Abstract
본 발명은 돌외 추출물 또는 지페노시드의 파킨슨병의 기억력 장애의 치료, 예방 또는 개선 용도에 관한 것이다. 본 발명의 돌외 추출물 또는 지페노시드는 파킨슨병의 진행 또는 L-DOPA 약물 치료에 의해 나타날 수 있는 기억력 장애의 치료, 예방 또는 개선 효과가 있다. 본 발명의 돌외 추출물 또는 지페노시드는 파킨슨 병 환자의 뇌에서 기억력과 관련되어 중요한 역할을 하는 부위인 해마 부위의 NMDA 수용체(NMDAR)의 증가된 활성을 감소시키고, ERK(extracellular signal-regulated kinase) 1/2 및 CREB(cyclic AMP-response element binding protein)의 감소된 활성을 증가시킴으로써, 기억력 개선 효과를 나타낸다. 본 발명의 돌외 추출물 또는 지페노시드는 천연물로부터 유래되었으므로 화학적으로 합성된 화합물과는 다르게 인체 적용시 부작용이 적다.
Description
본 발명은 돌외 추출물 또는 지페노시드를 유효성분으로 포함하는 파킨슨병에서 나타나는 기억력 장애의 치료 또는 예방용 약제학적 조성물에 관한 것이다. 보다 상세하게는, 본 발명은 돌외 추출물 또는 지페노시드를 유효성분으로 포함하는 파킨슨병 환자에게서 파킨슨병 치료약물인 L-DOPA(L-3,4-dihydroxyphenylalanine)의 장기간 투여 중에 나타나는 기억력 장애의 치료, 예방 또는 개선용 조성물에 관한 것이다.
파킨슨병(Parkinson's disease)은 대뇌 기저핵(basal ganglia)내 흑질(substantia nigra, pars compacta)의 도파민 신경세포(dopaminergic neuron)가 서서히 퇴행성 변화를 일으켜 선조체(corpus striatum)의 신경전달 물질인 도파민(dopamine)의 결핍으로 증상이 나타난다. 파킨슨병의 특징적인 운동장애는 진전(tremer), 서동(bradykinesia), 경직(rigidity) 등의 증상이 나타난다. 도파민 신경계의 도파민(dopamine)의 결핍은 신경세포사에 의하여 도파민 생합성 효소인 티로신 하이드록시라제(tyrosine hydroxylase, TH), 방향족 L-아미노산 디카복실라제(aromatic L-amino acid decarboxylase, AADC)의 활성 감소가 원인으로 알려져 있다.
파킨슨병의 병인론에 관해서는 유전적인 요인, 산화스트레스(oxidative stress), 유전적 민감성, 신경 세포의 괴사 등을 포함한 다양한 내적, 외적 발병 원인들이 보고되고 있다([1] Archibald and Burn, Medicine, 36, 630-635, 2008; [2] Yacoubian and Standaert, Biochim. Biophys. Acta, 1792, 676?687, 2009). 파킨슨병에서 나타나는 흑질의 도파민 신경세포는 질병이 진행되는 기간에 비례하여 계속 소실되는 것으로 알려져 있으며([3] Chen et al., Parkinson's disease, pp. 1033-1044, in Pharmacotherapy 8th Ed,, Eds., DiPiro et al., 2008), 현재까지 이러한 지속적인 신경세포의 손실을 막을 수 있는 뚜렷한 방법은 알려져 있지 않다.
파킨슨병의 주요 치료 방법은 L-DOPA(3,4-dihydroxyphenylalanine, levodopa, 레보도파) 약물 요법이다. L-DOPA 요법은 투여된 L-DOPA가 혈액-뇌 관문(blood-brain barrier)을 통과하여 뇌중에서 도파민으로 전환되어 작용하는 것으로서 파킨슨병의 증상(운동장애 등)을 호전시키는 치료 효과가 있다. 그러나, L-DOPA를 장기간 투여할 시 L-DOPA 치료에 대한 효능이 점점 없어지게 되고 증상은 악화되는 경과를 나타낸다([3] Chen et al., Parkinson's disease, pp. 1033-1044, in Pharmacotherapy 8th Ed,, Eds., DiPiro et al., 2008). 또한, 파킨슨병 환자에서 L-DOPA의 장기투여는 약물효능의 감소 뿐만 아니라, 운동이상증(dyskinesia), 운동동요(motor fluctuation) 및 다른 합병증을 유발한다([4] Jankovic, Mov. Disord., 20, S47-S55, 2005). 많은 파킨슨병 환자들은 운동장애 이외에 학습과 기억력 장애를 겪게 되며 질환이 진행될수록 이러한 증상들은 더 심해지게 된다([5] Buter et al., 2008; [6] Williams-Gray et al., 2007). 따라서 파킨슨병이 진행되는 동안 또는 L-DOPA 요법 등의 치료기간 동안 나타날 수 있는 다양한 부작용 중에서 기억력의 장애는 운동장애 증상을 포함하여 파킨슨병 환자에게는 삶의 어려움을 초래하므로, 환자에게 기억력의 개선효능을 나타내는 새로운 치료제의 개발이 필요하다.
