Abstract

개별 유방암의 작동자(driver)를 타겟으로 하는 치료, 특히 ER-양성 루미날 타입 암에 대한 내분비 치료 및 HER2 증폭을 갖는 암에 대한 트라스투주맙 치료는 유방암을 갖는 여성에서 실질적으로 개선된 임상 결과를 갖는다(2). 대조적으로, ER, PR 또는 HER2를 발현하지 않는 삼중-음성 유방암은 타모자이펜(tamoxifen) 및 아로마타제(aromatase) 억제제와 같은 호르몬 치료 또는 헤르셉틴(herceptin)과 같은 HER2 타겟 치료에 대하여 효과가 없다. 게다가, 삼중-음성 유방암은 증식성이 높고, 재발에서 사망까지 짧은 시간을 보인다(3). 따라서, 유방암의 상기 하위 그룹은 5년 내 원인-특성 생존(cause-specific aurvival)이 매우 불량한 가장 공격적인 암 타입으로 알려져 있다(4). 그러나, 삼중-음성 종양 성장을 촉진하는 종양 형성 작동자(driver) 및 인자(factor)들은 여전히 불명확하고, 이러한 유방암 타입에 대하여 적용가능한 효과적인 임상적 치료가 존재하지 않는다.
종양형성 유전자의 Ras 패밀리의 GTP 가수분해 효소(GTPase)는 세포 외부의 신호를 핵으로 도입하고, 세포 분화, 증식, 생존, 세포 주기의 진행, 이동 및 세포골격 변화와 연관되어 있는 하류 신호전달 경로의 다양한 배열에 다양한 상류 신호를 연결하는 신호 네트워크 내의 분자 스위치로서 행동한다(5, 6, 7). 신호전달 경로의 이러한 과잉 사이의 균형은 최종 세포 결과를 결정하는데 매우 중요하다. 이러한 "Ras-중독(addicted)" 암 세포내의 가장 빈번하게 활성화된 종양형성 유전자인 KRAS(Kirsten rat sarcoma viral oncogene homolog)를 갖는 모든 인간 악성 종양의 약 30%에서, RAS 돌연변이가 발견된다(8, 9). 비록 Ras의 돌연변이형이 대부분의 유방암(<5%)과 관련이 있는 것은 아닐지라도, 비정상의 Ras 활성 및 신호전달은 유방 암 진행을 촉진한다는 중요한 실험적인 증거가 있다(10). 이와 일치하여, Ras는 돌연변이 Ras가 없는 유방 암종(carcinoma) 세포에서 Ras는 빈번하게 활성화 된다. 상기 관찰과 같은 맥락에서, 임상적으로 주석이 잘 달린(well-annotated) 집단(cohort) 내에서, KRAS 종양형성 유전자(rs61764370 T>G)의 3` UTR(번역되지 않은 지역) 내의 기능성 생식 세포 이형이 최근 확인되었고, 침윤적인 상피성 난소 암(12) 및 유방암(13) 발병률 증가와 관련이 있는 것으로 보고되었다. 이러한 기능성 이형은 in vitro KRAS의 증가된 발현을 이끄는 let-7 miRNA 결합 부위를 방해하는 것으로 밝혀졌다. 특히 유방암의 KRAS 이형의 빈도는 삼중 음성 유방암과 연관이 있는 것으로 밝혀졌고, K-RAS가 삼중-음성 유방 암에 대한 새로운 치료적 타겟이 될 수 있다는 것을 제안한다. 그러나, K-Ras 타겟팅은 치료학적 접근으로서 광범위하게 성공적이지 못했고, K-Ras 돌연변이 암의 치료는 암 치료에 있어서 가장 극복하기 힘든 장애 중 하나로 남아있다(15, 16).
