실시예6 MTT assayExample 6 MTT assay

시료에 의한 macrophage Raw 264.7 cells 에 미치는 영향은 Carmichael 등의 MTT assay를 통해 평가하였다. 96well plate에 5×104 cells/well로 분주한 후, 80%의 confluency에 도달 할 때 serum free EMEM으로 교체하였다. 천연복합물을 농 도별로 첨가하여 배양 후 생존세포에 MTT (5mg/mL)를 4시간 처리 한 후 배지를 제거하고 생성된 formazan crystal을 DMSO에 녹여 흡광도 540nm에서 측정하였다. 본 발명에서 측정해본 결과 Chitosan, PVA등의 단일물질과 혼합 물질의 세포독성이 나타나지 않았다.     The effect of the sample on macrophage Raw 264.7 cells was evaluated by MTT assay of Carmichael et al. 실시예7 세포활성시험Example 7 Cell Activity Test

상기 실시예 1로 제조한 시트 위에 macrophage Raw 264.7 cells의 세포활성 시험을 실시하였다. T-bottle에 셀을 분주한 후, 80%의 confluency에 도달 할 때 serum free EMEM으로 교체하였다. 6well plate에 제조한 시트를 넣고, macrophage Raw 264.7 cells을 5×104 cells/well의 세포를 분주한 후 24시간, 48시간 세포활성을 측정하였다. 위와 같은 방법으로 측정한 결과 24시간 이후 48시간째 세포배양이 2~4배 가까이 증가한 것으로 보아 세포활성이 우수한 것을 알 수 있었다.   The cell activity test of macrophage Raw 264.7 cells was performed on the sheet prepared in Example 1. 실시예8 첩포시험Example 8 Patch Test

상기 실시예 1의 제조한 나노섬유 부직포를 이용한 마스크 시트의 첩포시험을 실시 하였는데, 피부 안전성 실험인 첩포시험은 Table 3와 같은 국제 접촉피부염 연구위원회 (International Contact Dermatitis Research Group)의 기준으로 판정하였다. 팔의 안쪽 부분에 부착하여 48시간 후의 피부 반응을 확인하였다.   The patch test of the mask sheet using the nanofiber nonwoven fabric prepared in Example 1 was carried out.
