한 일반적으로 ADCs라고도 함) 및 펩타이드-약물 콘쥬게이트(또한 일반적으로 PDCs라고도 함)를 제공할 강한 필요성은 여전히 남아 있다.

다중 약물 저항성을 진전시킬 암, 면역학적 질병 및 감염성 질환의 치료에서 사용하기 위한 약물 콘쥬게이트, 특히 항체-약물 콘쥬게이트 (또한 ADCs라고 함) 및 펩티드-약물 콘쥬게이트(또한 일반적으로 PDCs라 함)을 제공할 강한 필요성이 있다.
클로로히드린 크립토파이신-55이 자가-희생적, 펩티드-유래 링커를 가로질러 모노클로날 항체 또는 펩티드에 연결되는 콘쥬게이트가 종양 세포에 대하여 매우 효율적인 것을 발견하였다.
본 발명에 따른 콘쥬게이트는 하나 이상의 종양-관련 항원 또는 세포-표면 수용체에 결합하여, 이들은 치료되거나 진단될 수 있는 이들 질병의 진단 또는 치료에 사용될 수 있으며, 이는 종양-관련 항원 또는 세포-표면 수용체, 단백질 또는 항원에 대한 결합 덕분이다.

새롭게 제조한 PBS-EDTA-완충제 pH 7.2로부터, TCEP-PBS-EDTA-용액 [100 ㎕의 PBS-EDTA-용액 중 3 mM TCEP (Sigma-Aldrich)] 및 mAb-PBS-EDTA-용액 (0.8 mL PBS-EDTA-용액 중 9 mg/mL 트라스투주맵; 62 μM 트라스투주맵)을 제조하였다. 상기 TCEP-PBS-EDTA-용액을 mAb-PBS-EDTA-용액에 첨가하고, 얻어진 용액을 60min 동안 25℃에서 배양하였다. 시약의 최종 농도는 다음과 같다: 8 mg/mL 환원된 트라스투주맵 (55 μM), 290 μM TCEP.
PBS-EDTA-완충제 pH7.2에서 사전 평형화된 HiTrap Desalting Sephadex G25-5 mL (GE Healthcare/Amersham) 컬럼 상에서 탈염이 Akta instrument을 사용하여 수행되었고, 0.25mL 부피의 분획물들이 PBS-EDTA-완충제 내에서 수집되었다. 모든 분획물은 이들의 단백질-함량에 대해 시험되었다 (2 ㎕의 각 분획물은 200㎕ Bradford-용액에 첨가되었다). 상기 단백질-함유 분획물이 모아지고, 항체 농도는 Bradford 분석으로 재결정되어, 3.3 mg/mL (22.8 μM)를 얻었으며, 이는 총 항체의 5mg의 회수(70% 회수)이다. 유리-SH의 농도는 Elmann's 시약 DTNB (5,5'-디티올비스-(2-니트로벤조산), Sigma-Aldrich/Fluka) 및 L-Cys (40-50-100-200-400-800 μM)을 사용한 표준 곡선을 사용하여 결정되었다. 총 -SH 농도는 3개 유리-SH/항체의 평균으로 68.24 μM이었다.
사슬 간 디설파이드 결합의 부분적 환원은 상술한 바와 같이 반복되었고, 말레이미드-PEG-Val-Cit-Pro-Gly-크립토파이신-55-글리시네이트 (DMSO 중 1.5 mM; 100 ㎕) 용액을 4℃에서 2h 동안 계속 교반하면서 샘플(900μL)에 직접 적가하여, 최종 농도가 10% DMSO 중 150μM이 되게 하여, 유리-SH기에 대하여 1:1 몰비 및 항체(항체 최종 농도는 50μM이었고, 총 유리 SH의 150μM에 등가임)에 대하여 3: 1 몰비가 되게 하였다. 동시에, S-S 환원된 항체 대조군 샘플은 대조군으로서 콘쥬게이트되지 않게 두었다. 배양 시간 마지막에, 각 샘플은 즉각 HiTrap Sephadex G25 컬럼 위에 부하하였고 상술한 바와 같이 탈염을 수행했다. Bradford 및 Elmann 분석은 상술한 바와 같이 통합된 분획물에서 수행되었고 결과는 이하 요약되었다.
