기질로서 N-숙시닐-(L-Ala)3-p-나이트로이닐라이드(N-succinyl-(L-Ala)3-p-nitroanilide)를 사용하여 37℃에서 30분간 p-나이트로아닐라이드(p-nitroanilide)의 생성량을 측정함으로서 폴씬 판크리아스 엘라스타아제(porcine pancreas elastase) 저해 활성을 조사한다. 각 시험용액을 일정 농도가 되도록 조제하여 0.1 ml씩 시험관에 취하고 50 mM 트리스-염화수소 버퍼(Tris-HCl buffer) (pH 8.6)에 녹인 엘라스타아제, 판크리아틱 솔루션(pancreatic solution (Type I: From Porcine Pancreas 유래, 0.1 units/ml))용액 0.05 ml를 가한 후 기질로 50mM 트리스-염화수소 버퍼(Tris-HCl buffer, pH 8.6)에 녹인 N-숙시닐-(L-Ala)3-p-나이트로이닐라이드(N-succinyl-(L-Ala)3-p-nitroanilide, 0.1mg/ml)을 0.1 ml를 첨가하여 30분간 반응시키고 microplate reader를 이용하여 410 nm 에서 흡광도를 측정한다. 엘라스타아제 저해활성은 시료용액의 첨가구와 무첨가구의 흡광도 감소율로 나타낸다. 도 5는 용매별로 추출한 꾸지뽕 열매 및 씨앗 추출물의 엘라스타아제 저해 활성 측정 결과를 나타낸 것이다. 꾸지뽕 열매 및 씨앗 추출물에서 엘라스타아제의 저해활성을 측정한 결과, 6.25mg/ml의 농도에서 꾸지뽕 열매 물추출(CTFW) 49.3%, 꾸지뽕 열매 80%에탄올 추출(CTFE) 58.7%, 꾸지뽕 씨앗 물추출(CTSW) 64.9%, 꾸지뽕 씨앗 80%에탄올 추출(CTSE) 78.0%로, 꾸지뽕씨 80%에탄올추출에서 높은 엘라스타아제의 저해활성을 보이는 것으로 나타났다.Using N-succinyl-(L-Ala)3-p-nitroanilide as substrate, p-nitroanilide at 37°C for 30 min. 시험관에 pH 6.8의 1/15M 인산나트륨 버퍼(sodium phosphate buffer) 0.5 ml에 10 mM L-DOPA을 녹인 기질액 0.2 ml및 시료용액 0.1 ml를 넣은 혼합액에 110 Unit/ml 버섯 티로시나아제(mushroom tyrosinase) 0.2 ml를 첨가하여 25℃에서 2분간 반응시키고 반응액 중에 생성된 도파 크롬(DOPA chrome)을 마이크로플레이트 ㄹ리더(microplate reader)를 이용하여 475 nm에서 흡광도를 측정한다. 티로시나아제 저해활성은 시료용액의 첨가군과 무첨가군의 흡광도 감소율로 나타내었다. 도 6은 용매별로 추출한 꾸지뽕 열매 및 씨앗 추출물의 티로시나아제 저해 활성 측정 결과를 나타낸 것이다. 꾸지뽕 열매 및 씨앗 추출물에서 티로시나아제의 저해활성을 측정한 결과, 12.5mg/ml의 농도에서 꾸지뽕열매 물추출(CTFW) 2.4%, 꾸지뽕열매 80%에탄올추출(CTFE) 13.1%, 꾸지뽕씨 물추출(CTSW) 28.3%, 꾸지뽕씨 80%에탄올추출(CTSE) 60.7%로, 꾸지뽕씨 80%에탄올추출에서 높은 티로시나아제의 저해활성을 보이는 것으로 나타났다.110 Unit/ml mushroom tyrosinase was added to a mixture containing 0.2 ml of substrate solution and 0.1 ml of sample solution in which 10 mM L-DOPA was dissolved in 0.5 ml of 1/15M sodium phosphate buffer at pH 6.8 in a test tube. ) 0.2 ml was added, reacted at 25° C. for 2 minutes, and the absorbance of DOPA chrome generated in the reaction solution was measured at 475 nm using a microplate reader. 실시예Example 2. 2. 꾸지뽕kkujippong 추출물의 항알레르기 효능 확인을 위한 동물 모델 제작 Animal model production to confirm anti-allergic efficacy of extract

