Abstract
본 발명은, 아마씨 유래의 고리형 펩티드 혼합물을 유효성분으로 포함하는, 염증질환의 예방 또는 치료용 약학적 조성물에 관한 것이다. 본 발명에 의하면 LOMIX가 천연물에서 유래된 것이기 때문에 인체 독성이 적고 안전하다는 장점이 있으며, 급성 위염, 장염, 간염을 포함하는 다양한 염증성질환에 대하여 in vivo 치료 효과가 우수함을 확인하였는 바 차세대 치료제로 유용하게 이용될 수 있다.
Description
본 발명은, 아마씨 유래의 고리형 펩티드 혼합물을 유효성분으로 포함하는, 염증질환의 예방 또는 치료용 약학적 조성물에 관한 것이다.
염증(inflammation)이란 외부 감염원(박테리아, 곰팡이, 바이러스, 다양한 종류의 알레르기 유발물질)의 침입에 의하여 형성되는 농양의 병리적 상태를 뜻한다. 구체적으로, 외부 세균이 특정 조직에 침입하여 증식을 하게 되면 생체의 백혈구가 이를 인지하여 증식된 외부 세균을 활발히 공격하게 되는데, 이 과정 중 발생되는 백혈구의 사해가 균에 의하여 침입받은 조직에 축적됨과 동시에 백혈구에 의하여 사멸된 침입균의 세포 파괴물이 침입 받은 조직 내로 융해되어 농양이 형성된다. 염증에 의한 농양의 치료는 소염작용을 통하여 촉진될 수 있는데, 소염작용이란 항균제를 이용하여 침입균의 증식을 억제하거나 농양중에 축적된 이물질들을 탐식하는 대식세포(macrophage)를 활성화하여 상기 이물질들을 소화 및 배설하는 대식세포의 기능을 항진시키는 등의 염증치료 촉진작용이다.
일반적으로 염증반응은 생체의 세포나 조직에 기질적 변화를 가져오는 침습으로 인한 손상을 수복 및 재생하기 위한 생체방어 반응과정이고, 이 반응과정에는 국소의 혈관, 체액의 각종 조직세포 및 면역세포 등이 작용한다. 정상적으로 외부 침입균에 의하여 유도되는 염증반응은 생체를 보호하기 위한 방어 시스템인 반면, 비정상적으로 과도한 염증반응이 유도되면 다양한 질환들이 나타나게 되는데, 이러한 질환들을 염증질환이라 칭한다. 상기 염증질환은 외부자극에 의하여 활성화된 표적세포로부터 분비되는 다양한 염증 매개물질이 염증을 증폭 및 지속시켜 인체의 생명을 위협하는 질환으로서 상기 염증성 질환은 위염, 장염, 간염, 기관지염, 패혈증, 급성 호흡곤란 증후군, 다발성 장기부전 및 만성 폐쇄성 폐질환(chronic obstructive pulmonary disease)등을 포함한다.
많은 염증관련 질환의 발병에는 대식세포의 활성화와 이에 따른 염증관련 인자들의 과도한 생성이 연관되어 있는데, 대표적인 염증관련 인자들로는 IL-1β, TNF-α 등이 있다. 또한, 염증반응의 유도물질들 중에서 LPS(lipopolysaccharide)는 그람음성세균의 세포 표면을 구성하는 물질로서 백혈구와 같은 면역세포와 상호 작용을 하며, 염증관련 사이토카인의 조절에 관여한다.
한편, 아마과(Linaceae)에 속하는 식물인 아마씨(Linum usitatissimum L.)는 유럽과 아시아를 거쳐 캐나다로 전파되어 북위 55도 이상의 한랭한 지방에서만 식용으로 재배되는 까다로운 특성이 있어 많은 양이 생산되지 않는 귀한 씨앗이며, 단백질과 오메가-3 및 오메가-6와 같은 필수지방산과 리그난(lignan)과 같이 다른 씨앗에서 발견할 수 없는 항암물질 및 관상동맥, 심장질환과 같은 만성질환의 발병을 감소시켜주는 물질이 다량 함유되어 있다.
그러나, 아마씨로부터 분리 정제한 펩타이드의 면역연구는 1986년에 간세포에 콜레이트(cholate) 흡수 억제를 시작으로 폐상피 암세포에서 세포사멸 유도 및 T 세포증식 억제와 관련된 보고 뿐으로 (미국공개특허 2011-0111073), 항염증과 관련된 구체적인 약리활성 기전에 대하여는 거의 밝혀진 바가 없다.
이에, 본 발명자들은 염증질환 치료 효과가 우수하면서 부작용이 거의 없는 생약 성분을 찾고자 연구를 거듭한 결과, 천연물 유래의 고리형 펩타이드 혼합물(linusorb mixture; LOMIX)이 LPS 유도성 염증 사이토카인 생성을 억제하고, 그에 따른 신호전달 단백질의 발현도 억제함으로써 항염증 효능이 있음을 확인하고 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은, 아마씨 유래의 고리형 펩타이드 혼합물(LOMIX)을 유효성분으로 함유하는, 염증질환 예방 또는 치료용 약학조성물/건강기능식품 조성물을 제공하는 데 있다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
상기 과제를 해결하기 위하여, 본 발명은 하기의 성분 및 함량으로 이루어지는 고리형 펩티드 혼합물을 유효성분으로 포함하는, 염증질환의 예방 또는 치료용 약학적 조성물을 제공한다.