학습과 기억력은 뇌중의 해마(hippocampus) 부위가 중요하게 관여하는 것으로 알려져 있으며, 특히 해마 부위에 존재하는 N-메틸-D-아스파테이트(N-methyl-D-aspartate, NMDA)의 수용체(NMDARs)가 중요한 역할을 하고 있다([7] Bannerman et al., 1995; [8] Michaelis, 1998). NMDA 수용체(NMDARs)는 뇌의 해마(hippocampus) 부위에 존재하는 글루타메이트(glutamate) 수용체의 한 종류로서 NR1, NR2, NR3의 소단위로 구성되어 있다([9] Das et al., 1998). 또한, 학습과 기억력과 관련하여, 해마 부위에서 ERK(extracellular signal-regulated kinase) 1/2 와 CREB(cyclic AMP-response element binding protein)의 역할에 대하여 많이 연구되고 있다. 해마의 ERK1/2 인산화가 학습과 기억력 과정에 필수적인 역할을 한다고 보고되었으며([10] Blum et al., 1999), ERK1/2의 아래 단계인 CREB도 해마에서 기억력과 관련되어 있다고 보고되었다([11] Carlezon et al., 2005). 그리고, ERK1/2 경로는 NMDARs에서 기억력 과정의 필수적인 요소 중 하나임이 보고되고 있다([12] Krapivinsky et al., 2003).
파킨슨병 동물모델에서 기억력이 감소되는 경우, 해마의 NMDARs의 활성은 증가하며, 이와 관련하여 ERK 활성 및 CREB 활성은 감소한다. 또한 행동학적 측정은 EPM(Elevated plus-maze test)로 시행하며, EPM의 Retention transfer latency는 initial transfer latency time와 비교하여 백분율(%ITL)로 나타내며, 이 수치는 파킨슨병 동물모델에서 기억력이 감소하는 경우 대조군에 비하여 증가한다([13] Kumar et al., 2011). 그러므로, 파킨슨병에서 장기간 L-DOPA 치료 중에 나타나는 기억력의 악화에 대하여 생리활성 물질이 기억력의 개선효능을 나타내기 위해서는, 활성물질이 해마부위의 NMDA 신경세포에 대한 보호작용을 나타내야 한다.
돌외(Gynostemma pentaphyllum Makino, GP)는 박과(Cucurbitaceae)에 속하는 다년생 식물로 국내 및 아시아 지역에 분포되어 있다. 돌외의 주성분은 지페노시드(gypenosides, GPS), 플라보노이드(flavonoids), 폴리사카라이드(polysaccharides), 아미노산 및 비타민 등을 함유하고 있다. 플라보노이드 화합물로는 옴부오사이드(ombuoside), 옴부인(ombuin) 등을 함유하고 있으며, 이외에 루틴(rutin), 수종의 스테롤(sterols) 등을 함유하고 있다([14] Razmovski-Naumovski et al., 2005). 돌외의 주성분인 사포닌화합물은 Gynostemma total saponin [GP 총사포닌, 또는 지노사포닌(gynosaponins), 지페노시드(gypenosides, GPS)]이라 한다. 돌외는 자생지역에 따라 차이가 있으나, 약 90여종의 사포닌이 확인되어 화학구조가 보고되고 있으며, 돌외의 사포닌 활성분획을 총칭하여 지페노시드(자이페노사이드, gypenoside-enriched components, GPS)라고 한다([14] Razmovski-Naumovski et al., 2005).

