Abstract
본 발명은 퇴행성 신경질환의 바이오 마커 및 이의 용도에 관한 것으로, 본 발명에 따르면, 퇴행성 신경 질환에서 병리적 미토콘드리아단편화, 세포 변성 및 세포 사멸을 억제할 수 있는 주요 표적을 제시하여, 이러한 표적을 이용하여 퇴행성 신경세포 질환의 예방, 개선 또는 치료가 가능하고, 그뿐만 아니라. 퇴행성 신경세포 질환의 치료에 효과 있는 약물의 스크리닝에도 효과적으로 사용할 수 할 수 있다.
Description
본 발명은 퇴행성 신경질환의 예방 또는 치료를 위한 바이오마커와 이를 이용한 치료 후보약물의 스크리닝 방법에 관한 것이다.
미토콘드리아는 지속적인 융합과 분열을 반복하며 세포질에 그물망 형태로 존재하면서, 세포 호흡을 통하여 세포의 생존에 필요한 에너지를 생산하는 세포 내 소기관이다. 미토콘드리아는 세포 내 에너지 대사의 중추이므로 미토콘드리아의 기능 이상은 다양한 질병을 유발한다. 미토콘드리아의 기능 이상에 의해 발생하는 질병으로는 퇴행성 신경질환이 있고, 이러한 퇴행성 신경질환에는 근위축성 측삭경화증을 포함하는 운동뉴런질환, 알츠하이머병, 파킨슨병, 헌팅턴병 등이 포함된다.
운동뉴런질환(motor neuron disease)는 운동 신경에 점진적인 퇴행이 일어나는 중증 신경계 질환군으로 뇌에서 나와 연수 또는 척수로 전달되는 상위 운동신경이나, 척수에서부터 근육으로 전달되는 하위 운동신경에 모두 영향을 줄 수 있다. 상위 운동신경이 손상되면 신체의 과도한 반사 반응이 나타나고, 하위 운동신경이 손상되면 근육이 진행적으로 위축(atrophy)되고, 쇠약해 진다. 운동신경원 병은 여러 다른 형태로 나타나나, 일반적으로 희귀성 질환으로 분류되고 있다.
원인에 대하여 현재 많은 연구가 이루어지고 있지만 아직까지 정확한 원인은 밝혀져 있지 않고, 대게 유전에 의해서 발생하는 경우도 있지만, 특별한 원인 없이 산발적으로 발생하는 경우가 대부분이다. 유전적 소인이 있는 사람은 운동신경원 병이 생길 확률이 높을 것으로 추측되고 있지만, 환경적 요소들도 영향을 미칠 것으로 추정하고 있다.
특히, 유전 등에서 매우 복잡한 양상을 나타내는데, 예를 들어 적어도 5개의 유전자가 운동신경원 병의 활동인자로 알려져 있고, 척수성 근육 위축은 적어도 8개~12개의 다른 형태를 가진다. 진행성 구마비는 상염색체 열성 형질로 유전되고, 변형 유전자의 위치는 아직 밝혀지지 않았고, 근위축성 측삭경화증은 상염색체 우성 형질로 유전되는 것으로 추측되고 있다.
운동뉴런질환은 여러 가지 형태로 분류할 수 있고, 분류된 세부 질환들 사이에 공통적으로 나타나는 증상과 특징적 증상이 있다.
운동뉴런질환 중 하나인, 근위축성 측삭경화증(Amyotrophic lateral sclerosis-ALS)은 루게릭병(Lou Gehrigs disease)라고도 불리는 것으로, 상위와 하위 운동신경원에 모두 영향을 미친다. 처음으로 나타나는 증상으로 손의 사용이 서툴고 다리가 약해지며, 음식을 삼키는데 어려움을 겪고, 말이 느려진다. 그리고 질환이 진행됨에 따라 전신에 있는 다른 근육이 영향을 받게 된다. 호흡근에 영향을 미치게 되면, 기침, 호흡곤란 등이 나타날 수 있고, 점진적인 근육위축과 약화 그리고 근육강직이 나타나고, 결국은 말기에 호흡부전으로 사망하게 된다. 주로 40세에서 70세 사이의 성인에게 나타나며, 여성보다는 남성에게 좀 더 자주 발생하는 경향이 있다.
근위축성 측삭경화증의 병리적 현상 중, 운동신경세포 내의 대표적인 특징 중 하나는 세포 내 에너지 생산을 담당하는 미토콘드리아의 기능 및 형태학적 변형이다. 근위축성 측삭경화증의 병리적 현상이 일어나기 이전 시기부터 미토콘드리아의 비정상적인 단편화는 일어나기 시작하고, 이로 인해 미토콘드리아의 기능 저하뿐만 아니라, 더 나아가 세포 내 대사활동에 영향을 미치게 된다. 이러한 미토콘드리아의 비정상적인 형태적 변화는 근위축성 측삭경화증 뿐만 아니라, 퇴행성 신경질환인 알츠하이머에서도 나타난다고 알려져 있다(Hirai et al, J Neurosci, 21(9):3017-23, 2001; Santos et al, J Alzheimers Dis, 20:S401-12, 2010). 따라서 미토콘드리아의 비정상적인 형태적 변화는 미토콘드리아의 재생과 분열, 세포내의 이동과 분포에 밀접한 관련이 있으며, 이러한 형태적 변화와 관련된 단백질들의 비정상적인 활성은 신경세포의 손상을 초래할 수 있다(Su et al, Mol Neurobiol, 41(2-3):87-96, 2010).
또한, 근위축성 측삭경화증 발병 시, 신경세포 내의 많은 단백질들의 활성이 변화되고, 이로 인해 세포사멸이 유도된다. 단백질들의 활성은 대부분 인산화에 의해서 조절 받게 되는데, 이때 인산화를 조절할 수 있는 프로테인 포스파타아제 1(Protein phosphatase 1, PP1)의 활성 억제제를 이용하여 비정상적인 단백질들의 활성을 조절함으로써 근위축성 측삭경화증으로 인해 유도된 신경세포의 기능과 대사적 메커니즘의 흐름을 정상화 할 수 있으나, PP1은 매우 넓은 범위의 기질을 탈인산화 하고, 이에 따라 매우 다양한 생명 현상에 관여하기 때문에, PP1의 전반적 억제는 세포의 생존에 관련한 문제점을 유발할 가능성이 높다.
현대의학의 발달에도 아직까지 이러한 퇴행성 신경세포 질환의 병리기전조차 명확히 밝혀지지 않았고, 단지 증상의 진행을 늦추거나 완화시키는 수준에 머물러 있다. 현재, 근위축성 측삭경화증의 치료제로써 FDA 승인을 받은 리루졸(riluzole)이 유일하지만, 리루졸(riluzole)은 운동신경세포를 파괴하는 원인의 하나로 알려진 과도한 양의 글루타민산을 억제하는 것으로, 일단 파괴된 신경은 원래대로 되돌리 수 없고, 리루졸을 복용하더라도 환자의 증상이 멈추거나 원래의 건강한 상태로 돌아올 순 없어, 근위축성 측삭경화증에 대해 치료효과보다는 수명을 연장시키는 수단에 불과하다.