플로로글루시놀은, 히드록실 그룹으로 구성된 방향족성 페닐 고리를 포함하는 화학적 구조를 갖는 페놀계 화합물이다. 페놀계 화합물이 그러하듯, 플로로글루시놀은 다양한 생물학적 활성을 보이고, 의약, 화장품, 농약, 페인트, 시멘트 및 염료에서 다양하게 사용된다(17). 이전에, 촉매 활성을 통해 H2O2-유발 산화 스트레스로부터 세포를 보호하는 것으로 밝혀졌다(18). 또한, 플로로글루시놀은 산화스트레스를 억제하는 것에 의해 감마 방사선 유발의 세포 손상으로부터 방사선 방호를 한다(19). 더욱이 플로로글루시놀은 유사분열촉진 프로틴 카이네이즈(mitogenactivated protein kinases)에 대한 억제 활동을 통하여, 인간 케라티노사이트 내의 기질 메탈로프로티네이즈-1을 억제함으로써 콜라겐 파괴를 방지한다(20). 진피층 피부의 주요한 구조적 구성인 콜라겐의 변화는 자연적 노화 및 광노화피부에서 관찰되는 임상적인 변화의 원인으로 시사되었기 때문에(21), 플로로글루시놀은 피부 노화에 대한 잠재적인 예방 또는 치료제이다.
본 명세서 전체에 걸쳐 다수의 논문 및 특허문헌이 참조되고 그 인용이 표시되어 있다. 인용된 논문 및 특허문헌의 개시 내용은 그 전체로서 본 명세서에 참조로 삽입되어 본 발명이 속하는 기술 분야의 수준 및 본 발명의 내용이 보다 명확하게 설명된다.
본 발명자들은 K-Ras가 과발현 된 암세포의 활발한 이동 및 침윤 특성을 저해하여 암의 전이를 예방하고, 항암제(또는 방사선 조사)와 병용투여를 통해 높은 항암효과를 얻을 수 있는 약물을 개발하고자 예의 연구 노력하였다. 그 결과 플로로글루시놀(Phloroglucinol)을 이용하는 경우 상술한 효과가 우수하게 달성됨을 규명함으로써, 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은 K-Ras가 과발현된 암의 예방, 치료 또는 전이 억제용 약제학적 조성물을 제공하는데 있다.
본 발명의 다른 목적은 병용투여 형태로 사용하는, K-Ras가 과발현된 암 예방, 치료 또는 전이 억제용 약제학적 조성물을 제공하는데 있다.
본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명, 청구범위 및 도면에 의해 보다 명확하게 된다.
본 발명의 일 양태에 따르면, 본 발명은 (a) 유효성분으로서의 약제학적 유효량의 플로로글루시놀(phloroglucinol), 그의 염, 수화물 또는 용매화물; 및 (b) 약제학적으로 허용되는 담체를 포함하는, K-Ras가 과발현된 암 예방, 치료 또는 전이 억제용 약제학적 조성물을 제공한다.
본 발명자들은 K-Ras가 과발현 된 암세포의 활발한 이동 및 침윤 특성을 저해하여 암의 전이를 예방하고, 항암제(또는 방사선 조사)와 병용투여를 통해 높은 항암효과를 얻을 수 있는 약물을 개발하고자 예의 연구 노력하였다. 그 결과 플로로글루시놀(Phloroglucinol)을 이용하는 경우 상술한 효과가 우수하게 달성됨을 규명하였다.
본 발명에서 유효성분으로 이용하는 플로로글루시놀(phloroglucinol, PG)은 도 7에 나타낸 화학식으로 표시된다. 플로로글루시놀히드록실 그룹으로 구성된 방향족성 페닐 고리를 포함하는 화학적 구조를 갖는 페놀계 화합물이다. 페놀계 화합물이 그러하듯, 플로로글루시놀은 다양한 생물학적 활성을 보이고, 의약, 화장품, 농약, 페인트, 시멘트 및 염료에서 다양하게 사용되지만, K-Ras 과발현 암세포의 전이능 억제에 대하여 개시된 바 없다.
본 발명의 약제학적 조성물의 가장 큰 특징은 K-Ras가 과발현된 암의 예방, 치료 또는 전이를 억제하는 것이다.
본 발명의 K-Ras가 과발현된 암을 표현하기 위해 사용된 용어 "과발현(overexpression)"은 적합한 발현 분석법(예컨대, 웨스턴 블랏 분석, Peter B. Kaufma et al., Molecular and Cellular Methods in Biology and Medicine, 108-121, CRC press)을 통해 K-Ras의 발현 수준을 측정한 경우에 있어서, 정상세포(예컨대, 정상 인간 유방 상피 세포)의 K-Ras 발현 수준에 비하여 1.5배 이상, 바람직하게 1.7배 이상, 보다 바람직하게는 2배 이상, 보다 더 바람직하게는 2.3배 이상이거나, 비악성(non-malignant) 암세포(예컨대, 비악성 유방암 세포)의 K-Ras 발현 수준에 비하여 1.5배 이상, 바람직하게 1.7배 이상, 보다 바람직하게는 2배 이상, 보다 더 바람직하게는 2.5배 이상, 보다 더욱 더 바람직하게는 3배 이상인 것을 의미한다(도 1).
