Claims (10)
- - 항체(1)를 자기조립법 등을 이용하여 올린 기판(3);- 검출하고자 하는 항원(4)과 인터컬레이터(5)가 결합된 항원-인터컬레이터 복합체를 포함하는 측정용액(13);- 루테늄 이온(6)을 포함하는 수용액;- 상기 기판과의 사이에 일정한 최적 간격을 유지하면서 수용액 내에 위치하는 작업전극(working electrode, 9), 기준전극(reference electrode, 8) 및 대전극(counter electrode, 10);- 상기 수용액에 전압을 가할 수 있는 포탠시오스텟(potentiostat, 12); 및- 발광하는 빛을 측정하는 포톤카운터(11 또는 14)를 포함하여 구성되는 전기화학발광 방식의 인터컬레이터를 이용하는 항원 검출기.
- 제 1 항에 있어서, 상기 검출하고자 하는 항원이 단백질, 미생물, 바이러스 또는 화학물질인 항원 검출기.
- 제 1 항에 있어서, 상기 기판이 유리, 실리콘, PCB(printed circuit board) 또는 플라스틱류 위에 금을 얇게 코팅한 것인 항원 검출기.
- 제 1 항에 있어서, 상기 기판이 구멍이 뚫린 형태이며, 구멍의 내부 면에 금이 코팅되어 있고 여기에 항체가 고정화되어 있는 항원 검출기.
- 제 4 항에 있어서, 상기 구멍이 기판의 양면에 크기가 다른 입구를 가지도록 경사각을 가지고 비스듬하게 형성되어 있으며, 비스듬한 내부 경사면에 금이 코팅되어 여기에 항체가 고정화된 항원 검출기.
- 제 5 항에 있어서, 상기 기판의 경사면이 형성된 구멍의 넓게 뚫린 부분이 작업전극을 향하도록 되어 있고 좁게 뚫린 부분이 포톤 카운터를 향하도록 되어 있는 항원 검출기.
- 제 1 항에 있어서, 상기 인터컬레이터가 훽스트 33258 (hoechest 33258), DAPI(4',6-diamidino-2-phenylindole), 프롤린(proline), 노갈라마이신(nogalamycin), 디스타마이신 A(distamycin A), EtBr, 다우노루비신(daunorubicin) 및 독소루비신(doxorubucin)으로 구성된 군 중에서 선택된 것인 항원 검출기.
- 제 1 항에 있어서, 기판과 작업전극 사이의 간격이 0.5 ~ 2.0 mm 인 항원 검출기.
- 제 1 항에 있어서, 작업전극이 여러 개 설치되어 있고 설치된 작업전극에 대응하는 수의 서로 다른 종류의 항체가 고정화된 기판이 각각의 작업전극에 대응하는 위치에 위치함으로써, 여러 종류의 단백질을 동시에 검출할 수 있는 항원 검출기.
- (1) 기판 위에 항체를 고정화시키는 단계;(2) 루테늄 이온이 포함된 수용액 내에 상기 항체가 고정화된 기판을 위치시키고, 기판의 항체가 고정화된 위치와 적정 간격을 두고 작업전극, 기준전극 및 대전극을 위치시키는 단계:(3) 검출하고자 하는 항원과 인터컬레이터를 결합시켜 항원-인터컬레이터 복합체를 제조한 후, 이를 상기 기판이 있는 수용액에 투여하여 항원-항체 면역반응을 유도하는 단계; 및(4) 전원을 가하고, 인터컬레이터에 의하여 유도된 루테늄 이온의 전기화학발광을 측정하는 단계를 포함하여 구성되는, 제 1 항 내지 제 9 항 중 어느 한 항에 따른 항원 검출기를 사용하는 항원 검출 방법.