본 발명자들은 파킨슨병이 진행됨에 따라 또는 파킨슨병 환자에게 치료약물인 L-DOPA(3,4-dihydroxyphenylalanine)를 장기 투여하는 경우 나타나는 기억력 저하를 개선할 수 있는 천연물 유래 활성물질을 개발하기 위해 연구 노력하였다. 그 결과, 돌외 추출물 및 이로부터 유래하는 지페노시드가 파킨슨병 동물모델에서 이 질환이 진행됨에 따라 나타나는 기억력 장애의 증상을 개선하며, L-DOPA 장기 투여에 의해 악화되는 기억력 장애 증상을 현저히 개선할 수 있음을 실험적으로 증명하여 본 발명을 완성하였다.
따라서, 본 발명의 목적은 돌외(Gynostemma pentaphyllum Makino) 추출물 또는 지페노시드(gypenoside)를 유효성분으로 포함하는 파킨슨병에서 나타나는 기억력 장애의 치료 또는 예방용 약제학적 조성물을 제공하는 데에 있다.
본 발명의 다른 목적은 돌외 추출물 또는 지페노시드를 유효성분으로 포함하는 파킨슨병에서 나타나는 기억력 장애의 개선용 기능성 식품 조성물을 제공하는 데에 있다.
본 발명의 목적 및 장점은 하기의 발명의 상세한 설명, 청구의 범위 및 도면에 의해 보다 명확하게 된다.
본 발명의 일 양태에 따르면, 돌외(Gynostemma pentaphyllum Makino) 추출물 또는 지페노시드(gypenoside)를 유효성분으로 포함하는 파킨슨병에서 나타나는 기억력 장애의 치료 또는 예방용 약제학적 조성물을 제공한다.
본 발명의 다른 양태에 따르면, 돌외 추출물 또는 지페노시드를 유효성분으로 포함하는 파킨슨병에서 나타나는 기억력 장애의 개선용 기능성 식품 조성물을 제공한다.
본 발명에서 사용되는 돌외 추출물은 돌외 전체의 추출물, 또는 줄기, 뿌리, 잎, 꽃, 열매, 및 씨앗으로 구성된 군에서 선택된 돌외 일부의 추출물이며, 바람직하게는 돌외의 잎으로부터 얻은 추출물이다.
본 발명에서 돌외 추출물은 천연물로부터 추출물을 추출하는 당업계의 공지된 통상적인 방법에 따라, 즉, 통상적인 온도, 압력의 조건 하에서 통상적인 용매를 사용하여 추출할 수 있다. 바람직하게는 상기 돌외 추출물은 추출용매와 접촉시키는 공정을 통해 제조된다. 추출 단계 전에 상기 돌외 전체 또는 일부를 건조 및/또는 분쇄할 수 있다.
본 발명의 유효성분으로 이용되는 돌외 추출물을 얻기 위한 용매는 당업계에서 통상적으로 이용되는 어떠한 용매도 포함한다. 예를 들어, 상기 추출용매는 (a) 탄소수 1-4의 무수 또는 함수 저급 알코올 (예: 메탄올, 에탄올, 프로판올, 부탄올, 노말-프로판올, 이소-프로판올 및 노말-부탄올 등), (b) 상기 저급 알코올과 물과의 혼합용매, (c) 아세톤, (d) 에틸 아세테이트, (e) 클로로포름, (f) 1,3-부틸렌글리콜, (g) n-헥산, (h) 디에틸에테르, (i) 부틸아세테이트, (j) 디클로로메탄 또는 (h) 물을 포함하며, 바람직하게는 탄소수 1-4의 무수 또는 함수 저급 알코올, 아세톤, 또는 물이다. 한편, 본 발명의 추출물은 상기한 추출 용매 뿐만 아니라, 다른 추출 용매를 이용하여도 실질적으로 동일한 효과를 나타내는 본 발명의 추출물이 얻어질 수 있다는 것은 당업자에게 자명하다.
본 발명에서 추출공정은 추출용매를 부분적으로 또는 완전히 제거하는 단계를 통해 완성되며, 부분적 제거는 상당한 양의 유기용매가 없는 수성 농축물이 얻어질 때까지 농축하는 것을 의미하며, 완전한 제거에 의해서는 건조 잔류물을 얻을 수 있다.
