Abstract
Description
도 2는 벼 유래 소포자 또는 화분 특이적 프로모터에 GUS::GFP 유전자를 융합시킨 재조합 벡터를 이용하여 벼 형질전환체를 육성하고, T0 및 T1 세대의 벼 형질전환 식물체들의 약 내의 소포자 및 화분 발달단계별 GUS 유전자의 발현양상을 형광 및 광학 현미경에서 관찰한 결과이다. anther; 약, UC(Uninucleate micrpore); 1핵기 소포자, BC(Bicelluar microspore); 2핵기 소포자, TC(Tricellular microspore); 3핵기 소포자, MP(Mature pollen); 성숙화분.
도 3은 벼 유래 소포자 또는 화분 특이적 프로모터에 GUS::GFP 유전자를 융합시킨 재조합 벡터 pSK762를 이용하여 형질전환시킨 벼 형질전환체에서, 본 발명에 사용된 프로모터가 유래된 Os04g0585900 유전자의 발현양상을 벼 조직별(약, 엽, 줄기 및 뿌리조직)로 분석한 RT-PCR 결과(A) 및 T1 세대의 벼 형질전환 식물체의 잎, 줄기 및 뿌리 조직에서 GUS 유전자의 발현 양상을 분석한 결과(B)이다. MP; 소포자 및 성숙화분을 함유하는 약 조직, L1, L2 및 L3; 발아 후 2주, 3주 및 4주 생장한 잎 조직, S1, S2 및 S3; 발아 후 2주, 3주 및 4주 생장한 줄기 조직, R1, R2 및 R3; 발아 후 2주, 3주 및 4주 생장한 뿌리 조직.
도 4는 벼 Os04g0585900 유전자의 마이크로 어레이 분석 결과이다. Heatmap에서 노란색은 발현이 높은 것을 의미하고 파란색은 발현이 낮은 것을 의미한다.
Claims (8)
- 서열번호 1의 염기서열로 이루어진 벼(Oryza sativa) Os04g0585900 유전자 유래 소포자 또는 화분 특이적 발현 프로모터.
- 제1항의 소포자 또는 화분 특이적 발현 프로모터를 포함하는 소포자 또는 화분 특이적 식물 발현 벡터.
- 제2항의 소포자 또는 화분 특이적 식물 발현 벡터로 형질전환된 식물체.
- 제2항의 소포자 또는 화분 특이적 식물 발현 벡터에 외래 유전자를 재조합하는 단계; 및
상기 재조합 식물 발현 벡터를 식물체에 형질전환시키는 단계를 포함하는 외래 유전자가 소포자 또는 화분에 특이적으로 발현되는 형질전환 식물체의 제조 방법. - 제4항에 있어서, 상기 식물체는 단자엽 식물인 것을 특징으로 하는 방법.
- 제5항에 있어서, 상기 단자엽 식물은 화본과(Gramineae) 식물인 것을 특징으로 하는 방법.
- 제4항의 방법에 의해 제조된 외래 유전자가 소포자 또는 화분 특이적으로 발현되는 형질전환 식물체.
- 제7항의 형질전환 식물체의 종자.
