Abstract
본 발명은 소르빈산 칼륨을 이용한 골다공증 질환의 유발 방법에 관한 것으로, 본 발명에 따른 골다공증 질환의 유발 방법은 소르빈산 칼륨을 투여하는 단계만으로 골다공증을 용이하게 유발하여 골다공증 치료제 연구 및 개발에 유용하게 사용될 수 있는 효과가 있다.
특히, 조골세포 분화를 억제하는 소르빈산 칼륨을 투여하여 용이하게 골다공증 질환 동물 모델을 제조할 수 있으며, 제조된 동물모델은 인위적인 난소적출 시술이나 실험실 내 장기간 돌보는 과정이 요구되지 않아 시간적, 경제적인 면에서 유리한 효과를 갖는다.
또한, 조골세포 분화 감소로 인해 발생되는 골다공증 치료제의 개발 및 메커니즘 연구에 유용하게 사용될 수 있다.
특히, 조골세포 분화를 억제하는 소르빈산 칼륨을 투여하여 용이하게 골다공증 질환 동물 모델을 제조할 수 있으며, 제조된 동물모델은 인위적인 난소적출 시술이나 실험실 내 장기간 돌보는 과정이 요구되지 않아 시간적, 경제적인 면에서 유리한 효과를 갖는다.

Description
본 발명은 소르빈산 칼륨을 이용한 골다공증 질환의 유발 방법에 관한 것이다.
최근 각종 산업재해 및 교통사고가 증가하고 고령화 사회로 접어들면서 골다공증 및 그에 따른 골절등에 의한 근골격계 질환이 증가하고 있다. 이와 같은 골질환등의 예방과 치료를 위해 골 재생 능력유도를 위한 다양한 형태의 뼈이식제와 조골세포의 활성향상 및 파골세포의 활성을 저하시키기 위한 새로운 형태의 관련질환 치료제들이 개발되고 있다. 또한, 골종양, 골결손이 심한 부위를 채울 수 있는 신생골을 능동적으로 유도하기 위하여 줄기세포와 조골세포를 이용하여 다양한 연구들이 시도되고 있다.
골다공증 치료제를 개발하기 위한 통상적인 실험방법은 일차적으로 파골세포의 활성을 억제시키거나, 조골세포의 활성을 증가시키는 물질을 선택한 후, 인간에서 나타나는 골다공증과 유사한 증상을 보이는 동물을 사용하여 골량이 얼마만큼 회복되고, 골강도가 얼마만큼 회복되었는지 정도로 약효를 평가한다. 현재 사용되고 있는 골다공증 질환 동물모델은 인위적으로 난소를 적출하여 골소실을 유도한 동물을 사용하거나, 자연적으로 노화가 진행된 SAMP-6라는 마우스에 개발 약제를 투여한 뒤 그 약효를 평가하는 것이 일반적이다.
그러나 난소적출 골다공증 질환동물은 실험을 수행할 때 마다 난소적출 시술을 수행하여야 하는 시간적, 경제적인 면에서 단점이 있고 시술의 실패로 인한 실험의 오차 등이 유발될 가능성도 배제할 수 없으며, 노화성 모델인 SAMP-6와 같은 모델은 실험실 내에서 장기간 돌봐야 하는 단점과 실험적 오차가 생길 수 있는 단점이 있어 새로운 동물모델의 개발이 요구되고 있다.
한편, 소르빈산 칼륨(Potassium sorbate)은 식품을 장기적으로 보관하기 위한 식품 보존제로 알려져 있으며, 모든 식품에 사용하기 위한 항균제로 연구되어 왔다(Sofos, J.N., Busta, F.F. 1981. Antimicrobial activity of sorbate. J. 본 발명자들은 조골세포 분화에 영향을 미칠 수 있는 화합물에 대한 연구를 진행하던 중, 소르빈산 칼륨이 조골세포 분화를 억제함으로써 골다공증 치료제 연구 및 개발 과정에서 골다공증 질환 유도 물질로 사용할 수 있음을 확인하여 본 발명을 완성하였다.
따라서, 본 발명의 목적은 소르빈산 칼륨을 이용한 골다공증 질환의 유발 방법을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 소르빈산 칼륨(potassium sorbate)을 인간을 제외한 동물에 투여하는 단계를 포함하는 골다공증 질환의 유발 방법을 제공한다.