또한, 본 발명은 상기 고리형 펩티드 혼합물을 유효성분으로 포함하는 조성물의 염증질환 예방 또는 치료 용도를 제공한다.
본 발명의 일 구체예로, 상기 고리형 펩티드 혼합물은 아마씨에서 유래된 것임을 특징으로 한다.
본 발명의 다른 구체예로, 상기 염증질환은 위염, 장염 및 간염으로 이루어진 군에서 선택되는 것을 특징으로 한다.
본 발명의 또 다른 구체예로, 상기 고리형 펩티드 혼합물은 COX-2, TNF-α 및 iNOS의 발현을 억제하는 것을 특징으로 한다.
본 발명의 또 다른 구체예로, 상기 고리형 펩티드 혼합물은 NF-κB 및 AP-1 신호전달을 억제하는 것을 특징으로 한다.
본 발명의 또 다른 구체예로, 상기 고리형 펩티드 혼합물은 Src 및 Syk의 키나아제 활성을 억제하는 것을 특징으로 한다.
본 발명에 의하면 LOMIX가 아마씨라는 천연물에서 유래된 것이기 때문에 인체 독성이 적고 안전하다는 장점이 있다.
또한, 본 발명에 의하면 LOMIX가 in vivo 동물모델에서도 위염, 장염, 간염을 포함하는 다양한 염증성질환에 대하여 치료 효과가 우수함을 확인하였기 때문에 차세대 치료제로 유용하게 이용될 수 있다.
도 1은, RAW264.7와 HEK293 세포에 대한 LOMIX의 독성을 평가하기 위하여 MTT 어세이를 수행한 결과이다.
도 2는, LPS에 의해 유도된 염증조건하에서 LOMIX가 대식세포의 NO 생성능을 억제하는지 확인하기 위하여 Griess 어세이를 수행한 결과이다.