본 발명의 일 구현예에 따르면, 본 발명의 상기 유효성분은 암세포의 이동 및 침윤을 억제한다. 예를 들어, 유방암 세포에 있어서, 전이능력이 뛰어난 기저형(basal type)의 유방암 세포와 그렇지 못한 관강형(luminal type) 유방암 세포가 존재하는데, 기저형의 유방암 세포에서는 K-ras의 활성도가 높아져 있음을 알 수 있고, K-Ras를 억제하였을때, 암세포의 이동과 침윤 능력이 감소한다. 이는 K-Ras를 억제하는 경우 EMT 조절인자로 알려진 slug의 발현을 감소시키는 것에 의한다. 상피-중간엽 전환(Epithelial-mesenchymal transition)(EMT)은 정상적 배아발달 과정에서 발생하는 현상으로, 세포가 상피(epithelial)성 세포 표현형을 상실하고 이동성이 높은 중간엽(mesenchymal)성 세포 표현형으로 전환하는 과정이며, 암세포의 전이가 일어날 때 이동성과 침윤성을 증가시키는 기전으로도 잘 알려져 있다. 본 발명의 유효성분인 플로로글루시놀을 처리하는 경우, EMT 조절인자인 상기 slug의 발현을 감소시킨다는 결과를 도출하였으며, 이로 인하여 암세포의 이동 및 침윤을 억제시킬 수 있다.
본 발명의 일 구현예에 있어서, 본 발명의 조성물은 다른 항암제 또는 방사선과 병용 사용된다. 본 발명의 조성물의 유효성분인 플로로글루시놀이 K-Ras가 과발현된 전이성 암세포에 대하여, 이동 및 침윤 특성을 억제할 수 있다는 결과를 도출하였는 바, 종래 공지된 암세포를 억제하기 위한 함암제 및 방사선 치료 등과 병행하여 사용함으로써, 암세포의 사멸을 유도하고, 전이능력 또한 억제하여 보다 효율적인 암 치료를 할 수 있다. 본 발명의 상기 항암제는 특별한 제한이 없으나, 예를 들면, 택솔(taxol), 시스플라틴(cisplatin), 카르보플라틴(carboplatin), 프로카르바진(procarbazine), 메클로레타민(mechlorethamine), 시클로포스파미드(cyclophosphamide), 이포스파미드(ifosfamide), 멜팔란(melphalan), 클로라부실(chlorambucil), 비술판(bisulfan), 니트로소우레아(nitrosourea), 디악티노마이신(dactinomycin), 다우노루비신(daunorubicin), 독소루비신(doxorubicin), 블레오마이신(bleomycin), 플리코마이신(plicomycin), 미토마이신(mitomycin), 에토포시드(etoposide), 탁목시펜(tamoxifen), 트랜스플라티눔(transplatinum), 5-플루오로우라실(5-fluorouracil), 아드리아마이신(adriamycin), 빈크리스틴(vincristin), 빈블라스틴(vinblastin) 또는 메토트렉세이트(methotrexate)일 수 있다. 또한, 상기 방사선 치료는 특별히 제한되지는 않지만, 예를 들면 X-ray 조사 또는 γ-ray 조사를 이용한 치료일 수 있다.
본 발명의 일 구현예에 있어서, 본 발명에 의해 예방, 치료 또는 전이가 억제되는 암은 유방암, 뇌암, 신경 내분비 암, 위암, 폐암, 난소암, 간암, 기관지암, 비인두암, 후두암, 췌장암, 방광암, 부신암, 대장암, 결장암, 자궁경부암, 전립선암, 골암, 피부암, 갑상선암, 부갑상선암 및 요관암이다.