본 명세서에서 사용되는 용어 "추출물" 은 상술한 바와 같이 당업계에서 조추출물(crude extract)로 통용되는 의미를 갖지만, 광의적으로는 상기 조추출물을 추가적으로 분획(fractionation)한 분획물도 포함하는 의미이다. 즉, 본 발명의 돌외 추출물은 상술한 추출 용매를 이용하여 얻은 것 뿐만 아니라, 여기에 정제과정을 추가적으로 적용하여 얻은 분획물도 포함한다. 예컨대, 상기 추출물을 일정한 분자량 컷-오프 값을 갖는 한외 여과막을 통과시켜 얻은 분획, 다양한 크로마토그래피(크기, 전하, 소수성 또는 친화성에 따른 분리를 위해 제작된 것)에 의한 분리, 상기 설명된 추출 용매에 의한 2차 또는 3차의 추가적인 추출 분획 등, 추가적으로 실시된 다양한 정제 방법을 통해 얻어진 분획도 본 발명의 추출물에 포함되는 것이다.
본 발명에서 지페노시드(gypenoside)는 상기한 돌외 추출물로부터 분리 정제하여 얻은 것 뿐만 아니라, 화학적으로 합성하여 얻은 것도 동일한 효과를 갖는다는 것은 당업자에게 자명하다.

본 발명에서 파킨슨병에서 나타나는 기억력 장애는 보다 바람직하게는 파킨슨병이 진행됨에 따라 나타나는 기억력 장애 또는 파킨슨병 환자에게 치료약물인 L-DOPA (3,4-dihydroxyphenylalanine)을 투여함에 의해 유발되는 기억력 장애이다.
본 발명의 바람직한 일 구현예에 따르면, 상기 파킨슨병의 기억력 장애는 파킨슨병의 치료약물인 L-DOPA(3,4-dihydroxyphenylalanine) 투여에 의해 유도되는 기억력 장애이다.
본 발명의 다른 바람직한 구현예에 따르면, 상기 돌외 추출물 또는 지페노시드는 뇌의 해마 부위에 존재하는 N-메틸-D-아스파테이트(N-methyl-D-aspartate, NMDA)의 수용체(NMDAR)의 증가된 활성을 감소시킨다.
본 발명의 또 다른 바람직한 구현예에 따르면, 상기 돌외 추출물 또는 지페노시드는 뇌의 해마 부위에 존재하는 ERK(extracellular signal-regulated kinase)1/2 및 CREB(cyclic AMP-response element binding protein)의 감소된 활성을 증가시킨다.
본 발명의 약제학적 조성물은 (a) 돌외 추출물 또는 지페노시드의 약제학적 유효량; 및 (b) 약제학적으로 허용되는 담체를 포함하는 파킨슨병에서 나타나는 기억력 장애의 치료 또는 예방용 약제학적 조성물 형태로 제공될 수 있다.
본 명세서에서 사용되는 용어 “치료”는 (i) 질환 또는 질병의 발전의 억제, (ii) 질환 또는 질병의 경감, 또는 (iii) 질환 또는 질환의 제거를 의미한다.
본 명세서에서 사용되는 용어 “예방”은 질환 또는 질병을 보유하고 있다고 진단된 적은 없으나, 이러한 질환 또는 질병에 걸리기 쉬운 경향이 있는 동물에서 질환 또는 질병의 발생을 억제하는 것을 의미한다.
본 발명의 약제학적 조성물에 포함되는 약제학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 탄수화물류 화합물(예: 락토스, 아밀로스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 셀룰로스, 등), 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 염 용액, 알코올, 아라비아 고무, 식물성 기름(예: 옥수수 기름, 목화 종자유, 두유, 올리브유, 코코넛유), 폴리에틸렌 글리콜, 메틸 셀룰로스, 메틸히드록시 벤조에이트, 프로필히드록시 벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다.
본 발명의 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약제학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다. 한편, 본 발명의 약제학적 조성물의 경구 투여량은 바람직하게는 1일 당 0.0001 - 100 mg/kg(체중)이다.
본 발명의 약제학적 조성물은 경구 또는 비경구로 투여할 수 있다. 비경구로 투여되는 경우, 정맥내 주입, 피하 주입, 근육 주입, 복강 주입, 경피 투여 등으로 투여할 수 있다.
본 발명의 약제학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나 엑스제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
본 발명의 조성물은 돌외 추출물 또는 지페노시드를 유효성분으로 포함하는 파킨슨병에서 나타나는 기억력 장애의 개선용 기능성 식품 조성물의 형태로 제조될 수 있다.
