Abstract
본 발명에 따른 면역접합체는 표면에 다수의 약물을 결합시킬 수 있는 덴드론과 표적 지향적인 항체를 접합함으로써 표적 지향적인 약물치료에 이용될 수 있으며, 특히 다수의 항암제가 결합된 친수성 덴드론을 항체에 접합시킴으로써, 종양 특이적으로 높은 농도의 약물을 전달하여 강력한 항암효과를 나타낼 수 있는 약학적 조성물을 제공한다.
Description
본 발명은 항체에 덴드론이 접합된 면역접합체 및 이의 용도에 관한 발명으로, 보다 상세하게는 시스타민 중심을 갖는 덴드리머를 환원시켜 형성된 덴드론을 항체 또는 이의 단편과 접합시킨 면역접합체 및 상기 덴드론에 약물을 접합시켜 항체와 접합한 표적 지향적인 면역접합체에 관한 발명이다.
항암제로 사용되는 세포독성 약물인 독소루비신 등은 세포주기를 타겟으로 하므로 독성이 암세포의 증식정도에 의존적이다. 또한, 임상치료 효과를 얻기 위해 일반적으로 최대 허용량 근처에서 사용되고 있다. 그러나, 이러한 항암제들은 빠르게 증식하는 세포를 죽일 뿐, 정상 세포와 암세포 또는 암 조직을 차별화하지 못하여 암세포가 아닌 다른 세포를 사멸시키고, 고농도로 사용시 구토 등의 부작용을 일으킴이 보고되고 있다. 또한, 장기 치료 시 항암제에 대한 내성을 야기시킬 수 있어 세포독성 약물이 암세포만 표적하여 사멸시키는 향상된 치료 요법이 절실히 요구되고 있다.
한편, 종양 관련 항원에 대한 항체를 세포독성 약제와 접합시켜 특정 종양으로 전달시킴으로써 약제의 국부적 농도를 상승시켜 세포독성 약제의 효능을 개선시키고자 하는 많은 시도가 있어왔다. 많은 이들 방법은 제한된 성공을 거두었고 실패를 해명하기 위한 문제점들이 문헌에서 논의되어 왔다. 예를 들어, 독소루비신 또는 메토트렉세이트와 같은 화학양론적으로 작용하는 항암제에 대해, 요구되는 세포독성을 발휘하기 위해서는 비교적 높은 세포내 농도가 필요하다. 이들 농도는 (a) 일반적인 많은 항암제의 불충분한 효능, (b) 세포 표면의 낮은 항원 표적 농도, (c) 항원-항체 복합체의 표적 세포로의 비효율적인 내부화 및 (d) 표적 세포내에서 접합체로부터 유리된 약제의 비효율적인 방출 때문에 많은 항체-약제 접합체를 사용하여 달성되기가 어려운 것으로 사료된다.
따라서, 세포독성 약제를 표적 세포에 특이적으로 전달시킴으로서 정상 세포에 대한 약물의 부작용을 경감시키면서, 표적 세포 내에서 비교적 높은 약물 농도로 약물을 전달할 수 있는 표적 지향적 약물 전달용 면역접합체에 대한 요구가 증가되고 있다.
이에, 본 발명자들은 시스타민 중심을 갖는 덴드리머를 환원하여 형성된 덴드론은 항체와의 접합이 용이할 뿐만 아니라 그 표면에 다수의 약물을 접합할 수 있어, 표적 지향적인 약물 전달이라는 목적을 달성하면서 기존의 항체-약물 접합체와 비교해 높은 농도로 표적 세포에 약물을 전달할 수 있음을 발견하고 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은 중심에 티올(thiol)기를 갖는 덴드론(dendron), 링커 및 항체를 포함하는 면역접합체를 제공하는 것이다.
본 발명의 다른 목적은 상기 덴드론에 항암약물이 접합되어 있는 것을 특징으로 하는 면역접합체를 제공하는 것이다.
본 발명의 다른 목적은 상기 덴드론에 항암약물이 접합되어 있는 면역접합체를 유효성분으로 포함하는 암의 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 면역접합체의 덴드론 표면에 약물을 운반하는 것이 특징인 표적지향적 약물 전달용 면역접합체를 제공하는 것이다.
본 발명의 다른 목적은 (a) 시스타민 중심을 갖는 폴리아미도아민(PAMAM) 덴드리머를 환원시켜 중심에 티올(thiol)기를 갖는 덴드론을 형성하는 단계; 및 (b) 상기 덴드론을 항체에 접합시키는 단계를 포함하는 면역접합체 제조방법을 제공하는 것이다.
상기 목적을 달성하기 위해 본 발명은 중심에 티올(thiol)기를 갖는 덴드론(dendron) 및 항체를 포함하는 면역접합체를 제공한다.
본 발명의 다른 목적을 달성하기 위해 본 발명은 상기 덴드론에 항암약물이 접합되어 있는 것을 특징으로 하는 면역접합체를 제공한다.
본 발명의 다른 목적을 달성하기 위해 본 발명은 상기 덴드론에 항암약물이 접합되어 있는 면역접합체를 유효성분으로 포함하는 암의 예방 또는 치료용 약학적 조성물을 제공한다.
본 발명의 다른 목적을 달성하기 위해 본 발명은 상기 면역접합체의 덴드론 표면에 약물을 운반하는 것이 특징인 표적지향적 약물 전달용 면역접합체를 제공한다.