따라서 이러한 운동뉴런질환 및 퇴행성 신경 질환에 대한 근본적으로 질병에 대한 발생 기전의 이해를 바탕으로 치료 효과에 특이성이 높은 표적을 찾아서 조기 치료 및 치료를 위한 약물을 비롯한 치료법을 개발하는 것이 절실하게 요구된다.
따라서, 본 발명은 상기 문제점을 해결하기 위하여, 운동뉴런질환을 포함하는 퇴행성 신경 질환에 대한 특이적인 치료 타겟이 될 수 있는 표적을 제시하고, 이를 이용한 퇴행성 신경 질환의 치료를 위한 조성물과 퇴행성 신경 질환의 치료 약물의 스크리닝용 조성물, 키트 및 스크리닝 방법을 제공하는 것을 목적으로 한다.
미토콘드리아는 세포 내 에너지 생성을 위한 소기관으로, 근위축성 측삭경화증, 알츠하이머병과 같은 퇴행성 신경질환에서 비정상적인 형태를 나타낸다. 미토콘드리아의 형태적 변화는 미토콘드리아의 질적, 양적 변화의 항상성을 유지하기 위한 매우 중요한 과정 중 하나이다. 이에 본 발명에서는 퇴행성 신경질환에 대한 치료제 발굴을 위해 미토콘드리아의 형태를 조절할 수 있는 후보물질을 탐색하는데 초점을 맞추었다.
본 발명의 발명자는 PP1의 활성을 억제하면 미토콘드리아의 형태를 조절할 수 있고, 비정상적인 단백질들의 활성을 조절하여 운동신경세포의 기능과 대사적 메커니즘을 정상화 할 수 있음을 확인하였다. 그러나, PP1은 매우 넓은 범위의 기질을 탈인산화하고, 이에 따라 매우 다양한 생명 현상에 관여하므로, PP1의 전반적인 억제는 세포의 생존에 관련된 문제를 유발할 가능성이 높아, 퇴행성 신경질환의 치료에 효과적이면서도 특이성이 높은 표적이 필요하였다. 이에, PP1 촉매 소단위체로 알려진 α, β, γ 중에서 PP1 감마를 특이적으로 억제하면 미토콘드리아 단편화와 신경세포의 축삭 변성 등을 효과적으로 억제하여 신경 세포사멸을 감소시킴을 확인하여 본 발명을 완성하였다.
따라서, PP1 감마의 발현의 저해를 통해서 퇴행성 신경질환의 예방, 또는 치료 및 이에 대한 치료제 발굴에 활용할 수 있다.
상기 목적을 달성하기 위하여, 본 발명은 PP1 감마(Protein Phosphatase 1 gamma)의 발현 저해제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 PP1 감마(Protein Phosphatase 1 gamma)를 포함하는 시료에 운동뉴런질환 또는 퇴행성 신경질환 치료제 후보물질을 처리하는 단계; 상기 후보물질이 PP1 감마의 발현을 증가 또는 감소시키는지를 판단하는 단계; 및 상기 PP1 감마의 발현을 감소시키는 후보물질을 상기 퇴행성 신경질환 치료제로 선별하는 단계;를 포함하는 퇴행성 신경질환 치료제 스크리닝 방법을 제공한다.
또한, 본 발명은 PP1 감마(Protein Phosphatase 1 gamma)를 포함하는 퇴행성 신경질환 치료제 스크리닝용 조성물을 제공한다.
또한, 본 발명은 PP1 감마(Protein Phosphatase 1 gamma) 및/또는 PP1 감마의 발현량을 측정하는 제제를 포함하는 퇴행성 신경질환 치료제의 스크리닝용 키트를 제공한다.
본 발명에 따르면 퇴행성 신경 질환에서 병리적 미토콘드리아 단편화, 세포 변성 및 세포 사멸을 억제할 수 있는 주요 표적을 제시하였고, 이러한 표적을 이용하여 퇴행성 신경세포 질환의 예방, 개선 또는 치료가 가능할 뿐만 아니라, 퇴행성 신경세포 질환의 치료를 위한 약물의 스크리닝에도 효과적으로 사용할 수 있다.

본 발명의 발명자는 근위축성 측삭경화증과 같은 퇴행성 신경질환에서 특이성이 높은 표적의 필요성을 인식하고, 연구하던 중에 PP1의 촉매 소단위체인 PP1 감마를 특이적으로 억제하면 미토콘드리아 단편화, 신경세포 돌기 등을 효과적으로 억제하면서 세포사멸도 막을 수 있음을 확인하여 본 발명을 완성하였다.
따라서, 본 발명은 퇴행성 신경질환의 치료를 위한 표적, 그리고 치료를 위한 바이오 마커로서 PP1 감마(Protein Phosphatase 1 gamma)의 용도를 제공한다.
본 발명에서 "퇴행성 신경질환"은 신경세포의 사멸 또는 퇴행성 변화가 비정상적으로 진행되어 뇌 또는 뇌와 연결된 척수의 기능이 마비되는 질환을 의미한다. 구체적인 예로, 운동뉴런질환, 알츠하이머병, 파킨슨병, 헌팅턴병, 전두측두치매, 루이체치매, 피질기저퇴행증, 다계통 측위병 및 진행성핵상마비가 이에 포함된다. 본 발명에서는, 바람직하게는 운동뉴런(운동신경세포, motor neuron)의 사멸 또는 퇴행성 변화에 의해 유발되는 운동뉴런질환을 의미할 수 있다.
본 발명에서 "운동뉴런질환(motor neuron disease)"은 운동신경원 병, 운동뉴런 병, 운동신경원 증후군 등으로도 불리며, 신경계의 운동신경세포가 손상 받는 모든 질환을 포함하는 의미이다. 상기 운동뉴런질환은 침벙양상에 따라 척수근위축증(spinal muscular atrophy), 근위축성 측삭경화증(Amyotrophic lateral sclerosis-ALS), 진행성 구음장애(progressive bulbar palsy) 및 원발성 측삭경화증(Primary Lateral Sclerosis)으로 나뉠 수 있고, 구체적인 질병으로 베르드니히-호프만 병(Werdnig-Hoffman disease) 또는 유아성 척수성 근육위축(SMA1), 쿠겔베르그-벨란더 증후군(Kugelberg-Welander syndrome) 또는 청소년형 척수성 근육위축(Juvenile for m, type III, SMA 3), 진행성 척수성 근육위축(Progressive spinal muscular atrophy), 진행성 구마비(Progressive bulbar palsy) 및 가족성 운동신경원 병(Familial motor neuron disease) 도 포함한다.
