Description
도 2는 본 발명에 따른 앱타머 칩과 DAPI 염색법을 이용하여 표면에 특정 단백질을 발현하는 표적 세포의 검출 및 정량 결과를 나타낸 도이다.
Claims (15)
- 앱타머(aptamer) 칩을 이용한 표적 세포의 검출 방법에 있어서,
(a) 표적 세포를 포함하는 시료를 앱타머 칩과 접촉시키는 단계; 및
(b) 상기 표적 세포가 결합된 앱타머 칩과 세포 염색 용액을 반응시키는 단계를 포함하며,
상기 표적 세포는 세포막에 HER2(human epidermal growth factor receptor 2) 또는 PSMA(prostate-specific membrane antigen)를 발현하는 세포이고,
상기 앱타머는 HER2(human epidermal growth factor receptor 2) 또는 PSMA(prostate-specific membrane antigen)와 특이적으로 결합하는 것인, 앱타머 칩을 이용한 표적 세포의 검출 방법.
- 앱타머(aptamer) 칩을 이용한 표적 세포의 정량 방법에 있어서,
(a) 표적 세포를 포함하는 시료를 앱타머 칩과 접촉시키는 단계;
(b) 상기 표적 세포가 결합된 앱타머 칩과 세포 염색 용액을 반응시키는 단계; 및
(c) 세포 염색 용액과 반응한 부분의 세기를 측정하는 단계를 포함하며,
상기 표적 세포는 세포막에 HER2(human epidermal growth factor receptor 2) 또는 PSMA(prostate-specific membrane antigen)를 발현하는 세포이고,
상기 앱타머는 HER2(human epidermal growth factor receptor 2) 또는 PSMA(prostate-specific membrane antigen)와 특이적으로 결합하는 것인, 앱타머 칩을 이용한 표적 세포의 정량 방법.
- 제1항에 있어서,
상기 표적 세포의 검출은 육안으로 판별하는 것인 방법.
- 제1항 또는 제2항에 있어서,
상기 세포 염색 용액은 세포핵을 염색하는 DAPI(4',6-diamidino-2-phenylindole), 메틸렌블루(methylene blue), 아세트산카민(acetocarmine), 톨루이딘블루(toluidine blue), 헤마톡실린(hematoxylin) 또는 Hoechst; 세포질을 염색하는 에오신(eosin), 크리스탈바이올렛(crystal violet) 또는 오렌지 G(orange G); 및 미토콘드리아를 염색하는 로다민 123(rhodamine 123), 미토트랙커(MitoTracker), 야누스그린 B(janus green B), 테트라졸리움염(tetrazolium salt), DASPMI(Dimethylaminostyrylmethylpyridiniumiodine), DASPEI(2-(4-(dimethylamino)styryl)-N-Ethylpyridinium Iodide), DiOC6(3,3'-dihexyloxacarbocyanine iodide), DiOC7(3,3'-Diheptyloxacarbocyanine Iodide) 또는 JC-1(5,5,6,6-tetrachloro-1,1,3,3-tetraethyl-benzimidazolylcarbocyanine chloride)로 이루어진 군으로부터 선택되는 것인 방법.
- 제1항 또는 제2항에 있어서,
상기 앱타머 칩은 기판 상에 앱타머가 고정된 것인 방법.
- 제1항 또는 제2항에 있어서,
상기 앱타머는 5' 말단에 티올기(thiol group; -SH)가 결합된 RNA 앱타머인 것인 방법.

상기 기판은 유리, 알루미나, 세라믹, 탄소, 금, 은, 구리, 알루미늄 및 실리콘으로 이루어진 군으로부터 선택되는 것인 방법.

상기 기판은 실란 처리를 통해 표면 상에 반응기로서 티올기를 갖는 것인 방법.
