[Technical Field] The present invention relates to a composition for reducing wrinkles and promoting recovery of skin damage, which comprises extracts of Phellinus linteus, Saururus chinensis, and Saururus chinensis,

The present invention relates to a cosmetic composition for improving wrinkles and skin damage recovery and a composition for health functional foods, and more particularly to a cosmetic composition for improving wrinkle and skin damage recovery comprising a combination of herbal extracts, ≪ / RTI >

Skin is the primary barrier of the human body. It protects the internal organs of the body from external stimuli such as temperature and humidity changes, ultraviolet rays, and pollutants, and plays an important role in keeping the body homeostatic such as body temperature control. This important skin also becomes aged gradually as the age of the human body grows, and as a result, skin elasticity loss, keratinization, wrinkle formation, skin atrophy and the like appear. This aging of the skin not only causes wrinkles on the skin surface but also changes the entire line of the face. The appearance of wrinkles on the skin is a phenomenon accompanied by aging, which shows various structural changes due to the deterioration of vital function. That is, it is divided into intrinsic aging and aging caused by aging, and photoaging due to the addition of external factors, especially ultraviolet damage.

Much research has been carried out on the importance and role of collagen directly affecting such wrinkles and skin elasticity. When the majority of the skin, 77% of the skin, and 90% of the dermis are rich in quantity and quality of collagen, the skin can also be healthy and beautiful without wrinkles and sagging. Collagen is also known to promote regeneration of damaged skin.

In the dermis, collagen forms collagen fibers to maintain the shape of the skin and make the skin firm. Collagen is synthesized in fibroblasts. Progenitor collagen is synthesized in fibroblasts, and three precursor collagen molecules are twisted into a twisted shape and bonded together to form an initial shape of the collagen fiber and secrete it out of the cell. Outside the cell, both ends of the bulb collagen are cleaved by the enzyme, which then binds together to form thicker collagen fibers. Therefore, the collagen production necessary for the improvement of wrinkles and the repair of skin damage can be achieved by promoting the generation of bulb collagen in fibroblasts in dermis. It can also be achieved by inhibiting collagen production by collagenase (MMP-1) inhibition.

Conventionally, collagen has been commercialized in cosmetics. However, when collagen is applied to the skin surface as a cosmetic, it is difficult to absorb the transdermal absorption of collagen, which is a polymer, so that the function thereof can not be sufficiently expected. In addition, there was a method of injecting collagen directly into the dermis of the skin, but this method has not been a solution for improving the wrinkles of the skin due to side effects. In recent years, retinoic acid and proteins derived from animal placenta (Patent Document 1) have been known as substances promoting collagen synthesis. However, since retinoic acid is unstable, its formulation technique is complicated, and it is limited in terms of stability such as irritation to the skin. Animal placenta-derived protein has a fatal disadvantage that cattle extracts from mad cow disease can be used.

On the other hand, Orostachys japonicus. Is called "warsong". It is called "warsong", and it is found in sunny rocks and vegetation, and it is attached to tile and stone bricks of traditional houses. Nowadays, it reaches the extinction stage and it is artificially cultivated and proliferated and used for medicinal use. It has been used for treating schizophrenia, hepatitis, eczema, diarrhea, hemorrhoids, malignant boils, burns, etc., and it is known that it has a large effect of sucking pus when it is stuck on a boil or wound. It is known to be effective in promoting blood flow, pain, immunity, dermatitis, promoting hair growth, and skin rash. Patent Document 2 discloses a pharmaceutical composition for antioxidation containing, as an active ingredient, dichloromethane and / or hexane fraction of WASONG.

Saururus chinensis BAILL. Is a perennial plant of Saururus chinensis and is widely distributed in China, Japan and Korea. Sambucoptera is known to be effective for several species, jaundice, adult disease, fever, diuretic, liver cancer and anti-inflammation. Tetrahydrofuran, a lignan derivative isolated from Saururus chinensis, has been reported to inhibit cell adhesion, antiinflammation, liver protection and feeding inhibition activity. Patent Document 3 discloses the erythro-austrobailignan-6 inhibitory effect on fibroblast senescence isolated from Saururus chinensis.