본 발명의 일 구체예에 따르면, 본 발명에 의해 예방, 치료 또는 전이가 억제되는 암은 유방암이다. 더욱 구체적으로, 본 발명의 유방암은 삼중(triple)-음성 종양이다. 인간 유방암은 병리학, 분자적 프로필 및 치료적 민감성에 있어서의 차이에 의해 여러 다른 종류들로 이루어진(heterogeneous) 질병이다. 유방암은 에스트로겐 수용체(ER)-양성 및/또는 프로게스테론 수용체(PR)-양성, HER2(ERBB2)-증폭 및 삼중(triple)-음성(예를 들면, ER/PR-음성 및 HER2-음성) 종양으로 분류된다. 유전자 발현 및 수용체 프로파일링은 유방암을 4개의 생물학적 하위그룹으로 더 분류한다: 루미날 A(ER-양성 및/또는 PR-양성, HER2-음성), 루미날 B(ER- 및/또는 PR-양성, HER2-양성), HER2-양성(ER/PR-음성, HER2-양성), 및 기저층 유사(basal-like)(삼중-음성). ER, PR 또는 HER2를 발현하지 않는 삼중-음성 유방암은 타모자이펜(tamoxifen) 및 아로마타제(aromatase) 억제제와 같은 호르몬 치료 또는 헤르셉틴(herceptin)과 같은 HER2 타겟 치료에 대하여 효과가 없다. 게다가, 삼중-음성 유방암은 증식성이 높고, 재발에서 사망까지 짧은 시간을 보인다. 본 명세서에서 사용된 용어, 약제학적으로 허용되는 "염"은, 화합물이 투여되는 유기체에 심각한 자극을 유발하지 않고 화합물의 생물학적 활성과 물성들을 손상시키지 않는, 화합물의 제형을 의미한다. 용어 "수화물" 및 "용매화물" 역시 상기와 같은 의미를 가진다. 상기 약제학적으로 허용되는 염은, 플로로글루시놀을 염산, 브롬산, 황산, 질산, 인산 등의 무기산, 메탄술폰산, 에탄술폰산, p-톨루엔술폰산 등의 술폰산, 타타르산, 포름산, 시트르산, 아세트산, 트리클로로아세트산, 트리플루오로아세트산, 카프릭산, 이소부탄산, 말론산, 석신산, 프탈산, 글루콘산, 벤조산, 락트산, 푸마르산, 말레인산, 살리실산 등과 같은 유기 카본산과 반응시켜 얻어질 수 있다. 또한, 본 발명의 화합물을 염기와 반응시켜, 암모니움 염, 나트륨 또는 칼륨 염 등의 알칼리 금속염, 칼슘 또는 마그네슘 염 등의 알칼리 토금속염 등의 염, 디시클로헥실아민, N-메틸-D-글루카민, 트리스(히드록시메틸) 메틸아민 등의 유기염기들의 염, 및 아르기닌, 리신 등의 아미노산 염을 형성함으로써 얻어질 수도 있다.
본 명세서에서 사용된 용어, "수화물(hydrate)"은 비공유적 분자간력 (non-covalent intermolecular force)에 의해 결합된 화학양론적(stoichiometric) 또는 비화학양론적 (non-stoichiometric) 양의 물을 포함하고 있는 본 발명의 화합물 또는 그것의 염을 의미한다.
본 명세서에서 사용된 용어 "용매화물(solvate)"은 비공유적 분자간력에 의해 결합된 화학양론적 또는 비화학양론적 양의 용매를 포함하고 있는 본 발명의 화합물 또는 그것의 염을 의미한다. 그에 관한 바람직한 용매들로는 휘발성, 비독성, 및/또는 인간에게 투여되기에 적합한 용매들이다.

본 명세서에서 사용된 용어, "약제학적으로 허용되는 담체"는 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약제학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
본 발명의 약제학적 조성물은 경구 또는 비경구로 투여할 수 있고, 비경구 투여인 경우에는 정맥내 주입, 피하 주입, 근육 주입, 복강 주입, 경피 투여 등으로 투여할 수 있다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하며, 보통으로 숙련된 의사는 소망하는 치료 또는 예방에 효과적인 투여량을 용이하게 결정 및 처방할 수 있다. 본 발명의 일 구현예에 따르면, 본 발명의 약제학적 조성물의 1일 투여량은 0.0001-1000 ㎎/㎏이다.