본 발명에서 "PP1 감마(Protein Phosphatase 1 gamma)"는 PP1 감마 단백질, 유전자 및 상기 유전자를 발현하는 세포를 모두 포함하는 의미이다. PP1 감마는 프로테인 포스파타아제 1의 촉매 소단위체(catalytic subunit) 중 하나로, 인간의 PPP1CC 유전자(NM_002710.3) 또는 인간 PPP1CC 유전자에 의하여 코딩되는 단백질 효소 일 수 있다. 구체적으로 상기 PP1 감마는 서열번호 1의 염기서열을 갖는 것일 수 있다.
본 발명은 PP1 감마(Protein Phosphatase 1 gamma)의 발현 저해제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 치료용 약학적 조성물에 관한 것이다.
본 발명에서 상기 "PP1 감마(PP1 gamma)의 발현 저해제"는 PP1 감마의 mRNA 수준에서 발현을 억제하는 올리고 뉴클레오티드 또는 PP1 감마 단백질의 활성을 억제하는 물질을 통칭하는 의미이다. 보다 구체적으로 PP1 감마 유전자 또는 단백질에 직접적으로 작용하거나, 그의 리간드를 간접적으로 작용하는 등의 방식을 통해 PP1 감마의 발현을 전사 수준에서 감소시키거나, 그 활성을 방해함으로써 PP1 감마의 발현 또는 활성을 감소시키는 모든 물질을 포함할 수 있다.
상기와 같은 측면에서 본 발명의 약학적 조성물은 유전자 치료제, 단백질 치료제, 펩타이드 치료제 및 화학적 치료제 등에 포함되어 사용되는 것을 모두 포함한다.
상기 PP1 감마의 발현을 저해제는 PP1 감마를 표적으로 발현 또는 활성을 억제할 수 있는 화합물, 핵산, 펩타이드, 바이러스 또는 상기 핵산을 포함하는 벡터 등 그 형태에 제한 없이 이용 가능하다. 상기 PP1 감마의 발현 저해제는 바람직하게 PP1 감마의 mRNA 발현을 저해하는 올리고 뉴클레오티드, PP1 감마 단백질의 활성을 억제하는 항체 및 상기 항체의 항원 결합 단편으로 이루어진 군에서 선택된 것 일 수 있다.
상기 PP1 감마의 발현 저해제는 PP1 감마에 특이적으로 결합할 수 있고, 일 예로 PP1 감마의 염기서열에 상보적인 서열을 포함하는 것 일 수 있다.
본 발명에서 "올리고 뉴클레오티드"는 수 개 내지 수십 개의 뉴클레오티드가 인산에스테르 결합으로 중합되어 형성된 중합체를 의미한다. 상기 PP1 감마의 mRNA의 발현을 저해하는 올리고 뉴클레오티드의 예로, 바람직하게는 PP1 감마의 mRNA에 특이적인 안티센스 올리고 뉴클레오티드(antisense oligonucleotide), 앱타머(aptamer), siRNA(small interfering RNA), shRNA 및 miRNA를 포함하나, 이에 한정되지 않는다.
상기 바람직한 올리고 뉴클레오티드는 shRNA일 수 있다. 더욱 구체적으로는, 서열번호 2 내지 32로 이루어진 군으로부터 선택된 어느 하나의 염기서열을 포함하는 올리고 뉴클레오티드의 shRNA 일 수 있다. 구체적으로 상기 서열번호 2 내지 32로 이루어진 군으로부터 선택된 어느 하나의 염기서열을 포함하여 PP1 감마에 상보적으로 작용할 수 있는 shRNA라면 상기 염기서열의 앞 또는 뒤에 뉴클레오티드가 추가로 삽입된 형태도 모두 본 발명에 포함될 수 있다.
상기 shRNA는 본 발명의 구체적인 일 예에 따라, 바람직하게는 서열번호 2 내지 32로 이루어진 군으로부터 선택된 어느 하나의 염기서열로 이루어진 shRNA 일 수 있다.
본 발명의 일 실시예에서, PP1 감마의 mRNA 발현 수준을 억제하는 shRNA를 이용하여, 세포 사멸 억제, 미토콘드리아 단편화 억제 및 신경세포돌기의 변성이 억제되는 것을 실험적으로 확인하였다. 특히, PP1의 다른 촉매 소단위체인 알파 또는 베타와 비교해서 더욱 우수하게 신경세포의 대사를 정상화하면서, 세포 사멸 등의 부정적인 효과가 나타나지 않아, 퇴행성 신경질환의 특이적 치료 표적임을 확인하였다.
본 발명에서 "안티센스 올리고뉴클레오티드"는 특정 mRNA의 서열에 상보적인 핵산 서열을 함유하고 있는 DNA, RNA 또는 이들의 유도체를 의미하는 것으로, mRNA 내의 상보적인 서열에 결합하여 mRNA의 단백질로의 번역을 저해하는 작용을 한다. 본 발명에서 상기 PP1 감마 mRNA의 발현을 저해하는 올리고 뉴클레오티드는 PP1 감마 mRNA에 상보적인 서열을 갖는 DNA, RNA 또는 이들의 유도체로서 상기 PP1 감마의 mRNA에 결합하여, 번역, 세포질 내로의 전위(translocation), 성숙(maturation) 등을 방해하거나 또는 PP1 감마 mRNA의 그 밖의 다른 모든 생물학적 기능 또는 활성을 저해할 수 있다.
상기 안티센스 올리고 뉴클레오티드의 길이는 특별히 제한되는 것은 아니고, 바람직하게는 6 내지 100 염기일 수 있고, 보다 바람직하게는 8 내지 60 염기 일 수 있으며, 더욱 바람직하게는 10 내지 40 염기일 수 있다. 상기 안티센스 올리고 뉴클레오티드는 당해 기술 분야에서 사용되는 통상의 방법에 따라 생체 내(in vivo)에서 합성되거나, 시험관(in vitro)에서 합성되어 생체 내로 투여될 수 있다. 생체 내에서 안티센스 RNA를 합성하는 방법의 비제한적인 예로 다중 클로닝 부위(Multi-cloning site; MCS)의 기원(origin)이 반대 방향에 있는 벡터를 사용하여 안티센스 RNA가 전사되도록 하는 방법을 들 수 있다. 시험관 내에서 안티센스 RNA를 합성하는 방법의 비제한적인 예로 RNA 중합효소 I를 이용하는 방법이 있다.
본 발명에서, "앱타머"는 RNA, DNA, 변형된(modified) 핵산 또는 이들의 혼합물일 수 있으며, 그 형태가 직쇄상 또는 환상(環狀)일 수 있으나 이들에 한정되지 아니한다. 본 발명의 PP1 감마 mRNA의 발현을 억제하는 앱타머는 PP1 감마의 염기 서열을 참조하여 당해 기술 분야에서 통상의 지식을 가진 자가 공지의 방법에 따라 쉽게 제작할 수 있다.
