Abstract
본 발명은 돼지 인플루엔자 바이러스 A/Swine/Korea/S83/2004(H9N2)및 이의 백신에 관한 것이다. 특히 본 발명은 조류 인플루엔자 바이러스와 사람 인플루엔자 바이러스간의 조합으로 생성된 신규 인플루엔자 바이러스 A/Swine/Korea/S83/2004(H9N2) 및 이의 백신을 제공한다. 본 발명에 따라 돼지 인플루엔자 바이러스에 대한 면역을 효과적으로 형성시킬 수 있으며 따라서 돼지 농장에서의 인플루엔자 바이러스에 의한 질환 발병을 현저히 감소시켜 경제적인 손실을 방지할 수 있다.

도 1은 A/Swine/Korea/S109/2004(H9N2), A/Swine/Korea/S81/2004(H9N2) 및 A/Swine/Korea/S83/2004(H9N2) 인플루엔자 바이러스의 계통발생학적 관련성을 HA(Haemagglutinin) 유전자 염기서열로 분석한 것이다.

본 발명은 인플루엔자 바이러스 A/Swine/Korea/S83/2004(H9N2) 및 이의 백신에 관한 것이다.
20세기에 세계적으로 3종의 유행병이 발병하였었다(J. K. Taubenberger et al., Science 275, 1793 (1997) ; Schafer, J. R. et al.,
J. Virol. 194, 781 (1993) ; C. Scholtissek, W. Rohde, V. Von Hoyningen, R. Rott, Virology 87, 20 (1978) ; Y. Kawaoka, S. Krauss, R. G. Webster, J. Virol. 63, 4603 (1989)). 조류 인플루엔자계 H1N1 바이러스는 1918년에 "스페인 독감"을 야기하였고, 재조합 H2N2 인플루엔자 바이러스와 H3N2 인플루엔자 바이러스는 1957년 "아시아 독감'과 1968년 "홍콩 독감"을 발병시켰다. 재조합 H2N2 유행성 인플루엔자 바이러스와 H3N2 유행성 인플루엔자 바이러스는 사람과 조류 인플루엔자 바이러스간의 조합에 의하여 돼지에서 형성된 것으로 추정된다.
서브타입 인플루엔자 A형 바이러스는 15종의 HA(haemagglutinin)와 9종의 NA(neuraminidase)로 이루어져 있으며(Webster, R. G. et al., Microbiol. Rev. 56, 179 (1992)), 이중 H9N2 및 H5N1 서브타입은 사람 인플루엔자 유행병을 야기할 수 있는 가장 큰 가능성을 가지고 있다. H5N1 인플루엔자 바이러스는 중국, 베트남 및 타이완을 포함한 많은 아시아 국가에서 확인되는 고유종이며(S. H. Seo, E. Hoffmann, R. G. Webster Nat. Med. 8, 950 (2002) ; T. H. Tran, et al.,
N. Engl .J. Med. 350,1179 (2004) ; K. S. Li, et al.,
Nature 430, 209 (2004)), H9N2 인플루엔자 바이러스는 한국 및 중국과 같은 다수의 국가에 만연해있는 실정이다. 1999년 홍콩에서는 2명의 어린이가 H9N2 인플루엔자 바이러스에 감염된 바 있다(M. Peiris, et al.,
Lancet 354, 916 (1999)).
한편, 돼지는 유행성 인플루엔자 바이러스를 창조하여 사람에서 사람으로 전이시키는 "혼성관"으로 여겨진다(C. Scholtissek, Med. Princ. Pract. 2, 65 (1990)). 즉 돼지는 조류 인플루엔자 바이러스와 사람 인플루엔자 바이러스 모두에 감염가능하며(H. Kida, et al.,
J. Gen. Virol. 75, 2183 (1994)), 이들 바이러스간의 조합으로 새로운 형태의 인플루엔자 바이러스가 제조되기도 한다.
상기 종래기술의 문제점을 해결하기 위하여 안출된 것으로서, 본 발명은 조류 인플루엔자 바이러스와 사람 인플루엔자 바이러스 간의 재조합으로 생성된 신규한 돼지 인플루엔자 바이러스를 제공하는 것을 목적으로 한다.