본 발명의 다른 목적을 달성하기 위해 본 발명은 (a) 시스타민 중심을 갖는 폴리아미도아민(PAMAM) 덴드리머를 환원시켜 중심에 티올(thiol)기를 갖는 덴드론을 형성하는 단계; 및 (b) 상기 덴드론을 항체에 접합시키는 단계를 포함하는 면역접합체 제조방법을 제공한다.
이하 본 발명을 상세히 설명한다.
본 발명은 중심에 티올(thiol)기를 갖는 덴드론(dendron) 및 항체를 포함하는 면역접합체를 제공한다
바람직하게는, 상기 덴드론은 시스타민 중심을 갖는 덴드리머(dendrimer)의 환원에 의해 생성된 것임을 특징으로 할 수 있다.
본 발명은 또한 상기 덴드리머는 폴리아미도아민(PAMAM) 덴드리머인 것을 특징으로 하는 면역접합체를 제공한다.
덴드리머는 중심(core)를 갖고 그로부터 나오는 분지 구조의 다중 쉘을 가진 거대분자를 지칭한다. 덴드론은 중심점으로부터 나오는 분지들를 갖는 덴드리머의 일종으로 다수의 표면 작용기와 하나의 중심 작용기를 갖는 쐐기-형상의 덴드리머 분획을 의미한다. 덴드리머 및 덴드론의 두가지 모두에 대해, 분지들은 코어에 직접 또는 연결기를 통해 연결될 수 있다. 덴드리머는 일반적으로 다중 쉘 또는 세대 (generation), 예를 들어 G1, G2, G3, G4 등을 포함한다. 반복되는 반응 연쇄가 종종 덴드리머에 각 세대를 추가하기 위해 이용된다.
덴드리머라는 용어는 코어 및 상기 코어로부터 방사되어 나오는 분지 구조물들의 다중 쉘(shells)들을 갖는 거대 분자를 의미한다. 수지상(dendritic) 담체의 모양 및 크기는 다양할 수 있다. 일부 경우들에서, 상기 수지상 담체는 모양이 거의 구형 또는 공 모양일 수 있다. 나아가, 상기 수지상 담체는 약 15 옹스트롬(A) 내지 약 250A의 범위의 직경을 가질 수 있으며, 대응되는 분자량의 범위는 예로서 약 500달튼 내지 약 2백만 달튼이다. 덴드리머들은 각종 공급원들(Dendritech, Midland, Michigan)로부터 상업적으로 입수하거나 또는 당업자들에게 알려진 방법에 의하여 합성될 수 있다. 수지상 분자들은 크게 저분자량 종류 및 고분자량 종류들로 나눌 수 있으며 저분자량 분자들에 덴드리머들 및 덴드론들(dendrons)이 포함된다. 덴드리머들 및 덴드론들은 반복적으로 분지, 단분산되며, 대개 대칭성이 높은 화합물들이다.
덴드론은 대개 초점(focal point)이라고 불리는 화학적으로 다룰 수 있는(addressable) 단일한 기를 포함한다. 몰 질량 분포의 부재로 인하여, 고-몰-질량 덴드리머들 및 덴드론들은 거대분자이기는 하지만 폴리머는 아니다. 덴드리머드들의 성질은 분자 표면 상의 관능기들에 의하여 지배된다. 기능성 분자들의 수지상 캡슐화는 활성 자리의 분리, 수지상 골격들이 내부 기능과 외부 기능을 분리시키기 때문에 생체적합물질들 중 활성 자리들의 구조를 모방하는 구조의 분리를 가능하게 한다. 예로서, 덴드리머는 그의 말단기가 카르복실기처럼 친수성기인 경우, 수용성일 수 있다.
덴드리머들은 일반적으로 다음과 같은 특징들에 의해 구분될 수 있다: (i) 개시제 코어 (I), 이는 하나 이상의 반응 자리들을 가질 수 있으며, 덴드리머의 최종 위상(topology)에 영향을 미치도록 끝이 뾰족하거나 현저한 크기일 수 있다; (ii) 상기 개시제 코어에 부착된 분지된 반복 단위들의 하나 이상의 층들; (iii) 덴드리머의 표면에, 임의로 연결기들을 통하여 부착된, 예컨대 음이온성 또는 양이온성 기들과 같은, 기능성 말단기들.
본 발명에서 고려되는 덴드리머들은, 리신 또는 리신 유사체 건설 단위들을 포함할 수 있다. 리신 유사체(lysine analogue)라는 용어는 건축 단위들의 예비층에 대한 부착을 위한 하나의 아펙스(apex) 카르복실기, 및 건축 단위들, 차단기들, 링커들 또는 아릴산 기들이 거기 더 부착될 수 있는 2 또는 3 개의 일차 아민기들을 갖는 분자를 의미한다.
본 발명에서 고려되는 덴드리머들은, 폴리아미도아민 (PAMAM), 폴리(에테르히드록실아민) (PEHAM) 또는 폴리프로필렌이민 건축 단위들을 포함하는 것들을 포함하며, 바람직하게는 폴리아미도아민 건축 단위일 수 있으나 이에 제한되는 것은 아니다.
중심 부분은 건축단위에 대해 단지 한 부착 지점을 포함할 수 있거나 또는 2, 3, 또는 그 이상의 지점들을 포함할 수 있으며, 이들은 건축 단위들의 부착을 위해 더 사용되거나 또는 사용되지 않을 수 있다. 바람직하게는, 본 발명의 덴드리머는 시스타민 중심을 갖는 덴드리머일 수 있다.
시스타민 중심을 갖는 덴드리머를 환원시킬 경우, 티올(thiol) 중심기를 갖는 2 이상의 덴드론(dendron)이 형성이 되며, 상기 티올 중심기는 단백질, 펩타이드, 항체 또는 다른 벡터들과 용이하게 결합하여 접합체를 형성할 수 있다.