본 발명에서, "siRNA"는 RNA 간섭(RNA interference) 또는 유전자 사일런싱(silencing)을 매개할 수 있는 핵산 분자로서, 19 내지 27 뉴클레오티드 크기의 작은 RNA 조각을 의미한다. 상기 siRNA는 표적 유전자의 발현을 저해할 수 있기 때문에 효율적인 유전자 넉다운(knockdown) 방법 또는 유전자 치료 방법으로 사용될 수 있다. 이중 가닥의 RNA가 다이서(dicer)에 의해 절단되어 생성될 수 있는 상기 siRNA는 상보적인 서열을 갖는 mRNA에 특이적으로 결합하여 해당 mRNA의 발현을 억제할 수 있다.
본 발명에서, "shRNA(small hairpin RNA)"는 RNA 간섭(interference) 또는 유전자 사일런싱(silencing)을 매개할 수 있는 핵산 분자를 의미한다. 벡터를 이용하여 세포 내로 도입될 수 있고, 세포 내로 도입된 shRNA 발현 플라스미드는 짧은 헤어핀구조의 shRNA를 발현하게 되며, 이 shRNA는 세포 안의 절단 효소에 의하여 21 내지 23 뉴클레오티드 크기의 siRNA로 프로세싱된다. 이렇게 형성된 siRNA가 타겟으로 하는 mRNA의 발현을 저해하는 RNAi 현상을 유도한다.
상기shRNA는 본 발명에서 인간 PP1 감마 mRNA에 특이적으로 결합할 수 있는 서열을 갖는 것으로, PP1 감마 mRNA의 발현을 저해할 수 있는 것이라면 그 서열은 특별히 제한되지 않는다. 바람직하게는 서열번호 2 내지 32로 이루어진 군에서 선택된 염기서열을 포함하는 올리고 뉴클레오티드로 구성된 군으로부터 선택된 shRNA일 수 있다.
본 발명에서, "벡터"란 적당한 숙주세포에서 목적 단백질 또는 목적 RNA를 도입 시킬 수 있는 것을 의미하며, 도입 및 발현할 수 있는 발현벡터를 포함하며, 유전자 삽입물이 발현되도록 작동가능하게 연결된 필수적인 조절요소를 포함하는 유전자 작제물을 의미한다. 상기 벡터는 예를 들어 바이러스 벡터일 수 있다. 더욱 바람직하게는 유전자 발현이 안정적이고, 신경세포 등에 효과적으로 유전자를 전달할 수 있는 렌티 바이러스 벡터(Lenti Viral Vector), 아데노 바이러스 벡터(Adono Viral Vector) 또는 아데노-관련 바이러스 벡터(Adeno-associated Viral Vector, AAV) 일 수 있다. 바람직하게는 FUGW벡터 등을 포함한 렌티 바이러스 벡터 또는 아데노-관련 바이러스 벡터 일 수 있다. 렌티바이러스를 벡터로 사용하는 경우 유전자 전달 효율이 높고, 면역 반응 부작용이 거의 없다는 장점이 있다. 또한, 아데노-관련 바이러스 벡터를 사용하는 경우 신경세포(뉴런)에 더욱 효과적으로 전달할 수 있고, 바이러스의 크기가 작고 구조가 단순하여 숙주세포의 면역반응을 줄일 수 있는 장점이 있다.
본 발명에서 "프로모터"는 적당한 숙주 세포에서 작동 가능하게 연결된 다른 핵산 서열의 발현을 조절하는 핵산 서열을 의미하며, 본 발명에서 "작동가능하게 연결(operably linked)"은 일반적 기능을 수행하도록 핵산 발현조절 서열과 목적하는 단백질 또는 RNA를 코딩하는 핵산 서열이 기능적으로 연결(functional linkage)되어 있는 것을 말한다. 예를 들어 프로모터와 단백질 또는 RNA를 코딩하는 핵산 서열이 작동가능하게 연결되어 코딩서열의 발현에 영향을 미칠 수 있다. 발현 벡터와의 작동적 연결은 당해 기술분야에서 잘 알려진 유전자 재조합 기술을 이용하여 제조할 수 있으며, 부위-특이적 DNA 절단 및 연결은 당해 기술 분야에서 일반적으로 알려진 효소 등을 사용한다.
상기 프로모터는 구성적 또는 유도성일 수 있으며, 바람직하게는 특정한 조직에서 특이적으로 활성화될 수 있으며, 더욱 바람직하게는 신경세포에 대해 특이적으로 활성화되는 프로모터 일 수 있다.
본 발명에서, "miRNA(micro RNA)"는 세포 내에서 자연적으로 존재하는 물질로, RNAi 현상을 유도하여 특정한 유전자의 조절에 관여하는 물질을 의미한다.
상기 mRNA의 발현을 억제하는 물질은 PP1 감마 mRNA의 염기 서열을 참조하여 당해 기술분야에서 통상의 지식을 가진 자가 통상의 방법에 따라 쉽게 제작할 수 있다. 상기 제작 방법의 비제한적인 예로는 직접 화학적으로 합성하는 방법, 시험관 내 전사를 이용하여 합성하는 방법, 시험관 내 전사에 의하여 합성된 RNA를 전달하여 제작하는 방법 등이 있다.
본 발명에서 사용되는 용어 "항체"는 면역학적으로 특정 항원과 반응성을 갖는 면역 글로불린 분자를 포함하는 단백질 분자로, 체내에 특정 항원이 침입한 경우 이를 특이적으로 인식하여 반응하는 항원 수용체 역할을 하는 단백질 분자를 말한다. 하나의 항체 분자는 두 개의 중쇄(heavy chain) 및 두 개의 경쇄(light chain)로 구성되어 있으며, 이들 각각의 중쇄 및 경쇄는 가변 영역과 고정 영역을 포함한다. 상기 가변 영역은 3개의 상보성 결정 부위(Complementarity Cetermining Region: CDR) 및 4개의 구조 형성 부위(Framework Region; FR)를 포함하며, 상기 상보성 결정 부위들에 의해 항체와 항원 사이의 결합 부위가 형성되어 특정 항원에 대한 항체의 결합 특이성이 생성될 수 있다.
본 발명에서 사용될 수 있는 항체는 PP1 감마 단백질의 활성을 억제하는 항체라면 특별히 제한되지 아니하며, 예를 들어, 다클론 항체, 단일클론 항체 또는 항체 단편 등을 모두 포함할 수 있다. 또한, 키메라성 항체 또는 이종 결합 항체 등과 같이 유전 공학적으로 제작된 항체들도 모두 포함할 수 있다.
본 발명에서, "대상체"는 특정 질병, 장애 또는 질환에 걸린 포유동물과 같은 온혈 동물을 의미하며, 예를 들어, 인간, 오랑우탄, 침팬지, 마우스, 렛트, 개, 소, 닭, 돼지, 염소, 양 등을 포함하나, 이들 예에 한정되는 것은 아니다.
본 발명에서, "치료"는 증상을 경감시키거나, 증상의 원인을 일시적 또는 영구적으로 제거하거나 증상의 출현 및 상기한 질병, 장애 또는 질환의 진행을 예방 또는 둔화시키는 것을 포함하나 이에 제한되지 않는다.
본 발명에서, "예방"은 질환의 발병을 억제시키거나 지연시키는 모든 행위를 의미한다.