또한 본 발명은 조류 인플루엔자 바이러스와 사람 인플루엔자 바이러스 간의 재조합으로 생성된 신규한 돼지 인플루엔자 바이러스에 대해 면역을 형성시킬 수 있는 인플루엔자 백신 조성물을 제공하는 것을 목적으로 한다.
본 발명은 사람 인플루엔자 바이러스와 조류 인플루엔자 바이러스 간의 조합으로 생성된 신규 돼지 인플루엔자 바이러스 및 상기 바이러스에 대한 백신 조성물에 관한 것이다.
본 발명의 돼지 인플루엔자 바이러스는, A/Swine/Korea/S10/2004(H1N1), A/Swine/Korea/S109/2004(H9N2), A/Swine/Korea/S81/2004(H9N2) 및 A/Swine/Korea/S83/2004(H9N2)이다.
상기 A/Swine/Korea/S10/2004(H1N1) 인플루엔자 바이러스는 PB2(Polymerase basic protein 2) 유전자를 제외한 나머지 유전자들, PB1, PA(Polymerase), HA(Hemagglutinin), NP(nucleoprotein), NA(Neuraminidase), M(matrix) 및 NS (nonstructural) 유전자는 사람 인플루엔자 바이러스 A/WSN/33(H1N1)와 유사하고, PB2 유전자는 조류 H9N2 인플루엔자 바이러스로부터 유래된 것이다. A/Swine/Korea/S10/2004(H1N1) 인플루엔자 바이러스는 2005년 1월 7일자로 대한민국 대전시 유성구에 소재한 한국생명공학연구원에 KCTC 10758BP로 기탁하였다.
A/Swine/Kores/S10/2004(H1N1) 인플루엔자 바이러스의 PB2 유전자는 서열번호 1에 기재된 염기서열을 포함하며, PB1 유전자는 서열번호 2에 기재된 염기서열을 포함하며, PA(Polymerase) 유전자는 서열번호 3에 기재된 염기서열을 포함하며, HA 유전자는 서열번호 4에 기재된 염기서열을 포함하며, NP 유전자는 서열번호 5에 기재된 염기서열을 포함하며, NA 유전자는 서열번호 6에 기재된 염기서열을 포함하며, M(matrix) 염기서열은 서열번호 7에 기재된 염기서열을 포함하며, NS 유전자는 서열번호 8에 기재된 염기서열을 포함한다. 또한 PB2 단백질은 서열번호 33에 기재된 아미노산 서열을 포함하며, PB1 단백질은 서열번호 34에 기재된 아미노산 서열을 포함하며, PA 단백질은 서열번호 35에 기재된 아미노산 서열을 포함하며, HA 단백질은 서열번호 36에 기재된 아미노산 서열을 포함하며, NP 단백질은 서열번호 37에 기재된 아미노산 서열을 포함하며, NA 단백질은 서열번호 38에 기재된 아미노산 서열을 포함하며, M 단백질은 서열번호 39에 기재된 아미노산 서열을 포함하며, NS 단백질은 서열번호 40에 기재된 아미노산 서열을 포함한다.
상기 A/Swine/Korea/S109/2004(H9N2) 인플루엔자 바이러스는 사람 인플루엔자 바이러스와 조류 인플루엔자 바이러스 간의 조합으로 이루어진 H9N2 혈청형의 바이러스로, 이의 PB2 유전자, HA 유전자 및 NA 유전자는 조류 인플루엔자 바이러스로부터 유래된 것이며, PB1 유전자, PA 유전자, NP 유전자, M 유전자 및 NS 유전자는 사람 인플루엔자 바이러스로부터 유래된 것이다. A/Swine/Korea/S109/2004(H9N2) 인플루엔자 바이러스의 PB2 유전자, PB1 유전자, PA 유전자, HA 유전자, NP 유전자, NA 유전자, M 유전자 및 NS 유전자는 각각 서열번호 9 내지 16에 기재된 염기서열을 포함하며, 이들의 단백질은 각각 서열번호 41 내지 48에 기재된 아미노산 서열을 포함한다. A/Swine/Korea/S109/2004(H9N2) 인플루엔자 바이러스는 2005년 1월 7일자로 대한민국 대전시 유성구에 소재한 한국생명공학연구원에 KCTC 10761BP로 기탁하였다.
