Description
도 2는, 본 발명의 일 실시예에 따른, 본 발명에 의한 랩온어 칩의 미세유체 채널층에 대한 상면도를 예시적으로 나타낸 것이다.
도 3은, 본 발명의 일 실시예에 따른, 본 발명에 의한 랩온어 칩을 예시적으로 나타낸 것이다.
도 4a는, 본 발명의 일 실시예에 따른, 본 발명에 의한 랩온어 칩의 제조방법의 흐름도를 예시적으로 나타낸 것이다.
도 4b는, 본 발명의 일 실시예에 따른, 본 발명에 의한 랩온어 칩의 제조방법의 공정을 예시적으로 나타낸 것이다.
도 4c는, 본 발명의 다른 실시예에 따른, 본 발명에 의한 랩온어 칩의 제조방법의 공정을 예시적으로 나타낸 것이다.

도 7은, 본 발명의 실시예 2에 따라 제조된 랩오너칩에 Intact hCG, free beta hCG, AFP(Alpha fetoprotein) 이 포함 되어있는 다중 미세유체시료(Multiplex) GFP(Green fluorescent protein) 형광 분석 결과를 예시적으로 나타내었다.
도 8은 본 발명의 실시예 3에 따라 제조된 랩온어 칩의 단일 미세유체시료에 대한 GFP(Green fluorescent protein) 형광 분석에 따른 간섭현상 결과를 예시적으로 나타내었다.
Claims (20)
- 하부 기판;
상기 하부 기판 상에 형성된 미세유체 채널층; 및
상기 미세유체 채널층 상에 형성된 상부 기판; 을 포함하고,
상기 미세유체 채널층은 양면 접착소재를 포함하고, 그 두께가 1 ㎛ 내지 100 ㎛ 미만인 것인, 랩온어 칩.
- 제1항에 있어서,
상기 미세유체 채널층은, 모세관력 기반으로 유체가 이동하는 것인, 랩온어 칩.
- 제1항에 있어서,
상기 양면 접착소재는 고형 양면 접착제를 포함하고,
상기 접착제는, 고무, 아크릴(Acryl), 실리콘(Silicone), 폴리스틸렌(Polystyrene), 폴리부타디엔(Polybutadiene), 폴리이소프렌(Polyisoprene), 폴리에틸렌(Polyethylene), 폴리올레핀(Polyolefine), 및 에틸렌초산비닐(Ethylene vinyl acetate)으로 이루어진 군에서 선택된 1종 이상을 포함하는 것인, 랩온어 칩.
- 제1항에 있어서,
상기 양면 접착소재는, 기재; 및 상기 기재의 양면에 형성된 고형 양면 접착제를 포함하고,
상기 기재는, 종이, 고무, 레이온(Rayon), 면(Cotton), 폴리에틸렌, 폴리카보네이트, 폴리스틸렌, 아세테이트(Acetate), 폴리프로필렌(Polypropylene), 셀로판(Cellophane), 폴리비닐클로라이드(Polyvinyl chloride), 폴리에스테르(Polyester), 폴리테트라 플루오로에틸렌(Polytetrafluoroethylene), 폴리이미드(Polyimide), 폴리우레탄(Polyurethane), 알루미늄(Al), 및 구리(Cu)로 이루어진 군에서 선택된 1종 이상을 포함하는 것인, 랩온어 칩.
- 제1항에 있어서,
상기 상부 기판 및 하부 기판은, 폴리머 기판을 포함하고,
상기 폴리머 기판은, 폴리스틸렌, 폴리카보네이트, 폴리에스테르, 폴리이미드, 폴리에틸렌, 폴리프로필렌, 폴리비닐클로라이드, 폴리메틸메타크릴레이트(Poly methyl methacrylate), 폴리아미드(Polyamide), 폴리아크릴아미드(Polyacrylamide), 폴리에테르우레탄(Polyetherurethane), 폴리술폰(Polysulfone), 폴리비닐플루오라이드(Polyvinyl fluoride), 및 폴리부틸렌(Polybutylene)으로 이루어진 군에서 선택된 1종 이상을 포함하는 것인, 랩온어 칩.
- 제1항에 있어서,
상기 상부 기판, 하부 기판 또는 이 둘의 적어도 일부분은, 플라즈마 표면처리되고,
상기 상부 기판, 하부 기판 또는 이 둘의 적어도 일부분은, 소수성 및 친수성 표면 특성을 가지며,
상기 플라즈마 표면처리는, 산소(O2), 질소(N2), 수소(H2), 아르곤(Ar), 제논(Xe), 및 헬륨(He)으로 이루어진 군에서 선택된 1종 이상의 플라즈마를 이용하는 것인, 랩온어 칩.
- 제1항에 있어서,
상기 채널층 내에 노출된 하부 기판의 표면, 또는 상기 하부 기판 및 상부 기판의 표면에 시료분주 영역(Sample zone), 콘주게이션 영역(Conjugation zone), 반응 영역(Reaction zone) 및 시료분출 영역(Waste zone)을 포함하고,
상기 영역 중 적어도 하나 이상은, 플라즈마 표면처리된 기판 상에 형성된 것인, 랩온어 칩.