본 발명의 약제학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화 함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나 엑스제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실험 방법
1-1. 플로로글루시놀(Phloroglucinol)
플로로글루시놀은 시그마(sigma)사에서 구입한 것을 사용하였다.
1-2. 세포배양
인간 유방상피세포주 MCF-10A, 유방암세포주 MCF-7 및 MDA-MB-231, BT474, BT549, T47D(American Type Culture Collection)로부터 확립되었다. 세포들은 37℃의 가습된 5% CO2 조건에서 배양하였다. 인간 유방상피세포주인 MCF-10A는 5%의 열-비활성화 처리된 말혈청(Invitrogen 사), 10 μg/ml의 인슐린, 20 ng/ml EGF, 0.1 μg/ml 콜레라독소, 0.5 μg/ml 하이드로코티손, 페니실린(100 unit/ml) 및 스트렙토마이신(100 μg/ml)이 보충된 DMEM/F-12 배지에서 배양했다. MCF-7 세포는 10% 태아소혈청, 페니실린(100 unit/ml) 및 스트렙토마이신(100 g/ml)이 보충된 최소이글배지(minimum Eagle's medium)에서 배양했다. MDA-MB-231, T47D, BT474 와 SK-BR3, 세포는 10% 태아소혈청, 페니실린(100 unit/ml) 및 스트렙토마이신(100 g/ml)이 보충된 DMEM에서 배양되었다. BT549 세포는 10% 태아소혈청, 페니실린(100 unit/ml) 및 스트렙토마이신(100 g/ml)이 보충된 RPMI에서 배양되었다.
1-3. 시약 및 항체
포스포-AKT에 대한 폴리클로날항체(Ser-473), 포스포-AKT(Thr-308), 포스포-ERK1/2(Thr-202/Tyr-204), 포스포-p38(Thr-180/Tyr-182), ERK1, p38, 포스포-JNK1/2(Thr-183/Tyr-185) 및 N-cadherin은 Cell Signaling Technology(Beverly 사)로부터 얻었다. AKT, JNK1, Zeb1, Snail 및 Slug에 대한 폴리클로날항체는 산타 크루즈 바이오테크놀로지(Santa Cruz Biotechnology)사에서 구입하였다. 폴리클로날항체 비멘틴은 써모 사이언스(Thermo Science)사로부터 얻었다. 4,6-디아미디노-2-페닐인돌(DAPI) 및 β-액틴에 대한 모노클로날항체는 시그마사로부터 입수하였다. 항-마우스 Alexa Fluor 488 및 항-토끼 Alexa Fluor 488은 Invitrogen사로부터구입했다. JNK(SP600125), p38 MAPK(PD169316), MEK(U0126), PI3K(LY294002)와 Raf1(GW5074)의 특이적인 억제제는 칼바이오캠(Calbiochem)에서 입수하였다.
1-4. 웨스턴블랏(Western blot) 분석
세포용해물(lysates)은 단백질 분해효소 억제제가 보충된 용해버퍼(lysis buffer)[40 mM 트리스-HCl(pH 8.0), 120 mM NaCl, 0.1% Nonidet-P40]로 단백질을 추출하여 준비하였다. 단백질은 SDS-PAGE에 의해 분리하고, 니트로셀룰로스막 (Amersham)으로 이동시켰다. 막은 TBS(Tris-buffered saline) 내의 5% 탈지분유로 차단(blocked)하였고, 4℃에서 하루 동안 1차 항체와 배양하였다.
블랏은 퍼옥시데이즈가 접합된 2차 항체로 수행하였고, 단백질은 제조사의 지침에 따른 강화된 화학발광(ECL)절차(Amersham)를 이용하여 시각화하였다.
