Abstract
The present invention relates to a composition for periodontal disease and oral cleansing agent comprising the same. The composition according to an embodiment of the present invention may include a mold extract, sessin extract, peppermint extract and Angelica extract.
Description
The present invention relates to a composition for periodontal disease and oral cleansing agent comprising the same.
Periodontal disease is the highest prevalence of all dental diseases. According to the oral examination of the 4th National Health and Nutrition Survey conducted by the Korea Centers for Disease Control and Prevention in 2007, the prevalence of periodontal disease in Korean adults is 73.9%, and the prevalence increases with age.
Another major oral disease, tooth decay, improves patients' awareness and preventive measures such as fluoride application and sealants have been actively taken since childhood. In contrast, awareness of periodontal disease still needs to be improved, and oral hygiene management and regular dental check-ups require more attention to prevention.
Periodontal disease treatment typically uses antibiotics to remove subgingival bacteria remaining in the oral cavity after mechanical treatment. However, recently, the resistance of the antibiotics to oral bacteria in the oral cavity has been confirmed, and the need for proper antibiotic use is emphasized. Antibiotics can also cause side effects such as tooth surface pigmentation, exfoliative gingivitis, and mucosal hypersensitivity.
Oral care products currently on the market can have side effects as they have strong activity by chemical components. Therefore, there is a need for oral hygiene products that can manage oral health including periodontal disease without side effects.
An embodiment of the present invention is to provide a composition and oral cleaning agent comprising the same that can prevent or treat periodontal disease to promote oral health.
The composition according to an embodiment of the present invention may include a mold extract, sessin extract, peppermint extract and Angelica extract.
The mold extract, sessin extract, the peppermint extract and the Angelica extract may be obtained by extracting the mold extract, sessin, peppermint and donkey in a solvent at a predetermined first temperature for a predetermined time and then filtering.
The mold extract, sessin extract, the peppermint extract and the Angelica extract may be concentrated under reduced pressure at a predetermined second temperature.
The mold extract, sessin extract, the peppermint extract and the Angelica extract may be lyophilized at a predetermined third temperature.
The weight ratio of the mold extract, sessin extract, the peppermint extract and the Angelica extract may be 1: 1: 1: 1.
The composition according to the embodiment of the present invention may be included in the mouthwash.
According to an embodiment of the present invention, by providing a composition having little toxicity and little side effects to the human body can prevent and treat various oral diseases including periodontal disease to improve oral health. Embodiments of the present invention provide a composition capable of preventing and treating various oral diseases, including periodontal disease, and oral cleaning agents comprising the same. The composition according to an embodiment of the present invention includes a mold extract, sessin extract, peppermint extract and Angelica extract.
The mold extract is a substance extracted from the mold, sessin extract is a substance extracted from sessin, peppermint extract is a substance extracted from peppermint, Angelica extract is a substance extracted from Angelica. The composition of the present invention is made by mixing the hyunggwa extract, sessin extract, peppermint extract and Angelica extract.
The extract can be obtained from raw materials through heat extraction. Specifically, the mold extract, sessin extract, peppermint extract and Angelica extract can be obtained by extracting the raw material, sessin, peppermint and donkey as raw materials, and putting the raw materials in a solvent for extraction for a predetermined time at a predetermined temperature. have.
Here, the solvent used to extract the extract from the raw material may be an aqueous ethanol solution, for example 70% ethanol. The extraction temperature may be 60 ° C. and the extraction time may be 16 hours, but the extraction temperature and time are not limited thereto. Extract obtained through the above process can be concentrated under reduced pressure. Specifically, mold extract, sessin extract, peppermint extract and Angelica extract may be concentrated under reduced pressure at a predetermined second temperature. Here, the second temperature, which is a reduced pressure concentration temperature, may be 50 ° C.
The reduced pressure concentrated extract can then be lyophilized. Specifically, the mold extract, sessin extract, peppermint extract and Angelica extract may be lyophilized at a predetermined third temperature. Here, the third temperature, which is a freeze drying temperature, may be -70 ° C.
The extracts thus obtained may be mixed in a predetermined ratio to obtain a composition according to an embodiment of the present invention.
According to one embodiment of the present invention, the weight ratio of the mold extract, sessin extract, peppermint extract and Angelica extract extract included in the composition may be 1: 1: 1: 1. That is, in the composition of the present invention, hyunggwa extract, sessin extract, mint extract and Angelica extract may be included in the same weight ratio.
The composition according to the embodiment of the present invention may be included in an oral cleanser to be used for washing the oral cavity. According to an embodiment, the composition may be included in toothpaste in addition to mouthwashes, and used to prevent and treat oral diseases.
The inventors of the present invention by treating the composition of the present invention to the bacteria causing the periodontal disease confirmed the growth inhibition of the bacteria and confirmed the efficacy of the composition of the present invention against periodontal disease.