구체적으로, 본 발명의 약학적 조성물의 유효성분의 유효량은 질환의 치료를 이루는데 요구되는 양을 의미한다. 따라서, 질환의 종류, 질환의 중증도, 조성물에 함유된 유효 성분 및 다른 성분의 종류 및 함량, 제형의 종류 및 환자의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료 기간, 동시 사용되는 약물을 비롯한 다양한 인자에 따라 조절될 수 있다.
본 발명의 약학적 조성물은 유효 성분 이외에 약제학적으로 허용가능한 담체를 1종 이상 포함한다. 본원에서 사용되는 바와 같이, '약제학적으로 허용가능한 담체'는 투여용 약제학적 활성 화합물을 제형화할 경우에 유용하고 사용 조건하에 사실상 비독성 및 비민감성인 공지된 약제학적 부형제를 의미한다. 이러한 부형제의 정확한 비율은 유효성분의 용해도와 화학적 특성, 선택된 투여경로뿐만 아니라, 표준 약제학적 관행에 의해 결정된다.
상기 유효성분은 공지의 퇴행성 뇌질환에 대한 치료제와 병행하여 사용될 수 있다.
또한, 본 발명에서 PP1 감마의 특이적 억제 시 미토코드리아 단편화, 신경세포 돌기 변성 및 세포 사멸 등이 억제되는 것을 확인하였으므로, 미지의 물질들 중에서 PP1 감마의 발현 수준을 억제하는 물질을 선택함으로써 퇴행성 신경질환 치료제 후보 약물을 선별할 수 있다.
따라서, 본 발명은 PP1 감마(Protein Phosphatase 1 gamma)를 포함하는 시료에 운동뉴런질환 또는 퇴행성 신경질환 치료제 후보물질을 처리하는 단계; 상기 후보물질이 PP1 감마의 발현을 증가 또는 감소시키는지를 판단하는 단계; 및 상기 PP1 감마의 발현을 감소시키는 후보물질을 퇴행성 신경질환 치료용 물질로 선별하는 단계;를 포함하는 퇴행성 신경질환 치료제 스크리닝 방법을 제공한다. 본 발명에서 PP1의 촉매 소단위체 중에서 감마의 발현을 특이적으로 억제하면 미토콘드리아 단편화와 신경세포의 축삭 변성 등을 효과적으로 억제하여 신경세포의 사멸을 감소시킬 수 있음을 확인하였는바, 본 발명의 스크리닝 방법에 따라 선별된 상기 PP1 감마의 발현을 감소시키는 후보물질은 퇴행성 신경질환을 치료할 수 있는 물질로 효과적으로 사용될 수 있다.
구체적으로, 본 발명은 PP1 감마 단백질을 시험관 내에서 후보물질과 접촉시키고; 상기 후보물질이 상기 단백질의 기능 또는 활성을 증가시키는지 또는 감소시키는지를 판단하는 것을 포함하는 퇴행성 신경질환 예방 또는 치료용 의약의 스크리닝 방법을 제공한다.
마찬가지로, 본 발명은 PP1 감마 유전자의 mRNA 또는 이를 발현하는 세포를 시험관 내에서 후보물질과 접촉시키고, 상기 후보물질이 상기 유전자의 발현을 증가시키는지 또는 감소시키는지를 판단하는 것을 포함하는 퇴행성 신경질환 예방 또는 치료용 의약의 스크리닝 방법을 제공한다.
상기 PP1 감마의 발현은 PP1 감마의 단백질 발현량 또는 PP1 감마의 유전자의 mRNA 발현량을 측정함으로써 수행할 수 있다.
본 발명에서 "단백질 발현량 측정"이란 생물학적 시료의 유전자에서 발현된 단백질의 발현량을 확인하는 과정으로, 본 발명의 일 구현예에 따르면 상기 단백질에 대해 특이적으로 결합하는 항체 또는 그 항체의 항원결합단편을 이용하여 단백질의 발현량을 확인할 수 있다. 이를 위한 분석방법으로는 웨스턴블랏팅, ELISA(enzyme linked immunosorbent assay), 방사선 면역분석(RIA: Radioimmunoassay), 방사 면역 확산법(radioimmunodiffusion), 오우크테로니(Ouchterlony) 면역 확산법, 로케트(rocket) 면역전기영동, 조직면역 염색, 면역침전 분석법(Immunoprecipitation Assay), 보체 고정 분석법(Complement Fixation Assay), FACS, 단백질 칩(protein chip) 등이 있으나, 이에 한정되는 것은 아니며, 이러한 분석방법은 당해 기술분야에 통상의 지식을 가진 자가 공지된 기술을 이용하여 적절히 수행할 수 있다.
본 발명에서 "mRNA 발현량 측정"이란 생물학적 시료에서 퇴행성 질환 마커 유전자의 mRNA 발현 정도를 확인하는 것으로, 구체적으로 mRNA에 대한 프라이머 또는 프로브를 이용하여 측정할 수 있다.
본 발명에서 "프라이머"는 짧은 자유 3-말단 수산화기(free 3' hydroxyl group)를 가지는 핵산 서열로 상보적인 템플레이트(template)와 염기쌍을 형성할 수 있고 템플레이트 가닥 복사을 위한 시작 지점으로서 작용하는 짧은 핵산서열을 말한다. 프라이머는 적절한 완충용액 및 온도에서 중합반응을 위한 제제(즉, DNA 폴리머레이트 또는 역전사효소) 및 상이한 4 가지의 뉴클레오사이드 트리포스페이트의 존재 하에서 DNA 합성을 개시할 수 있다.
본 발명에서 "프로브"란 mRNA에 특이적으로 결합을 이룰 수 있는 짧게는 수 염기 내지 길게는 수백 염기에 해당하는 RNA 또는 DNA 등의 핵산 단편을 의미하며 라벨링되어 있어서 특정 mRNA의 존재 유무, 함량(발현량)을 확인할 수 있다. 프로브는 올리고뉴클레오타이드(oligonucleotide) 프로브, 단쇄 DNA(single strand DNA) 프로브, 이중쇄 DNA(double strand DNA)프로브, RNA 프로브 등의 형태로 제작될 수 있다. 적절한 프로브의 선택 및 혼성화 조건은 당해 기술분야에 공지된 기술에 따라 적절히 선택할 수 있다.
상기 프라이머 또는 프로브는 포스포아미다이트(phosphoramidite) 고체 지지체 합성법이나 기타 널리 공지된 방법을 이용하여 화학적으로 합성할 수 있다. 이러한 핵산 서열은 또한 당해 기술분야에 공지된 다양한 방법을 통해 변형시킬 수 있다. 이러한 변형의 예로는 메틸화, 캡화, 천연 뉴클레오티드 하나 이상의 동족체로의 치환, 및 뉴클레오티드 간의 변형, 예를 들면 하전되지 않은 연결체(예: 메틸 포스포네이트, 포스포트리에스테르, 포스포로아미데이트, 카바메이트 등) 또는 하전된 연결체(예: 포스포로티오에이트, 포스포로디티오에이트 등)로의 변형이 있다.