본 명세서에서 용어 “개선”은 질병으로 인한 증상 또는 질병의 합병증으로 인한 증상의 경감을 의미한다.
본 발명의 조성물이 기능성 식품 조성물인 경우, 본 발명의 조성물은 상기 유효성분 이외에, 식품 제조 시에 통상적으로 첨가되는 성분을 포함할 수 있으며, 예를 들어, 단백질, 탄수화물, 지방, 영양소, 조미제 및 감미제를 포함한다. 상술한 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스, 올리고당 등; 및 폴리사카라이드, 예를 들어 덱스트린, 사이클로덱스트린 등과 같은 통상적인 당 및 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 감미제로서 천연 감미제[타우마틴, 스테비아 추출물(예를 들어 레바우디오시드 A, 글리시리진 등) 및 합성 감미제(사카린, 아스파르탐 등)를 사용할 수 있다. 예컨대, 본 발명의 식품 조성물이 드링크제로 제조되는 경우에는 본 발명의 유효성분인 돌외 추출물 또는 지페노시드 이외에 구연산, 액상과당, 설탕, 포도당, 초산, 사과산, 과즙, 두충 추출액, 대추 추출액, 감초 추출액 등을 추가로 포함시킬 수 있다. 식품에 대한 용이한 접근성을 고려한다면, 본 발명의 기능성 식품 조성물은 파킨슨병에서 나타나는 기억력 장애의 예방 또는 개선에 매우 유용하다.
본 발명의 특징 및 이점을 요약하면 다음과 같다:
(i) 본 발명은 돌외 추출물 또는 지페노시드의 파킨슨병에서 나타나는 기억력 장애의 치료, 예방 또는 개선 용도에 관한 것이다.
(ⅱ) 본 발명의 돌외 추출물 또는 지페노시드는 파킨슨병이 진행하는 동안에 나타날 수 있는 기억력 장애 또는 치료 약물인 L-DOPA(3,4-dihydroxyphenylalanine)의 장기간 투여에 의해 유발되는 기억력 장애의 치료, 예방 또는 개선 용도에 관한 것이다.
(ⅲ) 본 발명의 돌외 추출물 또는 지페노시드는 파킨슨 환자의 뇌에서 기억력에 중요한 역할을 하는 부위인 해마 부위의 NMDA 수용체(NMDAR)의 증가된 활성을 감소시키고, ERK(extracellular signal-regulated kinase)1/2 및 CREB(cyclic AMP-response element binding protein)의 감소된 활성을 증가시킴으로써, 기억력 개선 효과를 나타낸다.
(ⅳ) 본 발명의 돌외 추출물 또는 지페노시드는 천연물로부터 유래되었으므로 화학적으로 합성된 화합물과는 다르게 인체 적용시 부작용이 적다.
본 발명은 돌외 추출물 또는 지페노시드의 파킨슨병의 기억력 장애의 치료, 예방 또는 개선 용도에 관한 것이다. 본 발명의 돌외 추출물 또는 지페노시드는 파킨슨병의 진행 또는 L-DOPA 약물 치료에 의해 나타날 수 있는 기억력 장애의 치료, 예방 또는 개선 효과가 있다. 본 발명의 돌외 추출물 또는 지페노시드는 파킨슨 병 환자의 뇌에서 기억력과 관련되어 중요한 역할을 하는 부위인 해마 부위의 NMDA 수용체(NMDAR)의 증가된 활성을 감소시키고, ERK(extracellular signal-regulated kinase) 1/2 및 CREB(cyclic AMP-response element binding protein)의 감소된 활성을 증가시킴으로써, 기억력 개선 효과를 나타낸다. 본 발명의 돌외 추출물 또는 지페노시드는 천연물로부터 유래되었으므로 화학적으로 합성된 화합물과는 다르게 인체 적용시 부작용이 적다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실험방법
1. 실험동물
실험동물(C57BL/6 마우스, 웅성, 20-25 g, Samtaco Co, 수원)를 사용하였으며, 주야 주기 12시간, 온도 23 2℃의 조건하에서 사육하였다. 모든 실험동물은 충북대학교 동물실험윤리위원회(CBNU Animal Care and Use Committee)의 취급기준에 의하여 수행되었다.