Antibacterial activity test was performed to confirm the growth inhibition and killing of bacteria. The inventors of the present invention prepared a sample of the composition of the present invention to assay the antimicrobial activity and assayed the antimicrobial activity against harmful periodontal disease bacteria through the measurement of viable cell number.
The test bacteria were cultured Streptococcus mutans in BHI broth medium, enterococcus faecalis gingivitis bacterium was used in BHI broth medium, and stomatitis Candida albicans in YM broth medium. The fungus Porphyromonas gingivalis was used in culture on Tryptic Soy Broth medium.
First, in order to prepare the composition of the present invention, hyunghyeong, sessin, peppermint and Angelica extracts were selected and pulverized, and then extracted for 16 hours at 60 ° C. in an incubator using ethanol as a solvent. Extracted.
Then, the extract was filtered and the filtrate was concentrated under reduced pressure, and lyophilized for 72 hours using a freeze dryer to obtain an extract powder. Hereinafter, the liquid extract made by dissolving the extract powder in DMSO (dimethyl sulfoxide) was used for the experiment.
Thereafter, hyeonggae extract, sessin extract, peppermint extract and Angelica extract were mixed in a weight ratio of 1: 1: 1: 1 to form a mixed extract.
The antimicrobial activity of the cavities, gingivitis, stomatitis and periodontal bacteria of the mixed extract according to an embodiment of the present invention was confirmed through the experiment as follows.
In this experiment, Streptococcus mutans was used as caries, Enterococcus faecalis as gingivitis, Candida albicans as stomatitis, and Porphyromonas gingivalis as periodontal bacteria.
Streptococcus mutans and Enterococcus faecalis were inoculated in BHI medium and cultured at 37 ° C. Candida albicans was inoculated in YM medium and cultured at 37 ° C. Porphyromonas gingivalis was inoculated in Tryptic soy media and anaerobic culture at 37 ℃.
Test strains incubated for 24 hours were inoculated with 100 μl of the strain to 24 well plates, and then the mixed extract was added at a concentration of 3 to 5 mg / ml in Streptococcus mutans and 2 to 4 mg / ml in Enterococcus faecalis. Candida albicans was added to each well plate at a concentration of 1 to 3 mg / ml and Porphyromonas gingivalis to a concentration of 2 to 4 mg / ml. The total volume of each medium was 1 ml in the bacterial culture solution to which a certain amount of the mixed extract was added.
After incubation at 37 ° C. for 6 hours, 1 ml each was plated on an agar plate. After incubating at 37 ° C. for 24 hours, the number of colonies observed visually on the plate was counted to determine the number of viable cells and the percentage (%) of viable cells. The percentage reduction of viable cell count used in the experiment is as follows.

As a result, the growth of Porphyromonas gingivalis was reduced to 3 log cfu / ml when added to 2 mg / ml compared to the control group, and to 4 log cfu / ml when added to 4 mg / ml. 1 and FIG. 2).
Enterococcus faecalis was reduced to 1 log cfu / ml when the mixed extract was added at 2 mg / ml, and decreased to 2 log cfu / ml when added at 4 mg / ml. Was confirmed (see FIGS. 3 and 4).
Candida albicans was reduced to 3 log cfu / ml when the mixed extract was added at 1 mg / ml compared to the control, and 4 log cfu / ml was decreased when the mixed extract was added at 3 mg / ml. 5 and 6).
In addition, when Streptococcus mutans was added to the mixed extract at 3 mg / ml, it was confirmed that the growth of the bacteria was inhibited at 3 log cfu / ml compared to the control group. The number was confirmed to decrease (see FIGS. 7 and 8).
Compared to other strains, Candida albicans bacteria were found to have the highest susceptibility to the composition of the present invention because the number of viable cells was reduced at low concentrations.
The percentage reduction of viable cell number for each strain of the composition of the present invention is shown in the following table.
According to the experimental results, when Porphyromonas gingivalis, Enterococcus faecalis, Candida albicans and Streptococcus mutans were treated with the composition of the present invention, Porphyromonas gingivalis was 3 mg / ml, Streptococcus mutans was 4 mg / ml, and Candida albicans was 2 mg. More than 99% of the bacteria were reduced at the concentration of mg / ml, while Enterococcus faecalis reduced more than 96% of the bacteria at the concentration of 4 mg / ml.
The composition according to the embodiment of the present invention is safe and harmless to the human body compared to oral care products made of conventional chemical components with little toxicity and little side effects. In addition, the periodontal bacteria porphyromonas gingivalis (porphyromonas gingivalis), including the effective removal of inflammation-inducing bacteria in the oral cavity can be expected to prevent and treat periodontal disease.