상기 인간을 제외한 동물은 개, 고양이, 토끼, 모르모트, 랫트, 마우스, 소, 양, 돼지, 염소, 원숭이 등을 포함하는 모든 동물이 이에 해당할 수 있고, 바람직하게는 마우스일 수 있다.
상기 투여는 개체에 소정의 물질을 도입하는 것을 의미한다.
상기 소르빈산 칼륨(potassium sorbate)은 목적 조직에 도달할 수 있는 일반적인 경로를 통하여 투여될 수 있으며, 복강내 투여, 정맥내 투여, 근육내 투여, 피하 투여, 피내 투여, 경구 투여, 국소 투여, 비내 투여, 폐내 투여, 직장내 투여를 통하여 투여될 수 있으나, 이에 제한되는 것은 아니다.
상기 소르빈산 칼륨(potassium sorbate)은 조골세포 분화를 억제하는 것을 특징으로 할 수 있다.
상기 소르빈산 칼륨(potassium sorbate)은 조골세포 분화를 억제하는 효과를 나타내어 골다공증 질환을 유발하기 위한 생체 시료로 이용될 수 있다.
상기 소르빈산 칼륨(potassium sorbate)은 Runx2(runt-related transcription factor 2) 및 ALP(alkaline phosphatase) 중 어느 하나 이상의 조골세포 분화 마커 유전자의 발현량을 감소시키는 것을 특징으로 할 수 있다.
상기 Runx2(runt-related transcription factor 2)는 조골세포 분화에 있어 중요한 조절자로 알려져 있으며, 다분화능세포 또는 조골전구세포에서 ALP(alkaline phosphatase), OC(osteocalcin) 및 골 시알로단백질(bone sialoprotein)과 같은 유전자를 조절함으로써 조골세포 분화를 촉진하는 유전자이다.
상기 ALP(Alkaline phosphatase)는 조골세포의 표면에서 합성되는 당 단백질로써 골 형성이 활발히 일어날때 그 활성이 증가하며, 무기 인산염의 축적을 유도하여 조골세포에서 세포외기질의 무기화를 증가시킨다.
상기 소르빈산 칼륨(potassium sorbate)은 Runx2(runt-related transcription factor 2), ALP(alkaline phosphatase)와 같은 조골세포 분화 마커 유전자의 발현량을 감소시킴으로써 골다공증 질환을 유발할 수 있으며, 조골세포 분화 감소에 따른 골다공증 질환 관련 연구에 효과적으로 사용될 수 있다.
특히, 연령 증가와 여성 폐경기 이후 조골 세포의 생성능이 급격히 감소함에 따라 발생되는 골다공증의 치료를 위해서는 조골세포 분화 감소에 따른 골 대사 관련 연구들이 요구되며, 상기 소르빈산 칼륨(potassium sorbate)은 조골세포 분화를 감소시키는 물질로 관련 메커니즘 분석을 위해 활용될 수 있다.
일례로, 상기 소르빈산 칼륨(potassium sorbate)은 MC3T3-E1 세포에 투여되어 골다공증 질환 관련 연구에 음성 대조군으로 사용될 수 있다.
또한, 상기 소르빈산 칼륨(potassium sorbate)은 Runx2(runt-related transcription factor 2) 억제제로 사용되어, Runx2(runt-related transcription factor 2) 관련 작용 기전 연구에 활용될 수 있다.
상기 소르빈산 칼륨(potassium sorbate)은 0.0001 내지 5 mM 농도로 투여되는 것을 특징으로 할 수 있다.
만일, 상기 기재된 범위 미만으로 투여될 경우 조골세포 분화마커 유전자의 발현 감소 효과가 미미하여 개체에 골다공증 질환을 유발할 수 없고, 상기 기재된 범위를 초과하여 투여될 경우 세포독성을 나타낼 수 있다.
또한, 상기 기재된 범위 내에서 상기 소르빈산 칼륨(potassium sorbate)은 MC3T3-E1 세포의 생존능을 향상시키는 효과를 발휘한다.
또한, 본 발명은 소르빈산 칼륨(potassium sorbate)을 인간을 제외한 동물에 투여하는 단계를 포함하는 골다공증 질환 동물모델의 제조방법을 제공한다.