본 발명에서 '염증'이란, 일반적으로 외부 물질 또는 해로운 자극으로 인한 숙주 침입에 관한 신체 조직의 국소화된 보호 반응으로 나타나는 결과로서, 이러한 염증의 원인은 박테리아, 바이러스 및 기생충과 같은 감염성원인, 화상 또는 방사선 조사와 같은 물리적 원인, 또는 독소, 약물 또는 산업적 시약과 같은 화학약품, 또는 알레르기 및 자가면역 반응과 같은 면역 반응, 또는 산화성 스트레스와 관련된 이상 상태일 수 있다.
또한, 본 발명에 따른 조성물에 의해 예방, 개선 또는 치료될 수 있는 '염증성 질환'에는 제한이 없으나, 위염, 장염, 간염, 기관지염, 패혈증, 급성 호흡곤란 증후군, 다발성 장기부전 및 만성 폐쇄성 폐질환(chronic obstructive pulmonary disease)등을 포함할 수 있으며, 특히 위염, 장염, 간염인 것이 바람직하다.
본 발명에서 사용하는 용어인 'LPS (lipopolysaccharide, 지질다당류)'란 그람음성세균 표층의 펩티드글리칸을 둘러싸는 외막의 중요 구성성분으로 세포 1개당 약 3×105분자(표면의 20~30%)가 존재한다. LPS는 숙주의 염증세포, 내피세포 등에 작용하여 사이토카인의 분비를 촉진한다.
본 발명의 치료 조성물은 기존 치료 활성 성분, 기타 보조제, 약제학적으로 허용가능한 담체 등의 성분을 추가로 포함할 수 있다. 상기 약제학적으로 허용가능한 담체는 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로스 용액, 말토 덱스트린 용액, 글리세롤, 및 에탄올 등을 포함한다.
본 발명에서 "개체"란 질병의 치료를 필요로 하는 대상을 의미하고, 보다 구체적으로는 인간 또는 비-인간인 영장류, 생쥐(mouse), 쥐(rat), 개, 고양이, 말 및 소 등의 포유류를 의미한다.
또한, 본 발명에서 "약학적 유효량"은 투여되는 질환 종류 및 중증도, 환자의 연령 및 성별, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정되며 상기 요소를 모두 고려하여 부작용 없이 최대 효과를 얻을 수 있는 양으로, 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 조성물은 목적 조직에 도달할 수 있는 한 투여방법에는 제한이 없다. 예를 들면, 경구 투여, 동맥 주사, 정맥 주사, 경피 주사, 비강 내 투여, 경기관지 투여 또는 근육 내 투여 등이 포함된다. 일일 투여량은 약 0.0001 내지 100mg/kg이고, 바람직하게는 0.001 내지 10mg/kg이며, 하루 일회 내지 수회 나누어 투여하는 것이 바람직하다.
본 발명의 조성물은 염증성 질환의 예방 및 개선을 위한 약제, 식품 및 음료 등에 다양하게 이용될 수 있으며, 분말, 과립, 정제, 캡슐 또는 음료 형태로 사용할 수 있다.
본 발명에서는, 세균 유래 염증유발인자인 LPS(lipopolysaccharide)를 이용하여 인위적으로 염증반응을 유도한 후, LOMIX이 염증관련 인자들, 이들을 유도하는 전사인자, 신호전달 단백질의 발현/활성에 미치는 효과를 평가하였다.
그 결과, 본 발명의 LOMIX는 RAW264.7(대식세포주)에서 염증의 발생원인인 NO 분비를 억제하고, 염증관련 인자인 NOS(Nitric Oxide Synthase), COX-2(Cyclo oxygenase-2), TNF-α의 발현(mRNA level)을 억제시키며, 이들 인자의 발현을 조절하는 전사인자인 NF-κB, AP-1의 활성 또한 억제시킴을 확인하였다. 나아가, 이들 전사인자의 상위 신호전달과정에서 NF-κB 관련 신호(p-50, IκBα, p-85, Src), AP-1 관련 신호(ERK, JNK)들의 전달과정을 억제시킬 뿐만 아니라, 이들의 상위 신호인 Src, Syk의 키나아제 활성을 직접적으로 억제시킴을 확인하였다.
또한, 본 발명의 LOMIX는 위염 동물모델에서 위 출혈의 정도 및 범위를 감소시키고, 장염 동물모델에서 장염의 특징 중 하나인 대장 길이의 감소 현상을 억제하고, 간염 동물모델에서 염증세포 이동/응집 및 간 수치(AST,ALT)를 낮추었는 바, in vivo 에서도 다양한 염증질환에 대하여 치료, 예방 및 개선 효과가 우수함을 확인하였다.
이하, 본 발명을 실시예에 의거하여 보다 구체적으로 설명한다. 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.