본 발명은 또한, 상기 면역접합체는 링커를 추가적으로 포함하는 것을 특징으로 하는 면역접합체를 제공한다.
본 발명에서 링커는 덴드론-항체 면역접합체를 형성하기 위하여 하나 또는 그 이상의 덴드론을 항체(Ab)에 연결시키는데 이용되는 이중기능 또는 다중기능 모이어티다. 덴드론-항체 면역접합체는 덴드론과 상기 항체에 공유적으로 부착되는 반응 기능을 보유한 링커를 이용하여 만들 수 있다. 예로써, 항체 (Ab)의 시스테인 티올은 링커 또는 약물-링커 중간생성물의 반응 기능기와 결합을 형성하여 면역접합체를 만들 수 있다.
한 측면에서, 링커는 공유 결합을 형성하기 위하여 항체 상에 존재하는 자유 시스테인과 반응할 수 있는 기능을 보유한다. 이러한 반응 기능기의 비제한적인 예시는 말레이미드, 할로아세타아미드, α-할로아세틸, 활성화된 에스테르 이를 테면 숙시니미드 에스테르, 4-니트로페닐 에스테르, 펜타플루오로페닐 에스테르, 테트라플루오로페닐 에스테르, 무수물, 산 염화물, 술포닐 염화물, 이소시아네이트, 트기로 이소티오시아네이트를 포함하며, 바람직하게는 말레이미드일 수 있다.
또한, 링커는 항체 상에 존재하는 친전자(electrophilic) 기와 반응할 수 있는 기능기를 가진다. 예시적인 이러한 친전자 기는 알데히드 그리고 케톤 카르보닐기를 포함하나 이에 한정되지 않는다. 링커의 반응 기능기의 헤테로 원자는 항체 상에서 친전자 기와 반응하여 항체 단위에 공유 결합을 형성할 수 있다. 이러한 반응 기능기의 비제한적인 예로는 하이드라지드, 옥심, 아미노, 하이드라진,티오세미카르바존, 하이드라진 카르복실레이트, 그리고 아릴하이드라지드를 포함한다.
링커는 하나 또는 그 이상의 링커 성분을 포함할 수 있다. 예시적인 링커 성분은 6-말레이미도카프로일 ("MC"), 말레이미도프로파노일 ("MP"), 발린-시투룰린 ("val-cit" 또는 "vc"), 알라닌-페닐알라닌 ("alaphe"), p-아미노벤질옥시카르보닐 ("PAB"), N-숙시니미딜 4-(2-피리딜티오) 펜타노에이트 ("SPP"), 그리고 4-(N-말레이미도메틸) 시클로헥산-1 카르복실레이트 ("MCC")를 포함한다.
링커는 덴드론의 방출을 용이하게 하는 절단가능한 링커일 수 있다. 비제한적인 예시적인 절단가능한 링커는 산-불안정한 링커 (예컨데, 하이드라존이 포함된), 프로테아제-민감성 (예컨데, 펩티다제-민감성) 링커, 광불안정한 링커, 또는 디설피드-함유 링커를 포함한다(Chari et al., Cancer Research 52:127-131 (1992); US5208020).
또한, 링커 성분은 "아미노산 단위"를 포함할 수 있다. 이러한 일부 구체예들에 있어서, 아미노산 단위는 프로테아제, 이를 테면 리소좀 효소에 의한 링커의 절단을 허용함으로써, 세포내 프로테아제에 노출될 때 상기 면역접합체로부터 덴드론의 방출이 용이하게 된다(Doronina et al. (2003) Nat. Biotechnol.21:778-784). 예시적인 아미노산 단위는 이펩티드, 삼펩티드, 사펩티드, 그리고 오펩티드를 포함하나 이에 한정되지 않는다. 예시적인 이펩티드는 발린-시투룰린 (vc 또는 val-cit), 알라닌-페닐알라닌 (af 또는 alaphe);페닐알라닌-리신 (fk 또는 phe-lys); 페닐알라닌-호모리신 (phe-homolys); 그리고N-메틸-발린-시투룰린(Me-val-cit)을 포함하나 이에 한정되지 않는다. 예시적인 삼펩티드는 글리신-발린-시투룰린 (gly-val-cit) 그리고글리신-글리신-글리신 (gly-gly-gly)을 포함하나 이에 한정되지 않는다. 아미노산 단위는 자연적으로 발생되는 아미노산 잔기 및/또는 소수 아미노산 및/또는 비-자연 발생적 아미노산 유사체, 이를 테면 시투룰린을 포함할 수 있다. 아미노산 단위는 특정 효소, 예로써, 종양-연합된 프로테아제, 카텝신 B, C 그리고 D, 또는 플라스민 프로테아제에 의해 효소 절단되고 최적화되도록 기획된다.

상기 종양성 항원의 유형은 또한 종양-특이적 항원(TSA) 또는 종양-관련된 항원(TAA)일 수 있다. TSA는 종양 세포에 대해 고유하고 신체의 다른 세포 상에서 발생하지 않는다. TAA 관련된 항원은 종양 세포에 대해 고유하지 않고 대신에 또한 항원에 대한 면역원성 내성 상태를 유도하지 못하는 조건 하에서 정상 세포 상에서 발현된다. 종양에 대한 항원의 발현은 면역 시스템이 항원에 대해 반응하게 하는 조건 하에서 발생할 수 있다. 본 발명은 상기 기술된 항체의 사용을 제한할 필요가 없고, 당업자에게 알려진 바와 같은 다른 그러한 항체는 본원에 기술된 면역접합체에 사용될 수 있다.