상기 소르빈산 칼륨(potassium sorbate)은 조골세포 분화마커 유전자의 발현량을 감소시키는 효과가 있어, 인간을 제외한 동물에 투여시 조골세포 분화 감소로 인해 골다공증이 유발할 수 있으며, 이를 통해 용이하게 골다공증 질환 동물모델을 제작할 수 있다.
나아가, 본 발명은 소르빈산 칼륨(potassium sorbate)을 유효성분으로 함유하는 골다공증 질환 동물모델 제조용 사료 조성물을 제공한다.
상기 사료 조성물은 개체에 특정 기간 공급되어 골다공증 질환 동물모델 제작에 사용될 수 있다.
상기 사료 조성물은 연구 목적 및 개체의 종류에 따라 소르빈산 칼륨의 함량 및 제공 기간을 특정 수준으로 조절하여 사용될 수 있다.
이하, 본 발명을 구체적으로 설명하기 위해 실시예 및 실험예를 들어 상세하게 설명하기로 한다. 그러나 본 발명에 따른 실시예들은 여러 가지 다른 형태로 변형될 수 있으며, 본 발명의 범위가 아래에서 상술하는 실시예들에 한정되는 것으로 해석되어서는 안 된다. 본 발명의 실시예들은 당업계에서 평균적인 지식을 가진 자에게 본 발명을 보다 완전하게 설명하기 위해서 제공되는 것이다.
<실험에 1> 소르빈산 칼륨 농도에 의한 세포독성 검사
(1) 세포배양
마우스 두개관 유래의 조골전구세포인 MC3T3-E1 세포를 사용하였고, 10% 소태아혈청(FBS, Atlas, Fort Collins, Colorado, USA), 100 units/ml 페니실린, 100 μg/ml 스트렙토마이신(Gibco, Grand Island, NY, USA)을 첨가한 알파-최소필수배지(α-MEM; Gibco) 배양액을 2일 간격으로 교체해주었으며, 37℃, 5% CO2 인큐베이터에서 배양하였다.
MC3T3-E1 세포를 조골세포로 분화시키기 위하여, 아스코르브산(A.A)과 베타-글리세로포스페이트(β-GP)(Sigma aldrich, St. Louis, MO, USA)를 이용하였다.
A.A와 β-GP는 각각 50 μg/ml와 5 mM의 농도로 α-MEM에 첨가하여, MC3T3-E1 세포의 조골세포 분화를 유도하였다.
(2) MTT 분석
소르빈산 칼륨의 세포독성 실험을 위하여, MC3T3-E1 세포를 48 well plate에 2 x 104 /well의 농도로 배양하였다. 소르빈산 칼륨을 각각 0, 0.05, 0.1, 0.2, 0.5, 1, 2, 5 mM 농도로 배양액에 첨가하여 37℃, 5% CO2 조건의 인큐베이터에서 24시간 배양한 후, 배양액을 제거하여 0.5 mg/ml농도로 MTT(3-(4,5-디메틸티아졸-2-일)-2,5-디페닐테트라졸리움 브로마이드)를 처리하고 37℃, 5% CO2 조건에서 1시간 동안 배양하였다.
이후, MTT를 제거한 뒤 다이메틸설폭사이드(DMSO)를 첨가하여 540 nm에서 흡광도를 측정하였다.
본 실험에서 얻은 결과는 표준편차로 표시하였고, 통계 처리는 Student's t-test를 이용하였으며, * P<0.05, ** P<0.01 *** P<0.001 인 경우 대조군과 소르빈산 칼륨 처리군 사이에 유의성이 있음을 인정하였다.
(3) 실험결과
MTT 분석 결과를 도 1에 나타내었다. 나타낸 바와 같이, 소르빈산 칼륨을 처리하지 않은 대조군과 소르빈산 칼륨을 처리한 군을 비교해 봤을 때, 소르빈산 칼륨 5 mM의 농도까지 세포의 생존에 영향을 미치지 않음을 알 수 있었다.
또한, 소르빈산 칼륨은 여러 세포에서 유전 독성을 나타낸다고 보고되어 있으나, MTT 분석 결과 소르빈산 칼륨 5 mM 농도까지는 조골전구세포에서 독성이 나타나지 않았고 오히려 세포의 생존을 높이는 것을 확인할 수 있었다.