선택된 면역접합체(immunoconjugation) 반응 조건하에서, SH/IgG의 평균 수는 2.7 내지 0.5로 떨어졌고, 따라서 이는 약물 링커에 대한 결합이 약 2의 약물-항체 비(DAR)로 일어난다는 것을 암시한다.
이후 조제용 겔 배제 크로마토그래피는, Akta instrument을 사용하여 수행되었다. 500㎕의 크립토파이신-55-글리시네이트-콘쥬게이트된 것과, 콘쥬게이트되지 않은 mAb 샘플을, 크로마토그래픽 완충제로서 PBS-EDTA pH7.2을 사용하여 Superdex 200 10/300 GL 컬럼 (GE Healthcare/Amersham) 위에 부하시키고, 흐름 속도 0.5 ml/min을 적용하였다. 250 ㎕-분획물들을 수집하였다. 약물 링커-콘쥬게이트된 항체의 정제는 2개 단백질 피크를 유지하였고, 피크 2는 크립토파이신-55-글리시네이트-콘쥬게이트된 항체의 모노머릭 형태에 대응하고 피크 1은 올리고머릭 형태에 대응하며, 이는 적당한 단백질 MW 표준과 비교하여 평가하였다(도 2, 왼쪽 패널). 대조적으로, 콘쥬게이트되지 않은 mAb 샘플은 단일 피크를 나타내고, 이는 상기 항체(mAb 대조군)의 모너머릭 형태에 대응한다(도 2, 오른쪽 패널). 피크 분획물은 통합되고 Bradford 분석에 의해 단백질 농도를 결정하였다. 결과는 이하에 요약되었다.

새롭게 제조된 PBS-EDTA-완충제 (137 mM NaCl; 2.7 mM KCl; 10 mM Na2HPO4; 2 mM KH2PO4; 10 mM EDTA; pH = 7.2)로부터, 2-MEA-PBS-EDTA-용액 (100 ㎕ PBS-EDTA-용액 중 6 mg 2-MEA) 및 mAb-PBS-EDTA-용액 (1 mL PBS-EDTA-용액 중 10 mg 트라스투주맵; 68.7 μM 트라스투주맵)을 제조하였다. 2-MEA-PBS-EDTA-용액 (300 ㎕)을 mAb-PBS-EDTA-용액 (1 mL)에 첨가하고, 얻어진 용액을 90min 동안 37℃에서 배양하였다. 시약의 최종 농도는 다음과 같다: 7.7 mg/mL 환원된 트라스투주맵 (53 μM), 13.8 mg 2-MEA (122 mM).
이후 HiTrap Desalting Sephadex G25 5 cm 컬럼 (PBS-EDTA-완충제) 상에서 탈염을 하고, 0.5mL 부피의 분획물들을 수집하였다. 모든 분획물들은 이들 단백질-함량에 대해 가시적으로 시험하였다 (2 ㎕의 각 분획물들을 200㎕ Bradford-용액에 첨가하였다). 단백질-함유 분획물은 수집되고 말레이미드-PEG-Val-Cit-Pro-Gly-크립토파이신-55-글리시네이트 (DMSO 중 1.82 mM; 150 ㎕) 용액을 첨가하였다. 2h 동안 6℃에서 배양하고 1/5mL 미만의 부피까지 용액을 농축하였다. Akta Purifier (Superdex 200 highload 16/60 컬럼; PBS-EDTA-완충제)로 정제하여, 2개 분핵물을 만들고 mAb-Cry-55-링커-피크 1(1.54 ㎍/㎕) 및 mAb-Cry-55-링커-피크2 (0.32 ㎍/㎕)으로 농축하였다. 피크 1 및 2는 상술한 2개 항체 응집 상태에 대응한다.