상기 A/Swine/Korea/S81/2004(H9N2) 인플루엔자 바이러스는 사람 인플루엔자 바이러스와 조류 인플루엔자 바이러스 간의 조합으로 이루어진 H9N2 혈청형의 바이러스로, 이의 PB2 유전자, HA 유전자, NA 유전자 및 NS 유전자는 조류 인플루엔자 바이러스로부터 유래된 것이며, PB1 유전자, PA 유전자, NP 유전자 및 M 유전자는 사람 인플루엔자 바이러스로부터 유래된 것이다. A/Swine/Korea/S81/2004(H9N2) 인플루엔자 바이러스의 PB2 유전자, PB1 유전자, PA 유전자, HA 유전자, NP 유전자, NA 유전자, M 유전자 및 NS 유전자는 각각 서열번호 17 내지 24에 기재된 염기서열을 포함하며, 이들의 단백질은 각각 서열번호 49 내지 56에 기재된 아미노산 서열을 포함한다. A/Swine/Korea/S81/2004(H9N2) 인플루엔자 바이러스는 2005년 1 월 7일자로 대한민국 대전시 유성구에 소재한 한국생명공학연구원에 KCTC 10759BP로 기탁하였다.
상기 A/Swine/Korea/S83/2004(H9N2) 인플루엔자 바이러스는 사람 인플루엔자 바이러스와 조류 인플루엔자 바이러스 간의 조합으로 이루어진 H9N2 혈청형의 바이러스로, 이의 PB2 유전자, HA 유전자, NA 유전자, M 유전자 및 NS 유전자는 조류 인플루엔자 바이러스로부터 유래된 것이며, PB1 유전자, PA 유전자 및 NP 유전자는 사람 인플루엔자 바이러스로부터 유래된 것이다. A/Swine/Korea/S83/2004(H9N2) 인플루엔자 바이러스의 PB2 유전자, PB1 유전자, PA 유전자, HA 유전자, NP 유전자, NA 유전자, M 유전자 및 NS 유전자는 각각 서열번호 25 내지 32에 기재된 염기서열을 포함하며, 이들의 단백질은 각각 서열번호 57 내지 64에 기재된 아미노산 서열을 포함한다. A/Swine/Korea/S83/2004(H9N2) 인플루엔자 바이러스는 2005년 1월 7일자로 대한민국 대전시 유성구에 소재한 한국생명공학연구원에 KCTC 10760BP로 기탁하였다.
본 발명에 따른 돼지 인플루엔자 바이러스는 한국 돼지의 비강에서 직접 분리한 것으로, 이들의 바이러스들은 도 1 내지 3으로 도시한 계통발생학적 연관성을 갖는다. 도 1은 A/Swine/Korea/S109/2004(H9N2), A/Swine/Korea/S81/2004(H9N2) 및 A/Swine/Korea/S83/2004(H9N2) 인플루엔자 바이러스의 계통발생학적 관련성을 HA를 이용하여 분석한 것이고, 도 2는 A/Swine/Korea/S10/2004(H1N1) 인플루엔자 바이러스의 계통발생학적 관련성을 HA로 분석한 것이고, 도 3은 A/Swine/Korea/S10/2004(H1N1), A/Swine/Korea/S109/2004(H9N2), A/Swine/Korea/S81/2004(H9N2) 및 A/Swine/Korea/S83/2004(H9N2) 인플루엔자 바이러스의 계통발생학적 관련성을 M 유전자를 대상으로 분석한 것이다. 도 1 내지 3에서, SW는 돼지를 의미하고, CK는 닭을 의미하고, DK는 오리를 의미하고, CD는 붉은갯도요(curlew sandpiper)를 의미하고, TK는 칠면조를 의미한다.
A/Swine/Korea/S81/2004, A/Swine/Korea/S83/2004 및 A/Swine/Korea/S109/2004의 HA 유전자는 아시아계 조류 인플루엔자 바이러스 계통에 속하며, A/Swine/Korea/S10/2004의 HA 유전자는 초기 사람 HA 유전자 계통에 속하여, 사람 인플루엔자 바이러스인 A/WSN/33(H1N1)과 군락을 이룬다. 또는 인플루엔자 바이러스의 종 특이성을 결정하는데 매우 중요한 인자인 M 유전자 분석시, 본 발명의 A/Swine/Korea/S10/2004(H1N1), A/SW/Korea/S109/2004(H9N2) 및 A/SW/Korea/S81/2004(H9N2) 인플루엔자 바이러스는 사람 인플루엔자 바이러스와 군락을 이루며, A/Swine/Korea/S83/2004(H9N2) 인플루엔자 바이러스는 아시아 조류 인플루엔자 바이러스와 군락을 이룬다.