상기 단백질 또는 유전자와 후보물질 간의 반응 확인은 단백질-단백질, 단백질-화합물, DNA-DNA, DNA-RNA, DNA-단백질, DNA-화합물, RNA-단백질, RNA-화합물 간의 반응여부를 확인하는데 사용되는 통상의 방법들을 사용할 수 있다.
예를 들면, 생체 외부에서(in vitro) 상기 유전자와 후보 물질 사이의 결합 여부를 확인하기 위한 혼성화 시험, 포유류 세포와 시험대상물질을 반응시킨 후 RT-PCR, 경쟁적 RT-PCR(competitive RT-PCR), 실시간 RT-PCR(Real-time RT-PCR), RNase 보호 분석법(RPA: RNase protection assay), 노던 블랏팅(Northern blotting), 및 DNA 칩 등을 통한 상기 유전자의 발현율 측정 방법, 또는 상기 유전자에 리포터 유전자를 연결시켜 세포 내로 도입한 후 시험대상물질과 반응시키고 리포터 단백질의 발현율을 측정하는 방법, 상기 단백질과 후보 물질을 반응시킨 후 활성을 측정하는 방법, 효모 이중 혼성법(yeast two-hybrid), 단백질에 결합하는 파지 디스플레이 펩티드 클론(phage-displayed peptide clone)의 검색, 천연물 및 화학물질 라이브러리(chemical library) 등을 이용한 HTS(high throughput screening), 드럭 히트 HTS(drug hit HTS), 세포 기반 스크리닝(cell-based screening), 또는 DNA 어레이(DNA array)를 이용하는 스크리닝법 등을 사용할 수 있다.
또한, 상기 단백질 발현량의 측정은 웨스턴블랏팅, 자석비드-항체면역침강법, ELISA, 및 질량분석기에서 선택된 하나 이상의 방법에 의한 것 일 수 있다.
본 발명의 스크리닝 방법에서, 시험대상물질(후보물질)은 통상적인 선정방식에 따라 신경세포에 대한 조절기능을 가진 의약으로서의 가능성을 지닌 것으로 추정되거나, 무작위적으로 선정된 개별적인 핵산, 단백질, 펩타이드, 기타 추출물 또는 천연물, 화합물 등이 될 수 있다.
본 발명에서 유전공학적 기술과 관련된 사항은 샘브룩 등의 문헌(Sambrook, et al. Molecular Cloning, A Laboratory Manual, Cold Spring Harbor laboratory Press, Cold Spring Harbor, N. Y. (2001)) 및 프레드릭 등의 문헌(Frederick M. Ausubel et al., Current protocols in molecular biology volume 1, 2, 3, John Wiley & Sons, Inc. (1994))에 개시되어 있는 내용에 의해 보다 명확하게 된다.
상기 기재 외에도, 당업계에 공지된 기술에 따라 PP1 감마의 발현의 측정에 사용하는 제제, 시약 및 방법이 적용될 수 있다.
또한, 본 발명은 PP1 감마를 포함하는 퇴행성 신경질환 치료제의 스크리닝용 조성물을 제공한다.
상기 스크리닝용 조성물은 PP1 감마의 발현량을 측정하는 제제를 더 포함할 수 있다.
본 발명에서 "PP1 감마의 발현량을 측정하는 제제"는 PP1 감마 mRNA 또는 단백질의 발현 수준을 확인함으로써 마커의 검출 및/또는 정량에 사용될 수 있는 분자를 의미한다. 따라서, 상기 PP1 감마의 발현량을 측정하는 제제는 상기 PP1 감마를 단백질 또는 mRNA 수준에서 측정할 수 있는 제제 일 수 있다.
구체적인 예로, 상기 단백질 수준에서 발현량을 측정할 수 있는 제제는 모노클로날 항체, 폴리클로날 항체, 기질, 앱타머, 수용체, 리간드 또는 보조인자 및 질량분광분석기 검출용 제제로 이루어진 군에서 선택된 것일 수 있다.
상기 "항체의 항원결합단편"이란 2 개의 경쇄 및 2 개의 중쇄를 갖는 완전한 형태의 항체의 구조를 갖지는 않으나, 항원성 부위에 대해 지시되는 특이적인 항원결합부위(결합 도메인)를 가져 항원 결합 기능을 보유하고 있는 항체 분자의 기능적 단편을 의미한다. 예를 들어 Fab, F(ab'), F(ab')2 및 Fv 등이 있다. 상기 항원결합 단편은 최소한 7개 이상의 아미노산을 포함한다.
상기 PP1 감마 mRNA의 발현량을 측정하는 제제는 역전사 중합효소 반응, RNase 보호 분석법 및 노던 블랏에 사용되는 제제로 이루어진 군에서 선택된 것 일 수 있다. 보다 구체적인 구현예에 따르면, mRNA에 특이적으로 결합하는 프라이머 쌍 및 프로브를 포함할 수 있다. 본 발명의 통상의 기술자는 이미 알려진 인간 PP1 감마의 염기서열을 바탕으로 이에 상보적으로 결합하는 프라이머 또는 프로브를 디자인 할 수 있다.
상기 스크리닝용 조성물은 상기 단백질 또는 유전자 외에도, 핵산 또는 단백질의 구조를 안정하게 유지시키는 증류수 또는 완충액을 포함할 수 있다. 또한, 상기 스크리닝용 조성물은 생체 내(in vivo) 실험을 위해, 상기 단백질 또는 유전자를 발현하는 세포, 또는 전사율을 조절할 수 있는 프로모터 하에 상기 유전자를 발현하는 플라스미드를 함유하는 세포 등을 포함할 수 있다.
또한, 본 발명은 PP1 감마 및/또는 PP1 감마의 발현량을 측정하는 제제를 포함하는 퇴행성 신경질환 치료제의 스크리닝용 키트를 제공한다.
상기 약학적 조성물, 스크리닝 방법 및 스크리닝용 조성물에 대한 내용은 스크리닝용 키트에도 준용될 수 있다.
상기 키트가 PP1 감마 mRNA의 발현량을 측정하기 위한 키트일 경우, RT-PCR을 수행하기 위해 필요한 필수 요소를 포함할 수 있다. RT-PCR 키트는 마커 유전자의 mRNA에 대한 특이적인 각각의 프라이머쌍 이외에도 테스트 튜브 또는 다른 적절한 컨테이너, 반응 완충액, 데옥시리보뉴클레오티드(dNTPs), Taq-폴리머라제 및 역전사효소와 같은 효소, DNase, RNase 억제제, DEPC-수(DEPC-water), 멸균수 등을 포함할 수 있다. 또한, 정량 대조군으로 사용되는 유전자에 특이적인 프라이머 쌍을 포함할 수 있다.
