상기 70% 에탄올을 이용하여 조제한 복분자 조추출물 50 g을 1ℓ의 70% 에탄올에 현탁시킨 후 헥산, 에틸아세테이트, 불포화부탄올로 단계적으로 용매의 극성을 높여가면서 각 용매의 가용분획물로 3회씩 분획한 다음, 에틸아세테이트 분획층을 모아 감압 농축기로 완전 농축하고 동결건조하였다. 그 결과 16.5g의 에틸아세테이트 분획 분말을 얻을 수 있었다.

복분자 추출물에 의한 골길이 성장의 측정을 위하여, 실험에 이용한 동물은 스프라규-다울리(Sprague-Dawley)계 수컷 흰쥐(3주령)로 샌타코(경기도, 대한민국)로부터 구입하여 사료와 물을 충분히 공급하면서 1주일간 실험환경에 적응시킨 다음 대조군과 각 시료 투여군으로 나누어 실험에 사용하였다. 동물사육실 외계조건은, 온도 20 ± 2℃, 상대습도 50 ± 10%, 조명시간 12시간( 07:00 점등, 19:00 소등)으로 조절하였다. 실험동물은 전 사육기간 동안 폴리카보네이트제 사육상자(10.25W x 18.75D x 8H inch)에 5마리씩 수용하였다. 사료는 실험동물용 고형사료(삼양사료, 한국)를 자유 섭취시키고, 물은 상수도를 물병을 이용하여 자유 섭취시켰다.

실험쥐들은 10마리씩 6개의 군으로 나누어, 대조군에게는 식염수를, 실험군에게는 100㎎/kg의 복분자 70% 에탄올추출물(실험군 1), 500㎎/kg의 복분자 70% 에탄올추출물(실험군 2), 100 ㎎/kg 복분자 70% 에탄올추출물의 에틸아세테이트 분획물(실험군 3), 100㎎ 복분자 물추출물(실험군 4) 및 500㎎ 복분자 물추출물(실험군 5)을 모두 실험개시일로부터 실험종료일까지 6일간 1일 1회 각각 생리식염수와 시료를 경구 투여하였다. 모두 실험군에 똑같이 실험종료 2일 전과 1일 전에 테트라사이클린(tetracycline : 10 ㎎/kg, i.p., Sigma, U.S.A.)을 투여하고 실험종료일까지 유지하였다. 실험 7일째에는 에테르로 마취하여 희생시킨 후 흰쥐로부터 분리한 족경골은 4% 인산염-완충액 파라포름알데히드(phosphate-buffered paraformaldehyde)에 48시간 고정한 후, 30% 수크로오스(Sucrose)용액에 침지시켜 탈수하였다. 이 후 고정된 골조직을 동결한 후 크라요컷(cryocut)을 사용하여 족경골(tibia) 근위부(proximal part)의 시상절편(sagital section)을 매 40㎛씩 절편한 후 수집하여 조직 표본으로 사용하였다. 그 후 절편은 길이 성장분석을 위해 사용되었다. 성장의 측정을 위해 성장판과 테트라사이클린(tetracycline)으로 침착되어 발색한 발광선과의 간격을 형광현미경으로 측정하였고(Hansson et al., Calcified Tissue Research, 10(3), 238, 1972, 도 1), 각 절편마다의 평균값을 산출하였다.The rats were divided into 6 groups of 10 rats, saline solution for the control group, 100 mg / kg bokbunja 70% ethanol extract (experimental group 1), 500 mg / kg bokbunja 70% ethanol extract (experimental group 2), 100 mg / kg Ethyl acetate fraction of 70% ethanol extract (Experimental group 3), 100 mg bokbunja water extract (Experimental group 4) and 500 mg bokbunja water extract (Experimental group 5) were all once a day for 6 days from the start of the experiment to the end of the experiment. 복분자 추출물 또는 에틸아세테이트 분획물에 대한 급성 독성을 알아보기 위하여, 하기와 같은 실험을 수행하였다.

4주령의 특정 병원체 부재(SPF, specific pathogens free) ICR계 마우스를 암수 각각 3마리씩 4개군(암수 각각 3마리/실험군)으로 나누어, 온도 22±3℃, 습도 55±10%, 조명 12L/12D 조건의 동물실 내에서 사육하였다. 마우스는 실험에 사용되기 전 1주일 정도 순화시켰다. 실험 동물용 사료(삼양사료, 한국) 및 음수를 멸균한 후 공급하였으며 자유 섭취시켰다.

상기 실시예에서 제조한 복분자 70% 에탄올 추출물 및 에틸아세테이트 분획물을 0.5% 트원(tween)80에 50㎎/㎖ 농도로 조제한 후, 마우스 체중 20g당 0.04㎖(100 ㎎/kg), 0.2㎖(500 ㎎/kg) 및 0.4㎖(1,000 ㎎/kg)씩 경구 투여하였다. 시료는 단회 경구 투여하였으며, 투여 후 7일 동안 다음과 같이 부작용 또는 치사 여부를 관찰하였다. 즉, 투여 당일부터 7일째까지는 매일 오전, 오후 1회 이상씩 일반 증상의 변화 및 사망 동물의 유무를 관찰하였다. 또한, 투여 7일째에 동물을 치사시켜 해부한 후 육안으로 내부 장기를 검사하였다. 투여 당일부터 1일 간격으로 체중의 변화를 측정하여 복분자추출물에 의한 동물의 체중감소 현상을 관찰하였다.Bokbunja 70% ethanol extract and ethyl acetate fractions prepared in Example were prepared at a concentration of 50 mg / ml in 0.5% tween 80, and then 0.04 ml (100 mg / kg) and 0.2 ml (500) per 20 g of mouse body weight.