본 발명의 면역접합체는 종양성 항원을 인식하는 종양 특이적 항체와 덴드론이 결합함으로써, 표적 지향적 약물 운송 등에 사용될 수 있어 약물의 부작용은 경감되고 치료효과가 극대화되는 효과를 나타낼 수 있는 것이다.
본 발명은 또한 상기 항체는 폴리클로날 항체, 모노클로날 항체, 키메라 항체, 인간화된 항체, Fv-단편, Fab-단편, F(ab’)2-단편 및 scFv 단편으로 이루어진 군에서 선택된 어느 하나인 것을 특징으로 하는 면역접합체를 제공한다.
본 발명에서 항체 (antibody)는 전체 항체 (whole) 형태일 뿐만 아니라 항체 분자의 기능적인 단편을 포함한다. 전체 항체는 2개의 전체 길이의 경쇄 및 2개의 전체 길이의 중쇄를 가지는 구조이며 각각의 경쇄는 중쇄와 다이설파이드 결합으로 연결되어 있다. 항체 분자의 기능적인 단편이란 항원 결합 기능을 보유하고 있는 단편을 뜻하며 Fab, F(ab'), F(ab')2, Fv 및 scFv 등을 포함한다.
항체 단편 중 Fab는 경쇄 및 중쇄의 가변영역과 경쇄의 불변영역 및 중쇄의 첫 번째 불변 영역(CH1)을 가지는 구조로 1개의 항원 결합 부위를 가진다. Fab'는 중쇄 CH1 도메인의 C 말단에 하나 이상의 시스테인 잔기를 포함하는 힌지 영역 (hinge region)을 가진다는 점에서 Fab와 차이가 있다. F(ab')2 항체는 Fab'의 힌지 영역의 시스테인 잔기가 디설파이드 결합을 이루면서 생성된다. Fv는 중쇄 가변부위 및 경쇄 가변부위만을 가지고 있는 최소의 항체조각으로 Fv단편을 생성하는 재조합 기술은 국제공개 특허 WO 88/10649, WO 88/106630, WO 88/07085, WO 88/07086 및 WO 88/09344에 개시되어 있다. 이중쇄 Fv(dsFv)는 디설파이드 결합으로 중쇄 가변부위와 경쇄 가변부위가 연결되어 있고 단쇄 Fv(scFv)는 일반적으로펩타이드 링커를 통하여 중쇄의 가변 영역과 경쇄의 가변 영역이 공유 결합으로 연결되어 있다. 이러한 항체 단편은 단백질 가수분해 효소를 이용해서 얻을 수 있고 (예를 들어, 전체 항체를 파파인으로 제한 절단하며 Fab를 얻을 수 있고 펩신으로 절단하면 F (ab')2 단편을 얻을 수 있다) 바람직하게는 유전자 재조합 기술을 통하여 제작할 수 있다. 본 발명에서 항체는 바람직하게는 Fab 형태이거나 전체 항체 형태이다. 본 발명의 목적상 상기 인간항체는 단일클론항체일 수 있다. 본 발명의 인간항체는 모든 구조가 인간으로부터 유래 되었기 때문에 기존의 인간화 항체 또는 마우스 항체에 비해서 면역화 반응이 일어날 확률이 적어서 인간에게 투여하였을 경우 원하지 않는 면역반응이 일어나지 않는 장점이 있다. 따라서 치료용 항체로서 매우 유용하게 사용될 수 있다.
본 발명은 또한 상기 항체의 중쇄 또는 경쇄의 불변영역 또는 가변영역에 존재하는 시스테인 잔기에 덴드론이 접합된 것을 특징으로 하는 면역접합체를 제공한다.
본 발명은 또한 상기 항체가 중쇄 또는 경쇄의 불변영역 또는 가변영역의 하나 또는 그 이상의 잔기가 시스테인 잔기로 치환된 항체로서, 상기 치환된 시스테인 잔기에 덴드론이 접합된 것을 특징으로 하는 면역접합체를 제공한다.
항체에 접합될 수 있는 덴드론의 수는 항체에 존재하는 자유 시스테인 잔기 수에 의해 제한된다. 덴드론-항체 면역접합체의 예시적인 구체예는 종양 세포를 표적으로 하는 항체, 링커 및 덴드론을 포함한다. 일부 구체예들에 있어서, 상기 항체는 하나 또는 그 이상의 아미노산, 잔기 이를테면 리신 및/또는 시스테인을 통하여 ,링커에 부착된다. 바람직하게는 시스테인일 수 있으나 이에 제한되는 것은 아니다.
하나 또는 그 이상의 자유 시스테인 잔기가 조작하지 않아도 항체 안에 이미 존재할 수 있고, 이런 경우 기존의 자유 시스테인 잔기가 이용되어 상기 항체를 약물에 접합시킬 수 있다. 일부 구체예들에 있어서, 항체는 하나 또는 그 이상의 자유 시스테인 잔기를 만들기 위하여 항체 접합에 앞서 환원 조건에 노출된다.