다른 예로, 상기 키트는 PP1 감마 단백질에 대해 특이적으로 결합하는 항체, 항체의 면역학적 검출을 위하여 기질, 적합한 완충용액, 발색 효소 또는 형광물질로 표지된 2차 항체, 발색 기질을 포함할 수 있다. 상기 기질은 니트로셀룰로오스 막, 폴리비닐 수지로 합성된 96 웰 플레이트, 폴리스티렌 수지로 합성된 96 웰 플레이트 및 유리 슬라이드글라스 등이 이용될 수 있고, 발색 효소는 퍼옥시다아제(peroxidase), 알칼라인 포스파타아제(alkaline phosphatase)가 사용될 수 있고, 형광물질은 FITC, RITC 등이 사용될 수 있고, 발색 기질은 ABTS(2,2'-아지노-비스(3-에틸벤조티아졸린-6-설폰산)) 또는 OPD(o-페닐렌디아민), TMB(테트라메틸 벤지딘) 등이 사용될 수 있다.
또 다른 예로, 본 발명의 키트는 단백질 또는 그 단백질을 코딩하는 유전자의 mRNA 발현량을 측정할 수 있는 마이크로어레이일 수 있다. 상기 마이크로어레이는 상기 본 발명의 마커를 이용하여 당해 기술분야에 공지된 방법에 따라 당업자가 용이하게 제조할 수 있으며, 일 예로 상기 PP1 감마 단백질을 코딩하는 유전자의 mRNA 또는 그의 단편에 해당하는 서열의 cDNA가 프로브로서 기판에 부착되어 있는 마이크로어레이일 수 있다.
또한, 상기 키트는 단백질 발현 수준 또는 mRNA 발현 수준을 측정하기 위한 하나 이상의 다른 마커, 구성성분, 시약, 용액 또는 장치 등을 더 포함할 수 있다. 상기 시약은 구체적으로, 질량분광분석기 검출용 시약, 또는 역전사 중합효소반응, RNase 보호 분석법, 노던 블랏에 사용되는 시약 일 수 있다.
다만, 본 발명은 다양한 변경을 가할 수 있고 여러 가지 형태를 가질 수 있는바, 이하에서 기술하는 특정 실시예 및 설명은 본 발명의 이해를 돕기 위한 것일 뿐, 본 발명을 특정한 개시 형태에 대해 한정하려는 것이 아니다. 본 발명의 범위는 본 발명의 사상 및 기술 범위에 포함되는 모든 변경, 균등물 내지 대체물을 포함하는 것으로 이해되어야 한다.
이하, 본 발명을 제조예 및 실험예를 통해 상세히 설명한다. 하기 실시예 및 실험예는 본 발명을 예시하는 것일 뿐 본 발명의 범위가 이들에 한정되는 것은 아니다.

프로테인
포스파타아제 1의 유전자 및 단백질 발현을 억제하기 위하여 shRNA 제작
근위축성 측삭경화증 생쥐 모델에서 프로테인 포스파타아제 1의 영향 및 주요 타겟을 찾기 위하여, PP1의 유전자 및 단백질 발현을 억제할 수 있는 shRNA를 제작하였다. 구체적으로, 프로테인 포스파타아제 1(Protein phosphatase 1, PP1)의 촉매 소단위체(catalytic subunit)는 알파(alpha, α), 베타(beta, β), 감마(gamma, γ)의 세 동형체(isoform)가 존재하므로, 이들에 대한 선별적으로 단백질 발현을 억제할 수 있는 각각의 shRNA를 제작하였다(도 2).
구체적으로, Thermofisher scientific의 BLOCK-iT™ RNAi Designer 프로그램을 이용하여 가장 유력한 후보 시퀀스 서열을 선별하여 각각의 시퀀스를 FUGW 벡터에 클로닝하였다. 클로닝된 벡터는 생쥐의 대뇌신경세포에서 발현시킨 뒤, PP1 알파, 베타, 감마의 발현을 효과적으로 감소시키는 shRNA를 선별하였다.
상기에서 제작된 shRNA의 서열을 사람의 PP1의 유전자 서열과 비교해본 결과, PP1 α와 γ는 매우 유사함을 확인 하였다.
또한, 각각 제작된 shRNA가 각각의 PP1 동형체의 단백질 형성을 억제할 수 있는지 확인하기 위하여, 웨스턴 블랏을 이용하여 단백질 발현 패턴을 관찰하였다.
구체적으로, PP1 알파, 베타 그리고 감마에 대한 shRNA를 포함하는 FUGW 벡터를 이용하여, 렌티 바이러스(lenti virus)를 제작하여, 마우스 대뇌 신경세포에 감염(infection) 시키고, 3일 뒤에 샘플을 용균(lysis) 하였다. 각각의 샘플을 BCA 방법을 이용하여 단백질을 정량 한 후, 12% 아크릴 아미드 젤에 로딩하였다. 로딩된 단백질을 PVDF 막(poly(vinylidenedifluoride) membrane)으로 트랜스퍼(transfer) 시키고, 각각의 PP1 알파, 베타, 감마의 일차 항체와 반응시켰다. 항체 반응이 끝난 후, ECL(electro chemical luminescence)을 이용하여 단백질의 발현량을 조사하였다. 그 결과를 도 3에 나타내었다.
도 3에 나타낸 바와 같이, shPP1-알파, shPP1-베타 및 shPP1-감마 모두 각각의 PP1 동형체에 대해서만, 특이적으로 단백질 발현을 억제함을 확인 하였다.

시험예
1] 미토콘드리아 단편화에 미치는 효과 확인
실시예 1에서 제조한 shRNA가 근위축성 측삭경화증의 병리적 현상중에 하나인 미토콘드리아 단편화에 영향을 미치는지 확인하기 위하여, 각각의 PP1 동형체(isoform)의 활성을 억제하는 shRNA를 이용하여 실험을 수행하였다.
구체적으로, 근위축성 측삭경화증을 유발하지 않는 그룹(GFP), SOD1에서 G93A 변이를 일으켜 근위축성 측삭경화증을 유발한 그룹(G93A), 그리고 TDP-43에서 Q331K 변이를 일으켜 근위축성 측삭경화증을 유발한 그룹(Q331K)으로 나누고, 각 군에서 shRNA 없이 GFP 벡터만 삽입한 대조군(control)과 shPP1-알파, -베타, -감마(FUGW-GFP vector)를 과발현 시킨 실험군으로 나누어 미토콘드리아의 형태 및 길이에 미치는 영향을 확인하였다.
4일 배양된 생쥐의 대뇌피질에 근위축성 측삭경화증을 유발하는 SOD1-G93A와 TDP-43-Q331K 클론과 shPP1-알파, shPP1-베타 또는 shPP1-감마를 동시에 과발현 시키고(FUGW-GFP vector 이용), 모든 그룹에 미토콘드리아를 표지하는 클론(dsRed-mito)을 같이 발현 시킨 뒤, 3일 후에 미토콘드리아의 길이를 관찰하였다. 형광현미경의 x1000배를 이용하여 미토콘드리아의 형태의 이미지를 확보하고 Image J프로그램을 이용하여 그 길이를 측정하였다. 그 결과를 분석하여 도 4에 나타내었다.