1: 세포배양 및 시약
1-1. 세포배양
마우스 대식세포주인 RAW264.7 세포를 페니실린(100 IU/ml) 및 스트렙토마이신(100 μg/ml)과 10%의 FBS를 함유하는 RPMI 1640 배지를 이용하여 100 mm 세포배양 접시에 70-80%의 밀도로 배양하였다.
인간 신장세포주인 HEK293 세포를 페니실린(100 IU/ml) 및 스트렙토마이신(100 μg/ml)과 10%의 FBS를 함유하는 DMEM 배지를 이용하여 100 mm 세포배양 접시에 70-80%의 밀도로 배양하였다.
1-2. 시약
아마씨 유래의 신규 고리형 펩타이드 혼합물(linusorb mixture, LOMIX)은, ㈜Prairie Tide Chemicals에서 Cyclolinopeptide Mixture 또는 Linusorb Mixture로 시판되는 물질(CAS No. MIX)을 구입하여 사용하였다.

2: 세포
생존률
평가
LOMIX의 세포독성을 확인하기 위하여, RAW264.7와 HEK293 세포에서 MTT 어세이를 수행하였다.
구체적으로, 96-well 플레이트에 1×106 cell/ml의 세포를 플레이팅하고 37℃에서 각 면역실험 조건에 상응하는 배양시간 동안 CO2 인큐베이터에서 배양하였다. 이후 10 ㎕ MTT 용액(stock 농도:5 mg/ml)을 첨가하고 3시간 동안 추가반응을 유도하였다. 반응 종료 및 formazan crystal 용해를 위해 각 well에 100 ㎕ MTT 스톱용액(10% SDS in 0.01M HCl)을 추가적으로 첨가하였다. 세포 생존율은 MTT가 formazan으로 환원된 양을 570 nm에서 흡광도를 측정하여 얻어진 OD 값을 통해 산출하였다.
그 결과, 도 1에 나타낸 바와 같이, LOMIX는 RAW264.7와 HEK293 세포에서 모두 세포독성이 나타나지 않았다.
실시예
3: 대식세포에서 NO
생성능
측정
LPS에 의해 유도된 염증조건하에서 LOMIX가 대식세포의 NO 생성능을 억제하는지 확인하기 위하여 Griess 어세이를 수행하였다.
구체적으로, 96-well 플레이트에 1×106 cell/ml의 RAW264.7 세포를 플레이팅하고, 5% CO2 및 37℃에서 18시간 동안 전배양하였다. 이후 배지를 제거하고 4배 농도로 조제된 50 μl의 LOMIX와 50 μl의 LPS(최종농도 1 μg/ml) 또는 Pam3 (최종농도 10 μg/ml) 또는 Poly I:C (최종농도 200 μg/ml) 함유 배지를 동시에 처리하여 배양하였다. 24시간 후 상층액을 100μl씩 다른 96-well 플레이트에 옮기고 NO 정량은 Griess 용액 (0.5% naphthylethylenediamine dihydrochloride, 5% sulfanilamide, 25% H3PO4)을 이용하여 540 nm에서 흡광도를 측정하여 실시하였다. 표준물질로 sodium nitrite (0 내지 100 μM)를 사용하여 검량선을 작성하였다.
그 결과, 도 2에 나타낸 바와 같이, LOMIX가 농도의존적으로 대식세포에서 NO의 생성능을 억제한다는 것을 확인하였다.
실시예
4: 대식세포 분화억제 효능평가
LPS에 의해 유도된 염증조건하에서 LOMIX가 대식세포의 분화를 억제하는지 확인하기 위하여 형태가 변형된 세포수를 측정하였다.
구체적으로, 12-well 플레이트에 1×105 cell/ml의 RAW264.7 세포를 플레이팅하고, 5% CO2 및 37℃에서 18시간 동안 전배양하였다. 이후 LOMIX와 LPS(최종농도 1μg/ml)를 처리하여 24시간 동안 배양하고, 이후 분화된 대식세포의 특징인 위족을 나타내는 세포수를 역위상차 현미경으로 측정하였다.
그 결과, 도 3에 나타낸 바와 같이, LOMIX가 대식세포의 분화를 억제한다는 것을 확인하였다.
실시예
5: 대식세포에서 사이토카인 발현억제 효능평가
LPS에 의해 유도된 염증조건하에서 LOMIX가 대식세포에서 염증관련 사이토카인의 발현을 억제하는지 확인하기 위하여, 사이토카인 mRNA 발현양을 semi-quantitative RT-PCR로 조사하였다.
구체적으로, RAW264.7 세포에서 추출한 total RNA를 First strand cDNA synthesis kit (Thermo scientific)를 사용하여 cDNA를 제조한 다음, 동량의 cDNA를 PCR로 증폭하였다. 이때 사용한 표적단백질의 프라이머쌍 염기서열은 하기 표 1에 나타낸 바와 같으며, 대조군으로는 GAPDH을 사용하였다.

