상기 실시예 1에서 얻어진 각 추출물 및 비교 대상 추출물을 이용하여 항균 활성을 시험하였다. 비교 대상으로는 도금양과(Myrtaceae) 시지기움속(Syzygium)의 식물인 도금양(잎) 및 정향나무(꽃봉오리)의 물 또는 에탄올 추출물을 이용하였다. 추출물은 각각 1.6 ㎎을 2% 디메틸술폭사이드(DMSO) 1 ㎖에 용해하였다. 이들을 다시 2% DMSO 수용액을 이용하여 순차 희석하여 2배 희석 계열을 작성하고, 96웰 플레이트에 100 ㎕씩 2연속 분주하였다. 이들 각 웰에 희석 균액 100 ㎕를 첨가하였다. 또한, 첨가한 희석 균액은 실시예 2에 나타낸 방법으로 액체 배지에서 전배양 후, 포르피로모나스 긴기발리스, 푸조박테리움 뉴클레아툼, 액티노마이세스 비스코서스에서는 2배 농도의 액체 배지에서 전배양액을 10배로 희석, 스트렙토코커스 뮤턴스, 스트렙토코커스 소브리너스의 전배양액을 2배 농도의 액체 배지에서 50배로 희석한 것이다. 왁스애플 및 워터애플 추출물의 각 웰의 농도는 800㎍/㎖, 400㎍/㎖, 200㎍/㎖, 100㎍/㎖, 50㎍/㎖, 25㎍/㎖, 12.5㎍/㎖, 6.25㎍/㎖, 3.13㎍/㎖, 1.56㎍/㎖, 0.78㎍/㎖이 되었다. 이 추출물과 포르피로모나스 긴기발리스, 푸조박테리움 뉴클레아툼 또는 액티노마이세스 비스코서스의 혼합액을 혐기 조건 하 37 ℃에서 24~48시간 동안 정치 배양하였다. 그 이외의 세균과 추출물의 혼합액에 대해서는 호기 조건 하 37 ℃에서 24~48시간 동안 정치 배양하였다. 각 웰의 탁도로부터 각각의 추출물의 최소 발육 저지 농도(MIC: Minimum Inhibitory Concentration)를 구하였다.Each extract obtained in Example 1 and the extract to be compared were tested for antimicrobial activity.