오브알부민(chicken egg albumin, grade V) 0.2 % 함유 용액에 수산화알루미늄겔[Al(OH)3 gel을 1:1로 혼합한 액을 0.2 mL 복강투여로 최초 감작시킨다. 복강투여 후 14일에 최초감작과 같은 방법으로 재 감작시킨다. 최초 복강투여 21일부터 30일까지 10일간 매일 네뷸라이저를 이용하여 0.2%의 OVA를 1시간 흡입시킨 뒤, 시험물질을 하기 표 3에 기재된 바와 같이 경구투여 한다. 마지막 OVA 흡입 시행한 12시간 후에 에테르를 이용하여 마취한다. 개복하여 대정맥으로부터 혈액을 채취한다. 채취한 혈액은 4℃에서 2~4시간 보관 후 원심분리(1,200 g, 15분)를 시행하여 혈청을 분리한다. 혈액을 채취하고 난 후 기관절개를 가하고 기관절개창을 통해 cut-down tube를 삽입하여 기관지 폐포세정액 (Bronchoal-veolar lavage fluid, BALF) 을 채취한다.Sensitize first by intraperitoneally administering 0.2 mL of a solution containing 0.2% of ovalbumin (chicken egg albumin, grade V) mixed with aluminum hydroxide gel [Al(OH)3 gel in a 1:1 ratio. 14 days after intraperitoneal administration, re-sensitize in the same way as the first sensitization. 실시예 2를 통해 분리한 혈청에서 총 IgE양을 엘라이자 키트(Mouse IgE ELISA Set Cat. No. 555248, BD)로 정량한다. 항체를 완충용액에 희석하여 96-well에 코팅한 후 4℃에서 20시간 방치한다. 각 well을 3회 세척하고 혈청(2배희석)을 100 μL씩 분주한다. 2시간 동안 실온에서 방치한 후 5회 세척한 다음 항체 Avidin-HRP conjugated 100μL를 처리하고, 1시간실온에서방치한후다시세척하고, 여기에 TMB 기질을 100 μL씩 분주하고 암소에서 30분간 방치한 후 50 μL의 용액을 처리한 후 ELISA reader 450 nm에서 흡광도를 측정한다. 도 7은 용매별로 추출한 꾸지뽕 열매 및 씨앗 추출물의 천식유발 마우스 혈청 IgE농도 억제 효과에 대해 나타낸 것이다. 천식모델동물에 시험약물 중 꾸지뽕씨 80%에탄올추출 200(CTSW200)투여한 경우 혈중 IgE 농도가 가장 많이 감소하였다.The amount of total IgE in the serum isolated in Example 2 was quantified with an ELISA kit (Mouse IgE ELISA Set Cat. 실시예 2를 통해 분리한 혈청에서 오브알부민 특이 IgE의 농도를 측정하기 위하여 엘라이자(ELISA) 를 실시하였다. 항원으로 OVA 20ug/mL 의 농도로 코팅완충용액에 희석하여 96-well에 코팅한 후 4℃에서 20시간 방치한다. 각 well을 3회 완충용액으로 세척한 후 혈청(2배희석)을 100 μL씩 분주한다. 2시간 동안 실온에서 방치한 후 5회 완충용액으로 세척한 다음 항체 Avidin-HRP conjugated 100μL를 처리하고, 1시간 실온에서 방치한 후 다시 세척한다. 여기에 TMB 기질을 100 μL씩 분주하고 암소에서 30분간 방치한 후 50 μL의 용액을 처리한 후 엘라이자(ELISA reader) 450 nm에서 흡광도를 측정한다. 도 8은 용매별로 추출한 꾸지뽕 열매 및 씨앗 추출물의 천식유발 마우스 혈청의 오브알부민 특이 IgE의 농도 억제 효과에 대해 나타낸 것이다. 천식모델동물에 시험약물 중 꾸지뽕씨 80%에탄올추출 200(CTSW200)투여한 경우 혈중 오브알부민 특이 IgE 농도가 가장 많이 감소하였다.ELISA was performed to measure the concentration of ovalbumin-specific IgE in the serum isolated in Example 2. 실시예 2를 통해 분리한 혈청의 IL-4사이토카인(IL-4 cyotkine) 농도의 양은 엘라이자(ELISA kit(BD))를 이용하여 측정한다. 도 9는 용매별로 추출한 꾸지뽕 열매 및 씨앗 추출물의 천식유발 마우스 혈청 내 IL-4사이토카인(IL-4 cyotkine) 농도 억제 효과에 대해 나타낸 것이다. 동물의 시험약물 중 꾸지뽕 씨앗 80% 에탄올 추출 200(CTSE200) 및 500(CTSE500) 모두 유의적으로 혈중 IL-4cytokine의 농도가 감소하였음을 알 수 있다.