제조
돌외시료는 원광식품(경남 거창군 남상면 둔동리)에서 구입하여 품종을 확인하였다(표준품 보관: 충북대학교 약학대학 생약학교실). 돌외의 잎(leaves, 10 kg) 부위를 채취하여 음건한 후 세절한 다음 에탄올(ethanol, 80%)로 추출하고 증발 농축하여 시료로 사용하였다(GP-EX, 0.96kg). 지페노시드(GPS)는 Ankang Dongke Maidisen Nautre Pharmaceutical Co.(순도>99%, confirmed by HPLC analysis, 중국)에서 구입하였다([16] Wang et al., 2010; [17] Shang et al., 2006).
3. 실험계획
돌외추출물(GP-EX)이 MPTP-투여 파킨슨병 모델에서 L-DOPA 투여에 의해 나타나는 기억력 저하에 대한 개선 효능을 가지는 지를 검토하기 위하여, 다음과 같이 대조군 및 실험군을 나누어 실험을 진행하였다. 그룹 1 (대조군): 0.9% 생리식염수 투여; 그룹 2 (MPTP 투여군): 1일 1회, 5일간 MPTP(30 mg/kg, i.p., Schober, 2004) 투여; 그룹 3 및 그룹 4(MPTP + L-DOPA 투여군): MPTP 투여군에 L-DOPA(10 mg/kg 및 25 mg/kg) 투여. L-DOPA는 MPTP를 5일간 투여 후부터 1일 1회 21일간 투여; 그룹 5 및 그룹 6(GP-EX 투여군): MPTP 및 L-DOPA 투여군에 GP-EX(50 mg/kg) 투여, 돌외 추출물(GP-EX, 50 mg/kg)는 MPTP + L-DOPA 투여군(그룹 3 및 그룹 4)에 1일 1회 21일간 투여. 마지막 GP-EX 투여 후 행동실험을 측정하였으며, 생화학적 실험을 위하여 뇌를 적출하였다.
또한, 지페노시드(GPS)가 MPTP-투여 파킨슨병 모델에서 L-DOPA 투여시 나타나는 기억력 저하의 개선 효능이 있는지를 검토하기 위하여 다음과 같이 대조군 및 실험군을 나누어 실험을 진행하였다. 그룹 1(대조군): 0.9% 생리식염수 투여; 그룹 2(MPTP 투여군): 1일 1회, 5일간 MPTP(30 mg/kg, i.p., Schober, 2004) 투여; 그룹 3 및 그룹 4(MPTP+L-DOPA 투여군): MPTP 투여군에 L-DOPA(10 mg/kg 및 25 mg/kg) 투여. L-DOPA는 MPTP를 5일간 투여 후부터 1일 1회 21일간 투여; 그룹 5 및 그룹 6(GPS 투여군): MPTP 및 L-DOPA 투여군에 GPS(50 mg/kg) 투여. 지페노시드(GPS, 50 mg/kg)는 MPTP + L-DOPA 투여군(그룹 3 및 그룹 4)에 1일 1회 21일간 투여. 마지막 GPS 투여 후 행동실험을 측정하였으며, 생화학적 실험을 위하여 뇌를 적출하였다.
4. Transfer latency time 측정
Elevated plus-maze 장치는 두개의 open arm과 두개의 closed arm으로 구성되어 있다(30 cm X 5 cm). 각각의 마우스를 바깥으로 얼굴이 향하게 open arm 놓고, closed arm에 들어간 첫 번째 시간을 initial transfer latency 라 부른다. 측정은 90 초간 측정하며 이 시간 동안 마우스가 closed arm에 들어가지 않는다면 30 초간 강제로 closed arm에 들어가도록 한다. Second trial은 첫 측정 24 시간 후에 측정하며 retention transfer latency라 부른다. Retention transfer latency은 initial transfer latency time와 비교하여 백분율(%ITL)로 나타내었다([13] Kumar et al., 2011).

(Western blotting)
해마(hippocampus)를 적출한 뒤에 즉시, 1% 프로테아제 억제제(protease inhibitor)와 1% 포스파타아제 억제제(phosphatase inhibitor)가 포함된 RIPA 용해(lysis) 용액(1 ml)을 넣어 균질화한 뒤에 원심분리하여 상측액만 사용하였다. 단백질 시료는 12-15% SDS-PAGE(sodium dodecyl sulfate-polyacrylamide) 겔을 이용하여 전기영동을 시행하고, 폴리비닐리덴 다이플루오라이드(polyvinylidene difluoride)막으로 이동시켰다. Phosphor-NMDAR1(Ser897), ERK(Thr201/Thr204), CREB(Ser133) 및 β-액틴(actin)에 대한 항체(1:1,000)를 가하여 배양한 다음 이동된 단백질은 ECL 기질용액(Amersham Pharmacia Biotech Inc., Piscataway, NJ, 미국)을 가하여 반응(blots)을 진행시키고, 방사선필름을 이용하여 인화하여 측정하였다.