또한, A/Swine/Korea/S109/2004(H9N2)에 면역시킨 토끼의 혈청은 A/Swine/Korea/S81/2004(H9N2) 및 A/Swine/Korea/S83/2004(H9N2) 바이러스에 대하여 혈구응집 억제 활성을 나타내는바, 상기 3종의 바이러스는 교차반응성이 있음을 확인하였다.
본 발명의 돼지 인플루엔자 바이러스들 중, A/Swine/Korea/S10/2004(H1N1)는 본 발명에서 확인한 200 두의 돼지 중 약 37%에서 양성반응을 나타내었고, A/Swine/Korea/S109/2004(H9N2)는 약 36%에서 양성 반응을 나타낸 바, 본 발명에서 분리한 돼지 인플루엔자 바이러스는 국내 돼지에서 유행하는 바이러스 종임을 알 수 있다. 따라서, 본 발명의 돼지 인플루엔자 바이러스에 대한 백신을 제조함으로써, 대부분의 돼지에서 인플루엔자 바이러스에 대한 면역을 형성시킬 수 있으며 또는 돼지를 통한 바이러스 전파 및 신종의 인플루엔자 바이러스의 생성을 억제시킬 수 있다.
또한 본 발명의 돼지 인플루엔자 바이러스들 중, A/Swine/Korea/S10/2004(H1N1), A/Swine/Korea/S109/2004(H9N2) 및 A/Swine/Korea/S81/2004(H9N2)는 항바이러스제인 아만타딘(amantadine)에 대하여 저항성을 갖는다. 상기 아만타딘은 인테그럴 멤브레인 단백질(이온 채널)을 표적으로 하나, 특히 인테그럴 멤브레인 단백질의 26에서 31번째 서열이 항바이러스 활성에 특이적으로 작용한다. 그러나 본 발명의 상기 3종의 바이러스는 31번째 위치에 Ser이 아닌 Asn이 위치하여 아만타딘의 항바이러스 활성에 영향을 받지 않으므로, 이들에 대한 백신이 필수적으로 요구된다.
한편, 본 발명은 돼지 인플루엔자 바이러스에 대한 백신에 관한 것이다. 상기 돼지 인플루엔자 바이러스는, A/Swine/Korea/S10/2004(H1N1), A/Swine/Korea/S109/2004(H9N2), A/Swine/Korea/S81/2004(H9N2) 및 A/Swine/Korea/S83/2004(H9N2)으로 이루어진 군으로부터 선택될 수 있다.
본 발명의 백신은 돼지 인플루엔자 바이러스 또는 이의 항원을 유효성분으로 포함한다. 상기 백신은 사균 백신, 생균 백신, 특이 항원 추출백신(subunit vaccine), 합성 백신(Synthetic vaccine) 또는 유전공학 백신(Genetic enginerring vaccine)일 수 있으나, 생균 백신의 경우 바이러스가 독성이 있는 균주로 환원되어 증식될 수 있으므로 인플루엔자 바이러스에 대한 백신은 사균 백신 특이 항원 추출백신, 합성 백신 또는 유전공학 백신이 바람직하다.
사균 백신은 불활화 백신 또는 사독 백신이라고도 하며, 죽은 바이러스를 포함한 백신이다. 이의 예로는 전 바이러스 백신(whole-virus vaccine)과 분할 백신(split vaccine)이 있으며, 이들은 공지된 방법으로 용이하게 제조가능하다. 예컨대 전 바이러스 백신은 바이러스 전체에 포르말린을 처리하여 제조할 수 있으며, 분할 백신은 바이러스에 에테르를 처리하여 외피만을 분쇄, 수득하여 제조할 수 있다.
특이항원 추출백신은 바이러스의 구성성분 중 면역기능을 일으킬 수 있는 항원 성분만을 추출하여 제조한 백신으로, 바이러스 방어에 필요한 항원 부위에 대해서만 면역형성을 유도함으로써 부작용을 최소화할 수 있다. 예켠대 돼지 인플루엔자 바이러스의 HA 단백질 및/또는 NA 단백질을 추출하여 사용할 수 있다.
