Description
Claims (15)
- 유사분열성 표지 및 정량적 DNA 염색액으로 유사분열 세포를 처리하는 단계, 및 정량적 DNA 염색액을 사용하여 상기 세포중의 DNA의 양을 정량화하는 단계를 포함하는, 수치적 염색체 이상을 검출하는 방법.
- 제 1 항에 있어서,DNA의 정량화 단계가 유세포 분석법을 사용하는 단계를 포함하는 방법.
- 제 1 항에 있어서,세포가 상기 정량화에 의해 정상 배수성을 갖는 세포에서보다 더 많은 DNA를 함유하거나 더 적은 DNA를 함유하는 것으로 결정되는 경우 상기 세포를 수치적 염색체 이상을 갖는 것으로 특징화하는 단계를 추가로 포함하는 방법.
- 세포의 개체군으로부터 수치적 염색체 이상을 갖는 세포를 확인하는 방법으로서, 유사분열성 표지 및 정량적 DNA 염색액으로 세포의 개체군을 처리하는 단계, 및상기 세포가 유사분열성 표지에 의해 유사분열 상태인 것으로 확인되고, 추가로 상기 세포가 정상 배수성을 갖는 세포에서보다 더 많은 DNA를 함유하거나 더 적은 DNA를 함유하는 것으로 상기 정량적 DNA 염색액에 의해 결정되는 경우 상기 개체군으로부터의 세포를 수치적 염색체 이상을 갖는 것으로 확인하는 단계를 포함하는방법.
- 제 4 항에 있어서,유세포 분석법을 사용하여, 상기 세포가 유사분열 상태인 것을 확인하고, 상기 세포가 정상 배수성을 갖는 세포에서보다 더 많은 DNA를 함유하거나 더 적은 DNA를 함유하는 것으로 결정하는 방법.
- 제 1 항 또는 제 4 항에 있어서,유사분열성 표지가 유사분열 동안에 발생하는 히스톤의 인산화를 검출하는 시약인 방법.
- 제 1 항 또는 제 4 항에 있어서,유사분열성 표지가 6G3 단일클론성 항체인 방법.
- 제 1 항 또는 제 4 항에 있어서,정량적 DNA 염색액이 삽입성(intercalating) 염료, 소홈 결합제(minor-groove binder), 사이아닌 염료 및 요오드화프로피듐으로부터 선택되는 방법.
- 제 1 항 또는 제 4 항에 있어서,정량적 DNA 염색액이 요오드화프로피듐인 방법.
- 세포의 개체군으로부터 수치적 염색체 이상을 갖는 세포를 단리하는 방법으로서, 유사분열성 표지 및 정량적 DNA 염색액으로 세포의 개체군을 처리하는 단계,상기 세포가 유사분열성 표지에 의해 유사분열 상태인 것으로 확인되고, 추가로 상기 세포가 정상 배수성을 갖는 세포에서보다 더 많은 DNA를 함유하거나 더 적은 DNA를 함유하는 것으로 상기 정량적 DNA 염색액에 의해 결정되는 경우 상기 개체군으로부터의 세포를 수치적 염색체 이상을 갖는 것으로 확인하는 단계, 및상기 세포를 단리하는 단계를 포함하는 방법.
- 제 10 항에 있어서,상기 세포 개체군이 상이한 수치적 염색체 이상을 각각 갖는 2 종 이상의 세포의 아형 개체군을 함유하고, 세포의 상기 아형 개체군을 각각으로부터 및 상기 세포 개체군으로부터 분리하는 단계를 추가로 포함하는 방법.
- 세포에서 수치적 염색체 이상을 유도하는 시약을 확인하는 분석법으로서,세포를 상기 시약으로 처리하는 단계,유사분열성 표지 및 정량적 DNA 염색액으로 상기 세포 또는 그의 자손을 처리하는 단계, 및상기 세포 및 그의 자손이 유사분열성 표지 및 정량적 DNA 염색액으로 처리하는 동안에 유사분열 상태인 경우 정량적 DNA 염색액을 사용하여 상기 세포에서 DNA의 양을 정량화하는 단계를 포함하는 분석법.
- 제 12 항에 있어서,상기 세포가 말초혈 단핵구, 임파구, 상피 세포 및 결합 조직 세포인 분석법.
- 세포에서 수치적 염색체 이상을 검출하는 키트로서, 유사분열성 표지 및 정량적 DNA 염색액을 포함하는 키트.
- 유사분열성 표지 및 정량적 DNA 염색액을 포함하는 조성물.