6. 통계적 분석
실험결과는 평균±S.E.M.으로 표시하였으며 유의성 검정은 Tukey 법에 의한 ANOVA 법에 의하여 계산하였다. 또한, 돌외 추출물 및 지페노시드(GPS)는 파킨슨병 동물 모델에서 50-400 mg/kg의 농도 범위에서 체중감소, 설사 그리고 구토 등의 독성작용을 나타내지 않는 것으로 보고되었으므로([15] Choi et al., 2013; [14] Razmovski-Naumovski et al., 2005), 독성이 낮은 치료물질임을 확인하였다.

추출물의 효과
Elevated plus maze 실험으로 initial transfer latency와 retention transfer latency를 통하여 공간적인 기억력을 측정하였다. MPTP 투여군은 대조군과 비교하였을 때 retention transfer latency(% of initial transfer latency, %ITL)는 140.9%(P < 0.05, n = 10)로 유의적으로 증가하였다. 그러나, 돌외추출물(50 mg/kg) 투여군은 MPTP 투여군과 비교하였을 때 %ITL가 114.7%(P < 0.05, n = 10)로 유의적으로 감소하였다(도 1의 패널 A).
또한 MPTP-투여 파킨슨병 동물모델에서 저용량 L-DOPA(10 mg/kg)의 투여는 미약한 기억력 개선효능을 나타내었으나(%ITL 값; MPTP 투여군 152.3%, L-DOPA 투여군 136.6%), 고용량 L-DOPA(25 mg/kg)의 투여는 기억력 개선효능을 나타내지 않았다(%ITL 값; 153.4%). 그러나 각 L-DOPA 투여군에 돌외 추출물(50 mg/kg)을 투여한 경우 기억력 개선 효능을 나타내었다(%ITL 값; 각 L-DOPA 투여군 121.9% 및 134.3%, P < 0.05)(도 1의 패널 B).

MPTP-투여 파킨슨병 동물모델에서 MPTP 투여군의 NMDAR1 인산화는 대조군에 비하여 2.4배(P < 0.05)로 유의적으로 증가하였으며, 돌외 추출물(50 mg/kg) 투여군은 MPTP 투여군에 비하여 감소하였다(대조군의 1.4배로 감소, P < 0.05)(도 2의 패널 A). 또한, MPTP-투여 파킨슨병 동물모델에서 저용량 L-DOPA(10 mg/kg)의 투여는 NMDAR1 인산화의 감소 효과를 나타내었으나(MPTP 투여군 1.65배, L-DOPA 투여군 1.22배), 고용량 L-DOPA(25 mg/kg)의 투여는 NMDAR1 인산화를 증가시켜 기억력 저하효능을 나타내었다(L-DOPA 투여군 1.71배). 그러나, 각 L-DOPA 투여군에 돌외 추출물(50 mg/kg)의 투여는 NMDAR1 인산화의 감소 효과를 나타내었다(L-DOPA 10 및 25 mg/kg 투여군, 1.10배 및 1.49배, P < 0.05)(도 2의 패널 B).

MPTP-투여 파킨슨병 동물 모델에서 MPTP 투여군의 ERK1/2 인산화는 대조군에 비하여 0.44배(P<0.05)로 유의적으로 감소하였으며, 돌외 추출물(50 mg/kg) 투여군은 MPTP 투여군에 비하여 증가하였다(대조군의 0.86배로 증가, P < 0.05)(도 3 패널 A). 또한, MPTP-투여 파킨슨병 동물모델에서 저용량 L-DOPA(10 mg/kg)의 투여는 ERK1/2 인산화의 증가 효능을 나타내었으나(MPTP 투여군 0.56배, L-DOPA 투여군 0.80배), 고용량 L-DOPA(25 mg/kg)의 투여는 ERK1/2 인산화의 증가효능을 나타내지 않았다(L-DOPA 투여군 0.52배). 그러나, 각 L-DOPA 투여군에 돌외 추출물(50 mg/kg)의 투여는 ERK1/2 인산화의 증가 효능을 나타내었다(L-DOPA 10 및 25 mg/kg 투여군, 0.87배 및 0.77배, P < 0.05)(도 3의 패널 B).