다른 한편으로는, 항체의 하나 또는 그 이상의 잔기가 시스테인 잔기으로 치환된, 시스테인 조작된 항체, 예컨데, "티오MAbs(thioMAbs)"를 만드는 것이 바람직할 수 있다. 특정 구체예들에 있어서, 치환된 잔기는 상기 항체의 접근가능한 부위에서 일어난다. 이들 잔기가 시스테인으로 대체됨으로써, 이로 인하여 반응성 티올 기는 상기 항체의 접근가능한 부위에 위치되며, 그리고 다른 모이어티, 이를 테면 덴드론 모이어티 또는 링커-덴드론 모이어티에 항체를 접합시켜, 본 명세서에서 더 설명되는 면역접합체가 탄생될 수 있다. 시스테인 조작된 항체는 예컨데, U.S. 특허 7,521,541에서 설명된 것과 같이 만들어질 수 있다.
이와 같이 추가된 시스테인은 디설파이드 결합 (disulfide bond)를 환원하여 자유 티올기(thiol group)가 노출되도록 하여 덴드론과 접합하기 위한 작용기로 사용될 수 있다. 상기 시스테인 잔기는 1개 이상이 추가될 수도 있으며, 티올-반응성 시약과 부위-특이적으로 커플링될 수 있다. 이와 같은 시스테인 잔기는 링커와 연결되어 상기 인간 항체와 덴드론이 접합할 수 있도록 한다. 상기 시스테인 잔기는 효과적으로 덴드론을 접합시키면서도 상기 항체가 표적으로 하는 종양성 항원과의 결합능력을 저해하지 않는 한 중쇄 가변영역 뿐 아니라, 경쇄 가변영역 또는 중쇄 또는 경쇄의 불변영역에 추가될 수 있다.
본 발명의 일실시예에 따르면 항체인 B12 항체 및 상기 항체의 중쇄 가변영역에 시스테인을 포함한 변형된 B12 항체 모두 FACS 분석 결과, 종양성 항원에 특이적으로 결합하는 것을 확인하였다. 이와 같은 결과는 덴드론 접합을 위해 시스테인을 추가하는 변형을 가하더라도 본 발명의 항체는 종양성 항원에 대한 결합력이 감소되지 않는다는 것을 뒷받침하는 것으로 약물 접합에 시스테인을 이용할 수 있다는 것을 시사한다.
본 발명은 또한 상기 덴드론은 링커를 통해 항체에 접합된 것을 특징으로 하는 면역접합체를 제공한다.

본 발명에 따른 면역접합체는 종양 특이적 항체에 덴드론을 접합함으로써 종양조직에 대해 표적 지향적으로 약물을 전달할 수 있으며, 상기 덴드론에 최소 둘 이상의 약물이 결합할 수 있어 기존의 항체-약물 접합체와 비교해 비교적 높은 농도로 약물을 종양 조직에 전달하여 치료효과를 극대화할 수 있다는 이점이 있다.
본 발명에 있어서 상기 항암약물은 독소루비신 (Doxorubicin), 카보플라틴 (파라플라틴)[Carboplatin(paraplatin)], 시스플라틴 (Cisplatin), 시클로포스파미드 (Cyclophosphamide), 이포스파미드(Ifosfamide), 니드란 (Nidran), 질소머스타드 (메클로에타민 염산염)[Nitrogen mustar(MechlorethamineHCL)], 블레오마이신 (Bleomycin), , 미토마이신 C (Mitomycin C), 시타라빈 (Cytarabine), 플루로우라실(Flurouracil), 젬시타빈 (Gemcitabine), 트리메트렉세이트 (Trimetrexate), 메토크렉세이트 (Methotrexate),에토포시드 (Etoposide), 빈블라스틴 (Vinblastine), 비노렐빈 (vinorelbine), 알림타 (Alimta), 알트레타민(Altretamine), 프로카바진 (Procarbazine), 탁솔 (Taxol), 탁소텔 (Taxotere), 토포테칸 (Topotecan) 및 이리노테칸 (Irinotecan)으로 이루어진 군에서 선택될 수 있으며, 바람직하게는 독소루비신일 수 있으나 이에 제한되는 것은 아니다.
상기 덴드론에 항암약물은 2개 내지 32개가 결합할 수 있으며, 보다 바람직하게는 3개 내지 15개의 항암약물이 결합되는 것을 특징으로 할 수 있으나 이에 제한되는 것은 아니다.
본 발명의 일실시예에 따르면, 독소루비신의 아민기에 시스-아코니틱 안하이드라이드(Cis-aconitic anhydride)를 반응시켜 Cis-aconitiyl-doxorubicin을 제조하였다. 상기 Cis-aconitiyl-doxorubicin과 PAMAM 덴드리머를 EDC(1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride)와 NHS (N-hydroxysuccinimide) 존재하에 반응시켜 PAMAM 덴드리머의 아민기와 Cis-aconitiyl-doxorubicin의 카르복실기가 결합된 PAMAM-doxorubicin을 제조하였다. PAMAM 덴드리머와 doxorubicin 간의 cis-aconityl 결합은 중성에서는 안정한 반면 산성에서 잘 끊어지는 특성이 있다. 따라서 체내에서 혈중에서는 안정한 결합을 유지하고 있다가 세포내 라이소좀에서는 잘 끊어져 독소루비신을 방출하는 기전을 갖고 있다(Zhu et al., Biomaterials, 2010, 31, 1360-1371).
본 발명은 또한 상기 면역접합체를 유효성분으로 포함하는 암의 예방 또는 치료용 약학적 조성물을 제공한다.
본 발명에 따른 면역접합체는 종양 조직에 표적 지향적으로 작용함으로써, 항암약물로 인해 정상조직에서 나타나는 부작용이 감소되며, 고농도의 약물을 종양 조직에 집중시킴으로 암의 예방 또는 치료에 유용하게 사용될 수 있다.