세포 생존도 분석
종양 세포주 (유방 암종 세포주 SKBR-3 및 MCF7 및 결장 암종 세포주 HCT116)는 American Type Culture Collection으로부터 얻어졌고, 표준 프로토콜에 따라 성장되었다. SKBR-3 세포는 높은 Her 2 준위를 발현하는 것으로 알려져 있으므로, 트라스투주맵-민감성이며, 반면에 MCF7 및 HCT116 세포는 낮은 Her 2 발현을 보이므로, 트라스투주맵-저항성이다. 종양 세포 생존도에 대한 크립토파이신-55-글리시네이트 ADC 콘쥬게이트의 효과는, 제조자의 프로토콜에 따라서 CellTiter-Glo® Luminescent Cell Viability Assay (Promega)을 사용하여 평가하였다. 콘쥬게이트되지 않은 트라스투주맵 및 트라스투주맵 ADC 사이의 차이를 검출할 수 있도록, 우리는 사전에 실험 조건들(농축 범위, 배양 시간)을 사전에 확인하였고, 이 조건 하에서 트라스투주맵만이 약 30%로 SKBR-3 세포 생존도에 영향을 미친 반면에, 유리 크립토파이신 55는 모든 종양 세포주에 동등하게 효과적이다. 세포들은 검은색-벽 96-웰 플레이트 내에 플레이트하였고(SKBR-3 및 MCF7에 대해 웰 당 5000 세포; HCT116에 대하여 웰 당 2000 세포), 5% CO2의 습기 조건 내에서 37℃에서 밤새 접착시켰다. 매질을 제거하고 트라스투주맵 ADC, 콘쥬게이트되지 않은 트라스투주맵, 또는 유리 크립토파이신 유사체 (CRY-55, CRY55-Gly)의 농도를 증가시키면서 이들을 포함하는 새로운 배양 배지로 대체하였고, 세포는 5% CO2 내에서 37℃에서 96h 동안 배양하였다. 5 내지 8개의 3배 데이터포인트는 각 실험 조건에 대하여 기록하였다. 배양 말기에 발광은 Perkin Elmer Wallac 1450 MicroBeta TriLux 검출기를 사용하여 측정하였다. 화합물 세포독성은 PBS (항체) 또는 0.1 % DMSO (화합물)로 처리한 세포에 비교하여 평가되었다. 4-펜티노일-옥트레오티드 (1):
옥트레오티드의 선형 전구제는 Tailhades et al. Angew . Chem . Int . Ed. Engl. 2010, 49, 117에 의해 이전에 설명된 것처럼 제조되었다. Fmoc-D-Phe-OH 결합 후, Fmoc 기는 피페리딘/DMF (3:7, 2 + 10 min)으로 제거되고, 이어서 DMF (6 x 1 min) 및 CH2Cl2 (1 x 1 min)으로 세척하였다. 4-펜티노익산(4 eq.)은 RT에서 4h 동안 교반하면서 Oxyma (4 eq.) 및 DIC (4 eq.)의 존재 하에서 결합되었다. 다음으로, 수지는 DMF (6 x 1 min), CH2Cl2 (1 x 1 min)으로 세척되고, Et2O으로 건조되었다. 상기 수지는 RT에서 2h동안 TFA/PhOH/H2O/TIS (88:5:5:2)을 사용하여 분열(cleave)되었다. RNA 정제 및 정량적 RT-
PCR
종양 세포주(랫트 췌장 종양 세포주 AR42J 및 인간 유방 암종 세포주 MCF7)는 American Type Culture Collection으로부터 얻고, 표준 프로토콜에 따라 성장되었다.