PCR 증폭은 Hipi PCR kit (Elpis biotech)을 사용하여 각 실험군 cDNA와 표적단백질들의 프라이머쌍, 대조군 GAPDH 프라이머쌍을 dNTP 250 uM, Tris-HCL(pH 8.3) 10 mM, KCl 50 mM, NgCl2 1.5 mM를 포함한 Hipi PCR kit 20ul에서 시행하였다. PCR은 95℃에서 45초간 denaturing; 55℃에서 45초간 annealing; 72℃에서 1분간 extension 하는 조건으로 시행하며, 총 20 cycles을 수행하였다. PCR로 증폭된 DNA는 1.5 % 아가로스겔에서 전기영동한 후 분획된 DNA 밴드의 강도를 측정하였다.
그 결과, 도 4에 나타낸 바와 같이, LOMIX가 대식세포에서 염증관련 사이토카인인 COX-2, TNF-α, iNOS 유전자의 발현을 억제함을 확인하였다.

6: 사이토카인 유전자 활성억제 효능평가
6-1. 전사인자 활성평가
LOMIX가 염증관련 유전자의 활성에 미치는 영향을 평가하기 위하여, 이들 유전자의 발현을 조절하는 전사인자인 NF-κB, AP-1의 활성을 Luciferase gene promotor 발현 측정법으로 측정하였다.
구체적으로, HEK 293 세포주를 Opti-MEM을 이용하여 24-well 플레이트에 분주한 후 37℃ 5% CO2 세포배양기에서 전배양하였다. PEI 법을 이용하여 NF-κB, AP-1 Luciferase DNA와 TRIF, MyD88, β-galactosidase DNA를 공트랜스펙션하였다. PEI법은 DNA 1 μg과 PEI 3 μg을 Opti-MEM에 각각 희석해 준 후 상온에서 20분 동안 배양한 후 DNA 희석액과 PEI 희석액을 혼합하여 다시 상온에서 20분 배양하는 방법으로 진행하였다. 배양 후 혼합액을 세포가 분주된 24-well 플레이트에 넣어준 후 6시간 후 세포배양 배지[10 % FBS, 1 % 페니실린/스트렙토마이신 in DMEM]로 교체해주고, 24시간 배양 후 시험물질을 농도별로 처리한 후 18시간 배양하였다. 이후, lysis buffer를 이용하여 세포를 용해시키고 기질과 1:1로 반응시키고 바로 Luminometer로 흡광도를 측정하였다. β-galactosidase의 경우는 기질인 X-gal과 1:1로 반응시킨 후 37℃ 배양기에서 5분 배양한 후 405 nm로 흡광도를 측정한다.
그 결과, 도 5에 나타낸 바와 같이, LOMIX가 MyD88/NF-κB 및 TRIF/NF-κB 매개의 루시퍼라제 활성을 억제하고, AP-1/MyD88 및 AP-1/TRIF 매개의 루시퍼라제 활성도 억제하였는 바, 이러한 결과는 LOMIX에 의해 결국 염증관련 유전자의 활성역시 억제됨을 의미하는 것이다.
6-2. 전사인자