바람직하게 본 발명의 치료 가능한 암은 본 발명의 면역접합체를 이용하여 선택적으로 사멸시킬 수 있는 암은 제한 없이 포함될 수 있으나, 그 예로 피부, 소화기, 비뇨기, 생식기, 호흡기, 순환기, 뇌 또는 신경계의 암이 있으며, 구체적으로 폐암, 비소세포성 폐암, 결장암, 골암, 췌장암, 피부암, 두부 또는 경부 암, 자궁암, 난소암, 직장암, 위암, 항문부근암, 결장암, 유방암, 나팔관암, 자궁내막암, 자궁경부암, 질암, 음문암, 호지킨병(Hodgkin's disease), 식도암, 소장암, 내분비선암, 갑상선암, 부갑상선암, 부신암, 연조직 육종, 요도암, 음경암, 전립선암, 만성 또는 급성 백혈병, 림프구 림프종, 방광암, 신장 또는 수뇨관암, 신장세포 암종, 신장골반 암종, 중추신경계 (central nervous system, CNS) 종양, 1차 중추신경계 림프종, 척수 종양, 뇌간 신경교종 또는 뇌하수체 선종일 수 있다.
본 발명에서 사용되는 용어 “예방”은 본 발명의 조성물의 투여로 암을 억제시키거나 진행을 지연시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어 “치료”는 본 발명의 조성물의 투여로 암의 증상이 호전 또는 이롭게 변경되는 모든 행위를 의미한다.
본 발명의 조성물은 약학적으로 허용 가능한 첨가제를 더 포함할 수 있으며, 약제학적으로 허용 가능한 첨가제로는 전분, 젤라틴화 전분, 미결정셀룰로오스, 유당, 포비돈, 콜로이달실리콘디옥사이드, 인산수소칼슘, 락토스, 만니톨, 엿, 아라비아고무, 전호화전분, 옥수수전분, 분말셀룰로오스, 히드록시프로필셀룰로오스, 오파드라이, 전분글리콜산나트륨, 카르나우바 납, 합성규산알루미늄, 스테아린산, 스테아린산마그네슘, 스테아린산알루미늄, 스테아린산칼슘, 백당, 덱스트로스, 소르비톨 및 탈크 등이 사용될 수 있다. 본 발명에 따른 약제학적으로 허용 가능한 첨가제는 상기 조성물에 대해 0.1~90 중량부로 포함되는 것이 바람직하나 이에 제한되는 것은 아니다.
또한, 본 발명의 조성물은 실제 임상 투여 시에 경구 및 비경구의 여러 가지 제형으로 투여될 수 있는데, 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다.
경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 락토코커스 중앙젠시스에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘 카보네이트, 수크로오스, 락토오스 또는 젤라틴 등을 섞어 조제될 수 있다. 또한 단순한 부형제 이외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용될 수 있다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제 및 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함될 수 있다. 비수성용제, 현탁용제로는 프로필렌글리콜, 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올 레이트와 같은 주사 가능한 에스테르 등이 사용 될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다.
한편, 주사제에는 용해제, 등장화제, 현탁화제, 유화제, 안정화제, 방부제 등과 같은 종래의 첨가제가 포함될 수 있다.
또한, 본 발명의 치료용 조성물은 임의의 생리학적으로 허용가능한 담체, 부형제 또는 안정화제(Remington: The Science and Practice of Pharmacy, l9th Edition, Alfonso, R., ed, Mack Publishing Co.(Easton, PA: 1995))를 추가로 포함할 수 있다. 허용가능한 담체, 부형제 또는 안정화제는 사용된 투여량 및 농도에서 수용자에게 비독성이고, 완충용액, 예를 들어 인산, 시트르산 및 다른 유기산; 아스코르브산을 비롯한 항산화제; 저분자량 (약 10개 미만의 잔기) 폴리펩티드; 단백질, 예를 들어 혈청 알부민, 젤라틴 또는 면역글로불린; 친수성 중합체, 예를 들어 폴리비닐피롤리돈; 아미노산, 예를 들어 글리신, 글루타민, 아스파라긴, 아르기닌 또는 리신; 단당류, 이당류, 및 글루코스, 만노스 또는 덱스트린을 비롯한 다른 탄수화물; 킬레이트제, 예를 들어 EDTA; 당 알콜, 예를 들어 만니톨 또는 소르비톨; 염-형성 반대이온, 예를 들어 나트륨; 및(또는) 비이온성 계면활성제, 예를 들어 트윈, 플루로닉스 또는 폴리에틸렌 글리콜 (PEG)이 포함된다.
본 발명의 의 인체에 대한 투여량은 환자의 나이, 몸무게, 성별, 투여형태, 건강상태 및 질환 정도에 따라 달라질 수 있으며, 일반적으로 0.01 - 100 mg/kg/day이며, 바람직하게는 0.1 - 20 mg/kg/day이며, 더욱 바람직하게는 5 - 10 mg/kg/day일 수 있다. 또한 의사 또는 약사의 판단에 따라 일정 간격으로 분할 투여할 수도 있다.
본 발명은 또한 상기 면역접합체의 덴드론 표면에 약물을 운반하는 것이 특징인 표적지향적 약물전달용 조성물을 제공한다.
표적지향형 약물전달시스템이란 약물을 치료부위에 선택적으로 전달함으로써 건강한 조직을 약물에 노출시키지 않음과 동시에 소량의 약물만으로도 우수한 치료효과를 나타낼 수 있도록 설계된 기술을 말한다. 표적지향적 약물전달시스템을 이용하게 되면 질병이 있는 인체의 특정 부위에 약물을 집중시킴으로써 약물치료 효과를 극대화 할 수 있으며, 항암제 등 독성이 강한 약물에 의한 부작용을 최소화시킬 수 있다.