1-5. PI3 키나아제분석
세포용해물(300 μg/500 μl)은 항-p85 항체(Upstate Biotechnology 사)를 사용하여 면역 침강을 실시하였다. 침전물은 1 % NP40/PBS로 2회 세척하고, 이어서 PBS, 0.1 mol/L 트리스-HCl(pH 7.5)/0.5 mol/L LiCl, 그리고 25 mmol/L HEPES(pH 7.5) / 100 mmol/L NaCl / 1 mmol/L EDTA로 세척하였다. 그 뒤 침전물은 50 μl의 전-초음파 처리한(presonicated) 포스파티딜이노시톨 기질에서 재현탁(resuspended)시키고, 상온에서 10 분간 배양하였다. 각 샘플은 10 μ Ci[32P]ATP로 상온에서 10분간 표지되었다. 반응은 클로로포름/메탄올/HCl(50:100:1) 100 μl를 첨가하여 중지시켰다. 지질은 200 μl 클로로포름으로 혼합하고 원심분리하여 추출하였고, 하부의 유기상은 새로운 튜브로 옮겼다.
유기상은 100 μl 메탄올 / 1 mol/L HCl (1:1)로 1회 세척하고, 상층을 제거하였다. 그 뒤 지질부는 건조하고 20 μl 의 클로로포름에 재현탁하였으며, 1 %의 포타슘옥살레이트에 미리 담지된 실리카겔 TLC(thin layerchromatography) 플레이트에 적용하였다. 인지질은 상온의 폐쇄된 유리체임버 내에서 45분 동안 새롭게 준비된 버퍼[클로로포름 / 메탄올 / 암모니아 / 물(43:38:5:7)] 상의 TLC에 의해 분석하였다.
1-6. 활성화된 K-Ras 친화성 침강분석(affinity precipitation assay)
활성화된 Ras 친화성 침강분석은 제조사의 지침에 따라 수행하였다. 요컨대, 세포 용해물은 Raf-1 RBD 아가로오스비즈(Upstate 사) 5 μg과 함께 4℃에서 30분간 배양하였다. 아가로오스 비즈를 세척 완충액(25 mM HEPES, pH 7.5), 10 mM MgCl2, 150 mM Nacl, 1 mM EDTA, 1% Nonidet P-40, 1 mM Na3VO4, 10% 글리세롤, 10 μg/ml 류펩틴, 10 μg/ml 아프로티닌 및 25 mM NaF로 3회 세척(extensive washing) 한 후, Raf-1 RBD 아가로오스비즈에 부착 된, 활성화 된 K-Ras(GTP-Ras)를 SDS-PAGE 샘플 완충액의 첨가에 의해 방출시켰다. 활성화 된 K-Ras의 양은 K-Ras 항체를 사용한 면역블랏(immunoblotting)에 의해 측정하였다.
1-7. 형질전환
발암성 K-Ras(G13D)를 레트로바이러스벡터 MFG 내로 도입하였다. 레트로바이러스 생산을 위해 HD29D 세포주를 10% 태아소혈청, 2 mmol/L GlutaMAX (Invitrogen 사), 50 unit/ml 페니실린 / 스트렙토마이신, 1 μg/ml 테트라사이클린, 2 μg/ml 퓨로마이신(puromycin) 및 0.6 mg/ml G418 설페이트(Calbiochem 사)가 보충된 DMEM(Invitrogen 사) 배지에서 배양했고, 리포펙타민(Lipofectamine) 2000 시약(Invitrogen)을 이용하여 MFG 또는 MFG-K-Ras에 감염시켰다. 감염 48시간 후, 바이러스 상청액(supernatant)을 새로운 배지의 보충에의해 5일 동안 매일 수득하였고, 0.45 μm 필터에 통과시켰으며, 바이러스 상청액은 영하 80℃에 냉동시켰다. 상청액은 4 μg/ml 폴리브린(Polybrene)(Sigma)을 첨가한 후 감염에 이용하였다.
1-8. 면역세포화학(EMT 분석)
세포는 4% 파라포름알데하이드로 고정하였고, PBS 상에서 0.1% 트리톤 X-100을 이용하여 투과시켰다. 세포고정 후, 세포는 4℃에서 하루 동안 1% 태아소혈청및 0.1% 트리톤 X-100과 PBS 용액상에 적절한 1차 항체와 함께 배양하였다. 사용된 항체는 하기와 같다: 인간 항-E-캐드헤린(anti-E-cadherin)(마우스폴리클로날항체, 1:200), 항-N-캐드헤린(마우스폴리클로날항체, 1:200) 및 항-비멘틴(Vimentin)(토끼폴리클로날항체, 1:200). 염색은 항-토끼 또는 항-마우스 Alexa Flour 488(Molecular Probes)를 사용해 시각화 하였다.