이동평가
LOMIX가 염증관련 유전자의 활성에 미치는 영향을 평가하기 위하여, 이들 유전자의 발현을 조절하는 전사인자인 NF-κB, AP-1의 핵내 이동여부를 웨스턴 블랏팅으로 측정하였다.
구체적으로, RAW264.7 세포를 90%의 밀도로 100mm의 dish에서 전배양시킨 후, LOMIX를 30분 동안 전처리하고 LPS로 자극하고, 약물에 따라 일정시간 후 ice-cold PBS로 세정하고 homogenization buffer A [20 mM Tris-HCl pH8.0, 10 mM EGTA, 2 mM EDTA, 2 mM DTT, 1 mM PMSF, 25 μg/ml Aprotinin, 10 μg/ml Leupeptin] 300 μl를 이용하여 세포를 모은다. Sonicator를 사용하여 Output 4의 세기로 세포를 파쇄한 후 8000 rpm으로 15분 동안 4℃에서 원심분리하여 상층액 (세포질 분획)을 분리하였다. 펠렛(핵 분획)은 300 μl homogenization buffer B [1% TritonX-100 in Homogenization buffer A]로 부유시킨 후 sonicator를 사용하여 Output 4의 세기로 세포를 파쇄하였다. 얻어진 분획을 SDS-PAGE와 웨스턴 블랏팅을 이용하여, 전사인자의 핵 내 이동 수준을 평가하였다.
그 결과, 도 6에 나타낸 바와 같이, 핵분획물에서 60분 정도후에 NF-κB의 서브유닛인 p65, p50와, AP-1의 서브유닛인 c-Fos, c-Jun의 단백질이 감소하였는 바, LOMIX에 의해 이들 서브유닛의 핵내 이동이 억제되었음을 알 수 있다.

신호전달 경로 확인
LOMIX의 타겟 전사인자인 NF-κB 신호전달 경로에 관여하는 타겟 단백질을 확인하기 위하여, 대식세포를 LPS로 자극하는 동안 LOMIX를 처리한 후 신호전달 인산화 단백질의 수준을 웨스턴 블랏팅으로 측정하였다.
구체적으로, RAW264.7 세포를 7×106 cell/ml 농도로 60mm의 dish에서 전배양시킨 후, LOMIX(200ug/ml)를 2, 3, 5분 동안 전처리하고 LPS(1ug/ml)로 자극한 다음, 약물에 따라 일정시간 후 세포를 수집하여 lysis buffer와 sonicator를 사용해 세포를 파쇄하고 단백질을 얻었다. 단백질들은 SDS-PAGE 수행후 공지의 방법으로 웨스턴 블랏팅으로 실시하였으며, 이때 사용된 항체로는 항 p-p85, p85, p-AKT, AKT, p-Src, Src, p-Syk, Syk 항체이다.
그 결과, 도 7a에 나타낸 바와 같이, LOMIX 미처리군에 비하여 LOMIX 처리군에서는, NF-κB의 상위 신호전달에 관련하는 p85와 AKT의 활성형(인산화형)인 p-p85 및 p-AKT 수준이 현저히 감소됨을 알 수 있었다.
또한, 티로신 키나아제인 Syk 및 Src에 의해 p85가 인산화된다고 알려져 있기 때문에, Syk 및 Src의 활성형(인산화형)인 p-Syk 및 p-Src 수준을 관찰한 결과, 도 7b에 나타낸 바와 같이, 이들의 수준 또한 현저히 감소됨을 알 수 있었다.
이러한 결과는, LOMIX이 NF-κB 관련 신호전달 경로에서 상위의 티로신 키나아제(Syk 및 Src)를 억제함으로써 결과적으로 NF-κB를 억제한다는 것을 의미하는 것이다.