총 RNA는 DNAse I 처리 (Qiagen)를 포함하는 RNeasy Mini Kit (Qiagen, Germantown, MD)사용하여, 제조자의 추천에 따라서 세포들로부터 추출되었다. 첫 단계 RT-PCR 반응은 96-웰 광학 플레이트 내에서 조립되었다. 정량적 RT-PCR은 다음과 같이 세번 수행되었다: 40 ㎕의 반응 혼합물은, 25 ㎕의 2X Master Mix, 0.5 ㎕의 100X RT (QuantiTect Probe RT-PCR Kit, Qiagen), 및 2.5 ㎕의 다음 qPCR 분석법 각각 (Applied Biosystems/Thermofisher): 랫트 SSTR2 (Rn01464950_g1), 인간 SSTR2 (Hs00265624_s1), 랫트 GAPDH (Rn01775763_g1), 및 인간 GAPDH (Hs04420632_g1)을 포함하여 제조되었다. 3 ng/㎕ 농도의 10 ㎕의 총 RNA를 최종 반응 부피 50μL로 첨가하고, ABI 7900 HT real time thermal cycler (Perkin Elmer)을 사용하여 정량적 RT-PCR를 수행하였다 (50℃ 30 min, 95℃ 10 min, 이어서 40 사이클: 95℃ 15 sec, 60℃ 1 min). 정략적 계산은 GAPDH-정규화된 mRNA 발현 값에서 2^(-ΔΔc-t) 방법을 사용하여 수행하였다. 종양 세포주 AR42J (STTR2높은) 및 MCF7 (SSTR2낮은)의 생존도에 대한 실시예 5의 크립토파이신-55-글리시네이트-옥트레오티드 콘쥬게이트의 효과는, 제조자의 프로토콜에 따라 CellTiter-Glo® Luminescent Cell Viability Assay (Promega)를 사용하여 평가되었다. 세포들을 검은색-벽 96-웰 플레이트(5000 세포/웰)에 플레이트하고, 5% CO2의 습기 대기 내에서 37℃에서 밤새 부착시켰다. 매질을 이후 제거하고, 크립토파이신-55-글리시네이트-옥트레오티드 콘쥬게이트 (CRY55-Gly-Oct), 콘쥬게이트되지 않은 옥트레오티드, 또는 유리 크립토파이신-55-글리시네이트 (CRY55-Gly)을 농도 증가시키면서 포함하는 새로운 배양 배지로 대체하고, 상기 세포들을 5% CO2 내에서 37℃에서 120h 동안 배양하였다. 9개의 3배 복제품 1:3 일련의 희석액 데이터포인트는 각 실험 조건에 대해 기록되었다(CRY55-Gly 및 CRY55-Gly-Oct에 대해 0.0015 nM 내지 10 nM, 콘쥬게이트되지 않은 옥트레오티드에 대하여 0.0015 μM 내지 10 μM). 배양 말기에 발광은 Perkin Elmer Wallac 1450 MicroBeta TriLux 검출기을 사용하여 측정되었다. 화합물 세포독성은 0.1 % DMSO으로 처리한 세포에 비교하여 평가되었다. H69 세포 및 약물-저항성 카운터파트 H69AR는 American Type Culture Collection에서 얻었고, 표준 프로토콜에 따라 성장되었다. 야생 타입 및 약물 저항성 종양 세포주의 생존도에 대한 크립토파이신 및 크립토파이신 콘쥬게이트의 효과는, 제조자의 프로토콜에 따라, CellTiter-Glo® Luminescent Cell Viability Assay (Promega)을 사용하여 평가되었다. 세포들을 검은색-벽 96-웰 플레이트(5000 세포/웰) 내에 플레이트시키고, 5% CO2의 습기 대기에서 37℃에서 밤새 부착시켰다. 매질을 제거하고, 크립토파이신 유사체 또는 대조군으로서 독소루비신의 농도를 증가시키면서 이들을 포함하는 새로운 배양 배지로 교체하였다. 세포는 5% CO2 내에서 37℃에서 120h 동안 화합물로 배양하였다. 8개 3배 복제품 1:3 일련의 희석액 데이터포인트는 각 실험 조건에 대하여 기록되었다(CRY55-Gly, CRY55-Gly-링커 73 및 CRY55-Gly-링커 76에 대해 0.05 nM 내지 10 nM, 독소루비신에 대해 1.2 nM 내지 2700 nM). 배양 말기에, 발광은 Perkin Elmer Wallac 1450 MicroBeta TriLux 검출기를 사용하여 측정되었다. 화합물 세포독성은 0.1 % DMSO으로 처리한 세포에 비교하여 평가되었다.