본 발명에 따른 면역접합체는 덴드론 표면에 목적하는 치료효과를 달성할 수 있는 약물을 접합시키고, 타겟 조직 또는 세포에 특이적으로 결합하는 항체를 상기 덴드론에 접합시킴으로써 약물을 효과적으로 목적하는 조직 또는 세포에 도달시킬 수 있는 표적지향적 약물전달용 벡터로서 이용될 수 있다.
상기 표적지향적 약물전달용 조성물에는 상기 약학적 조성물에 추가적으로 포함될 수 있는 담체 등이 동일하게 적용될 수 있다.
본 발명은 또한 (a) 시스타민 중심을 갖는 폴리아미도아민(PAMAM) 덴드리머를 환원시켜 중심에 티올(thiol)기를 갖는 덴드론을 형성하는 단계; 및 (b) 상기 덴드론을 항체에 접합시키는 단계를 포함하는 면역접합체 제조방법을 제공한다.
상기에서 살펴본 바와 같이, 본 발명에 따른 면역접합체는 표면에 다수의 약물을 결합시킬 수 있는 덴드론과 표적 지향적인 항체를 접합함으로써 표적 지향적인 약물치료에 이용될 수 있으며, 특히 다수의 항암제가 결합된 친수성 덴드론을 항체에 접합시킴으로써, 종양 특이적으로 높은 농도의 약물을 전달하여 강력한 항암효과를 나타낼 수 있다.

도 3은 시스타민 중심을 갖는 덴드리머를 이용해 본 발명에 따른 면역접합체를 제조하는 방법에 대한 모식도이다.
도 4는 항체와 덴드론-항체 면역접합체를 size-exclusion HPLC로 분리하고, UV 280 nm와 490 nm에서 검출하여 덴드론-항체 면역접합체를 확인한 크로마토그램이다.
도 5는 덴드론-항체 면역접합체에 염산과 메탄올을 처리한 후 유리된 독소루비시논 양을 분석하여 덴드론-항체 면역접합체에 함유된 독소루비신 함량을 측정한 크로마토그램이다.

이하 본 발명을 상세히 설명한다.
단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.
<실시예 1>
Cis
-aconityl doxorubicin의 제조
㈜ 동아제약으로부터 구입한 독소루비신(에이디엠 주사액)을 0.3% triethylamine (TEA)을 포함한 dimethylformamide (DMF)에 녹인 다음, 동일 용매에 녹인 Cis-aconitic anhydride와 차광하에서 2시간동안 반응시켰다(도 1). 이후 반응액을 C-18 silica가 충진된 카트리지에 로딩한 후 80% 메탄올로 용출시키는 solid-phase extraction method를 이용하여 미반응물을 제거하고 Cis-aconityl doxorubicin을 분리하였다. 분리된 Cis-aconityl doxorubicin이 담긴 용액의 메탄올을 진공을 이용한 원심분리기로 제거한 후 동결건조하였다.
제조된 Cis-aconityl doxorubicin은 DiKMA C-18 역상 컬럼에 주입한 후 3% (NH4)2CO3와 메탄올을 1:2 (v/v)로 혼합한 이동상으로 UV 490 nm에서 검출하여 분석하였다. 또한 MALDI-TOF 질량분석기를 이용하여 질량값을 측정하여 확인하였다. Cis-aconityl doxorubicin은 700.4 Da으로 질량값이 확인되었다(도 2).
<실시예 2>
덴드론-항체 면역접합체의 제조
Cis-aconityl doxorubicin에 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC)를 첨가한 후 15분간 반응시켰다. 이 용액에 시스타민 중심을 갖는 PAMAM 덴드리머를 첨가한 후 24시간 반응시키고, molecular weight cut-off가 3,000인 dialysis membrane으로 미반응물을 제거하여 독소루비신이 접합된 PAMAM 덴드리머를 제조하였다. 1 M dithiothreitol (DTT)를 이용하여 PAMAM 덴드리머의 디설피드 결합을 환원시켰다.
클라우딘-4 타깃 하는 항체의 시스테인기에 0.1 M phosphate buffer (pH 8.0) 에 녹인5,5‘-dithio-bis-(2-nitrobenzoic acid) (DTNB)를 반응시켜 -SH기를 활성화한 후, 미반응한 DTNB는 molecular weight cut-off가 30,000인 ultrafiltration tube를 이용하여 제거하였다. DTNB로 활성화된 항체와 독소루비신이 접합된 PAMAMA 덴드리머를 혼합한 후 3시간동안 반응시켜 덴드론-항체 면역접합체를 제조하였다.
제조된 덴드론-항체 면역접합체는 size-exclusion HPLC를 이용하여 분석하였다. 컬럼으로는 Superose 6 10/300 GL (GE, USA)을 사용하였고, 이동상으로는 10 mM phosphate buffered saline (pH 7.0)을 사용하였으며, UV 280 nm와 490 nm에서의 흡광도를 이용하여 물질을 검출하였다.
도 4의 size-exclusion HPLC 크로마토그램에서 확인되는 바와 같이, 독소루비신이 흡광도를 나타내는 490 nm에서의 피크 검출여부를 가지고, 덴드론-항체 접합체의 제조 여부를 확인하였다.