합성 백신은 바이러스의 항원 또는 항원결정기만을 화학적으로 합성하거나 또는 재조합 DNA 기술로 생산한 펩타이드를 포함한 백신으로, 예컨대 돼지 인플루엔자 바이러스의 HA 단백질 및/또는 NA 단백질을 합성하여 사용할 수 있다.
유전공학 백신은 바이러스의 병원성을 일으키는 특이 유전자를 변형하거나 제거한 것일 수 있다.
본 발명의 돼지 인플루엔자 바이러스 백신은, 일예로 (a) 돼지 인플루엔자 바이러스를 산란계 유정란에 주입하여 증식시키고, (b) 상기 유정란으로부터 요막 유강을 수득하여 여기에 포르말린, BPL(betapropiolactone) 또는 BEI(BINARYETHILIMINE)를 처리하고 및 (c) 상기 포르말린이 처리된 요막유강으로부터 사독화된 바이러스를 수득하고 이를 가금 바이러스 백신으로 제조하는 단계를 통하여 제조될 수 있다. 각 단계의 상세방법은 하기와 같다.
상기 (a) 단계는, 가금 인플루엔자 바이러스는 5 내지 15일령의 유정란에 주입하고, 유정란은 30 내지 40 ℃에서 24 내지 72시간 두어 바이러스의 증식을 유도한다.
상기 (b) 단계에서는 통상의 방법으로 요막유강을 수득하여 여기에 최종농도 0.005 내지 0.1 (v/w)%로 포르말린, BEI 또는 BPL을 가하고 저온에 방치하여 바이러스를 사독화시킨다.
상기 (c) 단계에서는 포르말린, BEI 또는 BPL이 처리된 요막유강을 원심분리 또는 여과를 통하여 사독화된 바이러스를 수득한다. 수득된 바이러스는 수산화알루미나 겔에 흡착시킨다.
상기한, 백신 제조방법은, 공지의 방법을 채택하거나 또는 일부 수정을 가하여 용이하게 실시가능하다.
또한 본 발명의 백신은 추가적으로 용매, 면역증강제 및 부형제로 이루어진 군으로부터 선택된 1종이상을 더욱 포함할 수 있다. 또한 상기 용매로는 생리식염수 또는 증류수가 있으며, 면역증강제로는 프레운즈 불완전체 또는 완전체 어드쥬번트가 있으며, 부형제로는 알루미늄 포스페이트, 알루미늄 하이드록사이드 또는 알루미늄 포타슘 설페이트(alum)가 있으나, 이에 한정되는 것은 아니며 공지의 물 질이 모두 포함될 수 있다.
본 발명의 백신은 돼지 인플루엔자 바이러스를 375 HA 내지 1000 HA 혈구응집 단위로 포함할 수 있으나, 이에 한정되는 것을 아니다. 다만, 백신의 혈구응집 단위가 375 HA 미만인 경우, 돼지에 효과적으로 항체형성을 유도하지 못할 수 있으며, 1000 HA를 초과하는 경우, 효율대비 비경제적일 수 있다.
본 발명의 백신은 경구형 또는 비경구형 제제로 제조할 수 있으며, 바람직하게는 비경구형 제제인 주사액제로 제조하며, 진피내, 근육내, 복막내, 정맥내, 피하내, 비강 또는 경막외(eidural) 경로로 투여할 수 있다.
본 발명의 백신은 A/Swine/Korea/S10/2004(H1N1), A/Swine/Korea/S109/2004(H9N2), A/Swine/Korea/S81/2004(H9N2) 또는 A/Swine/Korea/S83/2004(H9N2)에 감염될 수 있으며, 상기 인플루엔자 바이러스를 다른 개체에 전파시킬 수 있는 모든 종류의 가금, 조류 및 사람에 사용가능하다. 그 예로는, 돼지, 닭, 오리 또는 터키가 있다.
백신의 투여량은 적용 대상의 종류, 성장정도, 타 약물의 투여여부 및 건강상태에 따라 적절히 조절하여 결정할 수 있으며, 예컨대 체중 1 kg 당 15 HA unit 내지 60 HA unit로 접종할 수 있으며 1차 접종후 2 내지 1달 경과후 2차 접종을 더욱 실시할 수 있다.