MPTP-투여 파킨슨병 동물 모델에서 MPTP 투여군의 CREB 인산화는 대조군에 비하여 0.58배(P < 0.05)로 유의적으로 감소하였으며, 돌외 추출물(50 mg/kg) 투여군은 MPTP 투여군에 비하여 증가하였다(대조군의 0.89배로 증가, P < 0.05)였다(도 4의 패널 A). 또한, MPTP-투여 파킨슨병 동물모델에서 저용량 L-DOPA(10 mg/kg)의 투여는 CREB 인산화의 증가효능을 나타내었으나(MPTP 투여군 0.53배, L-DOPA 투여군 0.66배), 고용량 L-DOPA(25 mg/kg)의 투여는 CREB 인산화의 증가 효능을 나타내지 않았다(L-DOPA 투여군 0.48배). 그러나, 각 L-DOPA 투여군에 돌외 추출물(50 mg/kg)의 투여는 CREB 인산화의 증가효능을 나타내었다(L-DOPA 10 및 25 mg/kg 투여군, 0.85배 및 0.72배, P < 0.05)(도 4의 패널 B).

MPTP-투여 파킨슨병 동물모델에서 저용량 L-DOPA(10 mg/kg)의 투여는 미약한 기억력 개선효능을 나타내었으나(%ITL 값; MPTP 투여군 148.6%, L-DOPA 투여군 136.2%), 고용량 L-DOPA(25 mg/kg)의 투여는 기억력 개선효능을 나타내지 않았다(%ITL 값; 151.5%). 그러나, 각 L-DOPA 투여군에 지페노시드(GPS)(50 mg/kg)의 투여는 기억력 개선 효능을 나타내었다(%ITL 값; 각 L-DOPA 투여군 123.0% 및 136.1%, P < 0.05)(도 5).

결론
상기 실험결과에 따르면, 본 발명에 따른 돌외 추출물 및 지페노시드(GPS)는 파킨슨병 환자에게 L-DOPA 치료와 관련하여 나타나는 기억력 장애의 개선 및 치료제 개발 분야에 유용하게 응용할 수 있다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현 예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
Claims (8)
- 돌외(Gynostemma pentaphyllum) 추출물 또는 지페노시드(gypenoside)를 유효성분으로 포함하는 파킨슨병에서 나타나는 기억력 장애의 치료 또는 예방용 약제학적 조성물.
- 제 1 항에 있어서, 상기 파킨슨병에서 나타나는 기억력 장애는 파킨슨병 치료약물 L-DOPA (3,4-dihydroxyphenylalanine) 투여에 의해 유도되는 것임을 특징으로 하는 약제학적 조성물.
- 제 1 항에 있어서, 상기 돌외 추출물 또는 지페노시드는 뇌의 해마 부위에 존재하는 N-메틸-D-아스파테이트(N-methyl-D-aspartate, NMDA)의 수용체(NMDAR)의 증가된 활성을 감소시키는 것을 특징으로 하는 약제학적 조성물.
- 제 1 항에 있어서, 상기 돌외 추출물 또는 지페노시드는 뇌의 해마 부위에 존재하는 ERK(extracellular signal-regulated kinase) 1/2 및 CREB(cyclic AMP-response element binding protein)의 감소된 활성을 증가시키는 것을 특징으로 하는 약제학적 조성물.
- 돌외 추출물 또는 지페노시드를 유효성분으로 포함하는 파킨슨병에서 나타나는 기억력 장애의 개선용 기능성 식품 조성물.
- 제 5 항에 있어서, 상기 파킨슨병에서 나타나는 기억력 장애는 파킨슨병 치료약물 L-DOPA (3, 4-dihydroxyphenylalanine) 투여에 의해 유도되는 것임을 특징으로 하는 기능성 식품 조성물.
- 제 5 항에 있어서, 상기 돌외 추출물 또는 지페노시드는 뇌의 해마 부위에 존재하는 N-메틸-D-아스파테이트(N-methyl-D-aspartate, NMDA)의 수용체(NMDAR)의 증가된 활성을 감소시키는 것을 특징으로 하는 기능성 식품 조성물.