<실험예 2> 소르빈산 칼륨이 조골세포 분화 마커 유전자 발현에 미치는 영향 평가
(1) RNA 추출 및 실시간 중합효소 연쇄반응(RT-PCR, Real-time polymerase chain reaction)
MC3T3-E1 세포에서 조골세포 분화 마커 유전자의 mRNA 발현량을 분석하기 위하여 RNA를 추출하고 cDNA를 합성한 후 RT-PCR을 수행하였다. RNA 추출은 TRI-Solution™(Bio Science Technology)을 제조사의 지침에 따라 수행하였다. 분리한 RNA는 정량을 하여 TOPscript™ RT DryMIX(Enzynomics)를 제조사의 권장사항에 따라 50℃에서 60분 그리고 95℃에서 5분으로 역전사(reverse transcription) 반응을 수행하여 cDNA를 합성하였다. EmeraldAmp GT PCR Master Mix (TaKaRa Bio Inc, Tokyo, Japan)와 프라이머가 포함된 혼합물에 합성된 cDNA 3 μg를 혼합하여 RT-PCR을 수행하였다. PCR 수행시 사용된 프라이머의 염기서열은 표 1에 나타내었다.

(2) 실험결과
실험예 1의 결과에 따라 세포독성을 나타내지 않는 5 mM 농도의 소르빈산 칼륨을 사용하여, 조골세포 분화 마커인 Id1, Dlx5, Runx2 발현량을 RT-PCR을 수행하여 확인하였다.
도 2에 나타낸 바와 같이, 소르빈산 칼륨은 조골세포 분화의 핵심 조절인자로 알려진 Runx2의 발현을 억제하여 조골세포 분화 저해 효과가 있다는 것을 확인할 수 있었다.
<실험예 3> 소르빈산 칼륨 처리에 의한 알칼린 포스파타아제(ALP) 활성 확인
(1) 실험방법
알칼린 포스파타아제(Alkaline phosphatase, ALP) 효소를 염색하여 효소의 활성을 측정하기 위해, MC3T3-E1 세포를 24 well에 5 x 104 cells/well의 농도로 배양하고 소르빈산 칼륨을 5 mM 농도로 처리한 뒤, ALP 효소 활성 정도를 BCIP/NBT(sigma aldrich, St, Louis, MO)를 사용하여 측정하였다.
배지를 제거하고 고정액(4 % 포름알데히드)을 첨가하여 약 5분간 실온에서 보관하였다가 30초 간 증류수로 세척하였다. 준비된 BCIP/NBT 용액을 첨가하여 약 15분간 실온에서 반응한 뒤, 2분간 증류수로 세척한 후 염색 부위를 관찰하였다.
본 실험에서 얻은 결과는 표준편차로 표시하였고, 통계 처리는 Student's t-test를 이용하였으며, * P<0.05, ** P<0.01 *** P<0.001 인 경우 대조군과 소르빈산 칼륨 처리군 사이에 유의성이 있음을 인정하였다.
(2) 실험결과
알칼린 포스파타아제(Alkaline phosphatase, ALP)는 조골 세포의 표면에서 합성되는 당단백질로써 골 형성이 활발히 일어날 때 그 활성이 증가하여, 골 생성의 지표로써 사용된다.
MC3T3-E1 세포에 소르빈산 칼륨을 처리한 후, ALP 활성 변화를 ALP 염색을 통해 확인한 결과를 도 3에 나타내었다.
나타낸 바와 같이, 아무것도 처리하지 않은 대조군과 비교하여 소르빈산 칼륨을 처리한 실험군에서 ALP의 활성이 감소하는 것을 확인할 수 있었다.
또한, 조골세포 분화유도물질인 아스코르브산(ascorbic acid, AA)과 베타-글리세로포스페이트(β-glycerophosphate,β-GP)를 처리한 것과 비교하여, 아스코르브산, 베타-글리세로포스페이트, 소르빈산 칼륨을 함께 처리했을 때 ALP 활성이 감소됨을 확인할 수 있었다.
이러한 결과는 소르빈산 칼륨이 조골세포 분화과정을 억제하고 있음을 시사하는 것이다.