이하 본 발명의 실시예를 기재한다. 하기 실시예는 본 발명을 예시하기 위한 것일 뿐 본 발명이 하기 실시예에 한정되는 것은 아니다.
실시예 1: 돼지 인플루엔자 바이러스 검출
1-1. 돼지 인플루엔자 바이러스 검출
국내 돼지 농장에서 사육하는 500 마리의 돼지의 코로부터 시료를 채취하고, 암피실린, 젠타마이신 및 페니실린이 함유된 0.5 mL의 멸균된 포스페이트 완충액(PBS, pH7.4)에 현탁하였다
시료들은 10일령의 닭 수정란의 요막에 접종하여 35 ℃에서 48시간 배양하였고, 요막유액을 회수하였다. 요막유액은 0.5 %의 칠면조 적혈구와 반응시켜 HA(haemagglutination)분석을 통하여 바이러스 존재 여부를 확인하였다.
1-2. 돼지 인플루엔자 바이러스 동정
HA 반응이 확인된 바이러스의 게놈을 분석하기 위하여, RNeasy Mini Kit (Qiagen, Santa Clara, CA)를 이용하여 요막액에 존재하는 바이러스로부터 RNA를 추출하고, 인플루엔자 바이러스에 특이적인 프라이머로 RT-PCR(reverse transcription PCR; SuperScript Preamplification System, Life Technologies, Gaithersburg, MD)을 실시하였다. PCR 산물은 QIAquick PCR 정제 키트(Qiagen)으로 분리하고, pGEM-T 이지 벡터에 클로닝하여 서열분석을 실시하였다. 서열분석은 AmpliTaq DNA 중합효소, FS (Perkin-Elmer, Applied Biosystems, Inc., Foster City, CA) 및 합성 올리고뉴클레오티드를 사용한 BigDye Terminator Cycle Sequencing Ready Reaction Kit로 수행하였고, Lasergene sequence analysis software (DNASTAR, Madison, WI)으로 염기서열과 상기 염기서열로부터 암호되는 단백질 서열을 결정하였다.

염기서열 분석으로, 서로다른 염기서열을 갖는 돼지 인플루엔자 바이로러스 4종을 각각 동정하였다. 각각은 A/Swine/Korea/S10/2004(H1N1), A/Swine/Korea/S109/2004(H9N2), A/Swine/Korea/S81/2004(H9N2) 및 A/Swine/Korea/S83/2004(H9N2)으로 명명하였다.
4종의 바이러스 각각의 유전자 서열은 서열번호 1 내지 32로 기재하였으며, 이로부터 번역되는 단백질 서열은 서열번호 33 내지 64로 기재하였다(표 2).

1-3. 혈구응집 억제 반응(haemagglutination inhibition)
본 발명에서 동정한 4종의 돼지 인플루엔자 바이러스들중 동일한 H9N2 타입에 속하는 3종의 바이러스간의 교차반응성을 확인하고자 하였다.
A/Swine/Korea/S109/2004(H9N2)로 면역화시킨 토끼로부터 혈청을 수득하고, 이에 A/Swine/Korea/S109/2004(H9N2), A/Swine/Korea/S81/2004(H9N2), A/Swine/Korea/S83/2004(H9N2)과 돼지에서 분리된 조류 인플루엔자 바이러스 A/SW/Korea/S452/2004 및 닭에서 분리된 A/CK/Korea/S1/2003(H9N2) 및 A/CK/Korea/S24/2004(H9N2)을 각각 반응시켜, 혈구응집 억제 역가를 각각 측정하였다. 혈구응집 억제 역가는 40 이상인 경우 양성으로 판단하였다. 그 결과는 하기 표 3과 같다.

표 3에 나타낸 바와 같이, A/Swine/Korea/S109/2004(H9N2)로 면역화된 토끼의 혈청은, A/Swine/Korea/S109/2004(H9N2),A/Swine/Korea/S81/2004(H9N2) 및 A/Swine/Korea/S83/2004(H9N2)에 대하여 혈구응집 억제 역가를 나타내어,교차반응성이 있으나, 그 외 바이러스들은 교차반응성을 나타내지 않는다.