핵(nuclei)은 4, 6-디아미디노-2-페닐인돌(DAPI)(Sigma 사)을 사용하여 대조염색 하였다. 염색된 세포는 형광-현미경(Olympus IX71)을 사용하여 시각화하였다.
1-9. 침윤이동분석
세포(2 × 104cell/well)를 침윤 및 이동 분석을 위해 0.2 ml의 영양 배지에 현탁했다. 침윤분석을 위해 세포는 8 mm 포어크기의 트랜스웰 챔버(Transwell chamber)(Corning Glass)의 상부 웰(upper well) 상에 채워 넣었다.
상기 상부웰은 0.8 ml의 성장 배지로 채워진 하부 웰 챔버의 상면(upper side) 상에서 성장인자-감소된 매트리겔(BD Diosciences) 10 mg/ml로 프리코팅되어있다. 37℃에서 48 시간 배양 후 필터의 상부 표면 상의 비침습세포를 고정하고, Diff-Quick 키트(Fisher 사)로 염색하였고, 20 배율로 촬영하였다. 침습력은 웰 당 5 미시영역에서 세포의 수를 세는 것으로 결정하였고, 침습의 규모는 미세영역당 세포의 평균 수로 표현하였다. 세포는 위상차 현미경(Leica Microsystems)으로 이미지화하였다. 이동분석을 위하여, 같은 타입의 막을 포함하는 대조 인서트(inserts)를 갖지만 매트리겔 코팅은 갖지 않는 챔버를 사용하였다(24-웰플레이트의 웰당 하나의 챔버). 성장배지 0.2 ml 내의 2×104개의 세포를 각 인서트 상부 끝측에 첨가하고, 증식배지 0.8ml를 각 인서트의 기저 측에 첨가하였다. 침습분석을 위해 위와 같이 인서트를 처리하였다.
1-10. 통계분석
모든 실험데이터는 평균값으로 기재하였고, 에러 바는 실험적인 표준 오차를 나타낸다. 통계 분석은 비모수 스튜던트 t 검정(non-parametric Student t test)으로 수행하였다.
1-11. 동물 실험
마우스는 5주령차인 BALB/c 누드 마우스를 사용하였다. 각각의 개체는 5마리씩으로 진행을 하였다. 플로로글루시놀을 10 mg/kg으로 처리하였다. MDA-MB-231-LM2 세포를 사용하였다.

본 명세서 상의 도면이 나타내는 바를 중심으로 실험결과들을 설명하였다.

유방암 세포는 전이능력이 뛰어난 기저형(basal type)의 유방암 세포와 그렇지 못한 관강형(luminal type)의 유방암 세포로 나뉠 수 있다. 기저형의 정상 유방 세포인 MCF10A를 실험에 사용하였다. 암화유전자인 Ras 단백질은 활성화가 되면 기질인 Raf와의 결합능력이 증가하게 되는 것을 이용하여 Ras와 Raf의 결합정도를 측정함으로써 Ras의 활성도를 측정하였다. 전이능력이 뛰어나다고 알려진 관강형의 세포에서 K-Ras의 활성도가 높아져 있음을 확인하였다.

세포의 이동 및 침윤 능력을 측정하기 위하여 침윤 및 이동 분석(invasion & migration assay)을 실시하였다. 암화유전자인 K-Ras를 억제하였을 때, 이동과 침윤능력이 감소하는 것을 확인함으로써, 암화유전자인 K-Ras가 세포의 이동 및 침윤에 관여를 하고 있다는 것을 확인하였다.

기저형의 세포는 중간엽(mesenchymal)성 세포 표현형을 나타내는데, 중간엽성 세포의 표지자(marker)인 피브로넥틴과 비멘틴을 확인하였다. 암화유전자인 K-Ras의 억제는 중간엽성 세포의 특징을 잃어버리게 함을 확인하였다.

유방암세포는 폐(lung)로의 전이가 가장 잘 일어난다. 동물실험에서도 이미 확인이 되어있는 사실이다. 마우스의 꼬리혈관을 통하여 기저형의 유방암세포(MDA-MB-231)를 주입시켰다. 그 결과 K-Ras를 억제한 세포를 주입시킨 마우스는 폐로의 전이가 적게 일어남을 확인하였다.