Artemisia iwayomogi is a perennial herbaceous plant belonging to the family Asteraceae, which is also known as Vigorous Mugwort, Baby Rock Mugwort, and Mugwort. Stems and leaves have been medicinal since the Goryeo Dynasty because they are called Han - injin. The components of this herbal medicine are esculetin-6-methylether and esculetin-7-methylether. These components have been used as a summary of gallstone disease, infection and jaundice, ), Sedative effect and the like are known. Patent Document 4 discloses an antioxidative effect and a cytoprotective effect of the extract of Warmth.

Experimental Example

1. Cell culture

In order to confirm cytotoxicity and intracellular globular collagen synthesis of extracts, HDF-N cells were used with CCD-986Sk (human fibroblast cell line) to inhibit MMP-1 expression and inhibit elastase activity. Cells were cultured in DMEM (Gibco, USA) medium containing 10% (v / v) FBS (Gibco, USA), 100 U / mL streptomycin and 100 U / ml penicillin antibiotics (Gibco, USA) CO 2 and 37 ° C.

2. Cell viability measurement test ( MTT assay )

A cytotoxicity test was performed using an MTT assay.

The MTT assays, after absorption of MTT reagent into cells, form formazan by the succinate dehydrogenase of mitochondria. The intracellular accumulation of this substance indicates mitochondrial activity, broadly the activity of the cell Which is a representative method for measuring cell survival rate.

The cultured HDF-N cells were seeded at a density of 1 × 10 5 cells / well in 96 wells with 200 μl of the medium, cultured in a 5% CO 2 incubator at 37 ° C. for 24 hours, discarded and washed with PBS Next, the same amount of the medium and the sample dissolved in PBS were treated with various concentrations ranging from 0.1% to 5.0%, and the cells were cultured for 24 hours. Four hours before the end of the culture, 20 μl of 5 mg / ml MTT dissolved in PBS was added to each well, shaded with aluminum foil, and cultured in a 5% CO 2 and 37 ° C incubator for 3 hours. After removing the culture medium, 200 μl of DMSO solution was added, incubated at 37 ° C for 1 hour, and absorbance was measured at 570 nm using an ELISA reader. Cell viability was calculated by the following formula.

The results are shown in Figs. 1 and 2. Fig.

FIG. 1 is a graph showing cell survival rates obtained from the cytotoxicity tests on each of the extracts of Phellinseus sans suis extract, Saururus chinensis extract, and Radix scorpion extract prepared in Production Examples 1 to 3.

FIG. 2 is a graph showing the cell survival rate obtained from the cytotoxicity test on the mixed extract of rosacex, Saururus chinensis, and warm roe prepared in Example 3. FIG.

2. Synthesis effect of collagen synthesis

HDF-N cells were plated in 48-well plates at 1 × 10 4 cells / well and cultured at 37 ° C and 5% CO 2 for 24 hours. After the medium was removed, the cells were kept frozen for 24 hours. After that, the test substance was diluted in DMEM (excluding FBS) as a cell culture medium, and the concentration of the extract showed a cell viability of 100% -β was used, and cells were treated with a final concentration of 10 ng / ml and cultured for 24 hours. Subsequently, the culture medium was collected and the amount of collagen was measured using a precursor collagen type I c-peptide (PIP) EIA kit (TAKARA, MK101). On the other hand, the cells attached to the bottom were washed with PBS and lysed with 1N NaOH to measure total protein amount, and the amount of total collagen synthesis per protein was calculated.

The results are shown in Table 2 and Figs. 3 to 6.

3. Collagenase  ( MMP -1) expression inhibition test

CCD-986Sk cells were divided into 2x10 4 cells / well in a 24-well plate and cultured at 37 ° C and 5% CO 2 for 24 hours. After the medium was removed, the cells were washed with DPBS, and then the cells were treated at a concentration of 100% or more for 24 hours at 37 ° C and 5% CO 2 . After the culture, the amount of MMP-1 was measured using a human total MMP-1 ELISA kit (D & D system, DY901). UV-B was irradiated with 20 mJ / cm 2 as a negative control, and the inhibition rate of MMP-1 expression in the test group was expressed as a relative value with 0 as a suppression rate of MMP-1 expression in the negative control group.

The test results are shown in Table 3 below.