- 제7항에 있어서,
상기 시료분주 영역, 콘주게이션 영역, 반응 영역 및 시료분출 영역 중 적어도 하나 이상은, 미세 패턴이 형성되고,
상기 시료분주 영역, 콘주게이션 영역 및 반응 영역은, 복수개로 형성된 것인, 랩온어 칩.
- 제7항에 있어서,
상기 시료분주 영역은, 시료반응 용액(Sample buffer)이 상기 기판 상에 고정되며,
상기 콘주게이션 영역은, 콘주게이트 표면에 제1 탐침자(Probe)가 결합된 콘주게이트 복합체가 상기 기판 상에 고정되고,
상기 반응 영역은, 제2 탐침자가 상기 기판 상에 고정된 것인, 랩온어 칩.
- 제9항에 있어서,
상기 콘주게이션 영역은, 2종 이상의 콘주게이트 복합체가 고정되고, 상기 반응 영역은, 2종 이상의 제2 탐침자가 고정되어, 복합 미세유체시료를 동시다중(Multiplex)으로 분석하는 것인, 랩온어 칩.
- 제9항에 있어서
상기 콘주게이트는, 금(Gold), 은(Silver), 단백질, 폴리머 비드(Polymer bead), 형광물질(Fluorescent substances), 및 발광물질(Luminescent substances)로 이루어진 군에서 선택된 1종 이상을 포함하고,
상기 폴리머 비드는, 폴리스티렌, 폴리아미드, 폴리이미드 및 라텍스(latex)로 이루어진 군에서 선택된 1종 이상을 포함하는 것인, 랩온어 칩.
- 제9항에 있어서,
상기 제1 탐침자 및 제2 탐침자는, 항체, 단백질, 펩티드(Peptide), DNA, RNA, 동물세포, 식물세포, 박테리아(Bacteria), 및 압타머(Aptamer)로 이루어진 군에서 선택된 1종 이상을 포함하고,
상기 제1 탐침자 및 제2 탐침자는, 서로 상이하고, 각각 진단을 위해 반응하는 대상의 다른 부위에 결합하는 것인, 랩온어 칩.
- 제9항에 있어서,
상기 콘주게이트 복합체는, 형광물질을 더 포함하고,
상기 형광물질은, 아크리딘(acridine), 시아닌(cyanine), 플루로돈(fluorone), 옥사진(oxazine), 페나트리딘(phenanthridine), 로다민(rhodamine) 및 쿠마린(coumarine)으로 이루어진 군에서 선택된 1종 이상을 포함하는 것인, 랩온어 칩.
- 제9항에 있어서,
상기 콘주게이션 영역 및 반응 영역은, 연결자(linker)를 더 포함하고,
상기 연결자는, 아비딘-비오틴(avidin-biotin), EDC/NHS 또는 이 둘을 포함하는 것인, 랩온어 칩.
- 제9항에 있어서,
상기 시료분주 영역은, 침(Saliva), 전혈(Blood), 혈청(Serum), 혈장(Plasma), 소변(Urine), 대변(Stool), 정액(Semen), 양수(Amniotic fluid), 복수(Ascitic fluid), 흉막액(Pleural fluid), 뇌척수액(Spinal fluid), 및 복막액(Peritoneal fluid)으로 이루어진 군에서 선택된 1종 이상을 포함하는 시료가 사용되는 것인, 랩온어 칩.
- 제1항에 있어서,
상기 미세유체 반응 분석 시 보색대비 기법을 적용한 것인, 랩온어 칩.
- 제1항에 있어서,
상기 랩온어 칩은, 종이, 유리, 플라스틱, 금속 중에서 1종 이상을 포함하는 외형으로 포장된 것인, 랩온어 칩.
- 양면 접착소재를 미세유체 채널 형태로 가공하는 단계;
제1 기판과 상기 가공된 양면 접착소재를 결합하여 제1 기판 상에 미세유체 채널을 형성하는 단계;
상기 미세유체 채널 내에 노출된 제1 기판 상에 시료분주 영역(Sample zone), 콘주게이션 영역(Conjugation zone), 반응 영역(Reaction zone) 및 시료분출 영역(Waste zone)을 형성하도록, 각 영역에 따른 반응 시약을 각각 고정하는 단계; 및
상기 양면 접착소재와 제2 기판과 결합하는 단계;
를 포함하는, 랩온어 칩의 제조방법.
- 제18항에 있어서,
상기 제1 기판 상에 미세유체 채널을 형성하는 단계 이전 또는 이후에 상기 제1 기판의 적어도 일부분을 플라즈마 표면처리하는 단계; 를 더 포함하고,
상기 반응 시약을 각각 고정하는 단계는, 상기 시료분주 영역(Sample zone), 콘주게이션 영역(Conjugation zone), 반응 영역(Reaction zone) 및 시료분출 영역(Waste zone) 중 적어도 하나 이상을 패터닝하여 미세 패턴을 형성한 이후에 반응 시약을 고정하는 것인, 랩온어 칩의 제조방법.
- 제18항에 있어서,
상기 제2 기판과 결합하는 단계 이전에, 상기 제2 기판 상에 시료분주 영역(Sample zone), 콘주게이션 영역(Conjugation zone), 반응 영역(Reaction zone) 및 시료분출 영역(Waste zone) 중 1종 이상 또는 모두를 형성하도록, 각 영역에 따른 반응 시약을 각각 고정하는 단계; 를 더 포함하는 것인, 랩온어 칩의 제조방법.