인산화 억제 효과
상기 실시예 7의 결과를 토대로, LOMIX이 염증관련 신호전달 단백질인 Syk와 Src의 활성을 직접적으로 억제하는지 더욱 확인하기 위하여, kinase profiler service(Millipore)를 이용하여 인산화 활성을 평가하였다.
구체적으로, 최종 용액 부피를 25μl로 하여, Syk과 Src 1-5 mU을 반응버퍼와 배양시 MgATP를 첨가하여 반응을 개시하였다. 40분간 상온에서 반응한 뒤 3% 인산 용액을 5ml 첨가하여 반응을 정지시킨 후, 이 용액(10μl)을 P30 filtermat에 점적한 후 75mM의 인산으로 5분씩 3회 세척하고 메탄올로 건조시킨 뒤 섬광계수기로 측정하였다.
그 결과, 도 8에 나타낸 바와 같이, LOMIX은 Syk 키나아제 활성을 약 70% 억제시켰으며, Src 키나아제 활성을 약 50% 억제시켰음을 알 수 있었다.
실시예
9: 급성위염(acute gastritis)에 대한

vivo
치료 효과
급성위염 동물모델에서 LOMIX의 효과를 평가하기 위하여 하기와 같은 실험을 수행하였다.
우선, ICR mouse를 4군으로 분류한 뒤, 그 중 두군은 LOMIX의 vehicle인 0.5% CMC(음성대조군)으로, 나머지 두군은 각각 LOMIX 및 라니티딘(ranitidine) 40mg/kg(양성대조군)으로 3일간 경구투여하였다. vehicle을 주입한 두군 중 한군은 위염을 유발시키지 않았고, 나머지 세군은 에탄올과 염산(EtOH/HCl)으로 위염을 유발한 뒤 희생시켰다. 그 다음, 위를 절개하여 위벽 상태를 관찰함으로써 위염 경과를 확인하였다.
그 결과, 도 9에 나타낸 바와 같이, LOMIX 200mg/kg 처리군에서는 라니티딘 처리군과 마찬가지로 위출혈의 정도 및 범위가 감소하였는 바, 위염이 치료됨을 알 수 있었다.

10: 급성장염(acute colitis)에 대한

치료 효과
급성장염 동물모델에서 LOMIX의 효과를 평가하기 위하여 하기와 같은 실험을 수행하였다.
우선, ICR mouse를 4군으로 분류한 뒤, 그 중 세군에는 일주일간 3% DSS(dextran sulphate sodium)를 경구투여하여 장염을 유발시켰다. 이후, 장염을 유발시킨 세군 중에서 두군은 각각 LOMIX를 50mg/kg, 200mg/kg으로 3일간 경구투여하고, 나머지 두군은 LOMIX의 vehicle인 0.5% CMC를 경구투여한 뒤 희생시켰다. 그 다음, 장을 분리하여 장 길이를 측정함으로써 장염 경과를 확인하였다.
그 결과, 도 10에 나타낸 바와 같이, LOMIX 처리군에서는 음성대조군에 비하여 장 길이 감소가 억제되었는 바, 장염이 치료됨을 알 수 있었다.

11: 급성간염(acute hepatitis)에 대한

vivo
치료 효과
급성간염 동물모델에서 LOMIX의 효과를 평가하기 위하여 하기와 같은 실험을 수행하였다.
우선, ICR mouse를 4군으로 분류한 뒤, 그 중 두군은 LOMIX의 vehicle인 0.5% CMC(음성대조군)으로, 나머지 두군은 LOMIX 50mg/kg, LOMIX 200mg/kg으로 5일간 각각 경구투여하였다. 이후, vehicle을 주입한 두군 중 한군은 PBS를 복강주사하였고, 나머지 세군은 LPS(10 μg/kg), D-갈락토사민(1 g/kg)을 복강주사하여 간염을 유발한 뒤 희생시켰다. 그 다음, 간 조직은 헤마톡실린/에오신(H&E)으로 염색하여 염증세포 이동/응집 정도를 확인하고, 혈액을 채취하여 간 수치(AST,ALT)를 검사하였다.
그 결과, 도 11a에 나타낸 바와 같이, LOMIX 처리군에서는 음성대조군에 비하여 염증이 억제되었으며, 도 11b에 나타낸 바와 같이, LOMIX 처리군에서는 음성대조군에 비하여 간 수치(AST,ALT)가 현저히 저하되었는 바, 간염이 치료됨을 알 수 있었다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