상피-중간엽 전환(Epithelial-mesenchymal transition)(EMT)은 정상적 배아발달 과정에서 발생하는 현상으로, 세포가 상피(epithelial)성 세포 표현형을 상실하고 이동성이 높은 중간엽(mesenchymal)성 세포 표현형으로 전환하는 과정이며, 암세포의 전이가 일어날 때 이동성과 침윤성을 증가시키는 기전으로도 잘 알려져 있다. 암화 유전자 (K-Ras)를 발현한 세포는 세포와 세포간의 결합 및 EMT 마커로 알려진 단백질의 변화가 있음을 관찰하였다. 이는 플로로글루시놀에 의해 단백질 변화가 억제됨을 나타낸다. EMT 조절인자로 알려진 slug 의 발현이 플로로글루시놀에 의해 억제됨을 알 수 있었다.

면역형광염색법(immunofluorescence stain)을 이용하여 EMT 마커인 E-cadherin, N-cadherin, Vimentin 발현을 비교해 본 결과 플로로글루시놀 처리한 후 발현이 억제되는 것을 확인하였다.

암화유전자와 플루루글루시놀이 세포 내에서의 조건에서 어떠한 신호에 의한 조절인지 아니면 직접적인 결합을 통한 조절인지를 확인하고자 세포를 분해하여 단백질들을 추출, 그 단백질들에 직접 플루루글루시놀을 처리하여 암화유전자인 K-Ras의 활성을 측정하였다. 그 결과 활성이 감소하는 것을 확인, 직접적인 결합을 통한 효과임을 확인하였다.

마우스의 폐 조직을 IHC(면역조직화학염색)방법을 통하여 관찰하였다. 폐 조직에서 GFP의 발현을 확인하였지만, 플로로글루시놀을 처리한 마우스의 폐에선 관찰 할 수 없었다.

마우스의 생존 기간을 확인한 결과, 플로로글루시놀을 처리한 경우 실험 종료 시점까지 모두 생존하였으나, 처리하지 않은 마우스는 6주차에서부터 폐사하기 시작하여 실험 종료 시점엔 모두 폐사하였다.
이상과 같이 본 발명자들은 정상 세포에서 플로로글루시놀의 보호적인 역할과는 대조적으로, 플로로글루시놀이 K-Ras-유발 암 진행에 대항 항암 활성을 갖는다는 것을 밝혔다. 본 발명자들은 플로로글루시놀은 K-Ras-유도된 상피-중간엽 전환(EMT)를 약화시키고, 이에 의해 K-Ras-유발 암 전이를 억제한다는 것을 밝혔다. 또한, 본 발명자들은 플로로글루시놀이 K-Ras와 직접적으로 상호작용하고, 그 결과 PI3K(phosphoinositide 3-kinase)/AKT 및 Raf-1/ERK(extracellular signal-regulated kinase)와 같은 K-Ras의 하류 작용자를 억제한다는 것을 밝혔다. 이러한 결과는 플로로글루시놀이 정상 세포에는 손상을 주지 않고, 선택적으로 K-Ras-유발 암 진행을 타겟으로 하고, 유망한 치료제임을 시사한다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현 예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.

Claims (6)
- (a) 유효성분으로서의 약제학적 유효량의 플로로글루시놀(phloroglucinol), 그의 염, 수화물 또는 용매화물; 및 (b) 약제학적으로 허용되는 담체를 포함하는, K-Ras가 과발현된 암 예방, 치료 또는 전이 억제용 약제학적 조성물.

- 제 1 항에 있어서, 상기 암은 유방암, 뇌암, 신경 내분비 암, 위암, 폐암, 난소암, 간암, 기관지암, 비인두암, 후두암, 췌장암, 방광암, 부신암, 대장암, 결장암, 자궁경부암, 전립선암, 골암, 피부암, 갑상선암, 부갑상선암 및 요관암으로 구성된 군으로부터 선택되는 암인 것을 특징으로 하는 조성물.
- 제 4 항에 있어서, 상기 암은 유방암인 것을 특징으로 하는 조성물.
- 제 5 항에 있어서, 상기 유방암은 삼중(triple)-음성 종양인 것을 특징으로 하는 조성물.