<실시예 3>
덴드론-항체 접합체의 독소루비신 함량 측정
덴드론-항체 접합체에 함유된 독소루비신 함량을 측정하기 위해 검체에 2.5 M 염산과 메탄올을 처리한 후 방출되어 나온 doxorubicinone 양을 역상 HPLC 방법으로 정량하였다. 역상 HPLC는 컬럼으로 DiKMA Inspire C-18 역상 컬럼 (4.6 x 250 mm, 5 micron)을 사용하였고, 이동상으로 3% (NH4)2CO3와 메탄올을 1:2 (v/v)로 혼합한 용액을 사용하였으며, 유속은 1 mL/min으로 하였고, 검출은 UV 490 nm에서 실시하였다. Doxorubicin 표준품을 이용하여 5.0에서 200 μg/mL 농도 범위에서 검량선을 작성하였고, 이 검량선을 이용하여 덴드론-항체 접합체에서 유리된 doxorubicinone 양을 정량하였다.
이에 대한 결과를 도 5에 나타내었다.
도 5는 Doxorubicin 표준품과 덴드론-항체 접합체에서 유리된 doxorubicin을 검출한 HPLC 크로마토그램이다. 정량 결과 항체 1분자당 평균적으로 doxorubicin 8분자가 결합된 것으로 측정되었다.
<실시예 4>
덴드론-항체 면역접합체의 in vitro 항암효능 평가
본 발명에 따른 덴드론-항체 접합체에 함유된 독소루비신 기능을 확인하기 위하여 B12 항체단독 혹은 덴드론-항체 접합체 독소루비신 처리에 따른 세포독성 여부를 확인하였다. 세포독성을 확인하기 위하여 96-well plate의 각 well에 1x104개의 OVCAR-3 세포를 분주한 뒤 24 시간 동안 37. 5°C, 5% CO2 환경에서 배양하였다. 24시간 이후 세포 배양액을 제거하고 B12 항체단독, 덴드론-항체 접합체 독소루비신 10~10000ng/ml 농도로 처리한 뒤 24시간 혹은 48시간 동안 배양하였다. 24시간 혹은 48시간 배양한 뒤 배양액을 제거하고 PBS 워시를 2회 진행한 뒤 WST시약을 1:10으로 배양액에 섞어 각 well당 100μl씩 처리한 뒤 2시간 동안 37. 5°C, 5% CO2 환경에서 방치한 뒤 430nm파장에서 흡광도를 측정하였다.
그 결과, B12항체 단독은 세포독성이 나타나지 않았으나 덴드론-항체 접합체 독소루비신 처리한 세포군에서는 세포독성이 농도 의존적으로 나타나는 것으로 보아, 덴드론-항체 접합체 독소루비신이 세포 독성 활성을 나타냄을 확인하였다.
상기에서 살펴본 바와 같이, 본 발명에 따른 면역접합체는 표면에 다수의 약물을 결합시킬 수 있는 덴드론과 표적 지향적인 항체를 접합함으로써 표적 지향적인 약물치료에 이용될 수 있으며, 특히 다수의 항암제가 결합된 친수성 덴드론을 항체에 접합시킴으로써, 종양 특이적으로 높은 농도의 약물을 전달하여 강력한 항암효과를 나타낼 수 있는 약학적 조성물을 제공할 수 있다는 점에서 산업상 이용가능성이 높다.
Claims (13)
- 중심에 티올(thiol)기를 갖는 덴드론(dendron) 및 항체를 포함하는 면역접합체.
- 제1항에 있어서, 링커를 추가적으로 포함하는 것을 특징으로 하는 면역접합체.
- 제1항에 있어서, 상기 덴드론은 시스타민 중심을 갖는 덴드리머(dendrimer)의 환원에 의해 생성된 것임을 특징으로 하는 면역접합체.
- 제3항에 있어서, 상기 덴드리머는 폴리아미도아민(PAMAM) 덴드리머인 것을 특징으로 하는 면역접합체.
- 제1항에 있어서, 상기 항체는 종양 특이적 항체인 것을 특징으로 하는 면역접합체.
- 제1항에 있어서, 상기 항체는 폴리클로날 항체, 모노클로날 항체, 키메라 항체, 인간화된 항체, Fv-단편, Fab-단편, F(ab’)2-단편 및 scFv 단편으로 이루어진 군에서 선택된 어느 하나인 것을 특징으로 하는 면역접합체.
- 제1항에 있어서, 상기 항체의 중쇄 또는 경쇄의 불변영역 또는 가변영역에 존재하는 시스테인 잔기에 덴드론이 접합된 것을 특징으로 하는 면역접합체.
- 제1항에 있어서, 상기 항체가 중쇄 또는 경쇄의 불변영역 또는 가변영역의 하나 또는 그 이상의 잔기가 시스테인 잔기로 치환된 항체로서, 상기 치환된 시스테인 잔기에 덴드론이 접합된 것을 특징으로 하는 면역접합체.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 덴드론은 링커를 통해 항체에 접합된 것을 특징으로 하는 면역접합체.
- 제1항에 있어서, 상기 덴드론에 항암약물이 접합되어 있는 것을 특징으로 하는 면역접합체.
- 제10항의 면역접합체를 유효성분으로 포함하는 암의 예방 또는 치료용 약학적 조성물.
- 제1항 내지 제6항 중 어느 한 항에 기재된 면역접합체의 덴드론 표면에 약물을 운반하는 것이 특징인 표적지향적 약물 전달용 면역접합체.
- (a) 시스타민 중심을 갖는 폴리아미도아민(PAMAM) 덴드리머를 환원시켜 중심에 티올(thiol)기를 갖는 덴드론을 형성하는 단계; 및
(b) 상기 덴드론을 항체에 접합시키는 단계를 포함하는 면역접합체 제조방법.