도 4에 나타낸 바와 같이, shPP1-알파와 shPP1-감마가 근위축성 측삭경화증을 유발하는 두 가지 돌연변이(SOD-G93A, TDP43-Q331K)에 의해 일어나는 미토콘드리아 단편화를 억제함을 관찰할 수 있었다.

시험예
2] 세포사멸에 미치는 효과를 확인
실시예 1에서 제조한 shRNA가 근위축성 측삭경화증의 병리적 현상 중에 하나인 세포 사멸에 미치는 효과를 확인하였다. 실험군은 시험예 1과 동일하게 설정하였다.
구체적으로, 근위축성 측삭경화증을 유발한 그룹(SOD1-G93A, TDP-43-Q331K)에 shRNA 없이 GFP 벡터만 삽입한 대조군(control)과 shPP1-알파, -베타 또는 -감마를 과발현(FUGW-GFP vector 이용) 시킨 실험군으로 나누어 세포사멸을 분석하였다.
4일 배양된 생쥐의 대뇌피질에 근위축성 측삭경화증을 유발하는 SOD1-G93A와 TDP-43-Q331K 클론과 shPP1-알파, shPP1-베타 또는 shPP1-감마를 동시에 과발현(FUGW-GFP vector 이용) 시키고, 3일 뒤에 신경세포를 고정하였다. 고정 후 활성화된 Caspase-3의 발현을 보기 위하여, 항체를 이용하여 면역 염색을 실시하였고, 핵을 염색하기 위하여 Hoechst33342를 이용하여 색혈세포를 분석하였다. 형광현미경(200배)을 이용하여 염색된 신경세포의 이미지를 확보하였고, Caspase-3와 색혈세포의 수를 측정하였다. 그 결과를 분석하여 도 5에 나타내었다.
도 5에 나타낸 바와 같이, G93A 또는 Q331K 돌연변이에 의해 일어나는 세포 사멸을 관찰한 결과, shPP1-알파의 경우 근위축성 측삭경화증 유발 그룹에서는 세포사멸의 효과가 적고, 정상그룹(GFP)에서는 오히려 세포 사멸을 촉진하는 작용을 하는 것을 확인하였고, shPP1-베타 및 shPP1-감마가 세포 사멸을 효과적으로 억제하는 것을 확인하였다.

시험예
3] 신경돌기 변성 억제 효과 확인
실시예 1에서 제조한 shRNA가 근위축성 측삭경화증의 병리적 현상 중 하나인 신경돌기 변성 억제를 확인하는 실험을 수행하였다. 실험군은 시험예 1과 동일하게 설정하여 수행하였다.
구체적으로, 4일 배양된 생쥐의 대뇌피질에 근위축성 측삭경화증을 유발하는 SOD1-G93A와 TDP-43-Q331K 클론과, 각각의 그룹에 shPP1-알파, -베타 또는 -감마를 동시에 과발현(FUGW-GFP vector 이용) 시킨 뒤, 3일 후에 형광현미경을 이용하여 신경세포의 형태의 이미지를 얻었다. 상기 이미지에서 신경돌기의 길이는 Photoshop과 Image J 프로그램을 이용하여 측정하였다. 그 결과는 도 6에 나타내었다.
도 6에 나타낸 바와 같이, 각각의 PP1 동형체(isoform)에 대한 선택적으로 작용하는 shRNA를 이용하여 G93A 또는 Q331K에 의해 일어나는 신경돌기 변성을 관찰한 결과, shPP1-알파와 shPP1-감마가 총 신경돌기(total neurite)와 1차 축삭돌기(primary Axon)의 길이가 짧아지는 것을 감소시키는데 효과가 있음을 확인하였다.

Claims (17)
- PP1 감마의 mRNA 수준에서 발현을 억제하는 올리고 뉴클레오티드;, PP1 감마 단백질의 활성을 억제하는 항체; 및 상기 항체의 항원 결합 단편;으로 이루어진 군에서 선택된 PP1 감마(Protein Phosphatase 1 gamma)의 발현 저해제를 유효성분으로 포함하는 퇴행성 신경질환의 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서,
상기 퇴행성 신경질환은 운동뉴런질환, 알츠하이머병, 파킨슨병, 헌팅턴병, 전두측두치매, 루이체치매, 피질기저퇴행증, 다계통 측위병 및 진행성핵상마비로 이루어진 군에서 선택된 것인, 약학적 조성물. - 제1항에 있어서,
상기 운동뉴런질환(motor neuron disease)은 근위축성 측삭경화증(Amyotrophic lateral sclerosis-ALS), 원발성 측삭 경화증(Primary Lateral Sclerosis), 베르드니히-호프만 병(Werdnig-Hoffman disease) 또는 유아성 척수성 근육위축(SMA1), 쿠겔베르그-벨란더 증후군(Kugelberg-Welander syndrome) 또는 청소년형 척수성 근육위축(Juvenile for m, type III, SMA 3), 진행성 척수성 근육위축(Progressive spinal muscular atrophy), 진행성 구마비(Progressive bulbar palsy) 및 가족성 운동신경원 병(Familial motor neuron disease)으로 이루어진 군에서 선택된 것인 약학적 조성물. - 제1항에 있어서,
상기 PP1 감마의 mRNA 수준에서 발현을 억제하는 올리고 뉴클레오티드는 PP1 감마의 mRNA에 특이적인 안티센스 올리고 뉴클레오티드, 앱타머, siRNA, shRNA 및 miRNA로 이루어진 군에서 선택된 것인, 약학적 조성물. - 제1항에 있어서,
상기 PP1 감마는 서열번호 1의 염기서열을 갖는 것인, 약학적 조성물. - 제1항 또는 제4항에 있어서,
상기 PP1 감마의 mRNA 수준에서 발현을 억제하는 올리고 뉴클레오티드는 shRNA인, 약학적 조성물. - 제6항에 있어서,
상기 shRNA는 서열번호 2 내지 32로 이루어진 군으로부터 선택되는 염기서열을 포함하는 것 인, 약학적 조성물. - PP1 감마(Protein Phosphatase 1 gamma)를 포함하는 시료에 퇴행성 신경질환 치료제 후보물질을 처리하는 단계;
상기 후보물질이 PP1 감마의 발현을 증가 또는 감소시키는지 판단하는 단계; 및
상기 PP1 감마의 발현을 감소시키는 후보물질을 퇴행성 신경질환 치료용 물질로 선별하는 단계;를 포함하는 퇴행성 신경질환 치료제 스크리닝 방법. - 제8항에 있어서,
PP1 감마의 발현은 PP1 감마 단백질의 발현량 또는 PP1 감마 유전자의 mRNA 발현량인, 방법. - 제8항에 있어서,
상기 PP1 감마는 PP1 감마 단백질, 유전자 또는 상기 유전자를 발현하는 세포 인, 방법. - PP1 감마(Protein Phosphatase 1 gamma)를 포함하는 퇴행성 신경질환 치료제의 스크리닝용 조성물.
- 제11항에 있어서,