1-4. 혈청학적 역학조사
돼지에서 무작위적으로 혈액을 채취하여 200 개의 혈청 시료를 수득하고, 이를 대상으로 혈구응집 억제 양성여부를 판단하였다. 상기 혈청은 수용체 파괴 효소(receptor destroying enzyme, RDE, Denka Seiken Co., Ltd., Tokyo, Japan)로 처리하여 비특이적 반응을 상쇄시킨 후 혈구응집 억제 분석을 실시하였다. 그 결과, 표 4에 나타낸 바와 같이, A/Swine/Korea/S10/2004(H1N1)에 양성을 보인 개체율은 약 37%, A/Swine/Korea/S109/2004(H9N2)에 양성을 보인 개체는 약 36%를 차지하였고, 조류 인플루엔자 바이러스인 A/Swine/Korea/S452/2004에 양성을 보인 개체는 약 2%로 해당되었다. 즉, 돼지에서 A/Swine/Korea/S10/2004(H1N1) 인플루엔자 바이러스와 A/Swine/Korea/S109/2004(H9N2) 인플루엔자 바이러스의 감염율이 높음을 알 수 있다.

실시예 2: 항바이러스제에 대한 저항성 검증
2-1. TCID50 (tisse culture infective dose50)결정
바이러스를 10배수로 희석한 후 MDCK 세포가 자라고 있는 96 프레이트에 접종한 후 바이러스감염을 48시간후 측정하여 TCID50를 측정하였다.
2-2. 아만타딘(amantadine)에 대한 저항성 검증
MDCK 세포는 MEM 배지로 6-웰 플레이트에서 배양하고, 10 ug/mL의 아만타딘 하이드로클로라이드(amantadine hydrochloride, Sigma, St. Louis, Mo) 존재하에 A/Swine/Korea/S10/2004 (human-like H1N1), A/Swine/Korea/S109/2004 (reassorted H9N2) 및 A/Swine/Korea/S452/2004 (avian H9N2)를 각각 102 TCID50/ml으로 1시간 감염시키고, 세포는 0.5 ug/mL의 TPCK-처리된 트립신과 10 ug/mL의 아만타딘 하이드로클로라이드가 포함된 MEM 배지에 배양하였다. 감염 48시간 후 배양물의 상층액을 수득한 후 바이러스 역가를 TCID50으로 결정하였다.
도 4는 아만타딘의 A/Swine/Korea/S10/2004(H1N1), A/Swine/Korea/S109/2004 (H9N2) 및 A/Swine/Korea/S452/2004(H9N2)에 대한 항바이러스 활성을 나타낸 것으 로, □는 아만타딘이 처리되지 않은 상태에서의 바이러스 역가이고, ■는 아만타딘이 처리된 상태에서의 바이러스 역가이다. A/Swine/Korea/S10/2004(H1N1), A/Swine/Korea/S109/2004 (H9N2)는 아만타딘의 처리 여부에 관계없이 일정한 활성을 나타내어 아만타딘에 저항성을 갖는 바이러스임을 알 수 있으나, 조류 인플루엔자 바이러스 유전자를 가진 A/Swine/Korea/S452/2004(H9N2)는 아만타딘에 감수성을 가져 아만타딘 처리시 바이러스 역가가 현저히 감소하였다.
이상 살펴본 바와 같이, 본 발명은 신종의 돼지 인플루엔자 바이러스를 제공하여, 돼지를 인플루엔자 바이러스에 대한 면역을 효과적으로 형성시킬 수 있으며 따라서 돼지 농장에서의 인플루엔자 바이러스에 의한 질환 발병을 현저히 감소시켜 경제적인 손실을 방지할 수 있다.

- 인플루엔자 바이러스 A/Swine/Korea/S83/2004(H9N2)(KCTC 10760BP)의 사균 또는 이의 항원을 유효성분으로 포함하는 인플루엔자 바이러스 백신.
- 제 2항에 있어서, 상기 항원은 서열번호 60에 기재된 아미노산 서열을 포함하는 HA 단백질 또는 서열번호 62에 기재된 아미노산 서열을 포함하는 NA 단백질인 백신.
- 제 2항에 있어서, 상기 백신은 돼지 또는 사람에 적용되는 것인 백신.
